We performed diagnostic PCR assays and a phylogenetic analysis using partial sequences of TEF1 (translation elongation factor-1) to determine the trichothecene chemotypes and genetic diversity of F. graminearum isolates from maize and rice samples collected in 2009 in Korea. PCR using a species-specific primer set revealed a total of 324 isolates belonging to the putative F. graminearum species complex. PCR with trichothecene chemotypespecific primers revealed that the nivalenol (NIV) chemotype was predominant among the fungal isolates from rice (95%) in all provinces examined. In contrast, the predominant chemotype among the corn isolates varied according to region. The deoxynivalenol (DON) chemotype was found more frequently (66%) than the NIV chemotype in Gangwon Province, whereas the NIV chemotype (70%) was predominant in Chungbuk Province. Phylogenetic analysis showed that all DON isolates examined were clustered into lineage 7, while the NIV isolates resided within lineage 6 (F. asiaticum). Compared with previous studies, the lineage 6 isolates in rice have been predominantly maintained in southern provinces, while the dominance of lineage 7 in maize has been evident in Gangwon at a slightly reduced level.

Fungal occurrence during production of dried red pepper was investigated using red pepper samples collected at harvest, before and after washing, and before, during or after drying. Fungal incidence was evaluated by counting the number of fungal colonies grown after incubating random pepper cuts on potato dextrose agar plates. Washing with ground water had no significant effect on reduction of fungal contamination. Fungal increase was observed in some samples, and the insides of washer and containers were contaminated with fungi. Drying caused significant fungal increase regardless of drying method although the fungal incidence after machine drying was lower than that after greenhouse drying. Fungal increase was observed in the samples being dried in a greenhouse and some mycotoxigenic species were also detected. Therefore, the most important control point for fungal contamination during dried pepper production appears to be the drying process, especially in a greenhouse.

Sample preparation methods were evaluated for effectiveness in detecting foodborne pathogens from sprout seeds. The methods included: Rinse.-Test portions were rinsed with 0.1% peptone water, and the pellet after centrifugation was inoculated into pre-enrichment media; and Sprouting.-Seed samples were sprouted before pre-enrichment and sprouted seeds were inoculated into pre-enrichment media. In rinse method, E. coli was isolated from 13 of 280 sample units. In sprouting method, E. coli was isolated from 12 of 135 sample units. E. coli O157:H7, Salmonella spp., and L. monocytogenes were not detected in any of the samples. In the trials for recovering Salmonella enterica from artificially contaminated alfalfa seeds, the soak, rinse, and sprouting methods were evaluated. The detection rates of S. enterica were statistically different according to the amount of the sample tested and selective medium type (P < 0.05).

A total of 244 cereal samples (oat, sorghum, adlay, and proso millet) were collected from fields to examine the contamination of Fusarium mycotoxins in cereals during harvest season in 2017 and 2018. The contamination levels of deoxynivalenol (DON), nivalenol (NIV), and zearalenone (ZEA) were analyzed individually by using the immunoaffinity column clean-up method with ultra performance liquid chromatography, and fumonisins (FUM) were analyzed by using the QuEChERS method with liquid chromatography-mass spectrometry. Highest level of NIV contamination (120.0-3277.0 mg/kg) was observed in oat samples among the analyzed cereals. In the adlay samples, DON contamination was the highest (maximum level 730.0 ㎍/kg). The proso millet samples had a high frequency of detection of NIV and ZEA (61.5% and 57.9%, respectively), but the levels were low (average detection level of NIV, 75.6 ㎍/kg, for ZEA, 21.5 ㎍/kg). Among the cereal samples, sorghum had the highest contamination frequency of DON, ZEA, and FUM, and the co-occurrence of Fusarium mycotoxin was 70.0%, which was higher than the average of 29.9%. In order to safely manage Fusarium mycotoxin levels in cereals, continuous research on the development of contamination prevention technologies together with monitoring of mycotoxin contamination is needed.

To investigate the occurrence of fungi in dried ginseng products, we collected 24 white and 26 red ginseng samples from the retail market. Fungi were detected in 50% and 46% of white and red ginseng samples, respectively. The average level of fungal contamination was 0.5 and 0.2 log10 CFU/g in white and red ginseng, respectively. In white ginseng, Penicillium polonicum, P. chrysogenum, and Rhizopus microsporus dominated with each having an occurrence of 18.2%. In red ginseng, Aspergillus spp. was dominant with an occurrence of 87.6%, with A. chevalieri having the highest occurrence (50%). PCR screening for mycotoxigenic potential showed that the 13 isolates of 4 species (P. polonicum, P. chrysogenum, P. melanoconidium, and A. chevalieri) tested were negative for the citrinin biosynthetic gene. These results show that the samples tested in this study had low risk of mycotoxin contamination. However, there is a possibility of dried ginseng products, such as white and red ginseng, being contaminated with fungi.
Mulberry is considered a healthy food for antioxidants and many other beneficial nutrients. However, food safety concerns exist as this commodity scarcely passes through a sanitizing step due to the fragile nature of mulberry fruits. Fermented mulberry wort is one traditional way to utilize and preserve mulberries by mixing with sugars although hygienic practices are not often implemented. The purpose of this study was to investigate the fate of foodborne pathogens in fermented mulberry wort. Microbial population of inoculated E. coli in mulberry wort showed a decreasing pattern as the fermentation progressed. A quicker decrease was observed in the mulberry wort mixed with sugar at 1 to 0.8 ratio (w/w, mulberry: sugar) compared to 1 to 1 ratio, which could be due to the amount of acids generated during the fermentation process. When fully-fermented mulberry wort was contaminated with foodborne pathogens, they all decreased in population although their deceasing patterns varied depending on the species of tested bacteria. Steep deceases in population of inoculated foodborne pathogens were observed when the fermented wort was stored at $30^{\circ}C$ in comparisons to the other storage temperature, 5 and $20^{\circ}C$, regardless of bacterial species. This study necessitates the optimization of a sanitizing process during fermentation and storage of mulberry wort.
To investigate Fusarium mycotoxin contamination in rice samples from rice processing complexes (RPCs), paddy rice and rice-milling products such as husks, brown rice, blue-tinged rice, broken rice, rice bran, discolored rice, and polished rice were collected from nationwide in 2012. Three hundred seventy one samples of rice and its by-products were analyzed for three trichothethenes including nivalenol (NIV), deoxynivalenol (DON), and zearalenone (ZEA) by LC/MS. Discolored rice samples were found to have the highest contamination of DON, NIV or ZEA, followed by broken rice. Polished rice samples were largely free from mycotoxins, except three samples which were contaminated with NIV or DON at safety level. The rice byproduct samples were contaminated at higher level and frequencies than polished rice samples.
This study investigated reduction of microbial population, water soluble pigment, capsanthin content, surface color (Hunter L, a, b, ${\Delta}E$), and sensory properties of pepper powder by LED (red, yellow, blue, green) treatments. LED (red, yellow, blue, green) treatment were conducted in 1,000 lux storage at $25^{\circ}C$ for 10 days. The total aerobic bacteria was no significant difference among the control and treated with LED during 10 days. In yellow LED treatment, yeast and molds were decreased about 1.76 log. Surface color such as lightness (L), redness (a), yellowness (b) were showed a decreasing tendency as the storage period. In the overall color difference (${\Delta}E$) of yellow LED treatment was lower less than 3.0. Water soluble pigment was no difference in control and LED treated samples during storage period. Capsanthin content was significantly decreased as storage period was increased, but no significant differences were observed among red and yellow LED treatments. Sensory properties of control was significantly reduced by storage period but yellow and green LED treatments were no significantly differences.
Red pepper is widely used as a spicy flavor ingredient in the food industry and many households. The objective of this study was to assess the total aerobic bacteria count, coliforms count and incidence of Escherichiacoli, Salmonella spp., Escherichiacoli O157:H7, Listeria monocytogenes, and Bacillus cereus in red pepper and red pepper cultivated soil. The total aerobic bacteria number in red pepper and soil were in the range of 2.97 to 8.13 and 5.91 to 7.65 log CFU/g, respectively. The coliforms in red pepper and soil were in the range of 1.87 to 6.71 and 0.67 to 6.16 log CFU/g, respectively. E. coli was detected in 3 of 54 soil samples. In 3 out 63 red pepper and 53 of 54 soil samples, B. cereus was detected, while Salmonella spp., E.coli O157:H7, and L.monocytogenes were not detected. The results from this study provide an important basic information associated with the microbiological safety of fresh vegetables. Continuous caution is needed to prevent the contamination of pathogenic microorganisms during its farming.

The purpose of this study was to investigate hazardous microorganisms in mixed baby leafy vegetables and various baby leafy vegetables used as raw materials for fresh-cut produce in spring and summer. To estimate microbial loads, a total of 298 samples including 181 samples of mixed baby leafy vegetables purchased in a Korean market and 117 samples of various baby leafy vegetables from distribution companies were collected. Fecal indicators (coliform and Escherichia coli) as well as food-borne pathogens (E. coli O157:H7, Salmonella spp., Staphylococcus aureus) were enumerated. As a result, the mixed baby leafy vegetable samples showed significantly higher (P<0.05) coliform bacteria numbers in summer (5.59±1.18 log CFU/g) compared to spring (3.60±2.53 log CFU/g). E. coli was detected in 1.3% (1/79) and 42.2% (43/102) of samples collected in spring and summer, respectively. Only one sample collected from a market in spring was contaminated with S. aureus. In the experiment with baby leafy vegetables, the number of coliforms detected in baby leafy vegetables cultivated in soil in spring was 1.15±1.95 log CFU/g, and that in summer was 4.09±2.52 log CFU/g. However, the number of coliforms recovered from baby leafy vegetables cultivated in media was above 5.0 log CFU/g regardless of season. Occurrences of E. coli were 44.4% (12/27) and 19.0% (4/21) for baby leafy vegetables cultivated in soil and media, respectively. However, E. coli O157:H7 and Salmonella spp. were not detected. These results are in relation to microbial loads on mixed baby leafy vegetables associated with raw materials. Therefore, it is necessary to develop and implement hygienic practices at baby leafy vegetable farms to enhance the safety of fresh produce.
