Analysis of Sequence Type and Fluoroquinolone Resistance in Ciprofloxacin-Resistant Escherichia coli (Ciprofloxacin 내성 대장균에서 Sequence Type과 Fluoroquinolone 내성의 분석)

  • Fluoroquinolone (FQ) resistant gram-negative pathogens have emerged worldwide, and the recent increase in FQ resistant Escherichia coli is of great concern in Korea. This study investigated FQ resistance determinants and the epidemiological relationship of 56 ciprofloxacin-resistant E. coli isolated from a tertiary hospital in Daejeon, South Korea from June to December 2018. Molecular epidemiology was investigated by multilocus sequence typing (MLST). Polymerase chain reaction (PCR) and sequence analysis were performed to identify chromosomal mutations in the quinolone resistance determining regions (QRDR) of gyrA, gyrB, parC, and parE and to describe the occurrence of the following plasmid-mediated quinolone resistance (PMQR) genes: aac(6)-Ib-cr, qepA, qnrA, qnrB, qnrC, qnrD, and qnrS. MLST analysis showed 12 sequence types (STs) and the most prevalent ST was ST131 (31/56, 55.4%), followed by ST1193 (13/56, 23.2%), and ST405 (3/56, 5.4%). In 56 ciprofloxacin-resistant E. coli isolates, Ser83→Leu and Asp87→Asn in gyrA and Ser80→Ile and Glu84→Val in parC (51.8%, 29/56) were the most frequent amino acid substitutions and aac(6)-Ib-cr (33.9%, 19/56) was the most common PMQR gene. These results of FQ resistance determinants were more frequently observed in ST131 compared with other clones. Continuous monitoring of the epidemiological characteristics of ciprofloxacin-resistant E. coli isolates and further investigation of FQ resistance determinants are necessary.

  • The standard emission rate(ERs) of isoprene was quantitatively measured in situ from Quercus mongolica Fischer that dominates more than about 85% of domestic oak trees. The ERs values in spring and summer were similar to 64.4 and 58.1 (${\mu}gC/gdw/hr$), respectively. The ERs in autumn, 7.06(${\mu}gC/gdw/hr$), was about $8{\sim}9$ times lower than those in spring and summer. The coefficient of determination ($r^2$) between ERs and $C_L\;C_T$ ranged from 0.593 to 0.836. The correlation coefficients between the ERs and PAR. the ER and temperature suggested that ERs have strong correlation with PAR(photosynthetically active radiation) and temperature. In addition, the high values of PAR, temperature, and ERs were found in the time zone of $15:00{\sim}17:00$(spring), $15:00{\sim}16:00$(summer), and $14:00{\sim}15:00$(autumn).

Prevalence and Molecular Characterization of Quinolone Antibiotic Resistance in Escherichia coli Isolates from Raw Bulk Milk in Gyeonggi-do (원유시료에서 분리한 대장균의 퀴놀론 항생제 내성 기전)

  • The aim of this study was to investigate the prevalence of quinolone resistant E. coli from raw bulk milk and to characterize the resistance determinants. In this study, the gyrA, gyrB, parC, and parE quinolone resistance determining regions (QRDR) were sequenced from quinolone resistant E. coli isolates. Also, the presence of plasmid-mediated quinolone resistance (PMQR) and the expression of efflux pump genes based on quantitative real-time PCR (qRT-PCR) were investigated. Of the 487 coliform bacteria, 9 strains showed nalidixic acid resistance, and 6 of the 9 nalidixic acid resistant isolates were also ciprofloxacin resistant. These 9 strains had a single mutation at codon 83 (S83L) in gyrA, 2 of them had double mutations at codon 83 and 87 (S83L and D87N) in gyrA and 3 of the 9 isolates had single mutations at codon 80 (S80I) in parC. None of the 9 isolates harbored PMQR determinants. Compared with wild-type E. coli ATCC 25922, an over-expression of the acrB gene (2.15-5.74 fold), encoding the pump component of the AcrAB-TolC efflux pump was observed in 4 of 6 ciprofloxacin resistant isolates. This study identified the quinolone resistance mechanism of E. coli isolated from raw milk samples in Gyeonggi-do.

Analgesia after Epidural Dexamethasone is Further Enhanced by IV Dipyrone, but Not IV Parecoxibe Following Minor Orthopedic Surgery

  • Background: Epidural administration of dexamethasone has been suggested for pain control after minor orthopedic surgery. This study was conducted to assess its efficacy after such surgery, combined or not to IV dipyrone, IV parecoxibe or their combination. Methods: 91 patients were randomly assigned to seven groups. Patients were submitted to spinal bupivacaine anesthesia combined to epidural administration of either 10 ml saline or 10 mg dexamethasone diluted to 10-ml volume. Patients also received 10 ml IV saline or 1 gr dipyrone and/or 40 mg parecoxibe diluted to 10 ml with saline. Control group (CG) received epidural and IV saline. Dexamethasone group (DexG) received epidural dexamethasone and IV saline. Dipyrone group (DipG) received epidural saline and IV dipyrone. Dex-Dip G received epidural dexamethasone and IV dipyrone. Parecoxibe group (ParG) received epidural saline and IV parecoxibe. Dex-ParG received epidural dexamethasone and IV parecoxibe. Finally, Dex-Dip-ParG received epidural dexamethasone and IV dipyrone plus IV parecoxibe. Results: The CG expressed 4h of analgesia and sooner requested pain killer. DexG was similar to DipG or ParG or Dex-ParG (7-hours), and they requested less ketoprofen compared to the CG (P < 0.05). However, the Dex-DipG and the Dex-Dip-ParG resulted in longer time to demand pain killer (17-hours) and less ketoprofen consumption in 24-hours (P < 0.002). Adverse effects were similar among groups. Conclusions: The analgesia secondary to epidural dexamethasone was enhanced by IV dipyrone, while no effects were observed by the addition of IV parecoxibe.

  • La structure syllabique est un facteur important dan la prononciation d'une langue. Dans cette these, on a essaye de montrer que les caracteristiques de la syllabe varie d'une langue a une autre, et que les regles qui regissent la syllabe dan la langue maternelle a le dessue et done fait appliquer ses regles a la langue etrangere. Premierement, l'accentuation du coreen depend de la structure syllabique (qu'elle soit consonne-voyelle(CV) ou consonne-voyelle-consonne(CVC) et de la longueur de la voyelle. Ce qui fait que bien que le francais soit oxyton, et que la longueur et la qualite de la voyelle dependent de la structure syllabique, le francais, le francais parle par les coreens suit les regles du coreen. Autre caracteristique est que le coreen n'admet pas de suite de consonnes avant et apres la voyelle centrale comme dans "premier"[$pr{\partial}mje$] "autre"[o:tR]. D'ou l'insertion de voyelle superflue comme [${\varpi},{\;}{\wedge},{\;}{\partial}$]. Troisiemement, il existe une difference dans la coupe de la chaine parlee: en coreen la coupe ecrite(le blanc) egale a peu pres a la coupe orale(la pause) alors qu'en francais, c'est en groupe de mots que se fait la coupe. A l'interieur du groupe, les mots sont lies entre eux, soity par liaison, soit par enchainement. On peut remarquer donc une nette influence du coreen, ou la regle est de prononcer correctement par unites ecrites (equivalent des mots en francais) : la chaine parlee devient saccadee, avec un accent sur tous les mots, et des coups de glotte entre les mots, et l'une des voyelles [${\varpi},{\;}{\wedge},{\;}{\partial}$] inseree entre un mot se terminant par une consonne suivi d'un mot qui commence par une consonne.

Molecular Characterization of Quinolone Antibiotic Resistance in Escherichia coli Isolated from Retail Meat in Seoul (서울시내 시판 식육에서 분리한 대장균의 퀴놀론계 항생제 내성 기전 분석)

  • The aim of this study was to investigate the prevalence of quinolone resistant E. coli from retail meat and to characterize the resistant determinants. Determination of minimum inhibitory concentration, the sequence analysis of gyrA, gyrB, parC, and parE quinolone resistance determining regions (QRDR), the presences of plasmid mediated quinolone resistance (PMQR) and the expression of efflux pump genes were investigated. Of the total 277 retail meat samples, 67 coli form bacteria were isolated. 15 of 67 isolates showed nalidixic acid resistance and 7 of 15 nalidixic acid resistant isolates were also resistant to ciprofloxacin, moxifloxacin and levofloxacin. 11 of 15 nalidixic acid resistant strains were isolated from chicken, 2 of 15 were isolated from beef and 2 of 15 were isolated from pork samples. 11 of 15 nalidixic acid resistant strains have single mutation at codon 87 (D87N or D87G) in gyrA, 2 of 11 gyrA mutants have double mutations at codon 86 and 87 (L86A and L87I) in parC with mutations at codon 434+445+465 or 429 in gyrB. 2 of 15 resistant isolates harbored qnrS, a PMQR determinant. Over expression of the acrB gene, efflux pump gene (3.93~16.53 fold), was observed in 10 of 15 resistant isolates.

Molecular Characterization of Fluoroquinolone Resistant Escherichia coli Isolates from Chickens in Korea (닭에서 동정된 플르오르퀴놀론 내성 대장균 균주의 분자생물학적 성상에 관한 연구)

  • An aim of current study was to investigate the prevalence and the mechanism of quinolone-resistance in E. coli isolates obtained from chicken cecum in Korea. In addition, multilocus sequence typing (MLST) was also performed for the molecular characterization of E. coli isolates. In an antimicrobial susceptibility test by the disk diffusion method, the 63.5% (54/85) of E. coli isolates showed the resistance to quinolone group of antimicrobial agents. All of the 54 E. coli isolates showing resistant to quinolone group had sense mutations in gyrA gene and point mutations at the $57^{th}$, $80^{th}$, or $84^{th}$ residues in parC gene were detected in 90.7% of the isolates. Interestingly, E. coli ST was closely related to amino acid substitutions in parE gene. Our results indicated that the long-term use of antimicrobial agents in food-producing animals was strongly associated with a prevalence of antimicrobial resistance in commensal Enterobacteriaceae, suggesting the need for continuous surveillance and monitoring of antimicrobial resistant determinants in bacterial isolates from food animals.

Mutation in gyrA gene of nalidixic acid-resistant Salmonella isolates isolated from poultry slaughterhouse (닭 도축장에서 분리한 nalidixic acid 내성 Salmonella 균의 gyrA 유전자 돌연변이)

  • The objective of this study was to identify mutations in the quinolone resistance determining region (QRDR) of the gyrA, gyrB, parC and parE genes, and the presence of plasmid-mediated quinolone resistance (PMQR) genes: qnrA, qnrB, qnrS, aac(6')-lb-cr and qepA in 40 nalidixic acid- resistant ($NA^R$) Salmonella isolates isolated from poultry slaughterhouse. The MIC of NA and ciprofloxacin for 40 $NA^R$ Salmonella isolates was $128{\sim}512{\mu}g/mL$ and < $0.125{\sim}0.25{\mu}g/mL$, respectively. The Salmonella isolates were resistant to NA (100%), gentamicin (5.0%) and ampicillin (2.5%). All $NA^R$ Salmonella isolates represented point mutation in codons Aspartic acid(Asp)-87 (90%) and Serine(Ser)-83 (10%) of QRDR of gyrA gene: $Asp87{\rightarrow}glycine$, $Ser83{\rightarrow}tyrosine$. No mutations were observed in QRDR of the gyrB, parC and parE gene. Moreover PMQR genes was not found in any of the tested isolates. Our findings showed that DNA gyrase is the primary target of quinolone resistance and a single mutation in codon Asp87 and Ser83 of the gyrA gene can confer resistance to NA and reduced susceptibility ciprofloxacin in Salmonella isolates.

PDZ Domain-containing Proteins at Autotypic Junctions in Myelinating Schwann Cells (수초화 슈반세포 autotypic 세포연접의 PDZ 도메인 보유 단백질)

  • A type of cell junction that is formed between different parts within the same cell is called autotypic cell junction. Autotypic junction proteins form tight junctions found between membrane lamellae of a cell, especially in myelinating glial cells. Some of them have postsynaptic density-95/disks large/zonula occludens-1 (PDZ) domains, which interact with the carboxyl (C)-terminal PDZ-binding motif of other proteins. PDZ domains are protein-protein interaction modules that play a role in protein complex assembly. The PDZ domain, which is widespread in bacteria, plants, yeast, metazoans, and Drosophila, allows the assembly of large multi-protein complexes. The multi-protein complexes act in intracellular signal transduction, protein targeting, and membrane polarization. The identified PDZ domain-containing proteins located at autotypic junctions include zonula occludens-1 (ZO-1), ZO-2, pals-1-associated tight junction protein (PATJ), multi-PDZ domain proteins (MUPPs), membrane-associated guanylate kinase inverted 2 (MAGI2), and protease-activated receptor (PAR)-3. PAR-3 interacts with atypical protein kinase C and PAR-6, forming a ternary complex, which plays an important role in the regulation of cell polarity. MAGI2 interacts with ${\alpha}$-amino-3-hydroxyl-5-methyl-4-isoxazole propionate (AMPA) receptor at excitatory synapses. PATJ is detected in paranodal loops associated with claudin-1. On the other hand, MUPP1 is found in mesaxons and Schmidt-Lanterman incisures with claudin-5. ZO-1, ZO-2, and PAR-3 are found at all three sites. Different distributions of PDZ domain-containing proteins affect the development of autotypic junctions. In this review, we will describe PDZ domain-containing proteins at autotypic tight junctions in myelinating Schwann cells and their roles.

Cloning, Expression, and Characterization of a Family B-Type DNA Polymerase from the Hyperthermophilic Crenarchaeon Pyrobaculum arsenaticum and Its Application to PCR

  • The gene encoding Pyrobaculum arsenaticum DNA polymerase (Par DNA polymerase) was cloned and sequenced. The gene consists of 2,361 bp coding for a protein with 786 amino acid residues. The deduced amino acid sequence of Par DNA polymerase showed a high similarity to archaeal family B-type DNA polymerases (Group I), and contained all of the motifs conserved in the family B-type DNA polymerases for $3'{\rightarrow}5'$ exonuclease and polymerase activities. The Par DNA polymerase gene was expressed under the control of the T7lac promoter on the expression vector pET-22b(+) in Escherichia coli BL21-CodonPlus(DE3)-RP. The expressed enzyme was purified by heat treatment, and Cibacron blue 3GA and $Hirap^{TM}$ Heparin HP column chromatographies. The optimum pH of the purified enzyme was 7.5. The enzyme activity was activated by divalent cations, and was inhibited by EDTA and monovalent cations. The half-life of the enzyme at $95^{\circ}C$ was 6 h. Par DNA polymerase possessed associated $3'{\rightarrow}5'$ proofreading exonuclease activity, which is consistent with its deduced amino acid sequence. PCR experiment with Par DNA polymerase showed an amplified product, indicating that this enzyme might be useful in DNA amplification and PCR-based applications.
