This study was conducted to investigate the bacterial capability to accumulate phosphorus during liquid composting process of pig slurry. Storage liquid compost and pig slurry were analyzed by using MALDI-TOF technique, which showed the colonies of Acinetobacter towneri and Bacillus licheniformis. In addition, bacterial colonies were isolated under high phosphoric acid conditions using X-phosphate MOPS medium with the addition of 2 mM $K_2HPO_4$. Microbial growth was observed in high and low phosphoric conditions due to the growth of bacterial diversities in the liquid fertilizer and slurry. The colonies isolated in the high phosphoric acid medium were uncultured bacterium clone and Acinetobacter sp. were identified by analysis of 16S rRNA gene sequences. Uncultured bacterium showed higher growth rate and excellent phosphorus ability then Acinetobacter sp.. In addition to Paenibacillus sp. AEY-1 isolated from pig slurry performed excellent phosphorus utilizing capability.
An experiment of close contact melting of phase-change medium partially filled in an isothermally heated horizontal cylinder is performed which involves the volume expansion of liquid induced by the solid-liquid density difference. The solid-liquid interface motion and the free surface behavior of liquid were reported photographically. The experimental results show that the curvature of upper solid-liquid interface varied to flat as melting progresses. In addition to the varying interface shape, the melting rate increases with the lower initial height of solid and the free surface height of liquid increases linearly. The experimental results of molten mass fraction were expressed in a function of dimensionless time Fo.Ste$^{3}$4/ and agreed well with the analytical solutions.
Pathogenic Naegleria fowleri, Acanthamoeba castellanii, and Acanthamoeba polyphaga, are distributed worldwide. They are causative agents of primary amoebic meningoencephalitis or acanthamoebic keratitis in humans, respectively. Trophozoites encyst in unfavorable environments, such as exhausted food supply and desiccation. Until recently, the method of N. fowleri encystation used solid non-nutrient agar medium supplemented with heat-inactivated Escherichia coli; however, for the amoebic encystment of Acanthamoeba spp., a defined, slightly modified liquid media is used. In this study, in order to generate pure N. fowleri cysts, a liquid encystment medium (buffer 1) modified from Page's amoeba saline was applied for encystation of N. fowleri. N. fowleri cysts were well induced after 24 hr with the above defined liquid encystment medium (buffer 1). This was confirmed by observation of a high expression of differential mRNA of nfa1 and actin genes in trophozoites. Thus, this liquid medium can replace the earlier non-nutrient agar medium for obtaining pure N. fowleri cysts. In addition, for cyst formation of Acanthamoeba spp., buffer 2 (adjusted to pH 9.0) was the more efficient medium. To summarize, these liquid encystment media may be useful for further studies which require axenic and pure amoebic cysts.
In order to enhance shoot elongation and rooting of Anthurium andreanum 'Atlanta' in vitro, 15mL of liquid media containing various concentrations of activated charcoal, sucrose and MS salts were added in same vessels after small shoots were induced from the calli on mudium supplemented with 10.0mg/L BA and 0.1mg/L 2.4-D. The post-supplying of 15mL liquid medium containing MS macro and micro elements, 30g/L sucrose and 5.0∼10.0g/L activated charcoal was significantly stimulated the shoot elongation and rooting of regenerated shoots from calli. The medium addition was also resulted in the enhanced soil survival, elongation and rooting of plantlets in cultural soil mixed with perlite and vermiculite(1 : 1)
Conditions for isolation of protoplasts from conidiospores of Trichoderma koningii ATCC 26113 were tested. Maximum production of conidial protoplasts was obtained by preincubation of conidiospores on liquid minimal medium for 8 1/2 hrs. and by reaction with cell wall lytic enzyme for 3 hrs. Among effective cell wall lytic enzymes (Driselase, p-Glucuronidase, Novozyme and Zymolyase), Driselase was the most effective one on the production of conidial protoplasts. The production of conidial protoplasts was also enhanced by addition of 2-Deoxy-D-Glucose $(25{\mu}g/ml)$ into liquid minimal medium. Over 70% of the initial swollen conidia, preincubated in liquid minimal medium supplemented with 2-Deoxy-D-Glucose $(25{\mu}g/ml)$, were converted to protoplasts by incubation with 2% (w/v) commercial lytic enzyme Driselase at $28^{\circ}C$ for 3 hrs. The reversion frequency of the conidial protoplasts was about 30 times (25-50%) higher than that of mycelial protoplasts (0.6-1.3%).

In addition to these parameters, a variation of fluid properties such as phase density, phase viscosity due to a change of system pressure can be used for gaining insight into the nature of two-phase flow behavior up to dryout.
본 연구는 유채 꽃가루 분말의 영양성분 분석과 파쇄한 꽃가루 분말을 이용하여 효모 배양용 액체배지와 고체배지를 제조하여 효모 배양효율을 분석하였다. 유채 꽃가루 분말(100 g)은 탄수화물 58.9 g, 단백질 20.8 g, 지방 4.1 g, 회분 2.5 g 및 수분 13.7 g으로 구성되었다. 당질 중에서는 Fructose(13.7 g), Glucose(11.1 g)와 Sucrose(6.6 g), 미네랄 중에서는 K(606.7 mg)와 P(603.3 mg) 함향이 높았다. 유리 아미노산은 Glutamic acid(2,482.4 mg), Aspartic acid(2,136.5 mg), Lysine(1,648.3 mg), 및 Leucine(1,631.1 mg) 순으로 많았다. 유채 꽃가루 분말을 단독 또는 NaCl을 첨가하여 액체배지 및 고체배지 제조하고 효모를 배양한 결과, 꽃가루 분말이 15 g/L 이상 함유된 액체배지는 효모 배양 후 건물중이 YPD 배지(대조구)에 비해 많았고, 꽃가루 분말의 농도 의존적으로 효모 건물중이 증가하였다. 또한 액체배지에 NaCl을 1~20 g/L 추가 시 배양된 효모의 건물중이 한층 더 증가하였다. 유채 꽃가루로 제조한 고체배지를 이용하여 효모를 배양(48시간)한 결과, 콜로니(colonies) YPD 배지와 유사하게 잘 자랐다.

퇴비 숙성초기에 고온성 cellulose 분해이용균으로 분리된 Herpetosiphon geysericola CUM 317균주는 전분분해 효소인 ${\alpha}-amylase,\;{\beta}-amylase$ 및 glucoamylase를 모두 생성한다. 이 균을 사용하여 그 배양조건을 달리하여 각 amylase의 생성관계를 서로 비교한바 50℃의 밀기울 고체배지나 $40^{\circ}C$의 액체배지상에서 ${\beta}-amylase$는 배양초기 10시간만에 최대의 생성력가를 보였는 반면, ${\alpha}-amylase$와 glucoamylase는 30 내지 40시간 정도의 배양말기에 최대를 이루었다. Polypeptone을 함유한 액체배지에 탄소원의 첨가나 무기질소원의 첨가는 전반적으로 amylase들의 생성이 크게 저하되었으나 cellulose에 의해서 glucoamylase의 경우 150% 정도 증가되었다. 액체배지에 $CuSO_4$를 첨가해 줌으로서 ${\alpha}-amylase$만의 생성증가 효과를 얻었고 $CdSO_4$에 의하여 ${\beta}-amylase$만의 생성증가가 있었으며, 그리고 $CaCl_2$에 의하여 glucoamylase만의 증가효과가 있은 반면, 상대적으로 ${\beta}-amylase$의 급격한 감소가 일어났다. 이들 amylase들의 최적 효소생성 pH는 7.5였으며, 최적온도는 ${\alpha}-amylase$와 glucoamylase의 경우 $40^{\circ}C$였고 ${\beta}-amylase$는 $30^{\circ}C$였다.
Lilium asiatic hybrid 'Hae Hwa'의 인편 및 자구절편 배양으로 신초 cluster를 유도하여 자구를 대량증식하고, 형성된 신초 cluster에서 정상적인 자구를 대량생산하고자 일련의 실험을 실시하였다. MS배지에 BA 1.0mg/L와 IAA 0.5 mg/L가 첨가된 배지에서 신초 cluster의 유도는 인편보다 자구절편을 배양하는 것이 효과적이었다. MS배지에 BA 5.0 mg/L와 IAA 0.5 mg/L 첨가배지에서 신초수 및 신초무게가 양호하여 형성된 신초 cluster 절편에서 신초 cluster의 증식에 적합하였다. 또한 자구의 비대는 MS 배지에 sucrose 60∼90 g/L가 첨가된 배지에서 양호하였다. MS배지에 sucrose 30∼90 g/L를 주입한 5L airlift 생물반응기에 신초 cluster 절편체를 배양한 결과, 신초만 무성하게 자랐고 자구비대는 불량하였다. 신초 cluster를 8주간 배양한 후에 2배 MS 염류와 sucrose 250g/L, 활성탄 5 g/L를 포함한 액체배지를 동일용기에 첨가하여 배양한 결과, 명배양에서 형성된 자구수는 증가하였으나 소자구의 생장은 명, 암 모두 비슷하였다. 이로써 형성된 소자구의 무게가 53∼68 mg이며, BA 5.0 mg/L와 IAA 0.5 mg/L가 첨가된 MS 배지에서 신초 cluster를 증식한 후, 액체배지 첨가방법에 의하여 효율적인 소자구의 생산이 이루어졌다.
