본 연구에서는 pH 감응성 수화젤 입자를 이용하여 외부환경에 불안정한 활성물질을 화장품 제형 내에서는 안정하게 보존하고, 피부에 도포 시 빠른 방출로 피부에 흡수될 수 있는 지능형 전달시스템의 구현 가능성을 확인하기 위하여, 분산광중합을 이용하여 pH 감응성을 가지는 P(MAA-co-PEGMA) 수화젤 입자를 평균 크기 약 $2{\mu}m$의 구형 입자로 합성하였다. 합성된 P(MAA-co-PEGMA) 수화젤 입자는 수화젤의 MAA에 존재하는 카르복시기의 이온화에 의하여 pH 5를 전후로 한 급격한 팽윤비의 변화를 보여주었다. pH에 따른 수화젤 내부에 탑재된 알부민의 방출 및 피부투과 실험결과, P(MAA-co-PEGMA) 수화젤 입자는 pH 4.0에서는 소량의 알부민이 방출되어 피부 투과가 거의 일어나지 않은 반면, pH 6.0에서는 초기부터 다량의 알부민이 방출되어 상대적으로 높은 피부투과율을 나타내었다. 펩신을 이용한 알부민의 안정성 실험결과, P(MAA-co-PEGMA) 수화젤은 내부에 탑재된 알부민을 외부 환경으로부터 보호하여 알부민의 안정성을 유지시켜 주었다.
This paper presents a study into pH Indicator-Embedded hydrogel micro-particles which are encoded various shapes according to the captured indicator. We incorporate various pH indicators into a photo-curable hydrogel, PEG-DA(Poly(ethylene glycol) diacrylate). Using the latest fluidic lithography techniques, we can easily fabricate variously patterned hydrogel particles based on in-situ photopolymerization of the PEG-DA in a micro-fluidic channel. The shape of the particle is related to the pH indicator inside the particle. We demonstrate that the micro pH sensors change their colors according to the pH levels. The micro pH sensors have various characteristics that are related to the curing time, particle size, etc. By changing these conditions, we can adjust the long term stability and reaction time of the hybrid micro pH sensors.

Di-tyrosine photo-crosslinking of silk fibroin (SF) is recently highlighted as a biocompatible hydrogel fabrication process, because this method does not need potentially harmful chemical species. However, the resulting crosslinking density is often insufficient to obtain a mechanically stiff hydrogel unless additional oxygen is provided during the reaction. In this study, we proposed a combinational crosslinking method to form an SF hydrogel via the di-tyrosine photo-crosslinking with riboflavin (photoinitiator) and physical interaction of SF chains. In the UV light-irradiated SF solution, small particles formed and these particles promoted β-sheet formation of SF molecules, resulting in quick gelation. The di-tyrosine photo-crosslinking produced nuclei that might trigger regular assembly of SF molecules in high temperature condition. Conclusively, this process would contribute to a development of biocompatible hydrogel fabrication for biomedical uses of SF hydrogels.
본 연구에서는 알부틴, ascorbic acid, 아데노신 등과 같은 화장품 활성물질들을 주변 pH 변화에 따라서 선택적으로 방출하게 하는 지능형 전달시스템을 개발하기 위하여, pH 감응성 P(MAA-co-PEGMA) 수화젤을 분산 광중합을 이용하여 평균 크기 약 $2{\mu}m$의 구형 입자로 합성하였으며, 수화젤 입자는 수화젤의 $pK_a$인 PH 5를 전후로 하여 급격한 팽윤비의 변화를 보여주었다. P (MAA-co-PEGMA) 수화젤에 대한 활성물질들의 탑재에 영향을 미치는 요인들을 알아내기 위하여, 수화젤의 MAA와 EG의 조성과 탑재 pH에 따른 활성물질들의 탑재효율을 조사하였다. 수화젤을 구성하는 MAA와 EG의 조성 중 MAA의 함량이 감소함에 따라서 활성물질들의 탑재효율이 증가하였으며, 탑재 pH에 따른 실험에서는 수화젤과 활성물질들 사이에 형성되는 정전기적 반발력이 최소가 되는 pH보다는 수화젤의 팽윤비가 높게 되는 pH에서 탑재효율이 더 높게 나타났다.
Silver nanoparticles (AgNPs) can be used in the areas such as integrate circuit, cell electrode and antimicrobial deodorant. In this study, AgNPs have been prepared by using $AgNO_3$ aqueous solution in the carboxymethyl cellulose (CMC) hydrogel. CMC powders were dissolved in deionized water, and then irradiated by a gamma-ray with a radiation dose of 50 kGy to make CMC hydrogel. CMC hydrogels were dipped into $1.0{\times}10^{-2}M$$AgNO_3$ solution for 1 hour. After that, the swollen hydrogels were irradiated by gamma-ray for the formation of AgNPs. The characteristics of silver nanoparticles in the CMC hydrogels were monitored by UV-Vis and the morphological study and dispersed coefficient of particles were investigated by FE-SEM/EDX. It was observed that the sodium salt in the CMC is crucial to the formation of silver nanoparticle. Finally, antibacterial tests indiacted that the hydrogel containing silver nanoparticle has antibacterial activity.
pH감응형 P(MAA-co-EGMA) 수화젤을 분산 광중합을 이용하여 마이크로 크기의 미세입자로 합성하고 화장품 제형으로서의 응용 가능성을 평가하기 위하여 모델 탑재물질인 Rh-B와 화장품 분야에서 기능성 물질로 사용되는 ascorbic acid, adenosine, EGCG, arbutin을 이용하여 탑재 및 방출 거동을 조사하였다. Rh-B 탑재의 경우, pH 6.5인 수용액에서 이온화에 의한 P(MAA-co-EGMA) 수화젤의 음전하와 Rh-B의 양전하 사이의 정전기적 인력으로 인하여 가장 높은 탑재효율을 나타내었다. 그러나 화장품 기능성 물질들의 경우, pH 6.5 수용액에서 이온화된 P(MAA-co-EGMA) 수화젤의 음전하와 기능성 물질들이 나타내는 음전하 사이의 정전기적 반발력 때문에 상대적으로 낮은 탑재효율을 나타내었다. Rh-B를 사용한 방출실험 결과, p(MAA-co-EGMA) 수화젤 미세입자는 높은 pH에서는 다량의 Rh-B를 그리고 낮은 pH에서는 소량의 Rh-B를 방출하는 pH 감응성 방출거동을 나타내었다.
Hydrogels have gained considerable attention in various fields due to their easily transformative ability by different stimulation. In addition, metal-based conductive additives can enable the hydrogels to be conductive with dimension change. Although the development of the additives offered enhanced electrical properties to the hydrogels, correspondingly enhanced mechanical properties may limit the volume and electrical properties switching after stimulation. Here we prepared poly(N-isopropylacrylamide) (PNIPAM) thermo-responsive hydrogel that has a 32℃ of low critical solution temperature and added liquid metal particles (LMPs) as conductive additives, possessing soft and stretchable benefits. The LMPs enabled PNIPAM (PNIPAM/LMPs) hydrogels to be constricted over 32℃ with a high volume switching ratio of 15.2 when deswelled. Once the LMPs are spontaneously oxidized in hydrogel culture, the LMPs can release gallium ions into the hydrogel nature. The released gallium ions and oxidized LMPs enhanced the modulus of the PNIPAM/LMPs hydrogel, triggering high mechanical stability during repeated swelling/deswelling behavior. Lastly, highly constricted PNIPAM/LMPs hydrogel provided a 5x106 of electrical switching after deswelling, and the switching ratio was closely maintained after repeated swelling/deswelling transformation. This study opens up opportunities for hydrogel use requiring thermo-responsive and high electrical switching fields.
본 연구에서는 UV photo-lithography 방식의 particle replication in non-wetting templates(PRINT) 법을 이용하여 약물 전달에 운반체로 사용되는 $3{\mu}m{\times}3{\mu}m{\times}2{\mu}m$ 사이즈의 균일한 고분자 하이드로젤 입자를 제조하였다. 몰드(mold)와 기재(substrate)는 PRINT 방식을 통하여 탄성을 지닌 perfluoropolyethers(PFPE)로 제작하였으며 이를 반복적으로 사용할 수 있도록 하였다. 제작된 입자는 점착성이 있는 수용성 고분자를 이용하여 회수하였다. 입자의 주요 성분은 생분해성 고분자인 poly(ethylene glycol) diacrylate(PEG-diA)이며, 세포 uptake에 적합하도록 aminoethylacrylate(AEM)와 2-acryloxyethyltrimethyl ammonium chloride(AETMAC)를 첨가하였다. 본 연구를 통해 균일하고 원하는 크기의 생체분해성 고분자 입자를 제작하는 PRINT 기술이 약물 전달 및 유전자 전달에 필요한 수송체인 비바이럴 벡터를 제작하기 위한 효과적인 기술임을 제시하였다.
Incorporation of bioactive compounds such as vitamins, probiotics, bioactive peptides, and antioxidants into Nutrient Delivery System (NDS) for 'nanofood' provides simple way to develop novel functional foods that may have physiological benefits or reduce risks of diseases. As vital nutrient in nanofood, proteins possess unique functional properties including ability to form gels and emulsions, which allow them to be ideal nanofood materials for encapsulation of bioactive compounds. Based on protein physico-chemical properties, this review describes potential role of nanofood materials for development of NDS in hydrogel form, micro-or nano-particles. Applications of these nanofood materials to protect delivery-sensitive nutraceutical compounds are illustrated, and impacts of particle size on release properties are emphasized.
Silver nano-particles (AgNPs) have attracted much attention for centuries due to their unique optical properties, electrical conductivities, oxidative catalysis, and antibacterial effect. In this study, AgNPs have been prepared by using aqueous $AgNO_3$ solution in the poly(vinyl alcohol) (PVA)/poly(ethylene glycol) (PEG) hydrogels. PVA and PEG powders were dissolved in deionized water, and then irradiated by a gamma-ray with a radiation dose of 50 kGy to make hydrogels. PVA/PEG hydrogels were dipped into $1.0{\times}10^{-2}M$$AgNO_3$ solution for 1 hour. After that, the swollen hydrogels were irradiated by gamma-ray for the formation of AgNPs. FE-SEM is used to observe the formation of AgNPs as a function of the content of PEG and the irradiation dose. Also, AgNPs in the PVA/PEG hydrogels were monitored by UV-Vis. It is observed that the content of PEG and gamma-ray irradiation in the hydrogel is crucial to the formation of AgNPs. Finally, antibacterial tests indiacted that the hydrogel containing silver nanoparticle has antibacterial activity.
