The Major Postharvest Disease of Onion and Its Control with Thymol Fumigation During Low-Temperature Storage

  • Onion (Allium cepa L.) is one of the major vegetable crops in Korea that are damaged and lost by pathogenic fungal infection during storage due to a lack of proper storage conditions. The aim of this study was to determine an appropriate control measure using thymol to increase the shelf life of onions. To control fungal infections that occur during low-temperature storage, it is necessary to identify the predominant fungal pathogens that appear in low-temperature storage houses. Botrytis aclada was found to be the most predominant fungal pathogen during low-temperature storage. The antifungal activity of the plant essential oil thymol was tested and compared to that of the existing sulfur treatments. B. aclada growth was significantly inhibited up to 16 weeks with spray treatments using a thymol solution. To identify an appropriate method for treating onions in a low-temperature storage house, thymol was delivered by two fumigation treatment methods, either by heating it in the granule form or as a solution at low-temperature storage conditions (in vivo). We confirmed that the disease severity was reduced up to 96% by fumigating thymol solution compared to the untreated control. The efficacy of the fumigation of thymol solution was validated by testing onions in a low-temperature storage house in Muan, Jeollanam-do. Based on these results, the present study suggests that fumigation of the thymol solution as a natural preservative and fungicide can be used as an eco-friendly substitute for existing methods to control postharvest disease in long-term storage crops on a commercial scale.

  • Recent keywords of the heavy industries are large-scale structure and productivity. Especially, the sizes of the commercial vessels and the offshore structures have been gradually increased to deliver goods and explore or produce oil and natural gas in the Arctic. High heat input welding processes such as electro gas welding (EGW) have been widely used for welding thick steel plates with flux-cored arc welding (FCAW), especially in the shipbuilding industries. Because high heat input welding may cause the detrimental effects on the fracture toughness of the welded joint and the heat affected zone, it is essential to obtain the sufficient toughness of welded joint. There are well known that the fracture toughness like CTOD, CVN, and KIC were very important factors in order to secure the safety of the structures. Furthermore, the welding residual stress should be considered to estimate the unstable fracture in both EGW and FCAW. However, there are no references on the welding residual stress distribution of EGW and FCAW with thick steel plates. Therefore the welding residual stresses were very important elements to evaluate the safety of the welded structure. Based on the measurement results, the characteristics of residual stress distribution through thickness were compared between one-pass electron gas welding and multi-pass flux-cored arc welding. The longitudinal residual stress in the multi-pass flux-cored arc welding is tensile through all thicknesses in the welding fusion zone. Meanwhile, longitudinal residual stress of EGW is tensile on both surfaces and compressive at the inside of the plate. The magnitude of residual stresses by electron gas welding is lower than that by flux-cored arc welding.

Antiadipogenic Effects of Salvia plebeia R. Br. Extracts by Extraction Conditions in 3T3-L1 Preadipocytes (추출조건에 따른 배암차즈기 추출물의 지방세포 분화 및 지방축적 억제 효과)

  • This study was carried out to investigate the effects of Salvia plebeia R. Br. ethanolic extract with different aspects (stem/leaf and whole plant) on differentiation and lipid accumulation in 3T3-L1 preadipocytes. The morphological changes and the degrees of lipid accumulation in 3T3-L1 cells were measured by Oil Red O staining and intra-cellular triglyceride (TG) assay. The mRNA expressions of special peroxisome proliferation activated receptor- genes (PPAR), CCAAT/ enhancer-binding protein (C/$EBP{\alpha}$), fatty acid synthase (FAS) and lipoprotein lipase (LPL) were detected by reverse transcriptase polymerase chain reaction (RT-PCR). The 50% ethanolic extracts ($100{\mu}g/mL$) of stem and leaf (SALE) and 30% ethanolic extracts (100 g/mL) of whole plant (SAE) from Salvia plebeia R. Br. were significantly attenuated lipid accumulation during adipogenesis in 3T3-L1 cells. Ethyl acetate-soluble fractions ($50{\mu}g/mL$) significantly inhibited lipid droplet accumulation in 3T3-L1 cells. In addition, SALE induced down-regulation of specific adipogenic transcriptional factors (C/$EBP{\alpha}$ and $PPAR{\gamma}$) and target genes (FAS and LPL) during adipogenesis. Salvia plebeia R. Br. may be used as a safe and efficient natural substance to manage obesity.

Isolation of Surfactant-Resistant Pseudomonads from the Estuarine Surface Microlayer

  • Bioremediation efforts often rely on the application of surfactants to enhance hydrocarbon bioavailability. However, synthetic surfactants can sometimes be toxic to degrading microorganisms, thus reducing the clearance rate of the pollutant. Therefore, surfactant-resistant bacteria can be an important tool for bioremediation efforts of hydrophobic pollutants, circumventing the toxicity of synthetic surfactants that often delay microbial bioremediation of these contaminants. In this study, we screened a natural surfactant-rich compartment, the estuarine surface microlayer (SML), for cultivable surfactant-resistant bacteria using selective cultures of sodium dodecyl sulfate (SDS) and cetyl trimethylammonium bromide (CTAB). Resistance to surfactants was evaluated by colony counts in solid media amended with critical micelle concentrations (CMC) of either surfactants, in comparison with non-amended controls. Selective cultures for surfactant-resistant bacteria were prepared in mineral medium also containing CMC concentrations of either CTAB or SDS. The surfactantresistant isolates obtained were tested by PCR for the Pseudomonas genus marker gacA gene and for the naphthalene-dioxygenase-encoding gene ndo. Isolates were also screened for biosurfactant production by the atomized oil assay. A high proportion of culturable bacterioneuston was tolerant to CMC concentrations of SDS or CTAB. The gacA-targeted PCR revealed that 64% of the isolates were Pseudomonads. Biosurfactant production in solid medium was detected in 9.4% of tested isolates, all affiliated with genus Pseudomonas. This study shows that the SML is a potential source of surfactant-resistant and biosurfactant-producing bacteria in which Pseudomonads emerge as a relevant group.

Effects of Gamma-Irradiation on Biological Activities and Color Changes of Extracts of Schizandrae fructus (감마선 조사가 오미자의 생리 활성과 색상 변화에 미치는 영향)

  • This study was carried out to examine the effects of gamma irradiation on the color removal, antioxidation, DPPH radical scavenger, and antimicrobial activity of Schizandra fructus extracts by different solvents. Schizandra fructus was extracted by hot water, ethanol, acetone and methanol, and the extracts were irradiated 10, 20 and 30 kGy with gamma rays. Hunter color L-value was increased by irradiation in a dose-dependent manner, resulting in brighter color for all solvents used. The a and b values were decreased by irradiation in a dose-dependent manner. Antioxidant activity of the ethanol extract of Schizandra fructus against soybean oil was the highest. The free radical scavenging activity was the strongest in hot water extract. All solvent extracts from Schizandra fructus had the strongest antimicrobial activities to B. subtilis, B. natto, B megaterium, S. aureus, Sal. typhymurium and E. coli. These results suggested that Schizandra fructus extracts have a strong potential as natural food preservatives and cosmetic raw materials. Furthermore, irradiation may not influence adversely on biological activites of the extracts when irradiated up to 30 kGy.

Supplementation of Conjugated Linoleic Acid with γ-Oryzanol for 12 Weeks Effectively Reduces Body Fat in Healthy Overweight Korean Women

  • The enhancement of the human body fat reduction of conjugated linoleic acid (CLA) with the supplementation of $\gamma$-oryzanol (OZ) was investigated on overweight Korean women (n=51, BMI> 23). Subjects were divided into 4 groups of control, CLA, glyceride form of CLA (GCLA), and CLA plus OZ (CLA-OZ). The soft-gel capsule (500 mg) was used to deliver control (500 mg olive oil), CLA (500 mg CLA), GCLA (500 mg GCLA) and CLA-OZ (500 mg CLA plus 50 mg OZ). Three capsules were taken twice a day for 12 weeks. The CLA-OZ supplementation reduced 1.35% body fat that was 0.34% enhancement against CLA supplementation. As considered subject variations, CLA-OZ reduced body fat ranged from 7.9% to -2.7%, equivalent to 5.6 kg loss to 0.7 kg gain in body fat mass, against CLA. The CLA-OZ reduced body weight and body mass index (BMI), relative to control, but the reductions by CLA-OZ were not different from those by CLA and GCLA. All biochemical markers analyzed for safty were not significantly different within or between groups and were within the normal range. The CLA-OZ supplementation significantly reduced blood pressure, as compared to the supplementation of CLA, GCLA and control. These results suggest that OZ could be a useful ingredient to mix with CLA for the reduction of human body fat.

  • Recently, due to people's incontinent use all over the world, fossil fuels such as coal, oil, and natural gas were nearly to be exhausted and also causes serious environment pollutions. Therefore, there is a strong need to develop solar, wind, hydro, biomass, geothermal to replace fossil fuels to prevent suffering from above problems. Wish advances in sensor technology, such data is collected as a kind of stream data which arrives in an online manner so that it is characterized as high- speed, real-time and unbounded and it requires fast data processing to get the up-to-date results. Therefore, the traditional data processing techniques are not fit to deal with stream data. In this paper, we propose a kalman filter-based algorithm to process renewable stream data.

A Comparative Study on the Literature of the Cooking Product of the Korean Rice Cakes in Imowonshibyukji ("임원십육지"의 떡류(餠餌)조리가공에 관한 문헌 비교 연구)

  • Imwonshibyukji is a copying manuscript written around 1825, the 27th year of king Soonjo of the Chosun Period. It is a massive home encyclopedia of total 52 books and 100 volumes. It was quoted from many Chinese literatures The unpublished 'Jeongjoji'of its 17-20th volumes was analyzed to study the cooking methods on the Korean rice cakes (dduck) and studied for the degree of their usefulness and medical values in the light of the cooking and processing science. The book introduced 6 kinds of Korean rice cake: sirudduck (steamed rice cake), chinundduck (steamed and struck rice cake), zizinundduck (small oil fried rice cake), danggui (steamed rice cake mixed with rice power and honey), hondon (=danja: boiled and stuffed rice cake) and butu (fermented rice cake), all of which were composed of total 64 items. The main materials fur the Korean rice cake were as follows: sirudduck and butu were nonwaxy rice powder, chinundduct dinggui and hondon were glutinous rice powder, and zizinundduck was wheat flour. The side material was mainly natural food with the effect of a medicine. The literatures quoted in Imwonshibyukji are as follows: 4 Korean books including Ongheejabji were quoted total 28 times (41.5%), 5 Chinese books including Junsengpaljeon were quoted total 35times (52.2%), and I Japanese book Whawhansamjedohoi was quoted total 4 times (6.0%). The Korean rice cakes quoted in the Korean literature so far exist but the Chinese ones no longer exist. In the comparative analysis of Imwonshibyukji and other contemporary literatures, Chosunmoossangsinsikyorijebub was the most similar one to the book among others.

  • In the present maritime traffic conditions of mokpo harbor, there exists many hazardous factors which may lead to huge accidents including marine oil pollution We analyze marine traffic environments including traffic congestion, natural conditions, maritime traffic accidents of the last 10 years, the fishery status, operation of traffic routes and navigational aids of navigational aids and regulations relating ships' routeing both in and out of the country. Consequently, this work is to propose improved marine traffic system in future.

  • Heavy oil used as ship propulsion has a serious issue regarding exhaust emission of global warming. Recently, among large-scale merchant ships are using LNG as green ships so called ech-ships. In this study, an vaporizer and pipes under cryogenic and high pressure load were considered to evaluate structural integrity according to codes. Structural analysis of the vaporizer and pipes was performed using the commercial code, ANSYS. Integrity evaluation of the vaporizer based on von Mises stress was performed in accordance with allowable stress specified in ASME Boiler & Pressure Vesssel Section VIII Division 2. To assess structural integrity of the pipes, stress components were combined and compared with ASME B31.3. The calculated stresses for all load cases are lower than allowable stresses, therefore the structural integrity of equipments are verified.
