한약재 중 살충제 Endosulfan의 잔류분석을 위한 Macroporous Diatomaceous Earth 컬럼 적용 (Application of Macroporous Diatomaceous Earth Column for Residue Analysis of Insecticide Endosulfan in Herbal Medicines)

  • 현재 식품의약품안전청에서 고시하고 있는 한약재 중 잔류 농약 분석법에 따라 강활, 박하, 천궁 및 황기 시료 중 endosulfan과 그 분해산물인 endosulfan sulfate를 분석한 결과 많은 간섭물질의 등장과 낮은 회수율 등의 문제점을 확인하였다. 이에 고시된 시험법의 액-액 분배 과정을 대신하여 MDE column을 적용하여 ${\alpha}$-endosulfan 80.3 ~ 93.5%, ${\beta}$-endosulfan 81.0 ~ 100.3%, endosulfan sulfate 80.6 ~ 95.6%의 회수율과 1.1 ~ 3.4%의 변이계수(CV)를 얻었으며 이러한 결과는 잔류농약 분석기준을 만족하였다. MDE column의 사용으로 액-액 분배 과정을 대신하면서 발암가능 물질인 dichloromethane 사용을 배제할 수 있어 분석자의 안전성 향상, 노동력 및 전처리 시간의 절감, 발생되는 폐액 감소, 액-액 분배시 emulsion 현상의 해소, 분석자간의 재현성 양호 등의 이점도 얻을 수 있었다.

MALDI Mass Spectrometric Analysis of Nonderivatized Steroids Using Cyclodextrin-supported 2,5-Dihydroxybenzoic Acid as Matrix

  • Sex hormones are important metabolites in vertebrates' development and reproduction. For rapid screening sex hormones, matrix-assisted laser desorption/ionization (MALDI) mass spectrometry (MS) is one of the promising analytical platforms, but MALDI MS faces many challenges in detecting steroids such as low ionization efficiency and matrix background interference. One potential strategy to overcome matrix interference in the low m/z region is using a cyclodextrin (CD)-supported matrix for steroid analysis since CD-supported matrixes are known to effectively suppress matrix-related ion signals. In this study, we aimed to find the optimal CD-supported matrix for the analysis of the nonderivatized sex steroids. Our results showed that the ${\alpha}CD$-supported 2,5-dihydroxybenzoic acid (DHB) matrix efficiently ionized all three major classes of sex hormones, estrogens, androgens, and progestagens, with low or no matrix background and also with high sensitivity. In addition, the ${\alpha}CD$-supported DHB matrix mainly generated molecular ions or protonated ions of sex hormones, and this enabled us to obtain information-rich tandem mass spectra which potentially lead to unambiguous identification of steroid species from complex metabolite mixtures.

From proteomics toward systems biology: integration of different types of proteomics data into network models

  • Living organisms are comprised of various systems at different levels, i.e., organs, tissues, and cells. Each system carries out its diverse functions in response to environmental and genetic perturbations, by utilizing biological networks, in which nodal components, such as, DNA, mRNAs, proteins, and metabolites, closely interact with each other. Systems biology investigates such systems by producing comprehensive global data that represent different levels of biological information, i.e., at the DNA, mRNA, protein, or metabolite levels, and by integrating this data into network models that generate coherent hypotheses for given biological situations. This review presents a systems biology framework, called the 'Integrative Proteomics Data Analysis Pipeline' (IPDAP), which generates mechanistic hypotheses from network models reconstructed by integrating diverse types of proteomic data generated by mass spectrometry-based proteomic analyses. The devised framework includes a serial set of computational and network analysis tools. Here, we demonstrate its functionalities by applying these tools to several conceptual examples.

  • "Korea ginseng (Panax ginseng C.A Meyer) is an important medicinal plant. Its root has been used as an herbal medicine that provides resistance to stress and disease, and prevents exhaustion since the ancient time. Ginsenosides, glycosylated triterpene (saponin), are considered to be the main active compounds of the ginseng root. Despite of considerable commercial interests of ginsenosides, very little is known about the genes and their biochemical pathways for ginsenoside biosynthesis. This work will focus on the identification of genes involved in ginsenoside biosynthesis and the dissection of ginsenoside biosynthetic pathway using a functional genomics tool. Expression sequence tags (ESTs) provide a valuable tool to discovery the genes in secondary metabolite biosynthesis. We generated over 21,155 ginseng ESTs that is now sufficient to facilitate discovering the genes involved in ginsenoside biosynthesis such as oxidosqualene cyclase(OSC), cytochrome P450 and glycosyltransferase. With ESTs information, microarray technology will be used for the analysis of gene expression, and the identification of genes including transcription factors expressed in tissues under given experimental condition. Heterogous system such as yeast and plants will allow us to do the functional analysis. And selected ginseng hairy root which show variation in ginsenoside production will be used as a material for functional analysis of candidate gene. Functional genomics approach will successfully accelerate gene discovery, and also provide promises of metabolic engineering for the ginsenoside production."

Metabolomic approach for discrimination of processed ginseng genus (Panax ginseng and Panax quinquefolius) using UPLC-QTOF MS

  • Discriminating between two herbal medicines (Panax ginseng and Panax quinquefolius), with similar chemical and physical properties but different therapeutic effects, is a very serious and difficult problem. Differentiation between two processed ginseng genera is even more difficult because the characteristics of their appearance are very similar. An ultraperformance liquid chromatography-quadrupole time-of-flight mass spectrometry (UPLC-QTOF MS)-based metabolomic technique was applied for the metabolite profiling of 40 processed P. ginseng and processed P. quinquefolius. Currently known biomarkers such as ginsenoside Rf and F11 have been used for the analysis using the UPLC-photodiode array detector. However, this method was not able to fully discriminate between the two processed ginseng genera. Thus, an optimized UPLC-QTOF-based metabolic profiling method was adapted for the analysis and evaluation of two processed ginseng genera. As a result, all known biomarkers were identified by the proposed metabolomics, and additional potential biomarkers were extracted from the huge amounts of global analysis data. Therefore, it is expected that such metabolomics techniques would be widely applied to the ginseng research field.

Study of Metabolic Profiling Changes in Colorectal Cancer Tissues Using 1D 1H HR-MAS NMR Spectroscopy

  • Metabolomics is a field that studies systematic dynamics and secretion of metabolites from cells to understand biological pathways based on metabolite changes. The metabolic profiling of intact human colorectal tissues was performed using high-resolution magic angle spinning (HR-MAS) NMR spectroscopy, which was unnecessary to extract metabolites from tissues. We used two different groups of samples, which were defined as normal and cancer, from 9 patients with colorectal cancer and investigated the samples in NMR experiments with a water suppression pulse sequence. We applied target profiling and multivariative statistical analysis to the analyzed 1D NMR spectra to identify the metabolites and discriminate between normal and cancer tissues. Cancer tissue showed higher levels of arginine, betaine, glutamate, lysine, taurine and lower levels of glutamine, hypoxanthine, isoleucine, lactate, methionine, pyruvate, tyrosine relative to normal tissue. In the OPLS-DA (orthogonal partial least square discriminant analysis), the score plot showed good separation between the normal and cancer groups. These results suggest that metabolic profiling of colorectal cancer could provide new biomarkers.

전사체와 대사물질 구조분석을 통한 Novosphingobium pentaromativorans US6-1의 dibenzofuran 분해 경로 해석 (Investigation of biodegradation pathway of dibenzofuran by Novosphingobium pentaromativorans US6-1 via transcriptomic and mass-spectrometric analysis)

  • 다환 방향족 탄화수소(polycyclic aromatic hydrocarbon, PAH) 우수 분해균주인 Novosphingobium pentaromativorans US6-1의 dibenzofuran (DBF) 분해경로를 밝히기 위하여 중간대사물질 분석과 전사체 분석을 진행하였다. GC/MS로 중간대사물질을 분석한 결과, 3(2H)-벤조퓨라논이 검출되었는데 이 화합물은 측면 이산소화에 의해 생성된 중간대사산물들의 기본 골격이 되는 물질로써 균주 US6-1에 의한 DBF의 분해가 측면 이산소화로 진행될 가능성을 시사한다. RNA-Seq 분석 결과, 균주 US6-1이 DBF에 노출되었을 때 발현되는 유전자들의 대부분이 lateral dioxygenation과 관련이 있다는 것을 확인하였다. 이상의 결과로부터N. pentaromativorans US6-1에 의해 일어나는 측면 이산소화를통한 DBF 분해경로와 관련 유전자들을 제시하였다.

Comparative Analysis of Latex Plants by GC-MS using Methanol Extraction

  • Plants are able to produce a large number of diverse bioactive compounds. Solvent extraction is used for isolation of plant metabolites. The extract yield for plant metabolite extraction strongly depends on the nature of solvent. A review showed the methanol can yield more bioactive compounds. Drying of the sample material is also important for the extraction of plant material. The present study was carried out to analyze the phytocomponents of 5 different latex producing plants. The plants like Calotropis gigantea, Carica papaya, Nerium oleander, Ficus benghalensis and Plumeria alba leaves and latex. The GC-MS analysis of the metabolites revealed phytocomponents. Calotropis gigantea leaves showed 14 compounds and latex produced 5 compounds out of this 4,4,6A,6B,8A,11,11,14B-Octamethyl-1,4,4A,5,6,6A,6B,7,8,8A,9,10,11,12,12A,14,14A,14B-Octadeca-hydro-2 and 2R- Acetoxymethyl-1,3,3-trimethyl-4T-(3-Methyl-2-Buten-1-Yl)-1T-Cyclohexanol compound was present in both latex and leaf extraction. Beta. -carotene compound was present in both latex and leaf of Carica papaya. It was observed that Ficus benghalensis contained 2R-Acetoxymethyl-1,3,3-trimethyl-4T-(3-Methyl-2-Buten-1-Yl)-1T-Cyclohexanol was same in latex and leaf extraction.

Study on meat metabolites analysis during the oxidation process

  • This study was conducted to investigate the mechanism of the oxidation process of meat. In the instrumental color, redness, yellowness, and chroma showed significant differences during storage period (P<0.05), whereas hue angle increased (P<0.05), but no significant difference in lightness(P>0.05). The results of TBARS increased significantly during storage period (P<0.05). The thiol and carbonyl contents increased significantly during storage period(P<0.05). Metabolites analysis showed that lactic acid, proline, phenylalanine, mannose, talose, lysine, and tyrosine were significantly different with the storage periods (P<0.05). All the samples used in the experiment were able to confirm that sample went through normal oxidation process with indicators and components were increased or decreased. Further research is needed to study the correlation between metabolite materials.

Two New Species of the Family Acarosporaceae from South Korea

  • Acarosporaceae is a crustose lichen and is known as a species that has more than 50 multispores, and has hyaline spores. Those taxa are often found in rock and soil in mountain areas or coastal regions in Korea, and very diverse forms and species are known. However, after an overall genetic phylogenetic analysis of carbonized ascomata in 2015, species consisting only of the morphological base are newly divided, and several species of Acarosporaceae in Korea are also being discovered in this situation. As a result of analysis using internal transcribed spacer (ITS) and nuLSU gene analysis, Korean species belonged to Acarospora and Sarcogyne clade, and Acarospora classified as the Acarospora clade was mixed with the Polysporina group and the Sarcogyne clade is mixed with the Acarospora. We identified two new species (Acarospora beangnokdamensis J. S. Park & S. O. Oh, sp. nov., Sarcogyne jejuensis J. S. Park & S. O. Oh, sp. nov.) through morphological, molecular, and secondary metabolite substance and found one new record (Sarcogyne oceanica K. Knudsen & Kocourk). We have made a classification key for Acarospora and Sarcogyne in Korea and reported all information together here.
