Bee pollen is a valuable apitherapeutic product and has been known to have diverse biological activities, including antimicrobial, anti-inflammatory, and even anticancer activity. However, its effect on the immune system is not well studied and is rather controversial. This study intended to elucidate the biological activity of bee pollen on immunity. For this purpose, we used lyophilized bee pollen after wet grinding, which shows increased extraction of bioactive components and enhanced biological activity. First, lyophilized bee pollen after wet grinding significantly increased the proliferation of splenocytes isolated from normal mice. On the other hand, lyophilized bee pollen after wet grinding dose-dependently reversed splenocyte proliferation by concanavalin A or lipopolysaccharide. To clarify the activity of bee pollen on immunity lyophilized bee pollen after wet grinding was administered daily to mice for five weeks and isolated splenocytes. In this study, there was no significant difference in the population of immune cells and the size of spleen between bee pollen- and sterile water-treated groups. However, proliferation of splenocyte isolated from bee pollen-administered animals was boosted by both concanavalin A and lipopolysaccharide. Finally, kaempferol, a well-known flavonoid from bee pollen, dose-dependently increased splenocyte proliferation by both Con A and LPS. On the other hand, naringenin, another flavonoid in the bee pollen, dose-dependently inhibited the proliferation of splenocytes by Con A and LPS. Together, these data indicate that bee pollen may be able to prime the immunity to boost immune reaction after inflammation.
In order to investigate the effects of bisphenol A (BPA) on immune system in mice we examined the various immunological parameters. After single oral administration of BPA to female ICR mice, the weights of bodies and lymphoid organs, splenic cellularity and hematological parameters were examined on day 2 and 7. Among them WBC and splenic cellularity were slightly decreased on day 2. To assess the effects of BPA on humoral immune responses, splenic IgM plaque forming cell (PFC) and serum IgM were assayed. When BPA was administered after immunization with SRBC, but not before immunization, IgM PFC against SRBC was significantly lowered in a dose dependent manner. Serum IgM level was also decreased on day 4 when high dose (2000 mg/kg) of BPA was administrated after injection of OVA-antigen. The indexes of splenocyte proliferation (SP) to concanavalin A (Con A) and bacterial lipopolysaccharide (LPS) were measured in vitro by MTT assay. At low concentration BPA slightly increased splenocyte proliferation but at higher concentration it showed significant inhibitory effects on cell proliferation. Mitogen-stimulated SP was also determined with spleen cells from BPA treated mice. Con A-induced SP was slightly decreased and LPS-induced SP was especially inhibited at 1000 mg/kg and 2000 mg/kg of BPA. These results indicate that BPA is able to acutly evoke humoral and cell mediated immune suppression in mice.
Job's Tears(Yul-Moo) is a grass crop long-used as a traditional medicine; it is also a nourishing food. There are reports of its anti-inflammatory, stomachic, antiallergic activity, and antispastic effects and Job's Tears has been used in China to treat rheumatism, and neuralgia although its warts, rheumanism remains unclear. Thus, the present study was performed to investigate the in vitro effect of Job's Tears extracts on immune function. Here mouse splenocyte proliferation and cytokine production$(IL-1{\beta},\;IL-6,\;TNF-{\alpha})$ by peritoneal macrophages cultured with ethanol and water extracts of Job's Tears were examined. splenocytes proliferation increased with Job's Tears water extracts supplement at concentrations investigated The cytokine production$(IL-1{\beta},\;IL-6,\;TNF-{\alpha})$ by ELISA using a cytokine kit And $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ production increased water extracts supplementation. This in vitro study suggests that supplementation with Job's Tears water extracts may enhance immune function by regulating the splenocyte proliferation and enhancing cytokine production of activated macrophages.
Codonopsis lanceolata has long been used as a seasonal food and as a traditional tonic medicine with anti-inflammatory and anti-oxidation properties. The present study investigated the in vitro effect of Codonopsis lanceolata extracts on immune function in mice. After preparing a single cell suspension splenocyte proliferation was determined by the MTT(3-[4,5-dimethylthiazol-2-y]-2,5-diphenyl terazolium bromide) assay. The cytokines IL-1${\beta}$, IL-6, and TNF-$\alpha$ were not secreted by macrophages stimulated with or without LPS as determined by an ELISA cytokine kit assay. After a 48-hr incubation with the mitogens ConA or LPS there was an increase in splenocytes proliferation and in the production of IL-1${\beta}$, IL-6, and TNF-$\alpha$ in the suspensions supplemented with 50, 100, 250, 500 ${\mu}g/m{\ell}$ Codonopsis lanceolata water extract. The results suggest Codonopsis lanceolata water extract may enhance immune function by regulating splenocyte proliferation and stimulating cytokine production.
The present work was designed to investigate the effects of barzilin on ConAinducedd TCGF release, responsiveness to standardd IL-2, and mitogens-induced proliferation of splenocyte when administered intraperitoneally to 8 week-old C57BL/6 mice for 2 consecutive days. Immunological tests were performed 72 hours after the treatment of brailin. The administration of 50 mg/kg brazilin caused a noticeable increase in TCGF release and responsiveness to standard II-2 but inhibited mitogens-induced proliferation of splenocyte. These results that brazilin is able to modular immunological functions despite of its inhibitory effect on mitogen induced cell proliferation.
To investigate the effect of bovine colostral whey fractions on in vitro proliferation of mouse splenocytes, polypeptide fractions were separated from acid whey into 3 fractions depending on molecular weight by ultrafiltration: Fraction I, which contains the polypeptide larger than 10,000 Da, Fraction n, which contains the polypeptide ranging from 1,000 Da to 10,000 Da and Fraction III, which contains the polypeptide smaller than 1,000 Da. Fraction II showed the highest proliferative effect of mouse splenocytes among the colostral whey fractions and this proliferative activity increased in dose dependent manner. Unheated Fraction II and Fraction III showed significantly (p<0.01) higher proliferative effects than others but heated Fraction II showed the highest enhancing effect of mouse splenocyte among heated whey fractions (p<0.01). The supplementation of Fraction II and Fraction m showed greater proliferative effect of mouse splenocytes stimulated by concanavalin A (Con A) than that of whole whey or Fraction L Proliferative effect of mouse splenocytes stimulated by phytohemagglutinin (PHA) was the highest when Fraction II was supplemented Proliferative effect of the colostral whey fractions on mouse splenocytes by stimulation of lipopolysaccharide (LPS) was markedly enhanced by supplementation of Fraction II and Fraction m compared with whole whey and Fraction L It was estimated that colostral whey fraction containing IGF-I positively affected proliferation of mouse splenocyte.
Objectives : Silkworm is known as immunomodulatory substances and contain various bioactive compounds such as serine, tyrosine and alanine. The aim of this study was to investigated the immunopotentiating activity of combine extract that silkworm and food materials (Eucommia ulmoides, Angelica gigas, Acanthopanax, Allium hookeri, Cinnamomum cassia, Liriope platyphylla, Curcuma longa, Achyranthes japonica, Alpinia oxyphylla, Adenophora triphylla). Methods : Among 10 kinds of food materials, to select food materials with the effect of enhancing the immune function mouse splenocyte proliferation ability was measured by 3-[4,5-dimethylthiazol-2-y]-2,5-diphenyl terazolium bromide (MTT) assay. Then, combine extract of silkworm and food materials were evaluated that mouse splenocyte proliferation ability by EZ-cytox cell viability assay. Morever, cytokines production such as IL-2, IL-4, IL10, IL12, $IFN-{\gamma}$ on mouse T lymphocyte stimulated with concanavalin A (ConA) was measured. Results : Eucommia ulmoides, Acanthopanax, Allium hookeri, Cinnamomum cassia, Liriope platyphylla has high proliferation ability of mouse splenocyte compared with Curcuma longa, Achyranthes japonica, Alpinia oxyphylla, Adenophora triphylla. The silkworm and food material combined extract has a relatively high proliferation ability of mouse splenocyte proliferation when the silkworm and food materials are used as a single material. In particularly, combined extract of silkworm and Cinnamomum cassia was stimulate cytokine production on T lymphocyte such as IL12, $IFN-{\gamma}$. Combined extract of silkworm and Liriope platyphylla was stimulate cytokine production on T lymphocyte such as IL2, IL4, IL10. Conclusion : In conclusion, the combined extract of the silkworm and Cinnamomum cassia or Liriope platyphylla may enhance immune function by regulating mouse splenocyte proliferation and stimulating cytokine production.
Objectives : Samul-tang(SM). Sagunja-tang(SG). Pamul-tang(PM) and Sipjeondaebo-tang(SJ) was used many diseases such as sterility. menstrual disorder. general prostration. recruitment in Korea. We investigated the immune stimulatory activities of SG. SM. PM and SJ in in vitro. Methods: For comparision for effective of SM. SG. PM and SJ. this study examined anti-inflammation(NO. PGE2 assay). anti-oxidation(DPPH assay) and immune response in in vitro assay. For immune response activities. this study used NO synthesis on RAW 264.7 cells, splenocyte proliferation and cytokine assay(IL-2, IL-4) in splenocyte. Results: The results showed that SG. SM. PM and SJ were no significant effect anti-oxidation and anti-inflammatory effects. For immune response. they showed the splenocyte proliferation and macrophage proliferation. We confirmed that they synthesised NO a dose-dependent manner significantly and secreted the IL-4. Conclusions : These results suggested the SG, SM, PM and SJ had immune stimulatory activity. A efficacy of immune response in them had produced similar results.
Paeonia japonica var. pilosa $N_{AKAI}$, (PJ; Baek-Jak-Yak) is a medicinal plant which has been widely used as a component or blood-building decoctions. This study was performed to investigate the immunomodulative effects of PJ in mice, using in vitro and in vivo experiments. The immunomodulative effects were studied in vitro by determining the proliferation or mice splenocytes and the production of three kinds of cytokines (IL-1$\beta$, IL-6, TNF-$\alpha$) by mire peritoneal macrophages which were cultured with sequential fractions of PJ methanol extract (methanol, hexane, chloroform, ethylacetate, butanol and water). In an in vivo experiment using mice, different concentrations of PJ water extract were orally administrated every other day for two weeks. The production of cytokines (IL-1$\beta$, IL-6, TNF-$\alpha$) secreted by activated macrophages, and the proliferation of mice splenocytes, were used as indices for immunocompetence. In vitro supplementation using a hexane fraction of PJ in the range of 1 to 100 $\mu$ g/ml enhanced splenocyte proliferation by 1.8 to 12%, and by 10-15% using an aqueous fraction, compared to the control. IL-l$\beta$ production was significantly increased with the supplementation of butanol, hexane and water extracts of PJ Higher levels of IL-6 production were detected with supplementation of chloroform or water extracts. However, there were no significant differences in the production of TNF-$\alpha$ among the treated groups and the control. From the in vivo study, the highest proliferation of splenocytes was seen in the mice orally administrated with the PJ water extract at the concentration of 500 mg/kg body weight. In the case of cytosine production, IL-1-$\beta$, IL-6, and TNF-$\alpha$ released by activated peritoneal macrophages were augmented by the oral administration of a PJ water extract. These results indicate that Pl may enhance the immune function by regulating splenocyte proliferation and cytokine production capacity in mice.
Mancozeb, a polymeric complex of zinc and manganese salts of ethylene bisthiocarbamate (EBDC), is used widely in agriculture as fungicides, insecticides, and herbicides. Mancozeb can be occupationally and environmentally exposed to human and has been reported to induce estrogenic activity, therein it is considered as an endocrine disrupter, We investigated the effects of acute exposure of Mancozeb on the immune function in mice. After single oral administration of Mancozeb to female ICR mice, the immunopathological parameters (body- and organ-weight, splenic cellularity hematological parameters), mitogen (Con A, PHA+IL-2, LPS)-induced splenocyte proliferation (SP) and splenic IgM plaque forming cell (PFC). WBC and splenic cellularity were decreased, but liver-, kidney-, and spleen-weight were increased when compared with control group. Splenic IgM PFC against SRBC was slightly lowered. Mitogen-induced proliferation of spleen cells from Mancozeb-treated mice was not signifcantly changed ex vivo, however, SP in vitro were significantly lowered in concentration dependent manner. These results indicate that Mancozeb could affect the immune function in mice.
