Induction of Apoptosis with Kigelia africana fruits in HCT116 Human Colon Cancer Cells via MAPKs Signaling Pathway

  • Kigelia africana (Lam.) Benth. (Bignoniaceae) is a flowering plants in South, Central and West Africa and commonly known as the sausage tree (Eng.); worsboom (Afr.); umVunguta, umFongothi (Zulu); Modukguhlu (North Sotho); Muvevha (Venda). The dried, powdered fruits are used as dressing for wounds and ulcers, haemorrhoids, rheumatism, purgative, skin-firming, lactation in breast-feeding mothers. The aim of this study is to investigate the cytotoxic and apoptotic potentials of 70% ethanolic extracts of Kigelia africana fruits in HCT116 human colon cancer cells. Treatment of Kigelia africana fruits with various concentrations resulted in a sequence of characteristic of apoptosis, including loss of cell viability and morphological changes. Flow cytometry analysis showed Kigelia africana fruits increased the sub-G1 phase (apoptosis) population. Apoptosis confirmed by annexin V-fluorescein isothiocyanate and propidium iodide double staining in HCT116 human colon cancer cell lines. Moreover, analysis of the mechanism indicated that Kigelia africana fruits showed an increased Bax and Bcl-2 expressions in a dose-dependent manner, resulting in activation of hallmarks of apoptotic events, caspase-3, caspase-9 and cleaved poly-ADP-ribose polymerase. This is the first report to demonstrate the cytotoxicity of Kigelia africana fruits on HCT116 human colon cancer cells.

  • Oxidative stressand glia-associated chronic inflammation have been linked to the pathophysiology of Alzheimer's disease. Rhei rhizoma has been commonly used as a purgative and a haemostatic agent in traditional oriental medicine. Recently, the methanol extract from a dried root of Rheum undulatumhas been shown to have anti-allergic and anti-inflammatory effects. A Study on the Therapeutic Theory of Rumenshiqin (儒門事親) concerning Pediatrics (유문사친(儒門事親)에 나타난 소아(小兒)와 관련된 이론(理論)과 치법(治法)에 대한 고찰(考察))

  • Objective : Rumenshiqin(儒門事親) was compiled by Zhang Zi Huo(張子和) of the Jin(金) Dynasty of China. He invented unique medical theory and treatment such as three excretive methods(perspiratory, vomitory and purgative therapy). The aim of this study was to make the best use of the therapeutic theory of Rumenshiqin(儒門事親) connected with pediatrics. Methods: We conducted literature search in detail for contents of Rumenshiqin(儒門事親) and reviewed various kinds of books. Results : Zhang Zi Huo(張子和) recognized that children had pure-yang bodies(純陽之體) and emphasized that children need to prevent heat-stagnation(鬱熱) and asthenic yin(損陰). He instructed that a principle of raising such as wearing thin clothes, eating fresh food, harboring little desire and little anger, and taking temperate medicine. He described that pathogenic factors were mistake of rearing, misuse of medicine, exogenous factor, and excessive emotion. Conclusion : According to the above results, it is concluded that his therapeutic theory should be used at present.

Extractive Compounds of Ramaria formosa(Fr.) Quel (붉은싸리버섯의 추출성분)

  • To elucidate chemical strvacture of Ramaria formosa(Fr.) Quel was extracted with methanol for 72 hours, and then filtered twice. The extractive compounds were analyzed by the Libaries Search method, using GC and GC-MS Spectrometry. As a result, the five different compounds were analyzed, such as (1) methyl-hexadecanoate, (2) methyl-9,12- octadecadienoate, (3) methyl-cis-9-octadecenoate, (4) methyl-octadecanoate, and (5) 4-hexyl-2,5-dihydro-2,5-dioxo-3-furanacetic acid. The large quantity of compound, 4-hexyl-2,5-dihydro-2,5-dioxo-3-furanacetic acid, is expected that through experiments on animals the compound will be a potential agent which can be developed as a health food to relieve constipation or a laxative medicine.

  • The study has been carried out for the comparative analysis of relevant literature in order to highlight the present topic "Clinical study on effect on Virechana karma (therapeutic purgation)". Total no. of 40 cases has been selected from OPD and IPD of Panchakarma department, through proper history taking and clinical examination prior to VirechanaKarma. Only VirechanaKarmaYogya subjects were selected for the VirechanaKarma. Analysis of VirechanKarma on the basis of Vegiki (purgative bouts), Maniki (quantity), Langiki (clinical features) and Aantiki (end product) features were done in this study. In this study a highly significant relief was found in patients i.e. p<0.001 due to Virechana mainly vitiated PittaDosha (the heat energy in the body) and secondary Kaphadosha (mucus) and VataDosha (subtle energy associated with movement) are being expelled out which might have accounted for better relief in VirechanYogya (indicated) individuals (Pitta Pradhan Vyadhi (vitiated pitta), RaktajRoga (hemopoietic diseases), ShodhanYogyaAvastha (detoxification), etc.) in the above group of patients. VirechanaDravyas have properties like Tikshna (hyperfunction), Sukshma(subtle), Ushna (heat), etc. are described in Ayurvedic classics which play a vital role in the mode of action of VirechanaKarma has also been explained under probable mode of action.

Papaya: A gifted nutraceutical plant - a critical review of recent human health research

  • The plant kingdom is considered to be a repository of modern medicine, attributable to their rich source of bio-active molecules and secondary metabolites. It is indeed the Nutraceuticals that enhance immunity and ensure a healthier life because of their prophylactic and therapeutic values. Over centuries, papaya [Caricaceae; (Carica papaya Linn.)] is a renowned nutritious and medicinal plant. Each part of the papaya like root, stem, leaf, flower, fruit, seed, rinds, and latex has its own nutraceutical properties. It serves as food, cooking aid, and Ethnomedicine to prevent and treat wide-range of diseases and disorders. It has also been traditionally used as appetite enhancer, meat tenderizer, purgative, medicinal acne, abortifacient and vermifuge. Over decades, a series of scientific attempts were made to authenticate the nutraceutical properties of papaya. These studies validated that the papaya has antiplasmodial, antitrichochramal, antitrichomonal, antidengue, and anti-cancer activities. They have also exhibited that papaya possesses antiseptic, antiparasitic, anti-inflammatory, antidiabetic, and contraceptive features, and it helps in the management of sickle-cell anaemia, HIV, heart diseases and digestional disorders too. Nevertheless, the responsible bio-active molecules and their mode of actions remain indistinct and imprecise, and this calls for further pharmacological and clinical research on them. Conclusively, papaya is one of the naturally gifted plants; though its nutraceutical properties as a food or as a quasi-drug are poorly understood or undervalued by people. Accordingly, this scrutiny, demand for instigation of public health awareness campaigns to promote papaya consumption, so that the society shall acquire optimal benefits of papaya and in turn prevent and alleviate various diseases and illness.

High Prevalence of Haplorchiasis in Nan and Lampang Provinces, Thailand, Proven by Adult Worm Recovery from Suspected Opisthorchiasis Cases

  • Opisthorchiasis, a risk factor for cholangiocarcinoma in humans, is of public health importance in Thailand. The Annual Surveillance Reports from Nan and Lampang Provinces, Thailand, for the year 2011 showed an opisthorchiasis prevalence of over 70% by recovery of eggs in the feces. This study investigated whether most cases are actually due to minute intestinal flukes (MIF) rather than Opisthorchis viverrini, as the eggs of both can hardly be differentiated by morphology. Fifty and 100 cases from residents in Nan and Lampang, respectively, had stools positive for eggs initially assumed to be those of O. viverrini. Each patient was given praziquantel at 40 mg/kg in a single dose. After 2 hr, 30-45 ml of the purgative magnesium sulfate was given, and stools were collected up to 4 times sequentially. The stools were examined for adult worms by simple sedimentation. It was found that 39 of 50 cases (78.0%) from Nan Province had Haplorchis taichui, with intensities ranging from 5 to 1,250 with an average of 62 worms/case. Taenia saginata (7 cases) and Enterobius vermicularis (1 case) were other helminths recovered as the co-infectants. In Lampang Province, H. taichui was recovered from 69 cases (69.0%). The number of flukes recovered ranged from 1 to 4,277, with an average of 326 worms/case. Four cases had Phaneropsolus bonnei, and 10 T. saginata as the co-infectants. Adult specimens of O. viverrini were not recovered from any stool. Clearly, MIF infection, especially haplorchiasis, is more common in northern Thailand. These findings should encourage the Public Health Office to employ more specific tools than Kato's method for surveillance of opisthorchiasis in Thailand.

  • From the study of the definition of and intoxication, detoxication poisonous drug, the symptom and treatment of intoxication, andantidotes with DONG YUI BO GAM etc, it can be concluded as follows. 1. In ancint times, the poisonous drug was a drug which could treat diseases but after that it has been a drug which can injure one's health because it has severe property, poison or side effects 2. Intoxication means that poison goes and works in the body. Detoxication means treatment which clears the symptom of intoxication and removes the toxin in the body or on the body 3. At the intoxication's symptom, vomiting, madness, diarrhea, abdomianl pain, consumption are frequent in order 4. At the intoxication's treatment, vomiting method is used when poison is in the stomach, purgative method is used when poison is the intestine. 5. Mostly Hei-dou is used in drugs and Gan-kao. Qing-yan are used in order. 6. Mostly drugs of reliving fever are used in drugs. 7. Mostly cold drugs are used in drugs and warm, even, hot, cool drugs are used in order and mostly mild drugs are used in drugs and bitter, hot, salty, sore drugs are used in order.

Two Human Cases of Diphyllobothrium latum Infection through Eating Raw Freshwater Trout, Salmo gairdneri (송어를 감염원으로한 광절열두조충 인체감염)

  • A total of 31 human diphyllobothriasis cases have been reported in Korea. Authors experienced two more human cases of Diphyllobothrium latum infection, especially due to eating raw freshwater trout. Two cases were husband and wife in a famil residing in Chunchon city, Kangwon-do, who raw fish in October 1994. A 43-year old male(husband) was 69kg in body weight and healthy. A worm (6.65m in length; 8-l3mm in width) was expelled from him after, anthelmintic treatment. The second case(wife) was a 39-year old female who weighted 56kg. She complained about gastrointestinal trouble and abdominal discomfort. A worm (5.50m in length; 9-l3mm in width) was Obtained her after anthelmintic treatment. No scolex was collected from the two worms. Two patients were administrated with praziquantel(15mg/kg of body weight) and 15gm of magnesium sulfate as a purgative. Two worms identified as D. latum, .based on the following biological characters: extermal morphologies, coiling of uterus, the number of uterine loops, position of genital opening, morphologies of cirrus and cirrus sac and seminal vesicle on the histological sections, positions of vagina and uterine pore, distribution patterns of vitellaria, and microscopical and SEM morphologies of the egg.

Quantitative analysis of anthraquinones in Polygonum multiflorum Thunberg (하수오(何首烏)의 안트라퀴논 함량분석)

  • Objective: Polygonum multiflorum Thunberg (Polygonaceae) has been traditionally used as a tonic and the purgative in China and Korea. The genus Polygonum is a source of a wide range of phenolic compound, flavanoids, anthraquinones, stilbenes and tannins. In this study, three anthraquinones were isolated and quantitative determination of anthraquinones from P multiflorum has been developed for quality standardization. Methods : Three anthraquinone derivatives were isolated from a methanol extract of the radix of P. multiflorum by the chromatographic separation. Their structures were identified as emodin, physcion and ${\omega}$-hydroxyemodin on the basis of spectral data (MS, lH-NMR, 13C-NMR) and chemical analysis. HPLC analysis was performed to determine the contents of emodin, physcion, chrysophanol, rhein and ${\omega}$-hydroxyemodin in P. multiflorum from different specimens were collected from twenty Korean markets. Results: According to the results, the contents of emodin, physcion, chrysophanol, rhein and ${\omega}$-hydroxyemodin were 0.145%, 0.434%, 0.016%, 0.026%, 0.030% by HPLC, respectively. Conclusions : In these results, we have determined the contents of emodin, physcion, chrysophanol, rhein and ${\omega}$-hydroxyemodin in P. multiflorum, respectively. We hope that this study will contribute to the standardization and quality control of herbal medicine.
