Effects on antioxidant, whitening, and anti-wrinkle improvement of ethanol extracts from fermented Trigonotis radicans var. sericea by Lactobacillus brevis (Lactobacillus brevis 발효가 참꽃마리(Trigonotis radicans var. sericea) 에탄올 추출물의 항산화, 미백 및 주름 개선 활성에 미치는 영향)

  • This study was designed to compare the effects on antioxidant, whitening, and anti-wrinkle improvement of 60% ethanol extracts from Trigonotis radicans var. sericea (TR) and Lactobacillus brevis-fermented T. radicans var. sericea (FTR). The total phenolics contents of TR was 18.6 mg/g, and FTR was 18.2 mg/g. The HPLC analysis confirmed more phenolic compounds were produced in FTR by bioconversion than in TR. The FTR showed higher 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity than TR. The results of the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation decolorization assay, FTR and TR showed higher activity than BHT (positive control). The FTR (1.21-1.29 PF) showed higher antioxidant protection factor (PF) activity than the TR (1.15-1.22 PF) and showed higher tyrosinase, collagenase and elastase inhibitory activities. This results showed L. brevis -fermentation of Trigonotis radicans var. sericea changed the composition profile and has effects on antioxidant, whitening and anti-wrinkle improvement.

Physicochemical and Sensory Properties, and Bioactive Compounds of Blended Grape Juice from Different Grape Varieties (캠벨과 타품종 포도주스의 브렌딩에 의한 품질 및 기능특성)

  • Campbell juice (25%, 50%, 70% and 90%) was blended with different concentrations (10%, 25%, 30% and 50%) of juice from a range of grape varieties including Kyoho, Steuben and MBA. The concentrations of Campbell and the grape variety juices influenced the physicochemical characteristics of the blended grape juice. As the concentration of Campbell juice increased, there was an increase in the mean content of bioactive compounds including total phenolics, anthocyanin and flavonoids, and the radical scavenging activity increased. The treatment containing 25% juice from the various grape varieties had the lowest level of bioactive compounds. A sensory evaluation showed there was no significant difference among the blended juice samples in terms of aroma. The sample comprising 50% Campbell + 50% Kyoho was the most acceptable in terms of taste and overall acceptability, while the sample comprising 90% Campbell + 10% MBA was the most acceptable in terms of color and aroma.

Quantitative Assessment of the Relative Antineoplastic Potential of the n-butanolic Leaf Extract of Annona Muricata Linn. in Normal and immortalized Human Cell Lines

  • Natural products have been the target for cancer therapy for several years but there is still a dearth of information on potent compounds that may protect normal cells and selectively destroy cancerous cells. The present study was aimed to evaluate the cytotoxic potential of n-butanolic leaf extract of $Annona$ $muricata$ L. on WRL-68 (normal human hepatic cells), MDA-MB-435S (human breast carcinoma cells) and HaCaT (human immortalized keratinocyte cells) lines by XTT assay. Prior to cytotoxicity testing, the extract was subjected to phytochemical screening for detecting the presence of compounds with therapeutic potential. Their relative antioxidant properties were evaluated using the reducing power and $DPPH^*$radical scavenging assay. Since most of the observed chemo-preventive potential invariably correlated with the amount of total phenolics present in the extract, their levels were quantified and identified by HPLC analysis. Correlation studies indicated a strong and significant (P<0.05) positive correlation of phenolic compounds with free radical scavenging potential. The results revealed that the extract was moderately cytotoxic to normal cells with a mean IC50 value of 52.4 ${\mu}g$ when compared with those obtained for cancerous cells (IC50 values of 29.2 ${\mu}g$ for MDA-MB-435S and 30.1 ${\mu}g$ for HaCaT respectively). The study confirms the presence of therapeutically active antineoplastic compounds in the n-butanolic leaf extract of $Annona$ $muricata$. Isolation of the active metabolites from the extract is in prospect.

Phenolic Compounds and Antioxidant Activity in Cell Wall Materials from Deodeok (Codonopsis lanceolata) (더덕 부위별 세포벽 물질의 페놀성화합물과 항산화 활성)

  • This study measured the amounts of uronic acid, total sugars, non-cellulosic neutral sugars, phenolic compounds as well as antioxidants activity in cell wall materials (CWM) derived from different parts of deodeok (Codonopsis lanceolata). The values of the uronic acid (UA): neutral sugars (NS) ratio in polymers extracted from the CWM of the flesh and skin were 4 and 6, respectively. The total sugar contents of the flesh and skin were 788.6 and 824.9 ${mu}g/mg$ of CWM, respectively. Galactose and arabinose were the main noncellulosic neutral sugars. The chemical structure of five phenolic compounds from the CWM were analyzed and identified as vanillic acid, p-OH-benzaldehyde, vanillin, ferulic acid, and 8-O-4' diferulic acid by HPLC spectral data. Among them, p-OH-benzaldehyde, vanillin, and 8-O-4' diferulic acid were the first compounds identified from the deodeok. The content of 8-O-4' diferulic acid in the skin CWM was 56.1 ${mu}g/g$ AIR (alcohol insoluble residue). The ethanol-NaOH fractions from CWM had the highest oxygen radical absorbance capacity (ORAC) activities, followed by the AIR fractions and ethanol fractions.

NF-${\kappa}B$ Inhibitory Activities of Phenolic and Lignan Components from the Stems of Acanthopanax divaricatus var. albeofructus

  • Acanthopanax divaricatus var. albeofructus (ADA) is commonly ingested as a traditional medicine or as a component of a health drink in Korea. In this phytochemical study, nine phenolics (1 - 9) and three lignans (10 - 12) were isolated from the MeOH extract of the stems of ADA. Chemical structures were elucidated by comparing spectroscopic data with reported values. Nuclear factor kappa B ($NF-{\kappa}B$) inhibitory activity of the isolated compounds was evaluated using an $NF-{\kappa}B$ luciferase assay in HepG2 cells. Among them, compounds 1, 3 - 8, and 11 showed significant inhibitory effects on $TNF{\alpha}$-induced $NF-{\kappa}B$ transcriptional activity in a dosedependent manner, with $IC_{50}$ values ranging from 13.25 to $37.36{\mu}M$. Further studies on potential anti-inflammatory effects and the benefits of phenolic and lignan components from ADA are warranted.

Antioxidant Activity and Total Phenolic Compounds off P. eryngii spp. Extracts (버섯(P. eryngii spp.) 추출물의 항산화 활성과 총 페놀함량)

  • This objective of this study was to determined the activity of antioxidants of DPPH, SOD and total phenolic compounds in mushroom(P. eryngii spp.) extract. The antioxidant acitivity using DPPH showed that strain No. 2007$(31.78\%)$ and strain No.2014$(27.45\%)$ were the highest DPPH inhibition. Among the thirteen mushroom examined, strain No. 2014$(58.30\%)$ and strain No. 2010$(57.19\%)$ had the highest SOD activity. The highest total phenol content were in strain No.2013$(1465.03{\mu}g/g)$ and strain No.2005$(1401.09{\mu}g/g)$. Our study suggest that it may be possible to improve mushroom with high natural antioxidant.

  • Seven monoterpenes, three sesquiterpenes, three phenolics and one flavonoid were isolated from the MeOH extract of Amomum xanthioides. Their structures were determined by spectroscopic methods to be caryophyllene oxide (1), bornyl acetate (2), nerolidol (3), spathulenol (4), (-)-borneol (5), (+)-5-endohydroxycamphor (6), vanillic acid (7), protocatechuic acid methyl ester (8), betulabuside A (9), (1R,4S,6R)-6-hydroxyfenchan-2-one-6-O-$\beta$-D-glucopyranoside (10), (1S,4R,6S)-6-hydroxybornan-2-one-6-O-$\beta$-D-glucopyranoside (11), (1R,2S,4S,5R)-angelicoidenol 2-O-$\beta$-D-glucopyranoside (12), 1-O-vanilloyl-$\beta$-D-glucopyranoside (13), and quercetin-3-rhamnopyranoside (14). Compounds 6-14 were isolated for the first time from this plant source. Compounds 3 and 4 exhibited moderate cytotoxicity against four human cancer cell lines in vitro using a SRB bioassay.

  • We investigated the effect of inhibitory compounds derived lignocellulosic hydrolysates on cell growth, sugar consumption and ethanol productivity, and also we intended to identify the potential for ethanol production based on lignocellulosic hydrolysates. Cell growth and ethanol production in the presence of acetate were initiated after 12 hr. Furans showed a longer lag time and phenolics showed a significant effect on strain and ethanol production in comparison to other model compounds. In the case of lignocellulosic hydrolysates, the acetate strongly affected cell growth and ethanol production.

Evaluation of Chlorophyll Content, Antioxidant Activity and Phenolic Compounds in the Seedlings of Rice-type Tartary Buckwheat

  • Rice type tartary buckwheat is used as a substitute for rice in many Asian countries due to its easy dehulling character. The objective of the present study was to determine the chlorophyll, total polyphenol (TP), total flavonoid (TF), antioxidant activity and to quantify the bioactive compounds rutin, quercetin and chlorogenic acid in the seedlings of rice-type tartary buckwheat (RTTB). Young seedlings exhibited higher antioxidant (DPPH radical inhibition) activity in dose dependent manner. TP and TF content were highest ($3017.46{\pm}201.84{\mu}g$ TAE/100 mg dw and $1916.0{\pm}102.95{\mu}g$ QE/100 mg dw respectively) in 3 days after germination (DAG) seedlings compare to 6 and 9 DAG. The contents of rutin and quercetin increased with growing stage of seedlings. However, the chlorogenic acid decreased with increasing growth. Overall, RTTB seedlings can be regarded as a strong source of phenolics and have high possibility for food and nutraceutical application due to their efficient antioxidant properties, higher chlorophyll and phytochemical content.

  • Fourteen compounds were isolated from the n-BuOH fraction of the roots of Aralia cordata (syn. = A. continentalis). Through spectroscopic method, the chemical structures were elucidated as: caffeic acid (1), protocatechuic acid (2), thymidine (3), uridine (4), methyl-$\alpha$-D-fructofuranoside (5), a mixture (3 : 1) of $\beta$-D-fructopyranoside and $\beta$-D-fructofuranoside (6), 1-methyl 1,2,3,4-tetrahydro-$\beta$-carboline-3-carboxylic acid (7), methyl-$\beta$-D-fructofuranoside (8), sucrose (9), 5-caffeoylquinic acid (chlorogenic acid) (10), 3-caffeoylquinic acid (neochlorogenic acid) (11), 4-caffeoylquinic acid (cryptochlorogenic acid) (12), 3,5-di-O-caffeoylquinic acid (13), and 1-kestose [$\beta$-D-fructofuranosyl-($2{\rightarrow}1$)-$\beta$-D-fructofuranosyl-($2{\rightarrow}1$)-$\alpha$-D-glucopyranoside] (14). Among them, compounds 5, 7, 8, and 10 - 14 were isolated from this plant for the first time.
