Cloning and Expression of a Human Homolog of Mouse Gamml, MVGI, Localized in 12q13 (인간염색체 12q13에 내재한 마우스 Gamm1의 인간유전자 homolog, MYG1의 클로닝과 발현)

  • Isolation of a gene and determination of its expression pattern are essential in understanding its function. Among the genes localized in 12ql3, stSG3435 EST was chosen to study its expression pattern. The full-length CDNA was cloned by screening of human brain CDNA library and its sequence was determined by serial deletion followed by automated sequencing of the clones with overlapping fragments. The sequence analysis revealed that stSG 3435 CDNA displayed 100% identity to human MYGI and 86% identity to mouse melanocyte proliferation gene-1 (Gamm 1) originally identified from melanocyte, suggesting that MYGI determined by Northern blot analysis revealed the strongest expression in testes with ubiquitous expression in all the tissues tested. In order to investigate the cellular localization of its protein product, the green fluorescence protein gene was fused into the full-length coding sequence of MYGI, Transfection of the fusion construct followed by confocal microscopy resulted in the green fluorescence signal as a punctate state in cytoplasm indication that MYGI was localized in one of the cellular organelles.

Comparison of Pigments and Estimation of Production Period in Old and New Celestial Charts Folding Screens (신구법천문도 채색 안료 비교 및 제작시기 추정)

  • The pigments of three old and new celestial charts folding screens(『Celestial Chart(Folding Screen)』 and 『Old and New Celestial Charts, Eight-Panel Folding Screen』 of National Folk Museum of Korea and 『Koudou-Nanboku-Ryousouseizu』 of National Diet Library of Japan) were analyzed to estimate their dating. It was estimated that the 『Celestial Charts(Folding Screen)』 was painted using traditional pigments from the Joseon dynasty such as azurite, indigo lake, malachite, atacamite, vermilion, iron oxide, cochineal, gamboge, orpiment, lead white, talc and soot. The green and blue colors of the 『Old and New Celestial Charts, Eight-Panel Folding Screen』 and 『Koudou-Nanboku-Ryousouseizu』 were painted using artificial inorganic pigments such as emerald green and ultramarine blue. These pigments were imported from Europe post the mid-19th century. In the 『Old and New Celestial Chart, Eight-Panel Folding Screen』, only artificial inorganic pigments were used for green and blue colors. However in the 『Koudou-Nanboku-Ryousouseizu』, emerald green and atacamite in green color, and ultramarine blue and indigo lake in blue color were used together. Based on both the results of pigment analysis and the study of star charts and inscriptions, the 『Celestial Charts(Folding Screen)』 was painted post mid-18th century. The 『Koudou-Nanboku-Ryousouseizu』 and 『Old and New Celestial Charts, Eight-Panel Folding Screen』 were painted after green and blue artificial pigments were imported in the mid-19th century. The 『Koudou-Nanboku-Ryousouseizu』 in which both traditional and western artificial pigments were used, can be dated earlier than the 『Old and New Celestial Chart, Eight-Panel Folding Screen』.

Development of Miniaturized Culture Systems for Large Screening of Mycelial Fungal Cells of Aspergillus terreus Producing Itaconic Acid

  • The task of improving a fungal strain is highly time-consuming due to the requirement of a large number of flasks in order to obtain a library with enough diversity. In addition, fermentations (particularly those for fungal cells) are typically performed in high-volume (100-250 ml) shake-flasks. In this study, for large and rapid screening of itaconic acid (IA) high-yielding mutants of Aspergillus terreus, a miniaturized culture method was developed using 12-well and 24-well microtiter plates (MTPs, working volume = 1-2 ml). These miniaturized MTP fermentations were successful, only when highly filamentous forms were induced in the growth cultures. Under these conditions, loose-pelleted morphologies of optimum sizes (less than 0.5 mm in diameter) were casually induced in the MTP production cultures, which turned out to be the prerequisite for the active IA biosynthesis by the mutated strains in the miniaturized fermentations. Another crucial factor for successful MTP fermentation was to supply an optimal amount of dissolved oxygen into the fermentation broth through increasing the agitation speed (240 rpm) and reducing the working volume (1 ml) of each 24-well microtiter plate. Notably, almost identical fermentation physiologies resulted in the 250 ml shake-flasks, as well as in the 12-well and 24-well MTP cultures conducted under the respective optimum conditions, as expressed in terms of the distribution of IA productivity of each mutant. These results reveal that MTP cultures could be considered as viable alternatives for the labor-intensive shake-flask fermentations even for filamentous fungal cells, leading to the rapid development of IA high-yield mutant strains.

Toxic dinoflagellate Gymnodinium catenatum Graham(Dinophyceae) from the southern coast of Korea: morphology, phylogeny and effects of temperature and salinity on growth (남해안에서 분리한 유독 와편모조류 Gymnodinium catenatum Graham (Dinophyceae): 형태, 분자계통학적 특성 및 온도와 염분에 따른 성장 특성)

  • The toxic dinoflagellate Gymnodinium catenatum isolated from the southern coast of Korea was described under light and scanning electron microscopy, and its large subunit (LSU) rDNA was sequenced. In addition, the effects of temperature and salinity on its growth were investigated. The cells of G. catenatum, as viewed under the electronic microscope, were green-brown color, $38.1-77.4{\mu}m$ in length and $26.1-40.8{\mu}m$ in width. The epicone was conical, while the hypocone was trapezoidal. The nucleus was located at the central part of the cell. The apical groove was horseshoe-shaped and small pores were irregularly distributed on the cell surface. Molecular phylogeny based on LSU rDNA gene sequences showed that the Korean G. catenatum and previously reported species formed a monophyletic clade within Gymnodinium sensu stricto clade. The maximum growth rate of $0.37day^{-1}$, was obtained at $25^{\circ}C$ and 35 psu, and the maximum cell density of $1,073cells\;mL^{-1}$, was observed at $20^{\circ}C$ and 25 psu. However, G. catenatum did not grow at temperature < $15^{\circ}C$ and < $30^{\circ}C$. These results suggest that environmental conditions of summer and autumn in the southern coast of Korea may be favorable for the growth of G. catenatum.

In Vitro Antifungal Activity and Mode of Action of 2',4'-Dihydroxychalcone against Aspergillus fumigatus

  • 2',4'-Dihydroxychalcone (2',4'-DHC) was identified from a heat shock protein 90 (Hsp90)-targeting library as a compound with Hsp90 inhibitory and antifungal effects. In the presence of 2',4'-DHC ($8{\mu}g/mL$), radial growth of Aspergillus fumigatus was inhibited 20% compared to the control, and green pigmentation was completely blocked. The expression of the conidiation-associated genes abaA, brlA, and wetA was significantly decreased (approximately 3- to 5-fold) by treatment with 2',4'-DHC. The expression of calcineurin signaling components, cnaA and crzA, was also significantly reduced. The inhibitory effects of 2',4'-DHC on metabolic activity and mycelial growth were significantly enhanced by combination treatment with itraconazole and caspofungin. Docking studies indicated that 2',4'-DHC bind to the ATPase domain of Hsp90. These results suggest that 2',4'-DHC act as an Hsp90-calcinurin pathway inhibitor.

A Study of Analysis on the Noise Aspect of School District -A Case Study of Wonju City- (학교주변 소음실태 분석에 관한 연구 -원주지역 학교를 중심으로-)

  • In this study, noise analyses were performed toward ten school and one library-building adjacent to road. In particular, noise levels measured in the case-study city were compared to those from environmental noise standard as well as to encouraged noise level. In the analyses, the following conclusions are emphasized in order to make comfortable educational learning environment. 1) When windows were open, the measured noise level of 9 schools exceeded the encouraged noise level. In order to mitigate the exceeded level, it is necessary to plant trees or lawn and install soundproofing wall. 2) When windows were closed, the measured noise level of the most schools showed lower than that of encouraged level. For sound insulation, installing both double window and casement window are required. 3) Schoolhouse's vertical arrangement from the road in order to minimize the expected noise effect, and provision of green space by which visual hiding as well as psychological comfort are presumably achieved.

  • The Eco-performance assessment program was developed as part of Eco-performance management for buildings at the national level in developed countries. Cases of domestic Green Building Certification(GBC) system was developed but these program's destination of Eco-performance management for buildings needs systematic management by the limited GBC. Generally in the construction industry, Eco-performance management of Building Life-cycle is required. Therefore this paper develop the creation of Eco-performance information and management process, those are from design and maintenance management phases, through suggest the development of a information management for Eco-performance management of buildings. In addition, it makes to decrease the reproduction of information unnecessarily by analyzation of the related association between the information of facilities objects and Eco-performance management. Finally, study the previous reaserch and suggest the BIM by the method of information management and this paper purpose the development of web-based Eco BIM object library management system.

Verification and validation of ShipMo3D ship motion predictions in the time and frequency domains

  • This paper compares frequency domain and time domain predictions from the ShipMo3D ship motion library with observed motions from model tests and sea trials. ShipMo3D evaluates hull radiation and diffraction forces using the frequency domain Green function for zero forward speed, which is a suitable approach for ships travelling at moderate speed (e.g., Froude numbers up to 0.4). Numerical predictions give generally good agreement with experiments. Frequency domain and linear time domain predictions are almost identical. Evaluation of nonlinear buoyancy and incident wave forces using the instantaneous wetted hull surface gives no improvement in numerical predictions. Consistent prediction of roll motions remains a challenge for seakeeping codes due to the associated viscous effects.

  • For as to world, the concern about the environment problem is enhanced than any other time in the past because of being 21 century. And the environment problem is highlighted as the world-wide issue. The time of the environment problem intimidates the alive of the mankind and presence of an earth over the time. It becomes the essentiality not being selection in the personal living or the economical viewpoint now to prepare for the climatic modification. As to the company management, the green growth period which it excludes the environment management considering an environment, cannot carry on the company the continued management comes. That is, in the change center of the management paradigm, there is the environment management. Nearly, the greenhouse gas which the publication industry is the environmental toxic material like all industries is generated. The greenhouse gas is ejected in the process of running the manufacturing process and print shop of the various kinds material used as the raw material of the book. Particularly, the tree felling for getting the material of the paper is known to reach the direct influence on the global warming. This study does according to an object it considers and organizes the environment parameter based on this kind of fact as to the publication industry. And it is determined as the reference which is used as the basic materials preparing the case that carbon exhaust right transaction(CAP and TRADE) drawing are enforced in all industries and is sustainable the management of the publication industry and reduces the environmental risk among the company many risk management elements and plans and enforces the publication related policy that there is a value. In the printing publication industry, this study tried to inquire into elements discharging the environmental pollutant or the greenhouse gas. Additionally, in the printed publication production process, it tried to inquire into the effort for an environment-friendly and necessity at the printing paper and the printers ink, regarded as the element discharging the greenhouse gas all kinds of the printing materials, operation of the print shop and all kinds of the machines and recycle process, and etc. These considerations make these industrial field employees aware of the significance about a conservation and environmental protection. They try to give a help in the subsequent study producing quantitatively each environmental parameter emission of green house gas. This makes the calculation of the relative $CO_2$ output reproached ultimately possible. Meanwhile, in a sense, many research protects and improving an environment in connection with the contents of research at the printing publication industrial field is in progress. There will be the voluntary human face that it has to protect an environment but this can not do by the outside factor according to all kinds of environment related law and regulation. Anyway, because of acting on company management as the factor of oppression, the increase of this environment-related correspondence cost could know that the research that the environment loading relates with a procurement and development, environment management system introduction, quality control standard, including, normalizing including a material, and etc. through the part of the effort to reduce the cost low was actively in progress. As to the green growth era, as follows, this paper prescribed the subject and alternative of the print publication industry. It is surrounded by the firstly new digital environment and the generation of the subject. And secondly the printing industry is caused by the point of time when the green growth leaves by the topic which is largest in the global industry and it increases. The printing publication industry has to prepare the bridgehead for the environment-friendly green growth as the alternative for this resolution with first. The support blown in each industry becomes the obligation not being selection. Prestek in which the print publishing was exposed to spend many energies and which is known as the practice of the sustainable print publishing insisted that it mentioned importance of the green printing through the white pages in 2008 and a company had to be the green growth comprised through the environment-friendly activity. A study on vegetation and soil environmental characteristics of green roof in Daejeon Metropolitan City (대전광역시 옥상녹화 지역의 식생현황 및 토양환경 특성에 관한 연구)

  • This study was to analyze the soil environmental characteristics and vegetation status of green roof in Daejeon Metropolitan City. The investigated floras of vascular plants are 17 families, 26 genera, 28 species in Seo-Gu Daejeon District Office Building (SG), 25 families, 49 genera, 56 species in Galma Public Library (GP), and 34 families, 57 genera, 60 species in Daejeon City Hall (DC) respectively. Although the larger area shows the more numbers of species in introduced plants and naturalized plant, the naturalized plant ratios were similar with each other. They were 10.71%, 10.71%, and 11.67% at SG, GP, and DC respectively. As a result of analysis on soil physical property, soil depths including vegetation soil and drainage soil of 3 green roofs were 30cm. The depths of vegetation soil at SG, GP, and DC were 0~8cm, 0~10cm, 0~10cm respectively. As a results of soil chemical properties of our study, soil pH of vegetation soil and drainage soil were a range of 6.42 and 7.43, and a range of 6.55 and 7.43 on the average respectively. Available-P contents of vegetation soil and drainage soil were a range of 153.33 and 366.33mg/kg, and a range of 136.67 and 242.67 mg/kg which is very high, respectively. Carbon contents in soil at vegetation soil and drainage soil were a range of 3.16 and 6.38%, and a range of 1.63 and 2.47% respectively. Carbon storage per square meter within 30 cm were 2.76 kg, 2.99 kg, and 3.66 kg at SG, GP, and DC respectively.
