Photo-protective and Anti-melanogenic Effect from Phenolic Compound of Olive Leaf (Olea europaea L. var. Kalamata) Extracts on the Immortalized Human Keratinocytes and B16F1 Melanoma Cells

  • Ethylacetate and butanol fractions of leaf extracts (OLE) showed the higher contents of total phenolic compounds than hexane and water fractions. Oleuropein contents were $4.21{\pm}0.57,\;3.92{\pm}0.43,\;0.32{\pm}0.03,\;5.76{\pm}0.32$, and $32.47{\pm}0.25mg$/100g for ethanol extract, and hexane, chloroform, ethyl acetate, and butanol fraction, respectively. Treatment of ultraviolet-B (UVB) irradiated cells with 3 OLEs prepared by using ethylacetate and butanol at concentrations 0.001, 0.005, and 0.01% respectively showed significant recovery of cell viabilities. Treatment of dexametason 1 mM reduced tumor necrotic factor (TNF)-${\alpha}$ secretion by about 40%. UVB irradiated immortalized human keratinocytes (HaCaT) cells were treated with 3 different OLEs at the same concentrations. Ethylacetate fraction showed the strongest inhibition activity with respect of reduction of the elevated (TNF)-${\alpha}$. Cytotoxicity of OLEs on the B16-F1 cells was evaluated through thiazolyl blue tetrazolium bromide (MTT) assay. Ethylacetate fraction has no cytotoxicity in the range of 0.005-0.01%. A slight cytotoxicity was observed at the concentration of 0.1% butanol fraction of OLE that caused 10% decrease in cell viability.

  • PV module protective film plays an important role in protecting the solar cell from external environment by anti-hydrolysis polyester, UV resistance and mechanical properties. The backsheet was manufactured by using Roll-to-Roll dry laminating process. The backsheet structure is composed of 3 layers, which are PE, PET, and Fluorine polymer films. In this study, we have experimented the variation of thermal conductivities depending on MgO inputs 10% to 25% in order to confirm the dependence of the module efficiencies. High thermal conductive backsheet can increase the module output power efficiency because the heat is dissipated by spreading out the internal heat. Long-term environment weatherability tests were conducted for confirming 25 year reliability in the field such as PCT, UV, and power efficiency degradations. As the evaluation result, high thermal conductivity can be effective for increase of power efficiency of solar panel by using thermal conductive MgO masterbatch.

Protection of Skin Fibroblasts from Infrared-A-Induced Photo-Damage Using Ginsenoside Rg3(S)-Incorporated Soybean Lecithin Liposomes

  • Protection of skin cells from chronic infrared-A (IRA) irradiation is crucial for anti-photoaging of the skin. In this study, we investigated the protective activity of Rg3(S) and Rg3(S)-incorporated anionic soybean lecithin liposomes (Rg3/Lipo) with a size of approximately 150 nm against IRA-induced photodamage in human fibroblasts. The formulated Rg3/Lipo showed increased solubility in aqueous solution up to a concentration of 200 ㎍/ml, compared to free Rg3(S). In addition, Rg3/Lipo exhibited superior colloidal stability in aqueous solutions and biocompatibility for normal human dermal fibroblasts (NHDFs). After repeated IRA irradiation on NHDFs, elevated levels of cellular and mitochondrial reactive oxygen species (ROS) were greatly reduced by Rg3(S) and Rg3/Lipo. In addition, cells treated with Rg3/Lipo exhibited noticeably reduced apoptotic signals following IRA irradiation compared to untreated cells. Thus, considering aqueous solubility and cellular responses, Rg3/Lipo could serve as a promising infrared protector for healthy aging of skin cells.

Combination of Grapefruit and Rosemary Extracts Has Skin Protective Effect through MMPs, MAPKs, and the NF-κB Signaling Pathway In Vitro and In Vivo UVB-exposed Model

  • Long-term ultraviolet (UV) exposure accelerates the phenomenon of skin photo-aging by activating collagenase and elastase. In this study, we aimed to investigate the effects of a combination of grapefruit and rosemary extracts (cG&Re) on UVB-irradiated damage in HaCaT cells and dorsal mouse skin. In HaCaT cells, cG&Re recovered UVB-reduced cell viability and inhibited protein expression of mitogen-activated protein kinases (MAPKs), such as extracellular signal-regulated kinases (p-Erk), c-Jun N-terminal kinases (p-JNK), and a class of MAPKs (p-P38). Also, cG&Re suppressed UVB-induced collagen and elastin degradation by decreasing matrix metalloproteinases (MMPs) and nuclear factor kappa light chain enhancer of activated B cells (NF-κB) expression, which is a transcription factor. Similar results were observed in dorsal mouse skin. Taken together, our data indicate that cG&Re prevent UVB-induced skin photo-aging due to collagen/elastin degradation via activation of MAPKs, MMPs, and the NF-κB signaling pathway in vitro and in vivo.

Inhibitory Effects of a Combination of Grapefruit and Rosemary Extracts for Alleviating UV-Induced Skin Ageing

  • Ultraviolet (UV) radiation is associated with the development of extrinsic skin aging. We performed in vivo assays in order to investigate the protective effect of a combination of grapefruit and rosemary extracts (cG&Re) on UVB-induced skin aging. The results indicated that cG&Re displayed elastase inhibitory activity in a dose-dependent manner. Topical application of cG&Re mitigated photo-aging related lesions such as skin erythema and thickening in photo-aged BALB/c mice dorsal skin, by preventing UVB-induced collagen degradation. Immunohistochemical analyses revealed that cG&Re stimulated SIRT-1 expression, and suppressed MMP-1 and $IL-1{\beta}$ expression. It was observed that expression of MMP-1 and -13 mRNA was downregulated in the cG&Re-treated group. Furthermore, cG&Re treatment drastically suppressed protein expression of MMP-1 and regulated the phosphorylation of p-38 kinase. As expected, oral administration of cG&Re resulted in the same SIRT-1, MMP-1, and $IL-1{\beta}$ expression patterns observed upon topical application of cG&Re in the UV-induced mice model. Overall, the current results demonstrated that cG&Re attenuated both the downregulation of MMP-1 expression and up-regulation of SIRT-1 expression, as well as decreased phosphorylation of MAPK in UVB-induced skin ageing mice model, suggesting that cG&Re might be used as an internal food ingredient for beauty-purposes as well as a functional food material.

광용혈에 대한 Ketocarotenoids의 현저한 세포 보호작용에 관한 연구

  • ${\beta}$-Carotene has been known as an effective quenching agent of singlet oxygen and the carotenoid pigments in general are expected to protect cells against photosensitized oxidations. We are determined the quenching rate constants of several Ketocarotenoids including capsanthin, capsanthin diester, astaxanthin and fucoxanthin, and the relative quenching actiyities against singlet oxygen were compared with those of ${\beta}$-carotene and reported carotenoids. Nevertheless the ketocarotenoids exhibited lower quenching rate constants than ${\beta}$-carotene, they showed more pronounced protective activitives than ${\beta}$-carotene against photohemlysis induced by singlet oxygen. Among the ketocarotenoids investigated, fucoxanthin indicated a significant protective activity for the cell. The results suggested that. 1) 1O2 may be alikely initiator of photohemolysis, but this reaction is followed by slow dark reactions involving secondary reactive species. 2) For protection of RBC against photodynamic action with carotenoids, carotenoids having functional groups such as -C=0 and -OH groups are most efficient. This suggests that partition of carotenoids between the buck and the mombrane and/or their specific binding to membrane proteins are more critical for the photo-protection by carotenoids than is a diffusional quenching of 1O2.

미강유의 광산화와 항산화제의 방어작용 (Photo oxidation of Rice Bran Oil and Protective Action of Antioxidants)

  • To investigate the protective action of various antioxidants on the photooxidation of rice bran oil, it was irradiated with red and visible light in presence and absence antioxidants with or without sensitizer (methylene blue). 1. Rice bran oil with and without sensitizer in chloroform-ethanol (4:1, v/v) media was largely oxidized under visible light irradiation. on the other hand, rice bran oil without sensitizer was hardly oxidized under red light irradiation. 2. Rice bran oil with sensitizer was oxidized much faster than that without. And the absorbance of it irradiated with visible was increased more than that with red light. 3. The effectiveness of antioxidants on the rice bran oil photooxidation was same order as follows: Irradiation with visible light, no addition of sensitizer ${\beta}$-carotene > dl-{$\alpha}$-tocopherol > BHT > BHA Irradiation with visible light, addition of sensitizer ${\beta}$-carotene > BHT > dl-${\alpha}$tocopherol > BHA Irradiation with red light, addition of sensitizer ${\beta}$-carotene > BHA > BHT > dl-${\alpha}$-tocopherol From these results, we concluded that rice bran oil was largely oxidized under visible light irradiation, therefore it must be protected from photooxidative deterioration by the addition of antioxidants.

Hairless mice에서의 대두 홍삼 혼합 분말(APB-03)의 경구 반복 투여 시 피부 주름 생성 예방 효과 (Prevention of Photoaging and Wrinkle Formation in Hairless Mice Dorsal Skin by APB-03)

  • 본 실험은 대두 추출물과 홍삼 농축액 분말 혼합물인 APB-03(AmorePacific Beauty-03)의 경구반복투여에 대한 피부주름개선 효과를 평가하기 위하여 hairless mouse에서 경구 반복투여 및 UV 반복조사를 통한 효능평가시험을 실시하였다. 정상군(Control group)과 UV 대조군(UV control group), UV/APB-03 저용량군(UV/APB-03(XI) group), UV/APB-03 중간용량군(UV/APB-03(X5) group)과 UV/APB-03 고용량군(UV/APB-03(X10) group)을 두고, 경구로 주 5회 반복 투여하며, 광노화를 유발하기 위하여 주 3회 UV를 조사가 병행되었다. 시험 기간 중 임상증상, 체중변화 및 사료 음수 섭취량 등을 관찰하고, 2회의 피부주형을 채취하여 영상 분석을 실시하였으며 실험 종료 후 피부조직을 채취하여 병리조직학적 검사를 실시하였다. 시험물질을 투여한 모든 동물에서 시험 전 기간을 통해 사망한 동물은 없었으며, 본 시험물질에 의한 것이라고 인정되는 임상증상, 행동이상 및 체중변화 등도 관찰되지 않았으며, 부검시에도 본 시험물질의 투여에 기인한 것으로 사료되는 어떠한 육안적 병변도 관찰되지 않았다. UV 조사 기간 중, 육안으로 관찰 시 정상군에 비해 UV 대조군의 피부주름 증가가 뚜렷하였으며, APB-03을 투여한 군은 UV 대조군에 비해 주름 증가가 현저히 감소하였으며, 정상군과 비슷할 정도로 피부주름 상태가 양호하였다. 8주 및 12주에 제작된 replica를 image analyzer 분석평가를 실시한 결과, APB-03을 경구 반복 투여한 실험군은 UV 대조군에 비해 전체적으로 주름이 감소하였으며, 실험 12주의 UV/APB-03 고용량군의 피부주름은 UV 대조군에 비해 통계적으로 유의성 있게 감소하는 것을 알 수 있었다(p<0.01). H&E(Hematoxylin & Eosin) stain을 실시한 결과, UV 조사에 의한 표피 및 진피, 피하지방층에서의 손상에 대해 APB-03의 치유능력을 관찰할 수 있었다. 이상의 실험 결과를 종합해 볼 때, 암컷 hairless mouse에서 APR-03의 경구 반복 투여 시 UV 조사로 유도된 광노화의 억제 및 피부주름예방효과가 확인되었다.

  • This paper deals with the lightning surge counter. In order to install the effective surge protective devices, it is important to find the parameters of incident surges. For the purpose of observing the occurrence frequency as a parameter of the amplitude of surge, two type surge counters were designed and fabricated. One is operated by surge currents, and the other is operated by surge voltages. The former consists of current sensor, metal oxide varister (MOV), rectifier, capacitor and electromagnetic counter. The latter consists of rectifier, voltage divider, comparator, photo coupler and counter circuit, and is useful for detecting the surge voltages.

홍삼과 어유(魚油) 복합식이에 의한 UV-유도 광노화 억제효과 (Protective Effects of Complex Diet of Korean red ginseng and fish oil on UV-induced Photoaging)

  • Objective : UV irradiatiion causes skin-aging involving coarse wrinkles, thickning, dyspigmentation, and rough skin surface. This study was aimed to elucidate the anti-winkle activity of complex diet of Korean red ginseng (RG) and fish oil (FO) on UV-induced Photoaging. Methods : To investigated photo-protective effects of Korean red ginseng and fish oil on UV-induced damaged skins, SKH hairless female mice were randomly divided into six groups [control, UV, UV/RG, UV/FO, UV/RG/FO(low), UV/RG/FO(high)]and orally administered three times a week respectively. UV radiation was applied to the backs of the mice three times a week for 8 weeks. Expressions of matrix metalloproteinase (MMP)-3, MMP-13 and tissue inhibitor matrix metalloproteinase (TIMP)-1 in skin were measured by immunohistochemical staining. Results : In this study, UVB-induced epidermal hypertropy was diminished by RG group or FO group or complex group of RG and FO. Expression levels of MMP-3 and MMP-13 were reduced and expression level of TIMP-1 was increased by RG group or FO group or complex group of RG and FO. Especially MMP-3 and MMP-13 were markedly reduced by diet of FO and complex diet of RG and FO compared with untreated group. Conclusions : This results suggest that complex diet of RG and FO have a anti-wrinkle activity on UV-induced photo-aging and intrinsic aging.
