Optimization of Enzyme Digestion Conditions for Quantification of Glycated Hemoglobin Using Isotope Dilution Liquid Chromatography-Tandem Mass Spectrometry

  • Glycated hemoglobin (HbA1c) is used as an index of mean glycemia over prolonged periods. This study describes an optimization of enzyme digestion conditions for quantification of non-glycated hemoglobin (HbA0) and HbA1c as diagnostic markers of diabetes mellitus. Both HbA0 and HbA1c were quantitatively determined followed by enzyme digestion using isotope dilution liquid chromatography-tandem mass spectrometry (ID-LC-MS/MS) with synthesized N-terminal hexapeptides as standards and synthesized isotope labeled hexapeptides as internal standards. Prior to quantification, each peptide was additionally quantified by amino acid composition analysis using ID-LC-MS/MS via acid hydrolysis. Each parameter was considered strictly as a means to improve digestion efficiency and repeatability. Digestion of hemoglobin was optimized when using 100 mM ammonium acetate (pH 4.2) and a Glu-C-to-HbA1c ratio of 1:50 at $37^{\circ}C$ for 20 h. Quantification was satisfactorily reproducible with a 2.6% relative standard deviation. These conditions were recommended for a primary reference method of HbA1c quantification and for the certification of HbA1c reference material.

Effects of lethal concentration, hematological parameters and plasma components of common carp, Cyprinus carpio by waterborne acute nitrite exposure (수인성 아질산 급성 노출에 따른 잉어, Cyprinus carpio의 반수치사농도, 혈액학적 성상 및 혈장성분의 영향)

  • Common carp (Cyprinus carpio) (Weight 23.05 ± 6.98 g, Length 12.65 ± 1.38 cm) were exposed to waterborne nitrite at 0, 50, 100, 200, 400 and 800 mg NO2 -/L for 96 hours. The lethal concentration 50 (LC50) of C. carpio exposed to waterborne nitrite was 398.6 mg NO2 -/L. Hemoglobin, hematocrit and RBC count were significantly decreased by waterborne nitrite exposure. The MCV (mean mean corpuscular volume) (µl), MCH (mean corpuscular hemoglobin) (pg) and MCHC (mean corpuscular hemoglobin concentration) (%) were significantly increased. The inorganic component, plasma calcium, was significantly increased, and the organic components such as plasma cholesterol and total protein were significantly increased showing a similar tendency with calcium. In enzymatic components, the AST and ALT were also significantly increased by nitrite exposure. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of C. carpio.

Optimization of Microwave-Assisted Method for Accelerated Glycated Hemoglobin Quantification from Amino Acids to Proteins

  • Glycated hemoglobin ($HbA_{1c}$) has been commonly used to screen and diagnose for patients with diabetes mellitus. Here the accelerated procedure of microwave-assisted sample treatment from acid hydrolysis to enzyme digestion followed by isotope dilution liquid chromatography-tandem mass spectrometry (ID-LC-MS/MS) was optimized and applied to measure $HbA_{1c}$ in an effort to speed up analysis time. First, two signature peptides of $HbA_{1c}$ and hemoglobin $A_0$ were certified with amino acid analysis by setting optimized acid hydrolysis conditions to $150^{\circ}C$, 1.5 h and $10{\mu}M$ sample concentration in 8 M hydrochloric acid. Consequently, the accurate certified peptides above were used as calibration standards to implement the proteolytic procedure with endoproteinase Glu-C at $37^{\circ}C$, 700 W for 6 h. Compared to the traditional method, the microwave heating not only shortened dramatically sample preparation time, but also afforded comparable recovery yields. The optimized protocol and analytical conditions in this study are suitable for a primary reference method of $HbA_{1c}$ quantification with full SI-traceability and other similar proteins in complex biological samples.

  • This study was carried out to investigate the supplementary effects of ${\gamma}$-oryzanol on the blood glucose level in diabetic mice. We supplied 2 experimental diets (CO without and GO with 0.2% ${\gamma}$-oryzanol) to diabetic mice for 8 weeks. Diet intake, body weight and levels of blood glucose, hemoglobin $A_{lc}$ and insulin were measured. Though there was no significant difference in diet intake between experimental groups, the concentration of fasting blood glucose and blood glucose area from glucose tolerance test in diabetic mice was lower in GO group than CO group during the supplementary period of experimental diets. Hemoglobin Ai, was lower and serum insulin level was higher in GO group than CO group without significance. These results suggest that r-oryzanol decrease the blood glucose level, and ${\gamma}$ -oryzanol produced from residual product of rice may be developed with high value.

  • On protein equivalence base, fishmeal (FM) was replaced by lysine cell mass (LCM) in selected different diets in Korean rockfish, Sebastes schlegeli (Hilgendorf) Eight experimental diets were formulated to contain 100% FM (LC $M_{0}$), 90% FM+10% LCM (LC $M_{10}$),80% FM+20% LCM (LC $M_{20}$), 70% FM+30% LCM (LC $M_{30}$), 60% FM+40% LCM (LC $M_{40}$ ), 70% FH+30% LCM+lysine (LC $M_{+Lys}$), 60% FM+40% LCM+lysine (LC $M_{40+Lys}$), and 50% FM+50% LCM+lysine (LC $M_{50+Lys}$). Experimental individuals of the fish (12.6 g) were randomly fed on one of the experimental diets. After 6 weeks of feeding trial, weight gain (WG) and feed efficiency (FE) of fish fed LC $M_{0}$ diet was significantly (P〈0.05) higher than those of fish fed LC $M_{20}$, LC $M_{30}$, LC $M_{40}$ , LC $M_{30+Lys}$, LC $M_{40+Lys}$, and LC $M_{50+Lys}$ diets, however, there was no significant difference in WG of fish fed LC $M_{0}$ and LC $M_{10}$ diets. Supplementation of lysine has no effect on WG. There was no significant difference in condition factor (CF) of fish fed LC $M_{0}$, LC $M_{10}$ and LC $M_{20}$ diets. Hemoglobin (Hb) of fish fed LC $M_{0}$, LC $M_{10}$, LC $M_{20}$, LC $M_{30}$, LC $M_{40}$ , LC $M_{30+Lys}$, and LC $M_{40+Lys}$, diets were not significantly different from each other. No significant differences were observed in hematocrit (PCV) and hepatosomatic index (HSI) among all dietary treatments. Apparent digestibility of dry matter (ADM) and protein (ADP) of diets significantly decreased with increase in dietary LCM level, though there was no difference in ADM and ADP between LC $M_{0}$ and LC $M_{10}$.   • This study was carried out to investigate the supplementary effects of Suwon 464, which has over two times of dietary fiber content compared with normal rice (Ilpum), on blood glucose in diabetic mice. We supplied 5 kinds of experimental diets (corn starch diet as a control (CO), Ilpum polished rice diet (IP), Ilpum brown rice diet (IB), polished rice diet (SP) and brown rice diet (SB) of Suwon 464) to diabetic mice for 8 weeks, after analyzing dietary fiber contents of 5 experimental diets. Diet intake, body weight and contents of blood glucose, hemoglobin $A_{lc}$ and insulin were measured. The dietary fiber contents in CO, IP, IB, SP, and SB diets were 1.0, 1.2, l.4, l.4, and 2.0% respectively. Body weight was lower in SB group than the other groups though there was no significant difference in diet intake among experimental groups. The concentration of blood glucose in diabetic mice was lower in SB group than the other groups during the supplementary period of experimental diets. The hemoglobin Ale and serum insulin levels were lower in SP and SB groups. These results suggested that the brown rice of Suwon 464 with high dietary fiber can control diabetes in diabetic mice by reducing the blood glucose and hemoglobin Ale.   • To investigate the effects of the dissolved oxygen concentration (DO) of Tetillarca granosa (Linnaeus), We measured $LC_{50}$, survival, oxygen consumption rate, filtration rates and blood composition under $10{\cdot}{\pm}0.5^{\circ}C$ and $25{\cdot}0.5^{\circ}C$ as a function of DO. The 16 days-$LC_{50}$ of DO for T. granosa was 1.31 mg DO/L in large individual and 0.95 mg DO/L in small individual. At 25$^{\circ}C$, the $LC_{50}$ of DO in large and small individual was 1.13 and 1.M mg DO/L, respectively. With decreasing DO, oxygen consumption rate, and filtration rates of T. granosa decreased. Blood composition of T. granosa was analysed hemoglobin, glucose, total protein, total cholesterol, GOT and GPT, Hg was increased with decreasing DO, but glucose was decreased below 1.2 mg DO/L. Total protein, total cholesterol, GOT and GPT were investigated irregular and decreasing aspect.

  • Mirror carp, Cyprinus carpio nudus (weight 26.2±3.1 g, length 11.8±0.5 cm) were exposed to waterborne total ammonia nitrogen (TAN) at 0, 2, 4, 8, 16, 32, 64 and 128 mg TAN/L. The lethal concentration 50 (LC50) of mirror carp, C. carpio nudus exposed to waterborne ammonia was 60.38 mg TAN/L. The red blood cell (RBC) count was significantly reduced by ammonia exposure, whereas there was no significant changes in the hemoglobin concentration and hematocrit value. In the plasma components, glucose, total protein, aspartate aminotransferase (AST) and alkaline phosphatase (ALP) were significantly increased by ammonia exposure. The results of this study suggest that the ammonia exposure to C. carpio nudus affects the survival rates, hematological parameters and plasma components as toxicity.

  • Olive flounder Paralichthys olivaceus (total weight 216.4±14.6 g, total length 28.4±1.7 cm) were exposed to different pH levels (3, 4, 5, 6, 7, 8, and 9) for 96 h. At pH 4, hemoglobin decreased significantly, while plasma calcium, glucose, cholesterol, and ALP increased significantly. Exposure to pH 4 also induced stress responses, as evidenced by a significant decrease in heat shock protein 70 (HSP 70) and a significant increase in cortisol. The results of this study indicate that acute exposure to acidic or alkaline pH (pH 3 or 9) induced significant mortality, while exposure to pH 4significantly affected hematological parameters and stress responses in P. olivaceus.

  • Olive flounder (Paralichthys olivaceus) (Weight 110.9±17.1 g, length 22.3±1.2 cm) were exposed to waterborne nitrite at 0, 30, 60, 120, 240, 480 and 960 mg NO2-/L according to water temperature at 20℃ and 25℃ for 96 hours. The lethal concentration 50 (LC50) of olive flounder, P. olivaceus exposed to waterborne nitrite was 513.87 mg NO2-/L at 20℃ and 208.35 mg NO2-/L at 25℃, which means a significant difference in LC50 by the water temperature. Hemoglobin and hematocrit were significantly decreased by waterborne nitrite exposure. The inorganic component, plasma calcium, was significantly decreased, and the organic components such as plasma glucose and cholesterol were significantly decreased showing a similar tendency with calcium. In enzymatic components, the AST and ALP were also significantly decreased by nitrite exposure. The results of this study indicate that exposure to nitrite can affect the survival and hematological physiology of P. olivaceus, and the effect of exposure to nitrite had a significant effect on nitrite toxicity depending on the water temperature.
