Morphogenetic Behavior of Tropical Marine Yeast Yarrowia lipolytica in Response to Hydrophobic Substrates

  • The morphogenetic behavior of a tropical marine Yarrowia lipolytica strain on hydrophobic substrates was studied. Media containing coconut oil or palm kernel oil (rich in lauric and myristic acids) prepared in distilled water or seawater at a neutral pH supported 95% of the cells to undergo a transition from the yeast form to the mycelium form. With potassium laurate, 51 % of the cells were in the mycelium form, whereas with myristate, 32% were in the mycelium form. However, combinations of these two fatty acids in proportions that are present in coconut oil or palm kernel oil enhanced the mycelium formation to 65%. The culture also produced extracellular lipases during the morphogenetic change. The yeast cells were found to attach to the large droplets of the hydrophobic substrates during the transition, while the mycelia were associated with the aqueous phase. The alkane-grown yeast partitioned more efficiently in the hydrophobic phases when compared with the coconut oil-grown mycelia. A fatty acid analysis of the mycelial form revealed the presence of lauric acid in addition to the long-chain saturated and unsaturated fatty acids observed in the yeast form. The mycelia underwent a rapid transition to the yeast form with n-dodecane, a medium-chain aliphatic hydrocarbon. Thus, the fungus displayed a differential behavior towards the two types of saturated hydrophobic substrates.

Fatty Acid Composition of Domestic Seed Lipids (한국산 종자 지방질의 지방산 조성)

  • The seeds of 16 domestic plants were studied for their moisture, total lipids and fatty acid composition. Of the 16 seeds, chestnut, corn, mungbean and ginko nut yielded less than 9% by weight of total lipids compared to others that gave 20-73%. The identified fatty acids from the seed lipids ranged from lauric acid (12:0) to lignoceric acid(24:0). It was intended in this study to classify the seed lipids according to their major fatty acids: Group t-Oleic acid; Group 2-0leic acid and linoleic acid; Group 3-linoleic acid; Group 4-linolenic acid ; Group 5-erucic acid ; Group 6-ricinoleic acid. The saturated fatty acid content of mungbean (33%) was the highest among the seed lipids studied. The highest value for the P/S fatty acid ratio(10) was in perilla.

Fatty Acid Composition of Total Lipids from Needles and Pollen of Korean Pinus densiflora and Pinus koraiensis (소나무 및 잣나무의 잎과 꽃가루의 지방산 조성)

  • The needles and pollen of Pinus densiflora and Pinus koraiensis were studied for their lipid contents and fatty acid composition. The total lipid contents in needles of Pinus densiflora and Pinus koraiensis were 5.0 and 4.5%, whereas in pollen of Pinus densiflora and Pinus koraiensis 3.5 and 5.6%, respectively. Twenty-four fatty acids ranged from lauric acid to docosahexaenoic acid(22:6${\omega}$3) were identified in the needle lipids. In needles, linolenic acid and palmitic acid were the major fatty acids. The needles of Pinus densiflora showed higher proportions of docosahexaenoic acid and 5-olefinic nonmethylene-interrupted polyenoic acids than those in the Pinus koraiensis. Twenty fatty acids ranged from myristic acid to lignoceric acid were identified in the pollen lipids. Linoleic acid was the major fatty acid in the pollen followed by oleic and palmitic acid. The fatty acid profile of pollen of Pinus densiflora was similar to those of the Pinus koraiensis pollen lipids.

  • Effects of dark treatment and N1-dichlorophenyl-N3-dimethylurea (DCMU) on the desaturation of galactolipids of Dunaliella salina were investigated to see whether light-driven photosynthetic electron transport is involved in in vivo desaturation of galactolipids. The incorporation of radioactive fatty acid precursors ([14C]lauric acid) into galactolipids, mainly composed of prokaryotic molecular species, was most affected among different polar lipid classes by both treatments. The analysis of specific radioactivities of individual galactolipid molecular species revealed that their synthesis was greatly inhibited by the treatments except for eukaryotic molecular species, 18 : 3/ 18 : 3 digalactosyldiacylglycerol, whose desaturation occurs in endoplasmic reticulum.

Study on fatty acids composition by latent fingerprint deposition (유류된 잠재지문의 지방산조성에 관한 연구)

  • In order to investigate the information for effective detection and developing of latent fingerprints, we identified fatty acids composition of latent fingerprints on non-porous evidence surface and the chemical changes of latent fingerprint residue after print deposition during 7 months. Fingerprints from eight Korean male donors (aged 29-50 years) and one female donor (aged 36 years) were collected. All fingerprints were found to contain lauric acid (C12:0), myristic acid (C14:0), palmitic acid (C16:0), stearic acid (C18:0), elaidic acid (C18:1n9t), oleic acid (C18:1n9c), linoleic acid (C18:2n6c), arachidic acid (C20:0), linolenic acid (C18:3n3), erucic acid (C22:1n9) and docosadienoic acid (C22:2) and primarily palmitic acid (35.45-48.37%), oleic acid (14.84-28.49%), stearic acid (9.71-24.96%) and linoleic acid (7.68-18.8%) occupied 75% of total fatty acids. When the fingerprints were deposited at dark room for 7 months, total fatty acids components decreased about 12-25%. It can be explained that significant degradation of long-chain fatty acids such as elaidic acid (C18:1n9t), arachidic acid (C20:0), linolenic acid (C18:3n3), erucic acid (C22:1n9), and docosadienoic acid (C22:2) resulted in the generation of myristic acid (C14:0), myristoleic acid (C14:1) and pentadecanoic acid (C15:0).

  • Four fatty acid vinyl esters were synthesized by transesterification between vinyl acetate and lauric acid, myristic acid, palmitic acid, stearic acid, respectively. Fatty acid vinyl ester oligomers were prepared from polymerization of four fatty acid vinyl esters in the presence of potassium persulfate in methanol. The ${\alpha}-sulfonation$ of these four fatty acid vinyl ester oligomer were carried by direct addition of sulfur trioxide. Especially, molecular weights of sodium ${\alpha}-sulfo$ fatty acid vinyl ester oligomers were measured by boiling point method.

  • Allyl aliphatic carboxylates were synthesized by azotropic reaction with benzene between allyl alcohol and capric acid, lauric acid, myristic acid, palmitic acid or stearic acid respectively. allyl aliphatic carboxylates oligomers were prepared from polymerization giving allyl aliphatic carvboxylates in the presence of potassium persulfate in methanol, and the ${\alpha}-sulfonation$ of these five allyl aliphatic carboxylates oligomers were carried by direct addition of dry sulfur trioxide.   • To utilize deteriorated sweet persimmon(Diospyros kaki T.) effectively, this study was investigated about the effective (Dying method of the powder. Chemical components, minerals, fatty acids and amino acids of hot-air dried and freeze dried conditions were determined Sample conditions used on analysis as follow; pretreatments of hot-air dry and freeze dry were soft, soft+peel, mixer, mixer+peel. The contents of crude protein, crude lipid, crude ash of hot-air dry were 0.9-1.1%,15.0-39.0% and 2.3-3.3%, respectively. And those of freeze dry were 1.3-2.2%, 27.-49.0% and 2.5%, respectively. Potassium, magnesium, phosphorus, calcium and sodium content in hot-air dry and freeze dry were high. Other minerals were less than 3.00ppm in all conditions. The major fatty acid contents were detected capric acid(C$\^$10:0/), lauric acid(C$\_$l2:0)/, tridecanoic acid(C$\_$13:00/), palmitic acid(C$\_$l6:00/), palmitoleic acid(C$\_$l6:1/), oleic acid(C$\_$18:1), linoleic acid(C$\_$18:2/), linolenic acid(C$\_$18:3/). The essential amino acids such as aspartic acid, threonine, serine, glutamic acid, glycine, alanine, valine, methirmine, isoleucine, leucine, tryosine, phenylalanine, histidine, lysine, arginine in freeze dry were contained richly. On the basis of chemical analysis, hot-air dry method will have to supply the additional different method, because simple hot-air dry method was shorten the dry time but had many problems. Therefore, the effective drying method considering changed color and nutrition was shown freeze dry method.

  • A series of four sodium ${\alpha}-sulfo$ fatty acid allylester oligomers such as sodium ${\alpha}-sulfo$ lauric acid allylester oligomer, sodium ${\alpha}-sulfo$ myristic acid allyl ester oligomer, sodium ${\alpha}-sulfo$ palmitic acid allyl ester oligomer and sodum ${\alpha}-sulfo$ stearic acid allyl ester oligomer were examined for surface tension, defloculation effect and emulsifying power. Also critical micelle concentration (cmc) was evaluated. Consequently, these sodium allyl ${\alpha}-sulfo$ aliphatic carboxylate aligomers show o/w type emulsifying agent and dispersion effect in 1g/100ml soulution.

Increase of Conjugated Linoleic Acid (CLA) Contents in Milk by Fermentation with Bifidobacteria Isolated from Korean Infants (한국 유아에서 분리한 Bifidobacteria에 의한 발효유의 Conjugated Linoleic Acid (CLA) 함량 증가)

  • More than 200 Bifidobacterium sp. originated from human intestine were investigated for their ability to produce conjugated linoleic acid (CLA). Of the Bifidobacteria tested, 1 of culture type strain and 12 isolated strains from Korean infants showed CLA producing ability. cis-9, trans-11 octadecadienoic acid presented more than 90% of the total CLA isomers produced by the Bifidobacteria. CLA content in fermented milk by Bifidobacterium sp. KHU 141 increased by 39.6 mg/l00 g, which showed the potential use for producing fermented milk containing high content of CLA. In fermented milk, little changes showed in lauric acid, myristric acid, palmitic acid, oleic acid, and linolenic acid contents, whereas the content of linoleic acid (LA) decreased and the content of CLA increased. Bifidobacterium sp. KHU 141 converted 86.0% and 84.8% of LA consumed to CLA for 24 hr and 48 hr fermentation, respectively. Prolonging incubation from 24 to 48 hours did not appear to enhance CLA formation and CLA producing ability was stable whether bottle, test tube, or fermenter was used for making fermented milk by Bifidobacterium sp. KHU 141.
