Anti-pigmentation Effects of Panax vietnamensis Extracts via Tyrosinase Expression (Tyrosinase 발현 조절을 통한 Panax vietnamensis 추출물의 Anti-pigmentation 효과)

  • In this study, the anti-pigmentation efficacy of Panax vietnamensis (P. vietnamensis), a ginseng native to Vietnam, was confirmed. Melanin synthesis was repressed by ethanolic extracts of P. vietnamensis in B16F10 cells, melanocytes originated from mouse. At 250 ㎍/mL ethanolic extracts of P. vietnamensis, melanin contents were repressed by 64.04% compared to the control group. In addition, ethanolic extracts of P. vietnamensis downregulated tyrosinase activity and expression to 53.34% and 59.39%, respectively. As shown our result, ethanolic extracts of P. vietnamensis blocks α-MSH-mediated melanogenesis and is valuable whitening ingredients in cosmetics.

  • Methanol extract of plants in Jeju were investigated for biological properties related to whitening cosmeceuticals such as melanin contents on melanoma cell, mushroom tyrosinase activity inhibition. We found that extracts of leaves of Hypochoeris radicata, Solanum nigrum, Solidago serotica Gynostenmma pentaphyllum and Taxus cuspidata inhibit melanin synthesis in B16F10 melanoma cells. However they have no tyrosinase inhibitory activity.

  • The aim of this study was to investigate the effect of ethanol extract of Fagopyrum escuentum(FE) on the melanogenesis. To determine whether ethanol extract of FE suppress melanin synthesis in cellular level, B16F10 melanoma cells were cultured in the presence of different concentrations of FE ethanol extract. In the present study, the author examined the effects of FE ethanol extract on cell proliferation, melanin contents, tyrosinase activity. Cell proliferation was slightly increased by treatment with ethanol extract of FE (25-200 ${\mu}$g/ml). The ethanol extract of FE effectively suppressed melanin contents at a dose of 100 ${\mu}$g/ml. It was observed that the color of cell pellets was totally whitened compared with the control. The ethanol extract of FE inhibited tyrosinase activity, regulate melanin biosynthesis as the key enzyme in melanogenesis. These results suggest that the ethanol extract of FE exerts its depigmenting effects through the suppression of tyrosinase activity. And it may be a potent depigmetation agent in hyperpigmentation condition.

Comparative Analysis of Melanin Contents in Hairs among Cattle Breeds (소의 모발 멜라닌 색소 함량의 품종별 비교 분석)

  • To characterize the colorization patterns of bovine hairs, the melanin contents were quantitatively assayed and compared among cattle breeds. The total melanin levels measured by spectrophotometric assay (A500) from Jeju Black cattle were significantly lower than those from Holstein or Angus with black coat color but significantly higher than those from Hanwoo with yellow coat color or Angus and Holstein with red coat color (P<0.001). The total melanin levels from Hanwoo were significantly lower than those from Red Angus and Red Holstein but significantly higher than those from Hanwoo×Charolais crossbred (P<0.001). The relative ratios of eumelanin to pheomelanin (A650/A500) were 0.382, 0.359, 0.112 and 0.124 in Angus, Jeju Balck cattle, Red Holstein and Hanwoo, respectively. These results show that the spectrophotometric method provides a convenient way to qualitatively characterize hair melanin and may be useful for studying expression of major coat color genes in Hanwoo and Jeju Black cattle.

Inhibitory Effect on Melanin Synthesis of Radix Codonopsis Lanceolatae (더덕 추출물의 멜라닌 생성 억제 효과)

  • The aim of this study was to investigate the effect of Codonopsis lanceolatae on the melanogenesis of HM3KO human melanoma cells biologically. The cells were treated for 3 days and 5 days with Codonopsis lanceolatae at several concentrations. The effects on tyrosinase activity and melanin contents were examined. And DOPAchrome tautomerase(TRP-2) activity was also examined to search a pathway influencing this inhibitory effect. 1. It was examined mushroom tyrosinase activity was significantly inhibited by Codonopsis lanceolatae. 2. Treatment with Codonopsis lanceolatae did not affect cell viability at the highest concentration, 1 ㎎/ml, and suppressed melanin contents as a time and dose dependent manner. 3. It was investigated treatment with Codonopsis lanceolatae inhibited tyrosinase, a key enzyme forming melanin, activity in a dose-dependent manner. 4. Treatment with Codonopsis lanceolatae did not affect DOPAchrome tautomerase(TRP-2) activity. 5. There was not a morphological change by Codonopsis lanceolatae microscopically. These results suggest that Codonopsis lanceolatae is a candidate for an efficient whitening agent which supresses melanogenesis by a Raper-Mason pathway.

Effects of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini Extracts on Melanin Synthesis in Melanoma Cells (애기수영, 방가지똥 및 암대극 추출물이 Melanoma Cell에서 멜라닌 합성에 미치는 영향)

  • In this study, we investigated the effect of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini extracts on tyrosinase activity and melanin production as natural products of whitening functional cosmetics. To measure the melanin production, 50, 100, $200{\mu}g/mL$ of Rumex axetosella, Sonchus oleraceus and Euphoibia jolkini extracts were treated on ${\alpha}-MSH$ treated B16F10 melanoma cells, respectively. Melanin contents in ${\alpha}-MSH$ treated B16F10 melanoma cells were decreased by 41.5, 51.11, and 61% in $200{\mu}g/mL$ treatment compared to none treatment, respectively. In addition, the intracellular tyrosinase activity was decreased after treatments with all extracts. Furthermore, $100{\mu}g/mL$ of Euphoibia jolkini extract was decreased 81.5% of melanin production in B16F10 melanoma cells. When the three extracts were compared, Euphoibia jolkini extract was considered to be the most functional material for whitening effect.

Inhibitory Effect of Dalbergioidin Isolated from the Trunk of Lespedeza cyrtobotrya on Melanin Biosynthesis

  • Tyrosinase is a key enzyme for melanin biosynthesis, and hyperpigmentation disorders are associated with abnormal accumulation of melanin pigments, which can be reduced by treatment with depigmenting agents. The methanol extract of Lespedeza cyrtobotrya $M_{IQ}$ showed inhibitory activity against mushroom tyrosinase. The active compound was purified from the methanol extract of L. cyrtobotrya, followed by several chromatographic methods, and identified as dalbergioidin (DBG) by spectroscopic methods. The results showed that DBG exhibited tyrosinase inhibitory activity with an $IC_{50}$ of $20\;{\mu}M$. The kinetic analysis of tyrosinase inhibition revealed that DBG acted as a noncompetitive inhibitor. In addition, DBG showed a melanin biosynthesis inhibition zone in the culture plate of Streptomyces bikiniensis that has commonly been used as an indicator organism. Furthermore, $27\;{\mu}M$ DBG decreased more than 50% of melanin contents on the pigmentation using the immortalized mouse melanocyte, melan-a cell.

A Study of Melanin Synthesis Pathway of the Ethanol extract of Eclipta prostrata In Vitro Study (한련초 에탄올 추출물의 멜라닌 합성 경로에 대한 연구)

  • Objectives : This Study was conducted to investigate the melanin synthesis effect of Eclipta prostrata and to determine the relationship between melanin synthesis effect of Eclipta prostrata and cAMP/PKA melanin synthesis pathway. Methods : We measured melanin contents, tyrosinase activity, expression of TRPs, p-CREB in B16F10 cells cultured with Eclipta prostrata ethanol extracts(EEP). And after treatment with H89 and dibutyryl cAMP, which inhibit or promote the activation of PKA, we observed changes in melanin synthesis and tyrosinase activity stimulated by EEP. Results : EEP increased melanin synthesis by promoting the expression of tyrosinase and TRP-1. It also promoted expression of p-CREB. H89 suppresses melanogenic effect and expression of tyrosinase, TRP-1 in B16F10 stimulated by EEP. Dibutyryl cAMP promotes melanogenic effect of EEP. Conclusions : The results of this study suggest that melanin synthesis effect of EEP is related to induction of tyrosinase and TRP-1 expression through the cAMP/PKA pathway.

Effect of Glycyrrhizae Radix Water Extract on the Melanogenesis of Human Melanoma Cell (감초수추출물이 HM3KO 세포의 멜라닌 생성에 미치는 영향)

  • This study was conducted to evaluate the effects of Glycyrrhizae Radix water extract, known as depigmenting agent, on melanin biosynthesis in the HM3KO human melanoma cells. The inhibitory effect of Glycyrrhizae Radix water extract on melanogenesis was identified by mushroom tyrosinase assay in vitro. To determine whether Glycyrrhizae Radix water extract suppress melanin synthesis in cellular level, HM3KO cells were cultured in the presence of different concentrations of Glycyrrhizae Radix water extract and the effects on cell proliferation, melanin contents and tyrosinase activity were examined after 3 days. Treatment with Glycyrrhizae Radix at various concentrations did not exhibit any change of cell viability, and increased the cell proliferation. And the water extract of Glycyrrhizae Radix inhibited melanin contents and tyrosinase activity in a dose-dependent manner, compared with untreated group. These results suggest that the inhibitory effect of Glycyrrhizae Radix water extract on melanogenesis is due to the suppression of tyrosinase in HM3KO cells.
