Physicochemical Properties and Antioxidative Activities of Omija(Schizandra chinensis Bailon) (오미자의 이화학적 특성 및 항산화 활성)

  • To develop physiological functionality of Omija extracted with water was evaluated on antioxidative activity. Omija, high acid material with pH 3.6, contain $57.5{\pm}1.03%$ of moisture, and $18.8{\pm}0.12%$ of crude fat. This material have $12.6{\pm}0.04%$ of carbohydrate and $11.1{\pm}0.07%$ of crude protein as well, but ash and crude protein contents were found less than 10%. 10 mineral contents were also found, too; K and Ca showed the highest level, then Al, Mg, Na and Mn were followed. In composition amino acid contents, glutamic acid took the largest portion, $131.7{\pm}1.3$ mg/100 g, aspartic acid $51.5{\pm}0.6%$, and other composition amino acid under 50%. In case of free sugar contents, 7 types were found. Most of them were glucose and fructose. Total phenolic compounds showed the highest level, $2,862.6{\pm}31.7$ mg/100 g. $197.8{\pm}14.6$ mg/100 g of flavonoid and $225.6{\pm}18.2$ mg/100 g were included. In terms of electron donating ability, radical scavenging ability activated as the amount of Omija extract increased. In particular, Omija extract in 1,000 ${\mu}g/m{\ell}$ demonstrated almost similar electron donating ability, $72.4{\pm}0.21%$, to BHT. It was also found that antioxidant activities of electron donating ability, SOD-like ability, hydroxyl radical scavenging ability and nitrite scavenging ability were highly promoted as Omija extract concentration increased. The nitrite scavenging ability was significant when the extract belonged to strong acid region and doping concentrations increased.

Effective Components and Nitrite Scavenging Ability of Root and Leaves a Angelica gigas Nakai (당귀와 승검초의 기능성 성분과 아질산염 소거능)

  • This research was conducted to examine the usability of Dangui (Angelica gigas Nakai, Root) and Seungumcho (Angelica gigas Nakai, Leaf) as functional food in aspects of their functional components and nitrite scavenging ability. Analysis of proximate composition showed that Dangui contains more moisture, crude lipid and crude protein than Seungumcho. On the other hand, Seungumcho contains more than twice mont of crude ash and crude fiber than Dangui. Dangui showed higher contents in phosphorus, iron, magnesium, and Seungumcho showed higher contents in copper, calcium, sodium, potassium compare to each other. Substances such as calcium, magnesium and iron that showed high contents in Dangui and Seungumcho are the most important inorganic substances. Total dietary fiber(TDF) of Dangui, sum of IDF and SDF, was 24.2%, and Seungumcho showed 28.18%. vitamin C contents of a 29.690.33 mg/100g appeared only in Seunggumcho. Total phenol contents of Dangui was 0.100${\pm}$002% and that of Seungumcho 0.0900${\pm}$008%. Nnitrite Scavenging ability of both water soluble and methanol soluble extracts were more than 90% at pH 1.2, and it decreased as pH level adjusted to pH 4.2, pH and 6.0.

  • This study was performed to investigate the changes in quality properties of extract from roasted Pleurotus eryngii by microwave-assisted extraction (MAE). MAE was carried out under different conditions including ethanol concentration, extraction time and microwave power. Total phenolics compound contents, electron donating ability (EDA) and nitrite-scavenging ability at pH 1.2 were high at 50% ethanol concentration. Superoxide dismutase (SOD)-like activity was high at 75% ethanol concentration. Soluble solid and total phenolics compound content decreased as extending extraction time up to 7 min. EDA decreased as increased extraction time. SOD-like activity and nitrite-scavenging ability were not significantly different. The highest soluble solid content, total phenolics compound content and EDA were obtained at 100 W extraction of microwave power. SOD-like activity was high at 25 W. But microwave power did not significantly affect the nitrite-scavenging ability.

Antibacterial and Antioxidant activities of Bio-fermented Sanguisorbae Radix Extract (지유 발효추출물의 항세균 및 항산화 활성)

  • Objectives : The objective of this research was to investigate the antibacterial and antioxidant activities of bio-fermented Sanguisorbae Radix extract.Methods : The Sanguisorbae Radix extract was fermented byStreptococcus thermophilus,and their products was tested for antibacterial activity against pathogenic microorganisms namely,Bacillus cereus,Bacillus subtilis,Staphylococcus aureus,Escherichia coli,Vibrio parahaemolyticusandSalmonella typhimuriumby paper disc diffusion method and the antioxidant activities of extract was evaluated by five different assays as electron donating ability(EDA), superoxide dismutase(SOD)-like activity, polyphenol, flavonoid contents and nitrite scavenging ability.Results : The bio-fermented Sanguisorbae Radix extract was safe from heat. Antibacterial activity of fermented Sanguisorbae Radix extract appeared relatively highly againstBacillus cereusandStaphylococcus aureusand didn't show any difference. EDA in comparison to Vitamin C showed over 90% activity at about the same time of Sanguisorbae Radix extract expressed highly. SOD activity showed 15% in fermentation before and after. The nitrite scavenging ability of Sanguisorbae Radix extract before and after fermentation showed higher numerical value over 70% in pH 2.5 than that of butylated hydroxytoluene(BHT). But SOD activity, EDA and nitrite scavenging ability were not different between the Sanguisorbae Radix extract before and after fermentation. Total polyphenol content expressed over about 20 mg/g, and that of the Sanguisorbae Radix extracts was increased than that of the fermented Sanguisorbae Radix extracts.Conclusions : The results suggest the usefulness of developing functional materials using antioxidant active Sanguisorbae Radix extract was fermented bySalmonella typhimuriumwith high polyphenol contents and nitrite scavenging ability.

Functional Activities of Microwave-Assisted Extracts from Lyophyllum ulmarium (마이크로웨이브 추출공정에 의한 만가닥버섯의 기능적 특성)

  • Functional activities of Lyophyllum ulmarium microwave-assisted extracts under different conditions including electron donating ability, tyrosinase inhibition activity and nitrite scavenging effect were examined. Total polyphenol content increased as increasing microwave power up to 90 W in the water extracts. Electron donating ability increased with microwave power up to 90 W in 50% ethanol extract and 99% ethanol extract. Tyrosinase inhibition activity and nitrite scavenging effect in the extract increased as microwave power increased during extraction. Total polyphenol content increased as extending extraction time up to 5 min in the water extract. But the highest electron donating ability and tyrosinase inhibition activity was obtained after 10 min extraction. Significantly higher total polyphenol content and electron donating ability were found in the water extract whereas greater tyrosinase inhibition activity and nitrite scavenging effect were observed in 99% ethanol extract. The maxium nitrite scavenging effect was found at pH 1.2 and decreased as pH increased.

Antioxidative Activity and Nitrite Scavenging Ability of Ethanol Extract from Phyllostachys bambusoides (대나무 에탄올추출물의 항산화 효과 및 아질산염 소거작용)

  • Efficacy of antioxidative activity and nitrite scavenging ability of ethanol extract of Phyllostachys bambusoides S. et Z. (P. bambusoides) was investigated. Electron-donating ability of ethanol extract at $RC_{50}$ was $116.75\;{\mu}g/mL$. After addition of 0.92 mg/mL ethanol extract, autooxidation of pyrogallol decreased to 44% by superoxide dismutase-like activity. In antioxidative activity of ethanol extract against linoleic acid during incubation times of 4 and 6 day at $40^{\circ}C$, TBA values decreased by 74.76 and 58.48% with addition of 50 mg/mL, respectively. Nitrite scavenging ability showed the most remarkable effect at pH 1.2, decreasing to 43% by addition of 0.2 mg/mL. These results suggest that ethanol extract of P. bambusoides can be used in bioactive and functional material.

Nitrite Scavenging Ability and SOD-like Activity of a Sterol Glucoside form Chrysanthemum coronarium L. var. spatiosum (쑥갓 스테롤배당체의 아질산염소거작용 및 SOD 유사활성)

  • From the total methanolic extract of Chrysanthemum coronarium L. var. spatiosum (Compositae), nitrite scavenging ability and superoxide dismutase (SOD)-like activity were analyzed as antioxidative characteristics. After successive partitioning with chloroform, n-butanol, and water, the chloroform fraction showed the most significant nitrite scavenging ability with an $IC_{50}$ value of 39 ppm compared with the values of vitamin C and chlorogenic acid, 15 ppm and 36 ppm, respectively. The active fraction was subjected to silica gel and Sephadex LH-20 column chromatography, and the compound was isolated and identified as ${\beta}-sitosterol-O-{\beta}-D-glucoside$ using $^{1}H-NMR$ and $^{13}C-NMR$ spectral data. The glucoside was further hydrolyzed and confirmed as a glycosylated ${\beta}-sitosterol$. The compound and its aglycone, ${\beta}-sitosterol$, showed different nitrite scavenging and SOD-like activity. The $IC_{50}$ value of nitrite scavenging ability of the compound was 335 ppm at pH 1.5, while that of its aglycone was 41 ppm. As for the SOD-like activity, the $EC_{50}$ values of the sterol and the glucoside were 1,291 ppm and >2,000 ppm, respectively, compared with those of vitamin C and chlorogenic acid, 38 ppm and 449 ppm, respectively.

Electron Donating Ability and Nitrite Scavenging Activity of Materials in a Traditional One-dish Meal (Bibimbab) (한국 전통 한 그릇 음식(비빔밥) 및 그 재료들의 항산화성과 아질산염 소거능)

  • The electron donating ability and nitrite scavenging activity were measured in ethanol extracts from the raw, simple cooked and seasoned materials of Bibimbab (mixed rice), a Korean traditional one-dish meal. The ethanol extracts of raw fragrant edible wild aster exhibited the highest electron donating ability $(80.28\%)$, as measured by the DPPH method. When Bibimbab was compared with Kimbab and hamburgers, which are popular food, it showed the highest antioxidative activity by the DPPH method. The nitrite scavenging ability was measured in an environment similar to in human digestive organs, i.e. pH $1.2\~6.0$. Generally, the antioxidative activity decreased in the sequence pH 1.2, 3.0 and then 6.0. The simple cooked materials, with the exception of fragrant edible wild aster, increased the antioxidative potential. The seasoned materials also had a tendency to increase the antioxidative potential. In conclusion, the ethanol extract of Bibimbab showed the highest nitrite scavenging activity. Therefore, the nutritional superiority of Bibimbab was confirmed through its electron donating ability, as measured by the DPPH method, and nitrite scavenging effect.

The Electron Donating Ability, Nitrite Scavenging Ability and NDMA Formation Effect of Solvent Extracts from Yuza (Citrus junos SIEB ex TANAKA) (유자 용매추출물의 전자 공여능, 아질산염 소거능 및 NDMA 생성 억제능)

  • Methanol, ether, ethylacetate and water extracts from flesh and peel of yuna (Citrus junos) were investigated to analyze their ability of electron donating, nitrite scavenging and nitrosodimethylamine (NDMA) formation in model system. The electron donating ability of ynza flesh extract, when it added at 10 mg/ml or over, were more an $50\%$, except ether extract. The high electron donating ability ($99.2{\pm}0.37\%$) was observed in the methanol extract and its effect was similar to BHA and ascorbic acid, when 40 mg/ml of methanol extract was added in reaction solution. Nitrite scavenging ability of all extracts ken yuza was increased in proportion to sample concentration and more than $40\%$ when 1 mg/ml sample was added, under pH 1.2. Also nitrite scavenging ability was higher in peel extracts than flesh extracts of yuza. Inhibition ratio of NDMA formation from flesh methanol extract of yuza was $31.7{\pm}1.25\%$, when 40 mg/ml added under pH 1.2. In all samples, inhibition effects were lower than $20\%$, at pH 4.2 and 6.0.

Different of Total Phenolics and Flavonoids, Radical Scavenging Activities and Nitrite Scavenging Effects of Momordica charantia L. According to Cultivars (여주 품종별 폴리페놀, 플라보노이드 함량과 라디칼 소거활성 및 아질산염 소거능)

  • This study was carried out in order to determine the biological activities such as antioxidant activities in nine cultivars of Momordica charantia L. (Korea, China, Japan and Philippine native cultivars). Their antioxidant activities were measured using DPPH free radical scavenging, ABTS cation radical scavenging, and Nitrite scavenging ability. The highest total polyphenol content (16.82 ${\mu}g/m{\ell}$) measured in the native Korea cultivar, and this value was 4.0 ${\mu}g/m{\ell}$ higher than that of the 'Verde Beuhas' cultivar (12.82 ${\mu}g/m{\ell}$). The 'Peacock' cultivar had the highest total flavonoid amount which was 4.38 ${\mu}g/m{\ell}$. The free radical scavenging activity using DPPH method was the highest in the native China cultivar ($RC_{50}$ = 102.6 ${\mu}g/m{\ell}$). ABTS cation radical scavenging activity according to cultivars was significantly higher in 'Peacock'. Nitrite scavenging ability showed the most remarkable effect at the pH 1.2, exhibited to 81.5${\sim}$86.9% by addition of ethanol extract 1${\mu}g/m{\ell}$ from Momordica charantia L. These results suggest that Momordica charantia L. had the potent biological activities, and that their activities exhibited differently depending on cultivars.
