버찌와 꿀을 함께 발효한 버찌-꿀 술의 발효특성 (Characteristics of Wine Fermented with Fruit of Flowering Cherry and Honey)

  • 탄소원으로 꿀을 사용하여 제조한 버찌-꿀술의 이화학적 특성을 분석하였다. 버찌-꿀술의 발효는 Saccharomyces bayanus (EC-118)를 스타터균으로 하여 발효액 2 L를 사용하여 30일 동안 진행하였다. 발효전 발효액에서 가장 많은 탄수화물은 과당이었으며, 포도당과 설탕 순으로 나타났다. 발효기간 동안 포도당은 효모균에 의하여 과당보다 빠르게 이용되어 사라졌으며, 발효기간이 진행됨에 따라 과당/포도당 비는 증가함을 보였다. 발효액의 pH와 효모 생균수는 발효 초기 5일 동안 급격하게 변화하였으나 발효액의 $^{\circ}Brix$(%)는 30일 기간 동안 점진적으로 감소하였다. 최종적으로 완성된 버찌-꿀술은 산가 0.43%, pH 3.5, $9.7^{\circ}Brix$(%), 에탄올 농도 14%였다. 버찌-꿀 술의 양조특성을 규명하였으므로 추가적인 버찌의 기능성이 확보된다면 버찌-꿀 술의 개발가능성이 충분히 있다.

Anti-Inflammatory Activity of Acacia Honey through Inhibition of NF-κB and MAPK/ATF2 Signaling Pathway in LPS-Stimulated RAW264.7 Cells

  • Honey used as conventional medicine has various pharmacological properties. In the honey and anti-inflammatory effect, Gelam honey and Manuka honey has been reported to exert anti-inflammatory activity. However, the anti-inflammatory effect and potential mechanisms of acacia honey (AH) are not well understood. In this study, we investigated anti-inflammatory activity and mechanism of action of AH in LPS-stimulated RAW264.7 cells. AH attenuated NO production through inhibition of iNOS expression in LPS-stimulated RAW264.7 cells. AH also decreased the expressions of $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ as pro-inflammatory cytokines, and MCP-1 expression as a pro-inflammatory chemokine. In the elucidation of the molecular mechanisms, AH decreased LPS-mediated $I{\kappa}B$-${\alpha}$ degradation and subsequent nuclear accumulation of p65, which resulted in the inhibition of $NF-{\kappa}B$ activation in RAW264.7 cells. AH dose-dependently suppressed LPS-mediated phosphorylation of ERK1/2 and p38 in RAW264.7 cells. In addition, AH significantly inhibited ATF2 phosphorylation and nuclear accumulation of ATF2 in LPS-stimulated RAW264.7 cells. These results suggest that AH has an anti-inflammatory effect, inhibiting the production of pro-inflammatory mediators such as NO, iNOS, $TNF-{\alpha}$, IL-6, $IL-1{\beta}$ and MCP-1 via interruption of the $NF-{\kappa}B$ and MAPK/ATF2 signaling pathways.

Evaluation of Toxicity of Paper Mill Sludge to Honey Bees and Analysis of Volatile Organic Compounds

  • Large amounts of sludge produced by paper mill industries represent one of the most serious environmental problems in the world. Recently, beekeepers living in the neighborhood of the paper mill in Hwasan County, Youngcheon city, GB, Korea, became alarmed that honey bee colonies were dying off suddenly across the neighborhood. A preliminary study was conducted to evaluate the toxicity (oral, fumigation, repellent) of recycled solid paper mill sludge (SPMS) and leachate paper mill sludge (LPMS) to honey bee workers under laboratory conditions, and to analyze the volatile organic compounds(VOC). The SPMS and LPMS were separately subjected to a liquid-liquid extraction (LLE) at three temperatures to extract VOC(highest VOC yields: 1.52% SPMS and 0.34% LPMS). A total of 70 chemicals were detected in the VOC of paper mill sludges, of which 49 and 21 volatile organic compounds from SPMS and LPMS, respectively. The SPMS was dominated by high degree presence of stanols (saturated sterols), such as cholestanol, cholestan-3-ol and also saturated hydrocarbons. However, LPMS was characterized by the absence of sterols. Both SPMS and LPMS showed an influence on the olfactory behavior of honey bee on Y-tube assay, with repulsion rates of 72 and 68%, respectively. Both SPMS and LPMS at concentration of 100mg/mL caused higher honey bee oral mortality than the untreated controls at 48, 72, 96 and 120 hours after treatment(highest oral mortality at 120 hr: 85.74%(SPMS); 93.51 % (LPMS)). A similar pattern was observed when honey bees were tested to fumigant toxicity. Both SPMS and LPMS caused significant higher mortality than the untreated control 24 hour after the exposure (highest fumigation mortality at 120 hr: 69.4% (SPMS); 56.8% (LPMS)). These preliminary results indicated that paper mill sludge could be partly responsible for sudden death and disappearance of honey bees, especially in hot humid summer days. With climate change, the risk of environmental chemical exposure to honey bee would pose greater attention.

산지별 밤꿀에 함유된 Kynurenic Acid의 정량 분석과 분석법 검증 (Quantitative Analysis of Kynurenic Acid in Chestnut Honey from Different Regions and Method Validation)

  • Chestnut honey is a sweet dark-colored honey with a distinct bitter aftertaste. It contains numerous phenolic compounds and alkaloids and is noted for its antioxidant and anti-inflammatory activities. However, it has been established that there are differences in the composition and activity of chestnut honey constituents depending on the region of origin, the sources of which warrant further research. In this study, we analyzed the kynurenic acid (KA) contents in chestnut honey produced in nine different regions in Korea, using high-performance liquid chromatography in conjunction with ultraviolet detection, and validated the analytical method developed. Use of a reverse-phase column and detection at a wavelength of 240 nm were found to be optimal for the detection of KA. Similar evaluation of an optimal method for extracting KA from chestnut honey revealed that extraction using 10% EtOH at 20 times the sample volume over a 6 h period was the most suitable for obtaining a high content of KA. Among the nine regional chestnut honeys assessed, KA content was found to be highest in the "Gongju" sample (1.14 mg/g), followed by that in the "Cheongdo" and "Damyang" samples. Validation of the KA analytical method revealed a good analyte linearity, with a correlation coefficient (r2) of 0.9995, an accuracy of between 92.37% and 107.35%, and good precision (RSD ≤ 1.05%). Our findings in this study, based on a validated quantitative analytical method for KA, could make an important contribution to establishing a data profiling procedure for characterizing chestnut honeys produced in different regions, and may also provide basic data for the identification of functional honey.

LC-MS/MS를 이용한 벌꿀 중 grayanotoxin 분석법 연구 및 실태조사 (Determination and Monitoring of Grayanotoxins in Honey Using LC-MS/MS)

  • 본 연구는 야생꿀을 비롯한 벌꿀, 벌집채꿀 등 국내산 및 수입산 꿀의 안전관리를 위한 grayanotoxin(GTX)의 시험분석법 확립 및 실태조사를 위하여 수행하였다. GTX 표준품 I, II, III는 LC-MS/MS로 분자량을 확인한 후, 시판품인 III를 제외한 I과 II는 NMR을 이용하여 구조를 확인하였다. 총 111건(국내산 벌꿀25건, 국내산 야생꿀 21건, 벌집채꿀 13건, 수입산 벌꿀 44건, 수입산 야생꿀 8건)의 벌꿀시료는 메탄올을 사용하여 벌꿀-메탄올 용액을 만들어 tC18 cartridge에 loading 한 후, 여과된 액을 동량의 증류수로 희석하여, 이온화장치로 ESI를 장착한 triplequadrupole LC-MS/MS를 이용하여 분석하였다. LC의 용리액은 1% 포름산이 첨가된 "메탄올-물"을 사용하는 것이 10분 이내의 분석시간대에 나타나는 피크의 모양과 감도가 우수한 경향을 나타내었다. 본 방법을 이용하여 검체 중의 GTX I, II, III의 함유량을 조사한 결과 총 111건 중 수입산 야생꿀 3건(2.7%)에서 GTX I, II, III가 검출되었고, 수입산 야생꿀 1건에서 GTX I, III가 검출되었다. GTX I의 검출량은 최소 $3.13{\pm}0.00mg/kg$에서 최고 $12.93{\pm}0.01mg/kg$으로 나타났고 GTX II는 $0.84{\pm}0.01mg/kg$, $0.92{\pm}0.00mg/kg$, $1.08{\pm}0.01mg/kg$의 함량을 나타내어 GTX I 에 비해 낮은 수치를 나타내었다. GTX III는 최소 $0.25{\pm}0.01mg/kg$에서 최고 $3.29{\pm}0.74mg/kg$으로 함량에 큰 차이를 보였다. 본 방법을 이용한 총 111건의 벌꿀 시료의 GTX분석시 수입산 야생꿀 4건에서만 GTX가 검출됨을 알 수 있었다. 본 연구에서는 비 휘발성 또는 극성 때문에 GTX 분석시 GC 및 GC-MS에서 분석이 어려운 벌꿀시료를 대상으로 전처리 시간의 단축을 모색함과 동시에 LC-MS/MS를 이용한 시험분석법을 개발할 수 있었고, 모니터링을 통하여 네팔, 터키 등 특정 지역의 야생꿀의 섭취를 제한하는 과학적 근거를 마련할 수 있었다.

  • Poisonings caused by 'mad honey' are known to occur in response to grayanotoxins, which bind to sodium channels in the cell membrane, increasing membrane sodium permeability and preventing inactivation. Mild symptoms of mad honey intoxication are dizziness, weakness, hypersalivation, nausea, vomiting, and paresthesia. Severe intoxication, however, leads to serious cardiac manifestations such as atrioventricular block, dose-dependent hypotension, bradycardia, and respiratory depression. Atropine and vasoactive drugs improve symptoms of both bradycardia and respiratory rate depression. We report an unusual case of saliva-type hyperamylasemia in a mad honey poisoning patient who developed clinically significant bradycardia. She recovered fully within 3 days following atropine administration and medical treatment.

대규모 양봉농가의 소득 분석 (An Income Analysis of Large Scale Honey Bee Farming Farmhouseholds)

  • This study analyzed the income of large scale honey bee farming of Korea. Using a questionnaire, 44 large scale honey bee farming farmhouseholds were surveyed. The average hive sizes are 258. The results are as follows. First, gross receipts of bee farming farmhouseholds are 50,342 thousand won, and it's components are honey 71%, bee selling and/or lent 26%. Second, working expenses are 23,778 thousand won that is 47% of gross receipts. Feed costs are 38.2%, material costs are 23.9%, hired labor costs are 11.0%, moving costs are 10.1%. Finally, receipts per hive are 195.1 thousand won, working expenses per hive are 23,778 thousand won, so income per hive is 103.0 thousand won.

A Honey-Hive based Efficient Data Aggregation in Wireless Sensor Networks

  • The advent of Wireless Sensor Networks (WSN) has led to their use in numerous applications. Sensors are autonomous in nature and are constrained by limited resources. Designing an autonomous topology with criteria for economic and energy conservation is considered a major goal in WSN. The proposed honey-hive clustering consumes minimum energy and resources with minimal transmission delay compared to the existing approaches. The honey-hive approach consists of two phases. The first phase is an Intra-Cluster Min-Max Discrepancy (ICMMD) analysis, which is based on the local honey-hive data gathering technique and the second phase is Inter-Cluster Frequency Matching (ICFM), which is based on the global optimal data aggregation. The proposed data aggregation mechanism increases the optimal connectivity range of the sensor node to a considerable degree for inter-cluster and intra-cluster coverage with an improved optimal energy conservation.

전북지방 한봉꿀의 계절별 성분에 관한 연구 (Studies on the constituents of Korean honey harvested seasonally in Chonbuk area)

  • This study was to investigate the properties such as amounts of moisture, inverted sugar, cane sugar and 5-(hydroxymethyl)-2-furfural (HMF) in feeding-honey(n=45) and wild-honey (n=3) harvested in Chonbuk area from April to November 1995. The results obtained were summarized as follows ; The average percentages of moisture in both feeding- and wild-honey was 20.2%(range, 17.5~23.1) and 20.7%(19.4~22.2). And those of inverted sugar/cane sugar in both honeies were 64.4%(55.8~69.3)/6.8%(1.0~l5.7) and 68.9%(68.5~69.8)/3.1%(2.5~3.7), in seasonal analysis those were 65.9%/5.3% in spring, 65.7%/6.6% in summer, 62.1%/8.0% in autumn, respectively. On the other hand, the average amounts of HMF in both honeies were 44.86mg/kg(22.6~68.6) and 7.0mg/kg(6.2~7.6), its average difference between both honeies were about 37.86mg/kg regardless of region and season.

벌꿀-인삼분 혼합물의 유동학적 성질에 관한 연구 (Study on Rheological Properties of Honey Cinseng Powder Mixtures.)

  • The rheological properties of some honey-ginseng powder mixturs, used as pharmaceutical pills, were studied extensively by employing five kinds of honey, acacia, chestnut, buckwheat, green perilla and bush clover. Relation between the viscosity of the sample mixtures and the concentrations of ginseng powder and the temperature effect on the internal change of the structure were studied. The results obtained are as follows: 1) A complete gel formation of honey-ginseng mixtures was observed when about 50 days have passed after the sample preparation. 2) The viscosity of honey-ginseng powder mixtures was related with the concentration of ginseng powder, the temperature and the period of time required in gel formation as shown in the following equation. n=noexp$[{\kappa}_{1}c^{n1}+{\kappa}_{2}(T-273)^{-n2}+{\kappa}_{3} t^{n3}]$
