Sports impact on the nanomedicine absorption in drug delivery

  • Physical activities enhance blood flow in the vessels, which may increase the quality of medicine delivery. The emergence of revolutionary technologies such as nanoscience, made it possible to treat the incurable illnesses such as cancer. This paper investigates the impact of sport and physical exercises on the quality and quantity of the drug-delivery based on the mathematical modeling of a nanomotor made by nanotubes carrying the nano-drug capsules. Accordingly, the mathematical equations of rotating nanomotor are generated by considering the both of higher-order beam model and nonlocal strain gradient model, as a comprehensive continuum theory. Next, through the generalized differential quadrature together with Newmark-beta methods, the differential relations are discretized and solved. Finally, the impact of varied parameters on the dynamical behavior of the nanomotor is examined in detail. The outcomes of this investigation can be useful to achieve an excellent design of nanomotors carrying nano-drugs.

Development of wrinkled skin-on-a-chip (WSOC) by cyclic uniaxial stretching

  • The skin experiences constant physical stimuli, such as stretching. Exposure to excessive physical stimuli stresses the skin and can accelerate aging. In this study, we applied a method that allowed human fibroblasts and keratinocytes to be perfused with media to form 3D skin equivalents that were then uniaxially 10%-stretched for 12 h per day (at either 0.01 or 0.05 Hz) for up to 7 days to form wrinkled skin-on-a-chip (WSOC). There was more wrinkling seen in skin equivalents under 0.01 Hz uniaxial stretching than there was for non-stretched skin equivalents. At 0.05 Hz, the stratum corneum almost disappeared from the skin equivalents, indicating that stretching was harmful for the epidermis. At both frequencies, the production of collagen and related proteins in the skin equivalents, such as fibronectin 10 and keratin, decreased more than those in the non-stretched equivalents, indicating that the dermis also suffered from the repeated tensile stress. These results suggest that WSOCs can be used to examine skin aging and as an in vitro tool to evaluate the efficacy of anti-wrinkle cosmetics and medicines.

  • Gold nanoparticles have recognized a promising drug carriers in many diseases. These nanoparticles could carry anti-inflammatory drugs in the case of muscle injury and for fatigue relief. On the other hand, specific surface of this kind of nanoparticles could be critical in amount of drug they could carry. Therefore, in this study, we explore different methodology and influencing parameters on the specific surface of gold nanoparticles. After specifying the main parameters, different machine learning and artificial neural network are adopted to model the effects of different parameters. Furthermore, response surface methodology is utilized to obtain a quadrilateral relationship between different parameters and specific surface. The results indicate that concentration of the gold salt solution is the most important parameter in increasing the size of gold nanoparticle and, as a consequence, increasing specific surface. Moreover, the ability of gold nanoparticles in prolonging retention of the drugs is discussed in detail.

Effects of Nano-sized Carbon Black on the Lungs of High Fat-diet Induced Overweight Rats

  • Objectives This study was conducted to determine whether nano-sized carbon black exposure results in greater damage in high fat diet-induced overweight rats than normal weight ones and to identify the possible causes of any differences. Methods Two groups of Sprague-Dawley rats allocated by body weight (normal and overweight) were exposed to aerosolized nano-sized carbon black for 6 hours a day, 5 days per week over a 4-week period. Differential cell counts, lactate dehydrogenase (LDH) activities and albumin concentrations were measured in bronchoalveolar lavage (BAL) fluid, and histopathological findings in the lungs were evaluated. Tumor necrosis factor-alpha (TNF-${\alpha}$) and interleukin (IL)-6 were measured in BAL fluid and supernatants of lipopolysaccharide(LPS)-stimulated lymphocyte culture. Results Rats exposed to high concentrations of nano-sized carbon black showed significantly increased (p <0.05) polymorphonuclear leukocyte number and LDH activity in the BAL fluid from both overweight and normal rats. Mild histopathological changes were observed in normal rats irrespective of carbon black concentrations. However, severe histological scores were found in overweight rats ($1.75{\pm}0.46$, $2.25{\pm}0.46$, and $2.88{\pm}0.35$ after low, medium, and high concentration exposures). Proinflammatory cytokine levels of TNF-${\alpha}$ and IL-6 were significantly higher in the supernatant of LPS-stimulated lymphocytes of overweight rats, whereas there was no significant difference in the BAL fluid between normal and overweight rats. Conclusions Inflammation and damage to lungs exposed to nano-sized carbon black was more severe in high fat diet-induced overweight rats compared to normal rats.

Fabrication of Biodegradable Disc-shaped Microparticles with Micropattern using a Hot Embossing Process with Porous Microparticles

  • This paper demonstrates the development of a method for preparing micropatterned microdiscs in order to increase contact area with cells and to change the release pattern of drugs. The microdiscs were manufactured with hot embossing, where a polyurethane master structure was pressed onto both solid and porous microparticles made of polylactic-co-glycolic acid at various temperatures to form a micropattern on the microdiscs. Flat microdiscs were formed by hot embossing of porous microparticles; the porosity allowed space for flattening of the microdiscs. Three types of micro-grooves were patterned onto the flat microdiscs using prepared micropatterned molds: (1) 10 ${\mu}M$ deep, 5 ${\mu}M$ wide, and spaced 2 ${\mu}M$ apart; (2) 10 ${\mu}M$ deep, 9 ${\mu}M$ wide, and spaced 5 ${\mu}M$ apart; and (3) 10 ${\mu}M$ deep, 50 ${\mu}M$ wide, and spaced 50 ${\mu}M$ apart. This novel microdisc preparation method using hot embossing to create micropatterns on flattened porous microparticles provides the opportunity for low-cost, rapid manufacture of microdiscs that can be used to control cell adhesion and drug delivery rates.

The Effects of HAp and BCP Nano Powders Synthesized by Microwave-Assisted Synthesis on the Activation of Osteoblast and Osteoclast (마이크로웨이브법에 의해 제조된 HAp 및 BCP 분말이 뼈모세포 및 파골세포의 활성에 미치는 영향)

  • Hydroxyapatite (HAp) and biphasic calcium phosphate (BCP) nano powders were synthesized using the microwave-assisted synthesis process dependent on pH and microwave irradiation time. The average size of a powder was less than 100 nm in diameter. Through in-vitro cytotoxicity tests by an extract dilution method, the HAp and BCP nano powders have shown to be cytocompatible for L-929 fibroblast cells, osteoblastlike MG-63 cells and osteoclast-like Raw 264.7 cells. The activation of osteoblast was estimated by alkaline phosphatase (ALP) activity. When the HAp and BCP were treated to MG-63 cells, alkaline phosphatase activities increased on day 3, compared with those of the untreated cells. Also, the collagen fibers increased when the HAp and BCP powders suspension were treated to MG-63 cells, compared to those of the untreated cells. Quantitative alizarin red S mineralization assays showed a trend toward increasing mineralization in osteoblast cultured with powder suspension. In conclusion, hydroxyapatite and biphasic calcium phosphate appeared to be a bone graft substitute material with optimal biocompatibility and could be further applied to clinical use as an artificial bone graft substitute.

Association between oxidative stress and blood pressure in Korean subclinical hypertensive patients (경계성 고혈압 환자에서 혈압과 산화 스트레스 관련 지표 간의 상관성에 관한 연구)

  • This study was conducted in order to investigate the association between hypertension and oxidative stress-related parameters and to evaluate these parameters in subclinical hypertensive patients and normotensive subjects living in Korea. We attempted to determine whether oxidative stress-related parameters would differ between two groups of 227 newly-diagnosed, untreated (systolic blood pressure (BP) ${\geq}$ 130 mmHg and diastolic BP ${\geq}$ 85 mmHg) and 130 normotensive subjects (systolic BP < 120 mmHg and diastolic BP < 80 mmHg). General characteristics of the subjects were collected using a simple questionnaire. From subjects' blood, degree of DNA damage in lymphocytes, the activities of erythrocyte superoxide dismutase, catalase, and glutathione peroxidase, level of plasma total radical-trapping antioxidant potential (TRAP), glutathione, and anti-oxidative vitamins, as well as plasma lipid profiles and conjugated diene (CD) were analyzed. Evaluation of the associations of oxidative stress-related parameters with blood pressure of the subjects was performed using Pearson partial correlation and multivariate logistic regression analysis after adjusting for confounding factors. Several oxidative stress-related parameters were higher in subclinical hypertensive patients than in normotensive subjects. Plasma levels of ${\alpha}$-tocopherol, ${\beta}$-carotene, TRAP, and activity of GSH-px were significantly lower in subclinical hypertensive patients than in normotensive subjects. Increased levels of DNA damage, lipid peroxidation, triglyceride, total cholesterol, and LDL-cholesterol were observed in subclinical hypertensive patients. These results confirm an association between blood pressure and oxidative stress-related parameters and suggest that the pathogenic role of oxidative stress in hypertension might be significant.

  • The nano-surface modification techniques could be classified; internal modifications which enhance surface roughness and porosity in nano level and external modifications as nano particle coating. Nano-modified implant surface has various morphograpies such as nanotube, nanopit, nanonodule and polymorphic structures. Creating surface depends upon preparation method and material, however, there is no standard preparation technique not yet. The nano-modified surfacet is electrochemically stable comparing with the surface modified in micron level. Nano-modified surface has little cytotoxicity, stimulates osteoblast proliferation and differentiation. Moreover, it decreases soft tissue intervention by interrupting the proliferation of fibroblast. Nanostructure has similar size and shape with cells and proteins, consequently leads to good biocompatibility and enhanced osseointegration. However, the actual effect in vivo is limited, due to the distance of effect. Even if nano-modified surface has antibiotic property due to photocatalysis, short duration time makes clinical application questionable. Further investigations should focus on the optimal nano-modified surface, which has many potentials.

  • The retinal pigment epithelium (RPE) plays an important role in maintaining a healthy retina and the degeneration of RPE caused a number of retinal diseases. The transplantation of RPE has recently become a possible therapeutic modality for retinal degeneration. To transplant RPE cells securely, substrates are essential, and then as a substrate, we fabricated films using silk that has unique mechanical properties and biocompatibility. After the FTIR spectra, contact angle and biodegradation of silk films were confirmed, RPE cells were seeded and the influence of RPE cells on silk films was examined. We measured the cell adhesion, cell viability, morphology and specific mRNA expression by MTT assay, SEM, immunofluorescence and RT-PCR. In this study, we confirmed that attachment, proliferation and phenotype maintenance of RPE cells cultured on silk films were great, and thereby we were able to confirm the potential applications of silk films as tissue engineering carrier for regeneration of retina.

Improved Dissolution Behavior of Aceclofenac Loadings with Kollidon VA 64 Using Spray Drying and Rotary Evaporation Process (분무건조와 용매증발을 이용한 Kollidon VA 64에 포접된 아세클로페낙의 개선된 용출 거동)

  • In order to improve the poor water solubility of aceclofenac, it was loaded into solid Kollidon VA 64 dispersion prepared by spray drying and rotary evaporation methods using different drug and polymer ratios. Morphology and physicochemical behavior of the aceclofenac loaded solid dispersions was analyzed by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), X-ray diffractometry (XRD), and differential scanning calorimetry (DSC). Encapsulation efficiency and dissolution behavior in a simulated intestinal juice of aceclofenac in the solid dispersions was measured using HPLC and the latter was compared with that of the active pharmaceutical ingredient (API) and Airtal$^{(R)}$. It was demonstrated that two methods could significantly improve the dissolution behavior of aceclofenac.
