CHANGE OF THE INVASIVENESS WITH SELECTIVE COX-2 INHIBITION IN AN ORAL SQUAMOUS CELL CARCINOMA CELL LINE, KB ; PRELIMINARY IN VITRO STUDY (선택적 COX-2 저해를 통한 구강암세포주 KB의 침습성 변화에 관한 예비연구)

  • Cyclooxygenase-2 (Cox-2) is known as one of the critical factor in carcinomas of various organs. However, the importance of Cox-2 in oral squamous cell carcinoma has not been fully described yet. The purpose of this study is to evaluate the anti-cancer effect of selective cox-2 inhibitor, celecoxib in an oral squamous cell carcinoma cell line, KB with respect to cytotoxicity test, in vitro invasion and MMP-2 expression. In cytotoxicity test, celecoxib treated group showed definitely concentration dependent cytotoxicity. In addition, administration of celecoxib reduced the invasive potential of KB cell line significantly in invasion assay. However, there was no remarkable difference of the MMP-2 expression between the celecoxib treated group and the control group. Considering these data, celecoxib had a potential cytotoxic agent to oral squamous cell carcinoma cells. Also, it had anti-invasive property without acting on the MMP-2 expression mechanism. Therefore, it was postulated that celecoxib had the possibility of anti-cancer agent in treatment strategies of oral squamous cell carcinoma.

In vitro Screening of Natural Product Inhibitors on the UVB Phototoxicity (In vitro 실험법에 의한 천연물 중의 UVB 광독성 억제제 검색)

  • The phototoxicity inhibitory activity of 15 natural products having antiinflammatory effect was screened by three in vitro methods : yeast growth inhibition test with Candida albicans, RBC photohemolysis and MTT assay. We induced phototoxic reaction by irradiating UVB (312 nm) on chlorpromazine (CPZ) that has been widely documented as phototoxic agent in clinical and experimental studies and then observed the effects of the natural products after treating them with CPZ. In yeast growth Inhibition test, P. persica and E. officinalis showed the inhibitory effect on the UVB phototoxicity and E. officinalis, yeast, P. suffruticosa showed phototoxicity inhibitory effect in that their % hemolysis compared with control were 45.76 $\pm$ 0.91, 34.42$\pm$1.01, 35.30 $\pm$4.76 on UVB. In MTT assay, all tested natural products increased cell viability compared with the contort.

Cytotoxicity Evaluation of Elastomeric Impression Materials Using Different Fibroblasts Cell Lines

  • Purpose: Current common method of cytotoxicity evaluation for elastomeric impression materials use animal based cell lines, which the clinical relevance has been often questioned. Hence, the purpose of this study was to examine the difference in results with both human based and animal based fibroblast cell line. Materials and Methods: Three types of fibroblast cells were used in this study; conventional mouse fibroblasts of L929, human gingival fibroblasts (HGF-1), and immortalized human oral fibrobalsts (hTERT-hNOF). Test on extract and test by direct contact using different commercially available elastomeric impression materials were carried out according to the international standards. Result: There was significant difference in cell viability between types of fibroblasts cell used, where HGF-1 showed highest cell viability and L929 the lowest. Conclusion: Within the limitation of this study, careful consideration must be given when selecting the cells and interpreting the results for cytotoxicity evaluation of elastomeric impression materials, where use of human based cell lines such as hTERT-hNOF would be appropriate for both ease of cytotoxicity test and clinical relevance.

Comparison of the Two in Vitro Cytotoxicity Assays in Primary Cultured Rat Hepatocytes - the Neutral Red (NR) and Lactate Dehydrogenase (LDH) Tests (흰쥐의 배양 간세포를 이용한 세포독성시험에 있어서 뉴트랄레드 및 젖산 탈수소효소법의 비교)

  • The acute cytotoxicities of chloroquine sulfate, propranolol, ascorbic acid, acetylsalicylic acid and acrylamide on cultured adult rat hepatocytes were evaluated by the use of LDH leakage and NR uptake test. On the basis of $IC_{50}$ values, the rank order of cytotoxicities of these drugs in both tests was chloroquine sulfate > propranolol > acetylsalicylic acid > ascorbic acid. The $IC_{50}$ of LDH test was very similar to that of NR uptake test. Thus, we concluded that both tests are reliable and sensitive methods in detecting toxicity in adult cultured rat hepatocytes.

  • This study was conducted to assess a possible alternative method as replacements for in vivo test. The human fibroblasts were exposed to several photoxic chemicals (promethazine, neutral red, chlortetracyclone, amiodatone, bithional, 8-methyooxypsorale) and non-phototoxi substance, ammonium laureth sulfate and irradiatied with 5 J/$cm^2$ of UVA (3320~420nm). The cell viability was measured by NRU (neutral red uptake) assay. The photoxic potential of test chemicals in the NRU PT (phototoxicity test) was assessed by determining the PIF (photoirritancy Factor) by using a cut-off value of 5. The NRU PT responses of most chemicals showed a close agreement with in vivo response except bithinol. There was a relatively good agreement between in vitro NRU assay and in vivo data. These results suggest that NRU assay using fibroblast could be used to predict the phototoxicity.

In Vivo/In Vitro Properties of Novel Antioxidant Peptide from Pinctada fucata

  • Due to the potential of antioxidants to scavenge free radicals in human body, it is important to be able to prepare antioxidant peptides that meet the industrial requirements for cosmetics and food. Here, we determined in vivo/in vitro activities of antioxidant peptide from P. fucata (PFAOP) prepared by bio-fermentation method. The antioxidant property test results showed the DPPH, hydroxyl, superoxide radical-scavenging, and cellular antioxidant activity. EC50 values of PFAOPs were 0.018 ± 0.005, 0.126 ± 0.008, 0.168 ± 0.005, and 0.105 ± 0.005 mg/ml, respectively, exhibiting higher antioxidant activities than glutathione (p < 0.05). Moreover, anti-proliferation and cytotoxicity activity results illustrated PFAOP has a potent anti-proliferative activity against HepG2, Caco-2, and MCF-7 carcinoma cells with no cytotoxicity. Moreover, the protocols we developed in this work demonstrated several excellent advantages in PFAOP preparation compared to enzymatic hydrolysis or chemical synthesis methods and provide a theoretical foundation for higher-value application of marine-derived functional peptides.

In vitro Biocompatibility Evaluation of Biomaterial-elution Using Inflammatory Cell Lines (염증세포주를 이용한 생체재료 용출물의 체외 생체적합성 평가)

  • Various biometerials have been researched and have been developed for treatment of some disease through transplantation to body. They have been evaluated by in vitro cytotoxicity test using some skin-derived cell lines for prediction of their biocompatibility in vivo. However, the results of experiments using mesenchymal or epithelial cells could not be considered in vivo immune reaction. In this study, we evaluated the biomaterial-elution (elute from high density polyethylene film) using some cell lines (L929, Jurkat, U937) in vitro, and then that results were compared with in vivo results from guinea pig sensitization test. In sensitization test, saline and elution of syringe could not induce erythema, but only DNCB (hypersensitive chemical) induce erythema at guinea pig sensitization test. In cell experiment, the cytotoxicity results of inflammatory cells (Jurkat; T lymphocyte, U937; monocyte) was no difference with L929 (fibroblast) in the overall trend. However, inflammatory cell lines were only secreted inflammatory cytokine (TNF-${\alpha}$, INF-${\gamma}$) in some materials (biomateriallution, FAC, DNCB). And the biomaterial-elution did not have toxicity to the cells, but it induced the inflammatory cytokines in inflammatory cell lines only. So, we were predicted inflammatory reaction through the cytokine resultes of inflammatory cell lines, and it was more correlated with in vivo results than cytotoxicity test. Therefore, we suggested that the inflammatory cytokine assay using inflammatory cell lines are more effective method in vitro for evaluation of biocompatibility of biomaterials or chemicals.

Synergism of Cytotoxicity Effects of Triptolide and Artesunate Combination Treatment in Pancreatic Cancer Cell Lines

  • Background: Triptolide, extracted from the herb Tripteryglum wilfordii Hook.f that has long been used as a natural medicine in China, has attracted much interest for its anti-cancer effects against some kinds of tumours in recent years. Artesunate, extracted from the Chinese herb Artemisia annua, has proven to be effective and safe as an anti-malarial drug that possesses anticancer potential. The present study attempted to clarify if triptolide enhances artesunate-induced cytotoxicity in pancreatic cancer cell lines in vitro and in vivo. Methods: In vitro, to test synergic actions, cell viability and apoptosis were analyzed after treatment of pancreatic cancer cell lines with the two agents singly or in combination. The molecular mechanisms of apoptotic effects were also explored using qRT-PCR and Western blotting. In vivo, a tumor xenograft model was established in nude mice, for assessment of inhibitory effects of triptolide and artesunate. Results: We could show that the combination of triptolide and artesunate could inhibit pancreatic cancer cell line growth, and induce apoptosis, accompanied by expression of HSP 20 and HSP 27, indicating important roles in the synergic effects. Moreover, tumor growth was decreased with triptolide and artesunate synergy. Conclusion: Our result indicated that triptolide and artesunate in combination at low concentrations can exert synergistic anti-tumor effects in pancreatic cancer cells with potential clinical applications.

Antitumor Activities to Cytotoxicity of Phellinus linteus Ethanol Extract (목질진흙버섯 에탄을 추출물의 세포독성에 따른 항암활성)

  • We investigated antitumor activities of the ethanol extract from mushroom Phellinus linteus and Phellinus baumii on mulberry, oak and elm. in vitro test, the ethanol extract of mushroom cultivated on oak of Phellinus linteus showed highest activities about SK-OV-3, HCT15, XF498, SK-MEL-2 and A549. SK-OV-3 cell line showed 100% cytotoxicity in 100 $\mu\textrm{g}$/ml and HCT15 (98.39%), XF498 (89.62%), SK-MEL-2 (84.07%) and A549 (79.92%) cytotoxicity respectively. Also $IC_{50}$ showed 3.99 $\mu\textrm{g}$/ml to SK-OV-3 cell line and HCT15 (4.37 $\mu\textrm{g}$/ml), A549 (5.48 $\mu\textrm{g}$/ml), SK-MEL-2 (6.72 $\mu\textrm{g}$/ml), XF 498 (6.88 $\mu\textrm{g}$/ml). As those results, cultivated oak of Phellinus linteus showed a very low $IC_{50}$ value against SK-OV-3, HCT15, XF498, SK-MEL-2 and A549 cancer cell lines.

Interlaboratory Validation Study of In Vitro Alternatives to the Draize Eye Irritation Test : HET-CAM Test and Cytotoxicity Test for 20 Cosmetic Ingredients

  • 피부 전용 제재 개발을 위해 요구되는 동물 대체 시험법 중 가장 적극적으로 연구가 행하여지곤 실제 실용화가 예상되는 것은 안점막 자극 시험으로 지금까지 여러 가지 방법이 개발되었지만 그 중 계란 유정란의 응모요막(CAM)을 이용한 방법이 현재 가장 활발히 진행되고 있다. 이 방법이 일부 국가에서 이미 안점막 자극 시험 동물 대체 시험법으로 공인되었으며 현재까지도 validation 연구를 활발히 진행하고 있다. 본 연구에서도 국내에 적합한 안점막 자극 시험 동물 대체 시험법의 공인 시험법 개발 및 validation study를 목표로 계란 유정란의 응모요막을 이용한 방법 중 HET-CAM 방법을 시행하였으며 안점막 동물 대체 시험법으로 확립하고자 하였다. 틴ET-CAM 방법의 보완을 위해 배양된 세포를 통해 자극도를 측정할 수 있는 방법인 Cytotoxicity test를 도입하여 시행하였으며 두 방법의 data들을 분석하여 validation study를 수행하였다. 국내 유수의 6개 장업사가 본 연구에 참가하여 20가지의 화장품 전용제재를 대상으로 1차, 2차 validation study 를 진행하였다. HET-CAM test, Draize eye irritation test, Cytotoxicity test 측정 결과 HET-CAM 의 “Q” 수치는 대부분 강자극 수치인 2 이상이었고 10% sodium hydroxide가 가장 높은 수치를 보였으며 Tween 20(sorbitanpolyoxyethylene monolaurate) 100%가 가장 낮은 수치를 보였다. In vi패의 경우 10% sodium hydroxide가 가장 높은 수치를 보였으며 30군 propylene glycol 이 가장 낮은 자극수치를 보였다. HET-CAM test 와 Draize eye irritation test, Cytotoxicity test 간의 상관성 분석은 linear correlation coefficient 와 rank correlation coefficient를 구하여 비교하였으며 6개 장업사(A-F)의 실험실에서의 HET-CAM test 결과를 취합하여 각각 두 실험실간의 상관관계(linear correlation)를 분석하였다. Linear correlation coefficient 분석 결과를 보면 전반적으로 상관관계가 0.589 - 0.954의 범위였으며, 특히 A사와 B사 사이의 경우 0.954이었으며, E사와 D사 사이의 경우 0.942로 높은 상관관계를 보였다. 그 외에도 A사와 D사 사이의 경우(0.589)와 B사와 D사 사이의 경우(0.638)를 제외하고는 대체로 높은 상관관계를 나타내었다.
