공액리놀레산 함유 디글리세라이드 식용유지 조성물의 rat에 대한 단회 경구투여독성 및 항비만 효과 (Acute Oral Toxicity and Anti-obesity Effect of Diglyceride Preparation Containing Conjugated Linoleic Acid in Rat)

  • 단회 경구 투여 독성 평가를 위해 암 수 rat을 이용하여 시험물질 2,000 mg/kg과 5,000 mg/kg을 투여한 후 14일 동안 관찰한 결과, 투여에 기인한 사망례 및 체중변화, 특이한 일반증상은 관찰되지 않았다. 암컷 대조군 및 시험물질 2,000 mg/kg 투여군에서 투여 1일에 연변외 점액변이 관찰되었으나, 이는 투여에 기인한 일시적인 변화로 사료된다. 투여 2일부터는 암컷 대조군과 시험물질 투여군에서 특이한 일반증상이 관찰되지 않았다. 부검 시 암컷 대조군 및 암 수 시험물질 투여군에서 투여에 기인한 것으로 판단되는 육안소견은 관찰되지 않았다. 수컷 대조군 1례의 부검결과, 부고환, 정낭 및 양측 고환에서 소형 소견 관찰되어, 조직병리학적 검사를 실시한 결과, 부고환, 정낭 및 고환의 위축 소견이 관찰되었으나, 이는 자연 발생적인 소견으로 사료된다. 이상으로 공액리놀렌산이 함유된 디글리세라이드 식용유지 조성물을 rat에 단회 경구투여한 결과, $LD_50$값이 rat kg당 5,000 mg 이상으로 판단되며, 단회 경구 투여에 대한 독성을 나타내지 않는 것으로 판단된다. 항비만 효과를 조사하기 위해 obese Zucker rat에 saline, soybean oil, DG, DG+CLA를 8주간 투여한 후 효과를 비교하였다. 체중변화의 경우 DG+CLA 투여군은 대조군에 비하여 투여 후 3주째부터 통계학적으로 유의성 있게 체중 감소를 나타냈다. 1일 식이효율측정의 결과, soybean oil, DG 및 DG+CLA 투여군이 대조군에 비하여 통계학적으로 유의성 있는 1일 식이 효율의 증가를 나타냈다. 혈중지질분석의 결과, 총콜레스테롤 수치에서 soybean oil, DG 및 DG+CLA 투여군 모두 대조군에 비하여 통계학적으로 유의성 있게 총콜레스테롤 함량이 감소되었으며, 중성지방 수치는 soybean oil 및 DG 투여군에서만 대조군에 비하여 통계학적으로 유의성 있게 감소되었다. 지방조직무게 측정 결과, DG+CLA 투여군은 신장주위지방무게를 제외한 부고 환주위지방, 갈색지방 및 내장지방무게에서 대조군과 비교하여 통계학적으로 유의성 있게 지방조직무게가 감소되었다. 이상의 결과로 obese Zucker rat에 대한 5 mL/kg의 DG+CLA 경구투여는 CLA의 주요 이성질체인 c9, t11-CLA와 t10, c12-CLA가 hormone sensitive lipase와 carnitine palmitoyl transferase의 활성을 증가시켜 지방 분해를 촉진시켜 체내 지방의 축적을 막으며, 중성지방 콜레스테롤과 같은 지질 대사에 영향을 주어 항비만 효과를 나타내는 것으로 사료되어진다.

야관청혈탕이 고지방식이로 유도된 흰쥐의 비만 억제에 미치는 영향 (Effect of Yagwan-cheunghyeoltang on Obesity in Rats Induced by High Fat Diet)

  • Background : Obesity, the syndrome caused by a high fat diet, is a disease. At the same time, obesity causes diabetes mellitus, hyperlipidemia, and cardiovascular disease. Recently, its prevalence rate is increasing. Yagwan-cheunghyeoltang (YCT) used in this experiment is the prescription of Yagwanmoon added to Cheunghyeol-tang which is reported to be very effective in weight loss controlling and serum cholesterol. It is also reported that Yagwanmoon has significant antioxidant effects and YCT has a significant effect on blood glucose control. Objectives : This study was conducted to experimentally evaluate the effects of YCT on obesity in rats induced by high fat diet. Methods : The experiment was conducted with 4-week-old male rat s divided into 5 groups. They were a normal diet group, a high fat diet group, a positive drug control group, a 1% YCT group, and a YCT 3% group, and were tested for eight weeks. After four weeks of inducing obesity by a high fat diet, rats were allowed to lose weight by following the normal diet group, approximately 30% compared with 10 rats in each group were determined as still obese. Changes in body weight and organ weight and serum cholesterol, triglyceride, glucose-density, low density lipoprotein cholesterol, antioxidant activity were checked. Results : In the experimental groups, we observed weight loss and visceral fat reduction, improvement of liver function, reduction of serum glucose, activation of HMG-CoA reductase inhibitor, reduction of concentrations of leptin and it showed a significant effect on antioxidants and lipid peroxidation. Conclusions : YCT has significant effects on the regulation of hyperlipidemia and lipid peroxidation associated with obesity and has significant effects on, antioxidants and lipid peroxidation, too. Additional clinical studies are needed.

흰쥐에서 개량식 고추장과 발효 밀쌀의 항비만 효과 (Anti-Obesity Effect of Commercial Kochujang and Fermented Wheat Grain Products in Sprague-Dawley Rats)

  • The antiobesity effect of commercial kochujang and fermented wheat grains in Sprague-Dawley (SD) rats was studied. The experiment was consisted of 6 groups. Normal, high fat diet (HFD), HFD+raw wheat grains, HFD+first fermented wheat grains (FFWG, with Aspergillus oryzae) HFD+final fermented wheat grains (FiFWG, fermented more for 30~40 days), and HFD+ commercial kochujang. The results showed that final body weight, weight gain, food efficiency ratio, and adipose tissue weight were markedly decreased by the commercial kochujang and the fermented wheat grains, whereas non-fermented raw wheat grains had no such effect. Lipid contents such as total lipid, total triglyceride and total cholesterol decreased in the serum and organs of liver and adipose tissues by the commercial kochujang and the fermented wheat grains as well. These results also indicated that fermented wheat grains exhibited more suppressive effects on high fat induced-obesity than raw wheat grains. Increased fermentation time and adding the red pepper powder resulted in increased the anti-obesity effect. Especially, commercial kochujang showed higher antiobestic effects than fermented wheat grains. These in vivo findings suggested that well-fermented end products of the wheat grains and red pepper powder in kochujang could be useful in the prevention of obesity.

대청룡탕이 비만 유도 흰쥐의 체중 감량과 혈중지질농도 개선에 미치는 영향 (The Effects of Daecheongryoung-tang on Anti-weight Gain and Anti-hyperlipidemic in Obese Sprague Dawley Rat Induced by High Fat Diet)

  • Objectives: The aim of this study is to investigate the effects of Daecheongryoung-tang (DCR) therapy on body weight, serum total cholesterol, high density lipoprotein (HDL) cholesterol, low density lipoprotein (LDL) cholesterol, triglyceride, free fatty acid, total lipid, phospholipid level and complete blood cell count of obese rats. Methods: 34 rats are divided into 4 groups, the rats in the normal group are 7 and the rats in the other group are 9 per group; Normal group (general fat diet and no medication), Control group (high-fat diet and no medication), DCR_L group (high-fat diet and DCR 250 mg medication) and DCR_H group (high-fat diet and DCR 500 mg medication). DCR is administrated for 6 weeks. Results: There is significant statistical difference between Control group and DCR-H group for the body weight, the total cholesterol, LDL cholesterol, triglyceride, free fatty acid level. Also, there is significant statistical difference among Control group, DCR_L group and DCR_H group for body weight, triglyceride, free fatty acid and phospholipid level. Conclusions: These results suggest that medication of DCR_L and DCR_H is effective for the treatment of obesity.

비만 유도 흰쥐에 대한 양파의 항비만 효과 (Anti-obesity effects of onion juice in high fat diet-induced obese rats)

  • This study investigated the effects of onion juice on the serum lipid components and blood pressure in obese rats fed high fat diets. Thirty-nine of Sprague-Dawley rats were divided into six groups, and were treated for 8 weeks: (1) normal diet (ND); (2) high fat diet (HFD); (3) HFD for first 4 weeks and high fat diet with 40% onion juice for the last 4weeks (H-H+O); (4) HFD with 40% onion juice for 8 weeks (H+O); (5) HFD for first 4 weeks and ND for the last 4 weeks (H-N); (6) HFD for first 4 weeks and ND with 40% onion juice for the last 4 weeks (H-N+O). The rates of increasing body weight were reduced in H+O and H-H+O groups compared with HFD group. The levels of triglyceride, low density lipoprotein-cholesterol and total cholesterol in blood serum were significantly decreased in the H+O and H-H+O groups compared with the HFD group. Administration of onion reduced the size of adipocyte, steatosis, and serum hyperlipidemia in obese rats fed HFD. Moreover, the antihypertensive effects of onion were observed in obesity rat fed HFD. Overall results suggest that onion reduces the serum lipid components and improves hypertension in obese rat fed HFD.

Isaria sinclairii Extract Reduces Body Weight and Ameliorates Metabolic Abnormalities

  • Obesity is a major risk factor for cardiovascular disease. In our case study using animal models for disease states such as obesity or hypertension, we found that, Isaria sinclairii remarkably reduced body weight and ameliorated metabolic abnormalities in Zucker and SHR rats. Genetically obese (fa/fa) Zucker rats were one animal model chosen for this study. Four groups of rats received a standard diet and were treated orally with the following test samples daily for 8 weeks: saline (negative control), ethanol extracts of I. sinclairii, hot water extract of I. sinclairii, or $Xenical^{(R)}$ (30 mg/kg, positive control). Mild reductions (6.3%) in body weight gain were observed in the groups treated with the hot water extract of I. sinclairii compared to the control after 8 weeks. Interestingly, organ weight was greatly reduced by this Dongchunghacho (I. sinclairii), in parallel with the mild reductions in body weight gain and reductions in abdominal fat (adipose tissue). Also observed was a 4.1 % decrease in the ratio of heart weight/body weight compared to the control group. As a hypertensive animal model, SHR (spontaneously hypertensive rat) and WKY (Wistar Kyoto) rats were also administered these extracts for one month. Treatment with the hot water extract of I. sinclairii caused greater reductions in body weight gain for the SHR group (10.9%) compared to the WKY group's (5.2%). Based on these results, I. sinclairii extracts contain selective action for anti-obesity activity, naturally occurring candidate for regulation of body weight increase, as demonstrated in the present study.

산마늘 기능성 활성물질 탐색 및 자원화 (Effect of Allium victorialis var.platyphyllum Leaves on Triton WR-1339-induced and Poloxamer-407-induced hyperlipidemic Rats and on Diet-Induced Obesity Rats)

  • The herb of Allium victorialis var. platyphyllum (Liliaceae) has been used as an edible wild herb and to treat heart failure and gastritis. We have already reported antihyperlipidemic anti-tumor effects of this plant. To enlarge the commercial availability of this food, it was investigated whether the extracts of A. victorialis var.platyphyllum reduce hyperlipidemia and obesity or not. The plants tested in this experiment were collected from two eco-types of IS. Ullung and Mt. Odae cultivated at Pyongchang. Extracts were prepared by extracting the fresh leaves and those dried at $36^{\circ}C$ and $90^{\circ}C$, respectively. Pretreatment with the ethanolic extracts for two weeks (p.o.) reduced serum triglyceride-, total cholesterol- and LDL-cholesterol contents in rats induced by Triton WR-1339, respectively. Furthermore, oral administration of the extracts also inhibited the hyperlipidemia induced with oral diet of 30% corn oil. In the other attempt to find to alleviate the obesity, the model rats with obesity were induced by the high fat-diet for six weeks. Post-treatment with the extracts for two weeks significantly reduced the hyperlipidemia. Retroperitoneal-, epidymal- and total abdominal fat pad weights were considerably reduced at 100 mg/kg oral administration of the extracts. Increased feces lipid contents were also found in the rat treated with the extracts. The extract of Mt. Odae eco-type showed more potent activity than that of Is. Ullung one. These results suggest that use of the fresh leaves may lead to the higher activity in treatment of hyperlipidemia and obesity than of the dried one.

지방식이로 유도된 비만동물모델에서의 약침의 효과 (Effect of Herbal Acupuncture with Sang-hwang(Phellinus linteus) on High Fat Diet-induced Obesity in Rats)

  • Acupuncture has fairly good weight-reducing effect in treating simple obesity due to the neuroendocrine regulation. In this study, the antiobesity effects of herbal acupuncture(HA) with Sang-hwang(phellinus linteus) at Fuai(SP16) were investigated in the rat fed on high-fat(HF) diet. Sang-hwang mushroom has been proven to have anti-carcinogenic effects and Sang-hwang extracts are highly effective in treatment and preventive treatment of AIDS, diabetes and high blood-pressure. To determine whether the Sang-hwang herbal acupuncture may have the anti-obesity effect, male Sprague-Dawley(4-wk-old) rats were fed a HF diet for 5 wk, which produced significant weight gain compared to rats were fed a normal diet, and then herbal acupuncture were treated for 3 wk in HF diet group. The body weight, food consumption, food effeciency ratio(FER), body fat mass, plasma nitric oxide(NO) were investigated in rats fed on normal diet, HF diet, and HF diet with HA(HF-diet-HA) groups. NO has been proposed to be involved in the regulation of food intake. In addition, the expression of appetite peptides such as orexigenic peptide neuropeptide Y(NPY) and the anorectic peptide cholecystokinin(CCK) were observed in the hypothalamus. HF-HA group reduced body weight gain, FER, body fat contents and NO concentration compared to HF diet group. The expression of NPY was reduced in arcuate nucleus(ARC), and CCK was increased in the paraventricular nucleus(PVN) after treatment of HA. In conclusion, Sang-hwang HA reduced adipocity, plasma NO and hypothalamic NPY, but increased CCK expression in the HF dietinduced obesity rat, therefore HA may have anti-obesity action through regulating body weight and appetite peptide of the central nervous system.

갈근 추출물이 난소 적출로 비만이 유발된 Rat의 체중 변화와 유전자 발현에 미치는 영향 (Effects of Pueraria lobata on Body Weight and Gene Expression in Obese Rats Muscle with Estrogen Deficiency)

  • Objectives: GG is the EtOH fraction of extract of Pueraria lobata. In this study, we aimed to elucidate a possibility that GG reduce obesity and obesity-derived complications such as cardiovascular and metabolic disease. Methods: The effects of GG on the estrogen-deficient obese rats and the level of gene expression in muscle of rats were investigated. Results: GG decreased body weight in obese rats with estrogen deficiency. GG increased leptin gene expression in obese rats with estrogen deficiency. GG decreased TNFa gene expression in obese rats with estrogen deficiency. And GG increased PPAR-gamma, PGC-1a, Prdx6, FDFT1, and ACC gene expression of those in obese rats. Conclusions: We conclude GG might reduce body weight and regulate gene expression of muscle in obese rats.

고지방식이에 의한 비만 동물모델에서 유산균 마분말의 항비만 효과 (Anti-Obesity Effects of Lactic Acid Bacteria-Fermented Dioscoreae Rhizoma Powder on High Fat Diet-Fed Animal Model)

  • The purpose of this study was to examine the antiobesity effects of lactic acid bacteria-fermented Dioscoreae Rhizoma powder (LDR) in Sprague-Dawley rats with high-fat diet (HFD)-induced obesity. Rats were divided into 5 groups: normal diet group, control (high fat diet-vehicle) group, 0.4 g/kg LDR (high fat diet + 0.4 g/kg lactic acid bacteria-fermented Dioscoreae Rhizoma powder) group, 1 g/kg LDR (high fat diet + lactic acid bacteria-fermented Dioscoreae Rhizoma powder) group, 2.5g/kg LDR (high fat diet + lactic acid bacteria-fermented Dioscoreae Rhizoma powder) group. Our results indicate that LDR administration has effects on decreasing of body weight, fat weight, blood parameters and adipocyte size in the obesity animals. In addition, verified that fat degeneration and ballooning degeneration were alleviated in the experimental group fed on a high fat diet combined with LDR groups. These results indicate that lactic acid bacteria-fermented Dioscoreae Rhizoma powder may reduce elevating body weight and lipid accumulation in rat fed a high fat diet, suggesting its usefulness as a functional food for reducing body fat and obesity.
