Proliferation, Accumulation of Polyphenols, and Antioxidant Activities of Callus from the 'Anji Baicha' Cultivar of Tea [Camellia sinensis (L.) O. Ktze.]

  • Tea is one of the most consumed beverages worldwide and the relatively high levels polyphenols is benefit for health. In this study, we developed an efficient system for proliferation of callus from 'Anji Baicha', a cultivar of tea (Camellia sinensis). Callus tissue was initially induced by culturing leaf explants on medium containing different plant growth regulators. For callus induction, thidiazuron (TDZ) was more effective than 2,4-dichlorophenoxyacetic acid (2,4-D), ${\alpha}-naphthalene$ acetic acid (NAA), and $N^6-benzyladenine$ (BA). The frequency of callus induction from leaf explants reached 90.21% on $1.0mg{\cdot}L^{-1}$ TDZ and the developed callus was reddish and friable. We also tested the effect of different concentrations of NAA, 2,4-D, indole 3-acetic acid (IAA), BA, and TDZ, alone and in combinations, on callus proliferation. Medium supplemented with TDZ in combination with IAA was suitable for callus proliferation and accumulation of tea polyphenols. The growth index value and tea polyphenol content of callus cultured on MS medium containing $0.5mg{\cdot}L^{-1}$ TDZ and $1.0mg{\cdot}L^{-1}$ IAA was maximally 1,351% and 23.24%, respectively, and the relative abundance of epicatechin was as high as 17.449%. We also measured the antioxidant activity of all samples and the callus with the highest tea polyphenol content also exhibited high potential radical scavenging activity.

Polyphenol Analysis and Peroxynitrite Scavenging Effect of the Extracts from Eight Korean Mountainous Vegetable (한국 산채류 8종 추출물의 폴리페놀 함량분석 및 Peroxynitrite 소거효과)

  • Mountainous vegetables are usually used in the early stage of plant growth. We attempted to identify the quantity of polyphenols (caffoeoylquinic acids (CQs) and flavonoids) and peroxynitrite scavenging effect ($ONOO^-$) of eight Korean mountainous vegetables. The prominent characteristics were as follows: Only the roots of two plants Taraxacum platycarpum and Ixeris dentata contained chicoric acid. Five CQs were identified in the leaves of Cacalia firma whereas only 3-p-coumaroylquinic acid in the petioles of the plant was shown. The quantities of polyphenols such as quinic acid or tartaric acid derivatives in the mountainous vegetables were generally high, though those of flavonoids were very low. The $IC_{50}$ of chlorogenic acid, chicoric acid and kaempferol were 0.31, 0.12 and $0.25\;{\mu}g/ml$, respectively.

Construction of Data System on Seed Morphological Traits and Functional Component in Tartary Buckwheat Germplasms (쓴메밀 유전자원의 종자특성과 유용성분 변이에 관한 자원 정보 구축)

  • This study analyzed the phenotypes and chemotypes of 74 tartary buckwheat (Fagopyrum tataricum) germplasms using principal component analysis and cluster analysis. The average seed size of tartary buckwheat germplasm was 5.2 × 3.4 mm, which is smaller than the seed size of common buckwheat. The dark browned colored ovate or elliptic shape was mostly observed in collected germplasm. The average content of rutin was 1,393 mg per 100 g dry weight (DW) in tartary buckwheat seed. Similarly, the flavonoid and polyphenol contents ranged from 253 to 2,669 and 209 to 1,823 mg, respectively, per 100 g DW in the collected germplasm. The three components (PC1, 2, and 3) of principal component analysis revealed 68.55% of the total variance of the collected accessions. Cluster analysis using descriptors showed that 74 accessions were clustered into five groups. The study showed that the most interesting resources for functional breeding programs are: Five resources (HLB1004, HLB1005, HLB1007, HLB1009, and HLB1013) due to the rich rutin, polyphenol, and flavonoid.

Radical Scavenging Activities of Fruits of Crataegus pinnatifida BUNGE Major. from Korea

  • Screenings of potential antioxidant activities of Crataegus pinnatifida BUNGE Major. fruits extracted 80% methanol were performed using four antioxidant assays. Significant differences were observed both in total phenolic contents (TPC) and total flavonoid contents (TFC), DPPH radical scavenging activity, nitric oxide scavenging activity, ABTS radical scavenging assay, and reducing power assay. The total polyphenol content and total flavonoid content in the extract were measured to be $224.4{\pm}0.52$ mg GAE/100 g and $12{\pm}0.25$ mg QE/100 g, respectively. When the tested concentration was $500{\mu}g/mL$, DPPH and ABTS radical-scavenging activities of methanolic extracts were 84.15% and 88.8%, respectively. The reducing power and nitric oxide scavenging activity were increased at the manner of dose-dependently. These results suggest that methanolic extracts of Crataegus pinnatifida Bge. fruits possess excellent radical scavenging activities and may serve as a potential source of natural antioxidant.

Structure-Activity Relationship for Antidepressant Effect of Luteolin and Its Related Derivatives Isolated from Taraxacum mongolicum

  • The inhibitory effect and the structure-activity relationships of luteolin and its related derivatives isolated from Taraxacum mongolicum against MAO activities were investigated. The activity-guided isolation of extract from Taraxacum mongolicum led to the isolation of three flavonoids, luteolin, diosmetin, and luteolin-7-glucoside, a polyphenol, chlorogenic acid, a tyrosine and a uridine. The inhibitory activities of luteolin and its related derivatives against MAOs activities are dependent on their molecular structures. The presence of the phenolic hydroxy group at para-position is the active site for MAO-A inhibition as well as of MAO-B. The methoxy group has no potential on MAO-A inhibition. An additional phenolic hydroxy group at the ortho-position alleviates about 4-fold MAO-A inhibitory activity of phenolic hydroxy group at para-position. A carboxylic group seems to be critical for DBH inhibition and has no effects on MAO.

Growth Characteristics, Main Constituents and Antioxidant Activities in Local Accessions of Sophora flavescens AIT. (고삼 수집종의 생육특성, 주요성분 및 항산화 활성)

  • Background: Sophora flavescens AIT root is used as a herbal medicine in Asian culture. This study was aimed at selecting the best cultivars, by comparing growth characteristics, active ingredients, and antioxidant activities. Methods and Results: A total twenty nine accessions of S. flavescens were collected from five different regions in Korea. Plant height, leaf length, peduncle length, fresh root weight, and seed weight were the growth characteristics selected. Antioxidant activity was determined by measuring the total polyphenol (TPC) and total flavonoid contents (TFC) as well as the radical scavenging activity of DPPH, ABTS and FRAP. Oxymatrine and matrine, active ingredients in S. flavescens were analyzed by HPLC. Results indicated that accession YS-11 had the highest plant height (152.5 cm) and peduncle length (54.0 cm). The fresh root weight was highest in JA-01 at 4.9 kg, while the highest total seed weight was recorded in accession BH-04 at 77.7 g. The YS-03 accession contained the highest oxymatrine and matrine total contents (0.9616% and 0.9638%, respectively). Accessions BH-02, YS-11, YS-05, and MJ-03 had the highest levels of antioxidant activity (p < 0.05). Oxymatrine showed a high correlation with TPC and FRAP. TPC showed high correlations with TFC, DPPH, ABTS and FRAP. TFC showed a high correlation with DPPH. Conclusions: The superior lines can be selected for use as standard variety breeding material by comparing the growth characteristics of the accessions.

Evaluations on Antioxidant Effect of Methanol Extract from Immature Cotton Boll (미성숙 목화다래 메탄올 추출물의 항산화 효능 평가)

  • The results of the content of total polyphenol and flavonoid, DPPH (1-1-diphenyl-2-picryl-hydrazyl) radical scavenging activity, ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)] radical scavenging activity, nitrite scavenging activity and SOD-like activity of methanol extracts from immature cotton boll are follows. The contents of total polyphenol and flavonoid compound were higher in small size cotton boll, and DPPH and ABTS radical scavenging activity also showed a relatively high activity in the small size. These results indicate that there is a correlation between phenol content and DPPH radical scavenging, ABTS radical scavenging. The test concentrations of immature cotton boll extract for measuring DPPH and ABTS radical scavenging activities were set as 1.25, 2.5, 5, 10 and 20 mg/ml. Immature cotton boll has high radical scavenging activity at the concentration of 1.25~20 mg/ml and the result showed a tendency to increase in a concentration-dependent. The nitrite scavenging activity showed high activity in the pH 1.2, and the result in the pH 4.2 showed progressively less active, and in the pH 6.0 near neutral was confirmed that does not affect the nitrite scavenging. In addition, SOD-like activity showed somewhat lower activity compared with ascorbic acid, but tended to be higher when compared with the results of the other natural substances. Through this experiment, we confirmed that immature cotton boll was excellent antioxidant activity. Therefore, it is demonstrated that the cotton suggest the possibility of development of new material for cosmetic product or functional food in the future, and is expected to make a greater usability.

A Bioconversion Study on the Zanthoxylum schinfolium by Fermenting Bacteria and Their Functional Enhancement (유용 발효미생물 활용 생물전환 공정을 활용한 산초열매의 기능성 증대 방안 연구)

  • 생물전환(Bioconversion)은 천연소재의 기능성을 증대시키기 위한 방안으로 많은 연구가 진행되고 있으며 다양한 산업에서도 활용되고 있어 유용미생물을 기반 차세대 기술로 각광받고 있다. 이러한 기술의 도입은 식품은 물론 의약품 및 화장품 산업에서도 활발히 사용되고 있으며, 특히 최근 기능성 제품에 대한 소비가 급증함에 따라 그 중요성이 높아지고 있다. 산초(Zanthoxylum schinfolium)는 limonene, citronellal, phellandrene 등의 다양한 유효물질을 함유하고 있으며 유효성분들로부터 유래되는 항산화 활성과 항암활성, 항균활성 등의 효능을 지니는 것으로 보고된바 있다. 본 연구의 생물전환 공정에 사용된 유산균들은 전통 발효식품으로 알려진 다양한 젓갈류로부터 분리하였으며, 16S rDNA 염기서열 분석을 통해 유전학적 특성을 확인하였다. 또한 확보된 유산균들을 사용하여 산초(전북 진안군) 분말의 발효공정을 수행하였으며, 산초의 최적 추출조건을 선정하고 추출물을 제조하여 생물전환 공정 전 후 활성의 변화추이를 관찰하였다. 추출물의 활성평가는 항산화 효능 및 유효성분 함량을 평가하기 위하여 DPPH radical scavenging activity와 total polyphenol 함량을 평가하고 세포주를 활용해 MTT assay, Nitric oxide(NO) 생성억제 효능을 확인하여 세포독성 및 항염증 활성을 확인하였다. 실험결과, 생물전환 공정에 사용할 유용 미생물을 확보하기 위한 실험을 통해 다양한 젓갈류에서 다양한 미생물을 확보할 수 있었으며 약 16종의 유산균을 분리하였다. 분리된 미생물을 사용하여 산초 분말의 생물전환 공정을 실시한 결과, 5종의 유용미생물 처리에서 무처리 대비 DPPH radical 소거능 및 polyphenol 함량이 유의적으로 증가됨을 확인할 수 있었다. 그 중 가장 높은 활성을 나타내는 균주를 16S rDNA 염기서열 분석을 통해 확인한 결과 Weissella confusa D1로 확인되었다. 선별 균주를 활용한 생물전환 공정 후 항산화 활성은 대조군 대비 약 120%의 활성을 나타냈으며, polyphenol의 함량은 약 126%로 증가하는 것을 확인할 수 있었다. 더 나아가 생물전환 공정 후 산초추출물의 세포독성은 처리전과 비교하여 월등히 감소하는 경향을 확인할 수 있었으며, 항염효능 또한 증가하는 경향을 나타내는 것이 확인되었다.

Influence of Applied Pressure and Heat Treatment on Antioxidant Activities of Young Leaves from Achillea alpina and Solidago virgaurea subsp. gigantea (가압열처리가 톱풀과 울릉미역취 어린잎의 항산화 활성에 미치는 영향)

  • Present studies were conducted to investigate the effects of autoclaving on antioxidant activities of Achillea alpina and Solodago virgurea. At early April, young leaves of 2 species were collected, subjected to autoclaving ($121^{\circ}C$, 1.2 atmospheric pressure, 15 minutes), freezed-dried, grinded, and extracted with 80% ethanol. The same process was repeated with unautoclaved control. Total polyphenol and flavonoid contents, scavenging activities on DPPH and ABTS radicals and ferrous ion chelating effects were analyzed. Extraction yield of autoclaved S. virgurea was 39.55% and A. alpina 28.15%. In both species, autoclaving significantly reduced scavenging activities on DPPH and ABTS radicals. On the contrary, ferrous ion chelating effects increased after autoclaving, especially in young leaves of A. alpina. Autoclaving resulted in decrease of polyphenol and flavonoid contents, especially in the A. alpina. The present experiments demonstrated that autoclaving had negative effects on antioxidant activities of A. alpina and S. virgurea, except in ferrous ion chelating effects. Young leaves of former species were unstable to heat treatment, resulting in big lose of antioxidant activity.

Antioxidative Activity of Ethanol Extracts from Different Parts of Taraxacum officinale (민들레의 부위별 에탄올 추출물의 항산화 활성)

  • This study was conducted to investigate antioxidative and physiological activities of ethanol extracts from different parts (flower, leaf, root, and the whole plant) of Taraxacum officinale. The ethanol extracts from different parts were measured to examine total flavonoids content, total polyphenol content, elect ron donating ability,superoxide dismutase (SOD)-like activity, nitrite-scavenging ability and tyrosinase inhibition effects. Total flavonoids content in leaf extract (41.66 mg/g) and total polyphenol content in flower extract (71.91 mg/g) were higher than those of other parts. All assays were conducted at concentrations of 0.1, 0.3, 0.5, and 1 mg/mL ethanol extracts. The electron donating abilities of leaf, flower, the whole plant, and root extracts were 92.25%, 88.18%, 84.55% and 83.40%, respectively, at a concentration of 1 mg/mL. The activities were concentration dependent. The SOD-like activity of ethanol extracts from different parts was 8.40~11.20% at a concentration of 1 mg/mL. The nitrite-scavenging abilities of flower and leaf extracts measured at pH 1.2 were 47.37% and 47.18%, respectively, at a concentration of 1 mg/mL, which were higher than those of the whole plant and root extracts. Tyrosinase inhibition activity of the leaf extract at a concentration of 1 mg/mL was the highest (34.22%) and that of the whole plant and root extracts was shown to be more than 20%. These results suggest that ethanol extracts from different parts of Taraxacum officinale could be used as antioxidative functional food sources.
