자외선으로 유도된 Hs68 섬유아세포의 노화 반응에 대한 영실추출물의 억제 효능 (Inhibitory Effect of Rosa multiflora hip Extract on UVB-induced Skin Photoaging in Hs68 Fibroblasts)

  • 극심하고 지속적인 자외선에의 노출은 정상적인 피부구조를 파괴하는 다양한 피부 광노화 과정을 야기한다. 자외선은 인체 피부에서 세포외기질의 구성성분을 분해시키는 기질 분해효소인 matrix metalloproteinases(MMPs)의 발현을 활성화시키고, 콜라겐 합성은 감소시킴으로써 피부의 탄력과 구조적 치밀도를 약화시켜 궁극적으로 피부주름을 생성한다. 본 연구에서는 이러한 피부 광노화 현상을 완화시키는 소재로서 영실의 효능을 검증하고자 하였다. 먼저 인간 섬유아세포주인 Hs68을 이용하여 영실의 세포증식 촉진효능을 확인하였다. 여기에 더해 영실이 activator protein (AP)-1 전사인자의 억제를 통해 MMP의 발현을 감소시킴을 mRNA 및 단백질 수준에서 검증하였다. 또한, 진피층을 구성하는 타입 I형 콜라겐과 표피-진피 경계부를 단단히 고정시키는 역할을 하는 타입 IV형 콜라겐 역시 영실에 의해 발현이 증가하며, 자외선에 의한 염증반응의 억제에도 영실이 효과적으로 작용하는 것을 확인할 수 있었다. 결론적으로 본 연구를 통해 영실이 자외선에 의한 피부노화와 주름생성을 효과적으로 개선할 수 있는 가능성을 가짐으로써 항노화, 항염증 및 항주름 소재로서 화장품에 응용될 수 있을 것으로 기대된다.

천정기보단(天精氣保丹)의 자외선에 의한 세포 손상 억제 효과 (Protective Effect of Cheonjeongkibo-Dan UV-Induced Cellular Damage in Human Dermal Fibroblast)

  • In this study, we prepared CheonJeongKiBo-Dan(7 oriental medicinal plants, 7OMP: Astragalus Membranaceus root, Panax Ginseng root, Glycyrrhiza Glabra (licorice) root, Schizandra Chinensis fruit, Polygonatum Odoratum, Rehmannia Glutinosa root, Paeonia Albiflora root) by extracting them in one reactor and studied its efficacies on skin. UV irradiation has been suggested as a major cause of photoaging in skin. In order to investigate protective effects against UV-B induced cellular damage, 7OMP was extracted with 70% ethanol and dissolved in DMSO. The protective effect was detected by MTT assay, reactive oxygen species (ROS) generation, phosphorylation of ATR and p53 in human dermal fibroblast cell system after UV-B irradiation. 7OMP reduced UV-B-induced cellular damage in HDFs cells, and inhibited ROS generation. UV-B-induced toxicity accompanying ROS production and the resultant DNA damage are responsible for activation of ATR, p53 and Bad. In this study, 7OMP hampered phosphorylations of ATR and p53 in human dermal fibroblasts. Therefore, 7OMP may be protective against UV-induced skin photoaging.

Ellagic acid plays a protective role against UV-B-induced oxidative stress by up-regulating antioxidant components in human dermal fibroblasts

  • Ellagic acid (EA), an antioxidant polyphenolic constituent of plant origin, has been reported to possess diverse pharmacological properties, including anti-inflammatory, anti-tumor and immunomodulatory activities. This work aimed to clarify the skin anti-photoaging properties of EA in human dermal fibroblasts. The skin anti-photoaging activity was evaluated by analyzing the reactive oxygen species (ROS), matrix metalloproteinase-2 (MMP-2), total glutathione (GSH) and superoxide dismutase (SOD) activity levels as well as cell viability in dermal fibroblasts under UV-B irradiation. When fibroblasts were exposed to EA prior to UV-B irradiation, EA suppressed UV-B-induced ROS and proMMP-2 elevation. However, EA restored total GSH and SOD activity levels diminished in fibroblasts under UV-B irradiation. EA had an up-regulating activity on the UV-B-reduced Nrf2 levels in fibroblasts. EA, at the concentrations used, was unable to interfere with cell viabilities in both non-irradiated and irradiated fibroblasts. In human dermal fibroblasts, EA plays a defensive role against UV-B-induced oxidative stress possibly through an Nrf2-dependent pathway, indicating that this compound has potential skin antiphotoaging properties.

Fermented Acanthopanax koreanum Root Extract Reduces UVB- and H2O2-Induced Senescence in Human Skin Fibroblast Cells

  • The present study assessed the effects of an aqueous extract of Acanthopanax koreanum root (AE) and of AE following fermentation by lactic acid bacteria (Lactobacillus plantarum and Bifidobacterium bifidum) (AEF) on human skin fibroblast HS68 cells exposed to ultraviolet B (UVB) irradiation and oxidative stress. AEF effectively antagonized the senescence-associated β-galactosidase staining and upregulation of p53 and p21Cip1/WAF1 induced by UVB or H2O2 treatment in HS68 cells. It also exhibited excellent antioxidant activities in radical scavenging assays and reduced the intracellular level of reactive oxygen species induced by UVB or H2O2 treatment. The antioxidant and antisenescent activities of AEF were greater than those of nonfermented A. koreanum extract. AEF significantly repressed the UVB- or H2O2-induced activities of matrix metalloproteinase (MMP)-1 and -3, overexpression of MMP-1, and nuclear factor κB (NF-κB) activation. This repression of NF-κB activation and MMP-1 overexpression was attenuated by a mitogen-activated protein kinase activator, suggesting that this AEF activity was dependent on this signaling pathway. Taken together, these data indicated that AEF-mediated antioxidant and anti-photoaging activities may produce anti-wrinkle effects on human skin.

Photoprotective effects of topical ginseng leaf extract using Ultraflo L against UVB-induced skin damage in hairless mice

  • Background: Abnormal activation of matrix metalloproteinases (MMPs) plays an important role in UV-induced wrinkle formation, which is a major dermatological problem. This formation occurs due to the degeneration of the extracellular matrix (ECM). In this study, we investigated the cutaneous photoprotective effects of Ultraflo L treated ginseng leaf (UTGL) in hairless mice. Methods: SKH-1 hairless mice (6 weeks of age) were randomly divided into four groups (8 mice/group). UTGL formulation was applied topically to the skin of the mice for 10 weeks. The normal control group received nonvehicle and was not irradiated with UVB. The UV control (UVB) group received nonvehicle and was exposed to gradient-UVB irradiation. The groups (GA) receiving topical application of UTGL formulation were subjected to gradient-UVB irradiation on $0.5mg/cm^2$ [GA-low (GA-L)] and $1.0mg/cm^2$ [(GA-high (GA-H)] of dorsal skin area, respectively. Results: We found that topical treatment with UTGL attenuated UVB-induced epidermal thickness and impairment of skin barrier function. Additionally, UTGL suppressed the expression of MMP-2, -3, and -13 induced by UVB irradiation. Our results show that topical application of UTGL protects the skin against UVB-induced damage in hairless mice and suggest that UTGL can act as a potential agent for preventing and/or treating UVB-induced photoaging. Conclusion: UTGL possesses sunscreen properties and may exhibit photochemoprotective activities inside the skin of mice. Therefore, UTGL could be used as a potential therapeutic agent to protect the skin against UVB-induced photoaging.

자외선 조사 마우스에서 만성 피부손상에 대한 분죽(Phyllostachys nigra var. henenis Strapf)잎 추출물의 효과 (Photoprotective Effect of Bamboo (Phyllostachys nigra var. henenis Strapf) Leaf Extract against Ultraviolet Radiation-induced Chronic Skin Damage in the Hairless Mouse)

  • To evaluate the ability of Bamboo (Phyllostachys nigra var. henenis Strapf) leaf extract (BL) to protect the skin from photodamage, the gross and microscopic changes in the skin of hairless mice and BL-treated mice exposed chronically to ultraviolet (UV) were examined. The skin of the UV-irradiated mice showed characteristic signs of photoaging, such as deep wrinkles across the back, increased epidermal thickeness, numerous cell infiltration, and many enlarged keratinizing cysts. BL-treated mice showed a significantly decreased wrinkling score and lack of proliferation of cysts. By the 22nd week, 88.9% (i.p. with saline) or 60.0% (topical administration with cream base) of the UV-irradiated mice developed at least one tumor. BL delayed tumor onset significantly. BL (i.p.) was also effective in reducing the occurrence of UV radiation-induced skin tumors and reduced the number of tumors per mouse. After 22 weeks of treatment, 37.5% (i.p.) of the mice treated with BL were tumor-free. Tumor multiplicity was reduced by 81.2% (i.p.) in the BL treated groups. It is noted that skin that is chronically exposed to UV is subject to photoaging and photocarcinogenesis and regular use of BL would prevent these photodamaging effects of UV.

Pyrithione-zinc Prevents UVB-induced Epidermal Hyperplasia by Inducing HIF-$1{\alpha}$

  • Epidermal keratinocytes overgrow in response to ultraviolet-B (UVB), which may be associated with skin photoaging and cancer development. Recently, we found that HIF-$1{\alpha}$ controls the keratinocyte cell cycle and thereby contributes to epidermal homeostasis. A further study demonstrated that HIF-$1{\alpha}$ is down-regulated by UVB and that this process is involved in UVB-induce skin hyperplasia. Therefore, we hypothesized that the forced expression of HIF-$1{\alpha}$ in keratinocytes would prevent UVB-induced keratinocyte overgrowth. Among several agents known to induce HIF-$1{\alpha}$, pyrithione-zinc (Py-Zn) overcame the UVB suppression of HIF-$1{\alpha}$ in cultured keratinocytes. Mechanistically, Py-Zn blocked the degradation of HIF-$1{\alpha}$ protein in keratinocytes, while it did not affect the synthesis of HIF-$1{\alpha}$. Moreover, the p21 cell cycle inhibitor was down-regulated after UVB exposure, but was robustly induced by Py-Zn. In mice repeatedly irradiated with UVB, the epidermis became hyperplastic and HIF-$1{\alpha}$ disappeared from nuclei of epidermal keratinocytes. However, a cream containing Py-Zn effectively prevented the skin thickening and up-regulated HIF-$1{\alpha}$ to the normal level. These results suggest that Py-Zn is a potential agent to prevent UVB-induced photoaging and skin cancer development. This work also provides insight into a molecular target for treatment of UVB-induced skin diseases.

콜라겐 트리펩타이드를 함유한 콜라겐 가수분해물의 피부 광노화 예방 효과 (Beneficial effect of collagen hydrolysate containing collagen tripeptides on ultraviolet B-induced skin photoaging)

  • 본 연구는 식용 어류로부터 획득한 콜라겐 중 Gly-Pro-Hyp를 주요 지표로 하는 고함량 콜라겐 트리펩타이드에 대한 피부의 광노화 억제 효과를 확인하고자 HaCaT 세포와 SKH-1 hairless 마우스를 활용한 전임상 실험을 실시하였다. HaCaT 세포는 UVB 조사 이후 Collagen 1A와 MMP-1 그리고 -13의 변화를 관찰하였으며 마우스 실험은 총 50마리의 마우스를 5군으로 구분한 다음 NOR을 제외한 모든 마우스에 UVB를 조사하고 양성대조군인 retinoic acid 혹은 실험물질인 CTP (17, 34 mg/kg)를 섭취시키며 실험하였다. CTP는 HaCaT 세포의 Collagen 1A 증가 및 MMP-1와 -13 감소에 긍정적인 영향을 미쳤을 뿐만 아니라 UVB 조사를 통해 증가된 실험동물의 피부 주름 수와 깊이 그리고 넓이를 현저히 개선시키는 효과를 나타내었다. 특히, CTP의 섭취는 UVB 조사에 따라 파괴된 피부조직 내 콜라겐의 양과 밀도를 정상적으로 회복시켰을 뿐만 아니라 Collagen 1A의 발현 증가 및 MMP1과 -13의 발현 감소를 유도함으로써 UVB 조사에 의한 주름 생성을 감소시킨 효과를 보여주었다. 따라서, 본 실험은 CTP 섭취가 콜라겐 붕괴에 관여하는 MMPs 단백질 발현을 감소시키고 이와는 반대로 Collagen 1A의 발현은 증가시킴을 확인함으로써 피부 외관의 주름 수와 넓이 혹은 깊이를 개선시키는 것에 긍정적인 영향을 미치는 것으로 판단할 수 있다.

  • The clinical characteristics of photodamaged skin, such as coarse and fine wrinkle are not accurately evaluable with previous methods. Public awareness for wrinkle treatment and prevention which rely on proper assessment and evaluation of the underlying skin changes has been increased as the population ages. In this paper, we suggest an in-vivo method and instrument that allow us to acquire a wrinkle-enhanced image to improve the accuracy of quantitative and qualitative analysis for skin wrinkle. The method used involved white LED illumination and photography through polarizing filters. Finally, the polarized light photography yields an accurate and evaluable parameter of photodamaged skin, especially fine and coarse wrinkle.

Pueraria montana var. lobata Root Extract Inhibits Photoaging on Skin through Nrf2 Pathway

  • Pueraria montana var. lobata is a bioactive substance with various beneficial health effects and has long been extensively used as a traditional medication for the treatment of fever, acute dysentery, diabetes, and cardiovascular diseases in Northeast Asian countries. The purpose of this study was to evaluate the cytoprotective activity of Pueraria montana var. lobata ethanol extract (PLE) for ultraviolet B (UVB)-induced oxidative stress in human dermal fibroblasts (HDF). It was hypothesized that PLE treatment ($25-100{\mu}g/ml$) would reduce intracellular reactive oxygen species (ROS) levels as well as increase collagen production in UVB-irradiated HDF. The results confirmed this theory, with collagen production increasing in the PLE treatment group in a dose-dependent manner. In addition, regulators of cellular ROS accumulation, including HO-1 and NOQ-1, were activated by Nrf2, which was mediated by PLE. Hence, intracellular levels of ROS were also reduced in the PLE treatment group in a dose-dependent manner. In conclusion, PLE increases collagen production and maintains hyaluronic acid (HA) levels in human dermal fibroblasts exposed to UVB-irradiation, thereby inhibiting photoaging.
