참당귀(Angelica gigas Nakai) 중 Decursin 및 Decursinol Angelate의 분리 및 정제 (Isolation and Purification of Decursin and Decursinol Angelate in Angelica gigas Nakai)

  • 생약재로 사용 중인 Angelica gigas Nakai의 decursin 및 decursinol angelate를 60% ethanol로 최종적으로 추출 후 recycling HPLC 및 HPLC/MS에 의해 각각을 분리하였다. 60% ethanol로 $-20^{\circ}C$에서 추출하였을 경우 95% 순도의 decursin 및 decursinol angelate를 얻을 수 있었고 10번 이상의 recycle를 통해서 순수한 decursin 및 decursinol angelate를 분리할 수 있었다. 분리한 decursin 및 decursinol angelate를 HPLC/MS를 통해 분자량을 확인하였을 경우 m/z=329($[M+H]^+$) 및 m/z=351($[M+Na]^+$)과 같은 결과를 얻을 수 있었다. 이 분리 방법을 통해 decursin 및 decursinol angelate를 통한 의약품 및 식품 등에 관한 여러 연구에 있어 두 물질 각각에 대한 약리학적 효과를 도출하는데 기여할 것이며, 물질표준화에 기여할 것으로 사료된다.

천연물로부터 Quorum Sensing 저해제의 탐색 (Detection of a Quorum-Sensing Inhibitor from the Natural Products)

  • 인간이 서로 간의 의사소통을 위해 언어를 사용하듯이, 세균의 경우도 외부 환경 변화를 신속히 감지하여 서로 효과적으로 대응하기 위해서 주변 세포들과 소통할 수 있는 세균만의 독특한 화학적 언어를 사용하는 것으로 알려져 있다. 특히, 일정 세포 농도에 도달했을 때 자체적으로 생산된 화학적 신호를 통해 개체 수를 인지하고 그에 따라 특정 유전자의 발현을 동시에 조절하는 quorum sensing (QS) 기작은 다양한 세균 종들에서 광범위하게 존재한다. 본 연구는 다양한 천연물 추출물들을 대상으로 QS 저해 활성을 확인하였는데 QS 지시균주인 Agrobacterium tumefaciens NT1과 화학적으로 합성한 QS autoinducers을 사용한 bioassay를 수행하였다. 그 결과 양배추, 파, 양파의 추출물들에서 QS 저해 활성을 확인하였고, recycling preparative HPLC (prep-HPLC)를 통한 정제 과정을 통해, 83분 지점의 peak에 해당하는 성분들이 공통으로 QS 저해 활성을 가지고 있음을 확인하였다. 따라서 그 QS 저해 성분을 QSI-83으로 지정하고 thin layer chromatography (TLC)를 통해 P. syringae pv. tabaci의 autoinducers 합성을 저해하는 활성을 가지고 있음을 확인하였다. 또한 열에 대한 안정성과 세균 생장에서의 영향을 조사하였는데, 그 결과 QSI-83은 열에 안정하며 세균의 생장에는 영향을 끼치지 않는 물질임을 확인하였다. 따라서 우리는 천연물로부터 분리된 새로운 성분이 QS 저해제로서 이용될 수 있음을 제안한다.

  • A novel prepurification method was developed aiming at increasing yield and purity, also reducing solvent usage for purification of paclitaxel. The use of a micelle and precipitation in the prepurification process allows for rapid separation of paclitaxel from interfering compounds and dramatically reduces solvent usage compared to alternative methodologies. The prepurification process serves to minimize the size and complexity of the HPLC operations for paclitaxel purification. The process is readily scalable to a pilot plant and eventually to a production environment where multikilogram quantities of material are expected to be produced. As much as possible, the process has been optimized to minimize solvent usage, complexity, and operating costs.

반하에서 3,4-Dihydroxybenzaldehyde의 분리, 구조동정 및 Tyrosinase 활성 저해효과 (Tyronase Inhibitory Effect of 3,4-Dihydroxybenzaldehyde Isolated from Pinellia ternata)

  • Objectives : The purpose of this study is to isolate tyrosinase inhibitory material from Pinellia ternata and characterize its own structure and activity. Methods : Pinellia ternata (600g) was extracted with 95% methanol (1L) at $37^{\circ}C$ for 4 days, with shaking at 250rpm. The extract was further solvent-fractionated with n-hexane, chloroform, ethylacetate and water. The active fraction was subjected to JAI recycling prep-HPLC JAIGEL GS-320 column. The structure was identified for the active peak with NMR and GC. Results : Tyrosinase was potently inhibited by 95% methanol extracts from Pinellia ternata. The $IC_{50}$ value of the extracts was estimated to be 0.05mg/ml. The extracts was divided into four solvent-fractions, and the most potent tyrosinase inhibition was found in ethylacetate layer. $IC_{50}$ value of ethylacetate fraction was 0.001mg/ml. This fraction was further purified with JAI Recycling Preparative HPLC (Model: LC 9104). The isolated compound showing inhibitory activity was characterized on its chemical structure by NMR and the compound was identified as 3,4-dihydroxybenzaldehyde. $IC_{50}$ was found to be 7.74 ${\mu}M$ which is much lower than that of kojic acid $(66.5{\mu}M)$. Conclusions : The data suggest that 3,4-dihydroxybenzaldehyde isolated and identified from Pinellia ternata is very strong inhibitor to melanin biosynthesis.

Soraphinol C, a New Free-Radical Scavenger from Sorangium cellulosum

  • A new compound named soraphinol C (1) was isolated from myxobacteria Sorangium cellulosum KM1001 a soil isolate, together with a structurally related known compound, 4-hydroxysattabacin (2). These compounds were isolated by silica gel column chromatography and recycling preparative HPLC, consecutively. The structures of the compounds were determined on the basis of combined spectroscopic analyses. Compounds 1 and 2 exhibited antioxidant activity as a radical scavenger in the experiment using a hydrophilic free-radical initiator, 2,2'-azobis(2-amidinopropane)dihydrochloride with ORAC values of 0.956 and 0.617, respectively.

단삼(丹蔘)으로부터 항균물질의 분리 (Isolation of Antimicrobial Compounds from Salvia miltiorrhiza Bunge)

  • 단삼(丹蔘)을 bioassay-directed fractionation에 의한 에탄올 추출물의 클로로포롬 분획물을 silica gel column chromatography, recycling preparative-HPLC 등을 실시하여 항균활성을 나타내는 2개의 화합물을 분리하여 이화학적 성상 및 spectra data (IR,$^1H-NMR,\;^{13}C-NMR$ 및 Mass)에 의해 각각 Cryptotanshinone과 dihydrotanshinone I으로 동정하였으며, 이들 화합물의 항균성 (MIC)은 각각 S. epidermidis에 대하며 $6.3,\;3.1\;{\mu}g/ml$와 S. pyogenes에 대해 $6.3\;{\mu}g/ml$로 비교적 높은 활성을 나타내어 항생제로서 개발 가능성을 보여주었다.

한련초로부터 정제한 다당체의 혈당강하 효과 (Hypoglycemic Effect of Polysaccharide Isolated from Eclipta alba L.)

  • The major polysaccharide, named EAP, was purified from the aerial parts of Eclipta alba by Sepharose CL-2B ion exchange chromatography and recycling HPLC. The molecular weight of EAP was estimated to be 20911.9 D by MALDI-TOF MS. In addition, the sugar composition was determined to be arabinose (23.6%), mannose (24.8%), galactose (12.3%), and glucose (41.3%), respectively, by GC analysis. The EAP decreased the blood sugar level, which was induced by alloxan in rats, dose dependently.

  • Melanoidins, which were higher polymers with intense brown color, were investigated on their decolorization and degradation by ozonolysis. Amino acids linked up with melanoidins were readily separated by depolymerization of melanoidins with ozonolysis. The IR spectra of ozone-untreated MRPs showed a higher peak at $1665cm^{-1}$ (C=N) and $1600cm^{-1}$ (C=C) than the corresponding peaks of ozone-treated MRPs. Ozone-treated melanoidins with molecular weight of above 900 showed the highest nitrogen composition of all melanoidins tested. Ozone-treated melanoidins with molecular weight of 900 to 1000 were separated into five peaks on recycling preparative HPLC chromatogram. Major functional groups in ozone-treated melanoidins with molecular weight of 900 to 1, 000 were -CH$_2$-CO-, -CH$_2$-O- and CH$_2$-.
