A Convergent Study on Durability over the Exhaust Manifold Shape of Medium-sized Car (중형 자동차의 배기매니폴드 형상에 따른 내구성에 관한 융합 연구)

  • In this study, the configurations of the two engine exhaust manifolds were designed. And the strengths and durabilities were analyzed through the structural analysis and natural frequency analyses of these models. As the result of structural analysis, the strength of model A is much better than that of model B because the maximum stress and deformation of model B are considerably greater than those of model A by more than 9 and 39 times, respectively. It can also be confirmed that model A has the durability better than model B because the maximum frequency of model A is greater than the natural frequency of model B and its maximum deformation is smaller than model B. The result of this study can be used to investigate the durability due to the exhaust manifold shape of medium-sized car without actual test. It also seems to be helpful in the aesthetic convergent design of small car muffler.

Conditioned medium of E17 rat brain cells induced differentiation of primary colony of mice blastocyst into neuron-like cells

  • Background: Conditioned medium is the medium obtained from certain cultured cells and contained secretome from the cells. The secretome, which can be in the form of growth factors, cytokines, exosomes, or other proteins secreted by the cells, can induce the differentiation of cells that still have pluripotent or multipotent properties. Objectives: This study examined the effects of conditioned medium derived from E17 rat brain cells on cells with pluripotent properties. Methods: The conditioned medium used in this study originated from E17 rat brain cells. The CM was used to induce the differentiation of primary colonies of mice blastocysts. Primary colonies were stained with alkaline phosphatase to analyze the pluripotency. The morphological changes in the colonies were examined, and the colonies were stained with GFAP and Neu-N markers on days two and seven after adding the conditioned medium. Results: The conditioned medium could differentiate the primary colony, beginning with the formation of embryoid-body-like structure; round GFAP positive cells were identified. Finally, neuron-like cells testing positive for Neu-N were observed on the seventh day after adding the conditioned medium. Conclusions: Conditioned medium from different species, in this case, E17 rat brain cells, induced and promoted the differentiation of the primary colony from mice blastocysts into neuron-like cells. The addition of CM mediated neurite growth in the differentiation process.

Cultivation of Oyster Mushroom[Pleurotus ostreatus (Jacq. ex Fr.) Kummer] at Medium with Ca(OH)2 Treatment (수산화칼슘[Ca(OH)2] 처리 배지를 이용한 느타리버섯의 재배)

  • This research is aimed to investigate the cultivation method of oyster mushroom[Pleurotus ostreatus (Jacq. ex Fr.) Kummer] at the non-sterilized medium with $Ca(OH)_2$ treatment. Therefore, experiments were carried out to develop non-sterilization method of medium by addition of $Ca(OH)_2$ for omission of heat sterilization progress of medium. General components, minerals and amino acid in Jiri wild type No. 1 (Pleurotus ostreatus) and production cost were analyzed. For the purpose of omission of heat sterilization progress, treatment ratio of $Ca(OH)_2$ (purity 95%) was 5%(w/w) of dry medium. Initial pH of this medium was 11 and then the pH was changed by 9 after the uniform mixing of the medium for half an hour. The various germs occurred 50% and 100% at pH 8 and pH 7 of the non-sterilized medium, respectively. Production of oyster mushroom increased by $2,030\;ton\;ha^{-1}$ when the main raw material used corn pith instead of waste cotton. The time required of mycelium culture was 30 days when hypha was cultured at the non-sterilized medium, and pinhead occurred when 2 or 3 days was passed after the time required of mycelium culture. Occurrence of pinhead was most rapid at the condition of $22{\sim}26^{\circ}C$, 65% humidity and pH $6.5{\sim}7.0$ and required of $22{\sim}28$ days at $70{\sim}80\;mm$ thickness of non-sterilized medium. Ca content in 1st harvest oyster mushroom was higher than that in 2nd harvest one, and its difference was $30.3\;mg\;kg^{-1}$. Amino acid content by stipe thickness of oyster mushroom was ranged from 411.2 to $343.9\;mg\;kg^{-1}$ both in a pileus and a stipe of 1st harvest mushroom, and from 402.4 to $498.2\;mg\;kg^{-1}$ and from 442.6 to $470.4\;mg\;kg^{-1}$ in those of 2nd harvest one, respectively. The results of the present study suggest that the non-sterilization medium by addition of $Ca(OH)_2$ is usable with the cultivation of oyster mushroom.

  • This study was conducted to isolate and identify bacteria producing xylanase and cellulase from spent mushroom substrates and to determine the optimal medium conditions for their growth. Bacteria showing high xylanase and carboxymethyl cellulase activities and low protease and amylase activities were strain 201-3 and strain 206-3. Strain 201-3 was identified as Enterobacter ludwigii and named Ent. ludwigii KU201-3. 206-3 was identified as Bacillus cereus and named B. cereus KU206-3. The optimal medium condition of Ent. ludwigii KU201-3 was obtained when 1%(w/v) of soybean meal and 3%(w/v) of sucrose were used as nitrogen and carbon source, respectively. That of B. cereus KU206-3 was obtained when 3%(w/v) of soybean meal and 1%(w/v) of molasses were used as nitrogen and carbon sources, respectively.

  • The effects of subcooling and natural convection are studied numerically on the melting process of an initially subcooled phase-change medium filled inside a horizontal circular cylinder. It is postulated that melting continues with the tube wall kept at a constant temperature and with the unmelted solid core fixed. Primary emphasis is placed on the evolution of interface morphology, the local/overall heat transfer rate at the tube wall and at the interface, and the structure of natural convection. The numerical results are mainly presented in terms of the Rayleigh and subcooling numbers. As the degree of subcooling intensifies, the melting rate and the movement of the interface are impeded but the interfaces are of similar shape with the passage of time. The heat transfer characteristics are found to be mostly governed by the formation pattern of natural convection in the liquid phase. Good agreement with available experimental data is found.

  • Carotene, xanthophyll, carotenoid anthocyan, phycopyrine, chlorophyll, and monascus pigments are used as natural coloring agents since they are more stable to human body than synthetic coloring agents. Among them, monascus pigments are a natural red pigment produced by the Monascus purpureus. For the development of edible paint using natural pigment, Monascus purpureus strain was cultured at a temperature of $35^{\circ}C$ for 15 days on a PDYA plate and liquid medium to produce a red pigment. In addition, a large amount of the red pigment was extracted from Hongkuk Koji in parallel with water extraction and ultrasonic wave extraction. At this time, the yield of ultrasonic extract was 2~4 times higher. Thus, Monascus purpureus strains, etc. were prepared by freeze-drying powder. In conclusion, natural paints made with red pigments have enabled the development of been edible paints that can be used as eco-friendly materials with good viscosity, enhanced spread ability and coloration.

The Effect of Self-Leadership on the Lntention of Start-up in Women: The Mediation Effect of Self-Efficacy (여성의 셀프리더쉽이 창업의도에 미치는 영향 : 자기효능감의 매개효과)

  • This study analyzed the impact of women's self-preservation on their intention to start a business by using self-efficacy as a mediation, reflecting the importance of women's self-preservation to adapt to the new environment of start-up. In particular, in addition to the overall self-preservation, the sub-factors of behavioral delay, compensatory natural strategy, and constructive thinking strategy were also validated. As a research method, 198 people who responded to the online survey were analyzed using Model 4 of the PROCESS macro proposed by Hayes(2018), and indirect effects were analyzed by designating 5,000 bootstrapping samples. The findings were as follows. First, self-efficacy has been shown to have a mediating effect in which women's self-preservation influences their intention to start a business. At this time, the medium effect was shown as a partial medium. Second, the sense of self-efficacy was found to have a mediating effect on women's behavior-oriented strategies affecting their intention to start a business. At this time, the medium effect was shown as a partial medium. Third, self-efficacy was found to have a mediating effect in which women's natural compensatory strategies affect their intention to start a business. At this time, the medium effect was shown as a partial medium. Fourth, self-efficacy was found to have a mediating effect in which women's constructive thinking strategies affect their intention to start a business. At this time, the medium effect was shown as a complete medium. This shows that the higher a woman's self-preservation, the higher her self-efficacy and willingness to start a business. Based on the results of these studies, he made a developed proposal for women's start-ups.

Growth, Flowering, and Nutrient Composition of Salvia Grown in Peat moss Media Containing Pellets Processed with Poultry Feather Fibers at Different Mixing Ratios

  • The objective of this study was to determine the effect of replacing perlite (PL) with pellets processed with poultry feather fiber as an inert material to prepare growing medium. The growth and flowering of Salvia splendens 'Vista Red' grown in individual growing medium $Biosangto^{(R)}$, peat moss (PM), PL, and two pellets (P45-1 and P45-2) were evaluated. Peat moss was mixed with PL, P45-1, or P45-2 at various ratios (1:0 to 1:3 or 3:1 by volume) to investigate the feasibility of replacing PL with pellets. Nutrient composition of the growing medium and leaf tissues was analyzed. The number of florets, inflorescence length, plant height, and fresh weight of plants grown in media containing P45-1 or P45-2 were reduced compared to those grown in individual growing medium PM or PL. As the mixing ratio of P45-1 or P45-2 to PM was higher, the growth of salvia, such as inflorescence length, plant height, number of leaves, and fresh weight was inhibited. Our results indicate that mixing three parts PM with one part of P45-1 (PM/P45-1/3:1) or P45-2 (PM/P45-2/3:1) accelerated flowering and increased the number of florets and leaves compared to other mixing ratios of PM and pellets media. The concentrations of phosphorus (P), calcium (Ca), boron (B), iron (Fe), and copper (Cu) in individual growing medium PL, P45-1, and P45-2 were significantly lower than those in PM. The concentration of N was the highest in leaves of plants grown in P45-1 or P45-2 amended media, and the concentrations of P, Ca, and zinc (Zn) in leaves were lower in individual growing medium P45-1 or P45-2 than in PM and PL. The pH of PM/P45-1/3:1 or PM/P45-2/3:1 media was maintained at optimal level (5.8-5.9) and the concentrations of macro- and micro-elements in the media and leaves were considered to be optimal levels. Therefore, mixing three parts PM with one part P45-1(PM:P45-1/3:1) or P45-2 (PM:P45-2/3:1) is recommended for improved growth and flowering in salvia. This suggests that P45-1 or P45-2 can replace PL as an inert material to prepare growing medium.

Keratinase Production by Recalcitrant Feather Degrading Pseudomonas Geniculata and Its Plant Growth Promoting Activity (난분해성 우모분해 Pseudomonas geniculata에 의한 케라틴 분해효소 생산 및 식물성장 촉진 활성)

  • We investigated the optimal conditions for keratinase production by feather-degrading Pseudomonas geniculata H10 using one variable at a time (OVT) method. The optimal medium composition and cultural condition for keratinase production were determined to be glucose 0.15% (w/v), beef extract 0.08% (w/v), $KH_2PO_4$ 0.12% (w/v), $K_2HPO_4$ 0.02% (w/v), NaCl 0.07% (w/v), $MgSO_4{\cdot}7H_2O$ 0.03%, $MgCl_2{\cdot}6H_2O$ 0.04% along with initial pH 10 at 200 rpm and $25^{\circ}C$, respectively. The production yield of keratinase was 31.6 U/ml in an optimal condition, showing 4.6-fold higher than that in basal medium. The strain H10 also showed plant growth promoting activities. This strain had ammonification activity and produced indoleacetic acid (IAA), siderophore and a variety of hydrolytic enzymes such as protease, lipase and chitinase. Therefore, this study showed that P. geniculata H10 could be not only used to upgrade the nutritional value of feather wastes but also useful in situ biodegradation of feather wastes. Moreover, it is also a potential candidate for the development of biofertilizing agent applicable to crop plant soil.

A Cell-Based Assay System for Monitoring NF-$\kappa$B Activity in Human Epidermal Keratinocytes: A Screening Tool of the Antioxidants and Anti-inflammatories for Dermatological Purpose

  • A cell-based assay system for monitoring NF-$textsc{k}$B activity was developed to determine the influence of activated NF-$textsc{k}$B in human HaCaT cells. The pNF-$textsc{k}$B-SEAP-NPT plasmid that permits expression of the secreted alkaline phosphatase (SEAP) reported gene in response to the NF-$textsc{k}$B activity and contains neomycin phosphotransferase (NPT) gene for the geneticin resistance in host cells was constructed and transfected into human keratinocyte cell line HaCaT. Human HaCaT transfectant cells secreted the SEAP enzyme into the culture medium in a time-dependent manner until 72h. NF-$textsc{k}$B activities were measured in the SEAP reporter gene assay using a fluorescent detection method. The treatment of HaCaT cell transfectants with known antioxidants [e.g., N-acetyl-L-cysteine and vitamin C] showed inhibition of NF-$textsc{k}$B activity in a time-and concentration-dependent manner. The phorbol 12-myristate 13-acetate (PMA) known as a stimulator of NF-$textsc{k}$B expression demonstrated that it increased NF-$textsc{k}$B activity in a time- and concentration-dependent manner. This assay system could be used to determine the quantitative measurement of NF-$textsc{k}$B activity in the human skin and allow the screening of anti-inflammatory agents from various synthetic chemicals and natural products for dermatological purpose.
