Protective Activity Against Oxidative Stress of Plants Indigenous to Korea

  • We have screened the cytoprotective effect against $H_2O_2$ and $\gamma-ray$ radiation induced oxidative stress from 32 Korean plants. Betula ermani var.saitoana (caulis, leaves), Rosa wichuraiana (caulis), Sorbus commixta (caulis), Weigela florida (leaves), Cirsium rhinoceros (whole plant), and Viburnum erosum (caulis) were found to scavenge 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). As a result, extracts of six plants reduced cell death of Chinese hamster lung fibroblast (V79-4) cells induced by $H_2O_2$ treatment. In addition, these extracts protected cell death of V79-4 cells damaged by $\gamma-ray$ radiation. In addition, these extracts scavenged ROS generated by radiation. Taken together, the results suggest that Betula ermani var. saitoana, Rosa wichuraiana, Sorbus commixta, Weigela florida, Cirsium rhinoceros, and Vibumum erosum protect V79-4 cells against oxidative damage by radiation through scavenging ROS.

  • In this study, Tongue fern (Pyrrosia lingua) plants that have been used traditionally as medicines. Their traditional medicinal uses, regions where indigenous people use the plants, parts of the plants used as medicines. This study was designed to assess the antioxidant and inhibition activities of extracts from P. lingua. In the P. lingua extracts was measured ethanol activity, 80.0% ethanol was high activity. The antioxidant activity was measured in 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-Azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), assays. DPPH and ABTS radical in this experiment, solid and phenolic of extract were tested, but only an average concentration of 100 ㎍/mL was used. However, the phenolic extract is shown phenolic activity reached a peak. Also, phenolic extracts ware reached peak water and ethanol extracts. As a result, using the phenolic extracts did other antioxidant assays such as DPPH, ABTS, protection factor, and thiobarbituric acid reactive substances at 50-200 ㎍/mL concentrations. The activity of elastase and collagenase, inhibiting their activities may retard skin aging. α-Glucosidase and α-amylase, inhibitors need to be explored for the benefit of postprandial hyperglycemia in diabetic patients. Activities of tyrosinase, hyaluronidase and xanthine oxidase inhibitors of these enzymes are increasingly important ingredients in cosmetics and medications to protect the skin against hyperpigmentation and skin aging. Inhibition effects were investigated using the P. lingua extracts at 50-200 ㎍/mL concentrations. The expression levels of enzyme inhibitions activities were decrease in dependent-concentrations manner when P. lingua extracts were treated.

A Study on the Screening of 2, 4, 6-trinitntoluene Tolerant Indigenous Herbaceous Piano (2, 4, 6-trinitrotoluene에 대해 내성을 지닌 토착 식물종 선정에 대한 연구)

  • To select a suitable indigenous plant for the phytoremediation of TNT contaminated soil, eight representative species of native grasses were tested to identify TNT toxicity thresholds. The threshold was determined based on various factors including cumulative seed germination, root and shoot length, fresh biomass, and the amount of water uptake under various TNT concentrations. Phytotoxic effect of TNT on plants was increased with the increase in TNT concentration but the degree was varied between grass species. Concentrations up to 60-80mgTNT/liter did not affect germination of Abutilion avicennae, Echinochioa crusgalli var. frumentacea, and Aeschynomene indica. Phytotoxicity threshold inhibition (50%) of Abutilion avicennae, schinochioa crusgalli var. frumentacea, Aeschynomene indica were 5-40mgTNT/liter for root length, 50-73mg TNT/Liter for shoot length and 68-99mgTNT/Liter for fresh biomass during 14 days of seedling exposure. Root and shoot growth as well as fresh biomass decreased as TNT concentration increased. In addition, the amount of water uptake decreased with increasing TNT concentration in Abutilion avicennae and Aeschynomene indica. Comparison of toxicity thresholds for the tested grass species showed that sensitivity of plants to TNT was in the order of root length > shoot length > fresh biomass > germination rate. From these results, we concluded that Abutilion avicennae and Aeschynomene indica had tolerance to TNT among the species tested.

Plant Growth-promoting Ability by the Newly Isolated Bacterium Bacillus aerius MH1RS1 from Indigenous Plant in Sand Dune (해안 사구에서 서식하는 토착식물로부터 분리된 근권미생물 Bacillus aerius MH1RS1의 식물성장 촉진 능력 연구)

  • Coastal sand dunes have been seriously damaged caused by the development thoughtless for the environment and coastal erosion and destruction due to artificial structures like coast roads and breakwater. Hereupon, in this study we made a library of rhizobacteria that have the plant growth-promoting ability for plant rhizosphere of indigenous plants inhabiting in a coastal sand dune as well as the strong tolerance to salt, and evaluated the plant growth-promoting ability of these strains. Furthermore, we evaluated the effect of rhizobacteria on the growth rate of saline tolerant plants in sandy soil; selected out the most useful micro-organism for the restoration of a damaged sand dune. The effect of inoculation of strains selected from the first experiment on the growth of Peucedanum japonicum and Arundo donaxes planted in a coastal sand dune was evaluated. As a result, Bacillus aerius MH1RS1 had plant growth promoting activities: indole acetic acid (IAA) production, siderophores and 1-aminocyclopropane-1-carboxylate deaminase (ACC deaminase) activity, and also had a salinity tolerance. Also, in case of Peucedanum japonicum, the length of stems and weights of roots were enhanced by the inoculation of B. aerius MH1RS1. Fresh weights of stems and weights of roots in experimental group were, in particular, increased by 25% comparing with the control group. For an Arundo donax in experimental group, plant length increased by 18%, and weight of roots by 20% which is significant.

Plant Species Richness in Korea Utilizing Integrated Biological Survey Data (생물기초조사 통합자료를 활용한 우리나라 식물종 풍부도 분석)

  • The limitation in deriving the species richness representing the entire country of South Korea lies in its relatively short history of species field observations and the scattered observation data, which has been collected by various organizations in different fields. In this study, a comprehensive compilation of the observation data for plants held by agencies under the Ministry of Environment was conducted, enabling the construction of a time series dataset spanning over 100 years. The data integration was carried out using minimal criteria such as species name, observed location, and time (year) followed by data verification and correction processes. Based on the integrated plant species data, the comprehensive collection of plant species in South Korea has occurred predominantly since 2000, and the number of plant species explored through these surveys appears to be converging recently. The collection of species survey data necessary for deriving national-level biodiversity information has recently begun to meet the necessary conditions. Applying the Chao 2 method, the species richness of indigenous plants estimated at 3,182.6 for the 70-year period since 1951. A minimum cumulative period of 7 years is required for this estimation. This plant species richness from this study can be a baseline to study future changes in species richness in South Korea. Moreover, the integrated data with the estimation method for species richness used in this study appears to be applicable to derive regional biodiversity indices such as for local government units as well.

The Effects of the Biodiversity Increase after Creation of the Artificial Wetland -The Case of Ecological Pond at Seoul Technical High School- (인공습지 조성후 생물다양성 증진 효과에 관한 연구 -서울공고 생태연못을 중심으로-)

  • The purpose of this study is to evaluate the creation techniques of artificial wetland, one of biotopes developed to promote biodiversity in urban areas, and to look for improvement steps. Specifically, artificial wetland creation techniques were categorized into living environment and living creature classification. Being living conditions for creations, habitat environment was reviewed with a focus on water and soil environments. Living creatures were classified into plants, insects, fish, and birds. The evaluation of creation techniques was done in post-construction evaluation while considering the creation of habitats for living creatures. Intervention by users, changes in living environment and living species, and relevance of creation techniques were reviewed. Key results of this study are as follows. (1) Water environment for the living environment of creatures provides a suitable environment conditions for the living of creatures through a process easing the use of piped water. Various water depths and embankment appear to have a positive impact on the living of aquatic life. In particular, embankment covered in soil naturally played an important role as a place for the activities of aquatic insects and young fish as well as the growth of aquatic plants. (2) Various aquatic and ground plants to promote insect-diversity, shallow water, and old-tree logs had contributed greatly in increasing the types and number of insects. Aquatic insects. Aquatic insects were seen much particularly in areas where aquatic plants are rich but water is shallow than any other areas. (3) A space piled with stone to provide habitats for fish was not much used. However, it was observed that fish used embankment built with natural stones and embankment using logs in areas where water is deep. In addition, it was confirmed that 1,500 fish that had been released propagated using various depths and places for birth. (4) It was analyzed that techniques (creation of island, log setting, and creation of man-made bird nests) to provide habitats and to attract birds are not serving their roles. In such a case, it is believed that species had not increased due to the smallness as well as isolated features of the area. Based on theoretical review, they are judged to be areas that are likely to be used when a greater variety of birds is introduced. It is judged that attracting and keeping more birds at the site, such spaces need to be linked systematically in the future in terms of building eco-network while ensuring an adequate living areas. (5) In the study areas, users intervened greatly. As a result, a blockage was created preventing the normal growth of plants and non-indigenous plants were introduced. In order to limit the intervention by users, setting enough buffer zones, and environment education programs were urgently required. D/H=1>Hyangkyo> houses on the river>temples>lecture halls. Protective Activity against Ionizing Radiation of Antioxidative Plants Indigenous to Korea

Carpinus laxiflora (caulis), Quercus salicina (caulis), and Castanopsis cuspidata (caulis) were found to scavenge 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical and intracellular reactive oxygen species (ROS). As a result, extracts of three plants reduced cell death of Chinese hamster lung fibroblast (V79-4) cells induced by $H_2O_2$ treatment. Studies on the Crude Drugs of Atractylodis Species (출류(朮類) 대조 생약에 관한 연구)

  • Atrartylodis Rhizoma(蒼朮)'s origin plants are Atratylodes lancea and A. Chinensis in Chinese, Japanese and Korean pharmacopoeia. A. Japonica is only indigenous in Korea, it is actually used as Atractylodis Rhizoma in Korean market. A. lancea is used in Hunan province, China and A. Chinensis is used in Hubei province, China. It is impossible to distinguish with species differency as macro- and micro-morphology. We tried to distinguish with species differency by HPLC and GC-Mass spectra. Atractylone(mw. 216) which is a marker compound in Atractylodis Rhizoma Alba(白朮) was detected in A. japonica. Atractylodin (mw.182) was detected in A. lancea and two eudesmadien derivatives (mw. 204) were detected in A. chinensis. HPLC chromatogram showed the same patterns. As a result, we propose that A. japonica will be added as Atractylodis Rhizoma (蒼朮)'s origin plant in Korean Pharmacopoeia. Atractylodis Rhizoma Alba(白朮)'s origin plants are A. macrocephala in China, and A. Japonica and A. ovata in Korea and Japan. In GC-Mass analysis, all samples showed same patterns and the main compound was atractylone.

An analysis of natural products patents (천연물 신약 특허 동향 분석)

  • Objectives : This study aims to investigate the current situation of domestic patents in Korea on natural products and to provide directions for developing and applying herbal medicinal products by in-depth analyses. Methods : Combinations of words, such as "A61K" or "A61P", "herb" or "Korean medicinal herb" or "Korean medicine" or "traditional" or "natural" or "$botanic^*$" or "China", and "medicine" or "treatment" or "prevention" or "improvement", were used to search patents in the World Intellectual Property Service (WIPS) database. Three methods were applied to analyze natural products patents. First, the number of patent registrations was sorted by year. Second, the assignees were analyzed by count and country of origin. Finally, cluster map analysis was conducted to explore frequently emerging words in natural products and the relationship amongst those words to treat corresponding diseases/technologies. Results : Results showed that, first, the total number of patents had been increasing. Among the patents, 76.4% were registered by domestic institutes/companies, and 23.6% by foreign institutes/companies. Second, USA, Japan, and China possess a considerable number of Korean patents and, therefore, domestic institutes/companies can seek joint technological development opportunities with their counterparts from those countries in the future. Finally, a total of four clusters were identified by cluster map analysis. Each of the clusters includes natural products related to diseases involving skin, aging, and blood sugar, as well as adult diseases. Conclusions : In this study, natural products patents registered in the Korean Intellectual Property Office were analyzed. The analyses results showed the kinds of natural products that had been employed for the treatment of certain diseases. However, natural products included in existing patents were minimal given that 4,174 species of indigenous plants are found in Korea. Thus, Korean institutes/companies should utilize unexplored plants to develop more value-added drugs.

Characteristics and Management of Complementary Pond of Village - in Case of Dalmaji Village in Gangjin-gun - (마을의 비보연못 특성 및 관리 - 강진 달마지 마을을 사례로 -)

  • The purpose of this study is on identifying charactreistics and management direction of complementary pond of Dalmaji village in Gangjin. Accordingly, this village was indicated as Experience and Resort village by Korean government. This study describes the construction background and characteristics and identifies species diversity and characteristics through field survey. For this, literature and field survey was performed. In field survey, vegetational survey was performed in Aug. of 2015. D(Dominant degree) and S(Sociability degree) was measured by Brown-Blanquet's method. The answer was searched from Myungsanron(明山論), a textbook of Fengsui(風水) in Joseon dynasty. Of these causes, it was considered that the representative one is a protection of fire and infectious disease because the Wolgak mountain is a fire mountain(火山) as a stone mountain in Fengsui. The results demonstrate decline of aquatic ecosystem because of neglect management. The total number of species was 24. Of the species, Zelkova serrata is a natural vegetation of this complementary forest. This is nominated as Lagerstroemia indica and Camellia japonica community. Species composition demonstrate the characteristics of Korean southern region as mixed forest of evergreen trees and deciduous ones which are indigenous plants. It was considered Juniperus chinensis KaizukaVariega in central round island was planted in Japanese colonial period and it have to be removed. It was considered that name of moon pond and moon island etc. projects the mind of village ancestors which are going to reach an ideal condition of moon. And management of quantity and depth of pond water was critical for restoration of disappearing lotus from related literature. Periodic management was needed for ecological structure and function improvement because of ecological disturbance by the invasive plants like Humulus japonicus.
