A Combined Pharmacophore-Based Virtual Screening, Docking Study and Molecular Dynamics (MD) Simulation Approach to Identify Inhibitors with Novel Scaffolds for Myeloid cell leukemia (Mcl-1)

  • Chemical feature based quantitative pharmacophore models were generated using the HypoGen module implemented in DS2.5. The best hypothesis, Hypo1, which was characterized by the highest correlation coefficient (0.96), the highest cost difference (61.60) and the lowest RMSD (0.74), consisted of one hydrogen bond acceptor, one hydrogen bond donor, one hydrophobic and one ring aromatic. The reliability of Hypo1 was validated on the basis of cost analysis, test set, Fischer's randomization method and GH test method. The validated Hypo1 was used as a 3D search query to identify novel inhibitors. The screened molecules were further refined by employing ADMET, docking studies and visual inspection. Three compounds with novel scaffolds were selected as the most promising candidates for the designing of Mcl-1 antagonists. Finally, a 10 ns molecular dynamics simulation was carried out on the complex of receptor and the retrieved ligand to demonstrate that the binding mode was stable during the MD simulation.

  • Management of indoor air quality of underground subway station is an important issue. The air handling unit (AHU) installed in the underground subway station is the main facility determining the air quality of station. Especially for removing particulate matters, it is important to operate the appropriate air filter. However, there is no quantitative test method for existing air filter in site. It is rarely known that the collection efficiency of air filter can satisfy the designed criteria or not. In this study, we set the reference method using the mini-volume air sampler inside the AHU to evaluate the collection efficiency of different types of air filters. In addition, we applied the portable instrument based on the light-scattering method to the efficiency test. The method we applied in this study can be used in the evaluation of air filter in site.

  • Boiler equipment in power plant is always being operated in harsh conditions and severely exposed to the extreme environment of high temperature. Therefore periodically the state of devices should be checked, diagnosed, and analyzed to ensure the reliability of the equipment. Traditionally, such a diagnosis is based one or two physical properties of the sample taken from the equipment like hardness, microstructure, etc.. However, to enhance the confidence of the diagnosis, it is necessary to synthesize those properties together. This paper is to propose such a synthetic procedure for SA213-T12(1Cr-0.5Mo) which was developed to be used in $565^{\circ}C$ below condition. Creep test and accelerated degradation test were performed simultaneously, and the physical properties such as microstructure, tensile strength, yield strength, hardness, and indentation properties were measured. This paper proposes a method of determining the remaining life by quantitative comparison. It will provide the basis of evaluating life assessment more objective and reliable.

A Study on the Characteristics of Smoke Control using PIV in Tunnel Fires (터널내 화재시 PIV를 이용한 연기제어 특성에 관한 연구)

  • This study aims to measure a smoke density and velocity by using the PIV method in case a fire occurs in tunnels. By doing so, this will estimate a critical velocity, examine its appropriateness, and present the basic materials necessary for designing a smoke control equipment. For this study, a visualization test was conducted based on the 1/20 miniature of a real tunnel according to the Froude scaling. As a part of basic experiments, a correlation between smoke density and brightness was analyzed here, and a critical velocity was estimated on the condition that a fire breaks out in tunnels. As a result, this study finds that there is a correlation between smoke density and brightness within a range of 100% to 30% transmittance, from which a quantitative smoke density can be obtained. The study also suggests that a critical velocity calculated from the Kennedy formula shows about 10% difference from that estimated in the test.

  • The purpose of this study is to analyze the existing studies about the effect on private tutoring and EBS broadcasting which is considered as the alternative which was presented by education authorities. And this study attempts to measure the degree of the effects of private tutoring and EBS engagement on mathematics achievement using various quantitative methodologies. T-test and OLS multiple regression indicates some selection-bias which has positive direction compared to the PSM method closed to experimental design. So, it will be required to consider the methodology which measures the exact effect of institution or participation when we conduct the observational studies.

  • Purpose: The purpose of this study was to analyze the published articles in the Journal of Korean Academy of Adult Nursing from 2004 through 2006. Methods: Two hundreds and ten articles were analyzed focusing on research methodology and key words using descriptive statistics. Results: The proportion of quantitative research was 88.1%, while the proportion of qualitative research was 5.2%. The majority of the qualitative research design was survey(67.1%). Seventy-four percent of the research had verbal consent and 8% had written consent from the participants. Eight percent of the research provided conceptual framework. The prevailing data collection settings were hospitals(50.5%) and community(37.1%). For the data analysis, 95% used parametric analysis methods; descriptive statistics(26.2%), chi-square test(18.3%), t-test(18%) and ANOVA(17.4%). Key words were categorized into four nursing domain: human, health, nursing, and environment. The most frequently used domain was health. Conclusion: The number of the published articles in the Journal of Korean Academy of Adult Nursing has been increased and quality has been improved compared with the articles published before the 2000 year. Varied research methodology and data analysis methods were utilized.

Application of DNA Microarray Technology to Molecular Microbial Ecology

  • There are a number of ways in which environmental microbiology and microbial ecology will benefit from DNA micro array technology. These include community genome arrays, SSU rDNA arrays, environmental functional gene arrays, population biology arrays, and there are clearly more different applications of microarray technology that can be applied to relevant problems in environmental microbiology. Two types of the applications, bacterial identification chip and functional gene detection chip, will be presented. For the bacterial identification chip, a new approach employing random genome fragments that eliminates the disadvantages of traditional DNA-DNA hybridization is proposed to identify and type bacteria based on genomic DNA-DNA similarity. Bacterial genomes are fragmented randomly, and representative fragments are spotted on a glass slide and then hybridized to test genomes. Resulting hybridization profiles are used in statistical procedures to identify test strains. Second, the direct binding version of microarray with a different array design and hybridization scheme is proposed to quantify target genes in environmental samples. Reference DNA was employed to normalize variations in spot size and hybridization. The approach for designing quantitative microarrays and the inferred equation from this study provide a simple and convenient way to estimate the target gene concentration from the hybridization signal ratio.

  • A small loop-loop multi-frequency electromagnetic(EM) induction method is a useful technique to map a resistivity distribution efficiently and non-destructively. However, for quantitative interpretation and depth sounding, the quality of measured data is crucial. In this paper, we propose a bias correction of measured data by using background noise measurements to obtain reliable data, and propose an evaluation technique of apparent that can provide a resistivity image easily. We have performed small loop-loop EM measurements to detect water saturation in a man-made test site. The application of our proposed techniques to the measured data was successful.

  • In this paper, we evaluated the character of speech of hearing impaired person (HIP) who acquire his hearing loss after the youth. It is usually observed that severe HIP decreased not only speech perception but also vocalization. so there is a need for sensitive and quantitative measures or the assesment of the speech of the HIP to serve both diagnostic and prognosic purposes, 7 HIP and 12 normal hearing person(NHP) were studied with pure tone test and speaking test using word/sentence table which consists of vowel(a:), mono and two syllables and a sentence. we analyzed formant frequency, pitch, sound intensity, speech duration of HIP and NHP speech. According to the results, in the HIP's speech we find that formant frequency was shifted, first-formant prominence was reduced, the dynamic range of sound intensity was decreased, speech duration was prolonged. In the next, we expect the correlation between hearing and speech character of HIP is cleared through analysis of more acoustic parameters and precise selection of HIP group.
