Inhibitory Effect of Muscat Bailey A Seed Extract on Melanin Production in $\alpha$-Melanin Stimulating Hormone-stimulated B16 Cell (머루포도 씨 추출물의 $\alpha$-Melanin Stimulating Hormone으로 자극한 B16세포에서 melanin 생성억제 효과)

  • Inhibitory effect of skin and seed of three species grape cultivated in Korea on melanogenesis was investigated. Melanin generation was examined in $\alpha$-Melanin Stimulating Hormone-stimulated B16 cell, mouse melanoma, in the presence of samples. All skin sample did not show the inhibitory effect. Seed extract of Campbell early and Neo Muscat had negative effect on cell viability. When $50{\mu}g/ml$ seed extract of Muscat Bailey A was treated, amount of generated melanin and cell viability were $51.6{\pm}20.5%$ and $90.4{\pm}11.3%$ compared to control, respectively. Seed extract of Muscat Bailey A reduced the tyrosinase protein induced by $\alpha$-Melanin Stimulating Hormone, which suggests that inhibitory effect of seed extract of Muscat Bailey A on melanin is partly due to suppression of tyrosinase that is responsible for melanin production.

  • Free radicals and oxidative stress playa crucial role in the pathophysiology of a broad spectrum of cardiovascular diseases including congestive heart failure, valvular heart disease, cardiomyopathy, hypertrophy, atherosclerosis and ischemic heart disease. We have demonstrated that IH 636 proanthocyanidin extract (GSPE) provides excellent protection against free radicals in both in vitro and in vivo models, and exhibits significantly better efficacy as compared to vitamins C, E and $\beta$-carotene.(omitted)

Antioxidant and Lipid Peroxidation Inhibition Effects of Catechin Compounds Isolated from Ethyl Acetate Fraction of Grape Seed Ethanol Extract (포도종자 에텔아세테이트 분획물로부터 분리한 카테킨 화합물의 항산화 및 지질과산화 억제효과)

  • This study evaluated the isolation and identification of biologically active compounds from the ethyl acetate fraction of grape seed extract (Campbell early). Ethyl acetate fraction was further purified with sephadex LP-20 column chromatography. Each biologically active compound for free radical scavenging effect and lipid peroxidation inhibition was isolated and identified with ${1}^H$ and${12}^C$-NMR. Major compounds were identified as (+)-catechin and (-)-epicatechin respectively. The amounts of (+)-catechin and (-)-epicatechin in grape seed were 45.7$\%$ and 35.1$\%$, respectively. The purified (+)-catechin and (-)-epicatechin showed more strong free radical scavenging effects ($RC_{50}$= 11.1 $\mu$g/mL and 10.4 $\mu$g/mL) than ethyl acetate fraction ($RC_{50}$= 15.4 $\mu$g/mL). However, ethyl acetate fraction showed much stronger lipid oxidative inhibition effect than the purified (+)-catechin and (-)-epicatechin.

Isolation and Identification of an Antioxidant Substance from Ethanol Extract of Wild Grape (Vitis coignetiea) Seed (머루종자 에탄올 추출물로부터 항산화활성물질 분리 및 동정)

  • Antioxidant compound(s) were identified from the ethanol extract of wild grape (Vitis coignetiea) seed. Organic solvent fractions of n-hexane, chloroform, ethyl acetate and butanol were obtained from the ethanol extract of wild grape seed, among which ethyl acetate fraction showed the strongest reducing power. Ethyl acetate fraction was further purified through ODS column chromatography and HPLC, and isolated antioxidative active compound was identified through $^1H-NMR$ as (+)-catechin (52.7 g/100 g). (+)-Catechin and ethyl acetate fraction both showed approximately 80% scavenging effect. These results indicated (+)-catechin in the ethyl acetate fraction synergetically interacts with unknown antioxidative compound(s).

Effects of Antioxidant Activities in Ethanol Extract of Apple Peel, Grape Peel, and Sweet Potato Peel as Natural Antioxidant (천연 항산화제로의 활용을 위한 사과, 포도, 및 고구마 껍질 에탄올 추출물의 항산화 효과)

  • This study examined the antioxidant capacities of apple peel, grape peel, and sweet potato peel. The antioxidant activities were evaluated using total phenolic contents, total flavonoids contents, DPPH radical scavenging activity, ABTS radical cation scavenging activity, FRAP reducing power, and ORAC assay. The total phenolic (7.76 ${\mu}M$ quercetin equivalent/g peel) and total flavonoids (1.03 ${\mu}M$ quercetin equivalent/g peel) contents in apple peel were significantly higher than in grape peel and sweet potato peel (P<0.05). The scavenging activities of DPPH and ABTS radicals of a 70% ethanol extract of apple peel was 3.2-4.6 and 2.8-5.4 times high than those of grape and sweet potato peel, respectively. In addition, the FRAP reducing power and ORAC assay of 70% ethanol extraction from apple peel were significantly higher than those of the other samples. Therefore, apple peel can be used efficiently as a natural antioxidant.

Total Polyphenol Content and Antioxidative Activity of Wild Grape (Vitis coignetiae) Extracts Depending on Ethanol Concentrations (에탄올 농도에 따른 머루(Wild Grape, Vitis coignetiae) 추출물의 폴리페놀 함량 및 항산화 활성)

  • As an oriental medicinal plant, wild grape (WG, Vitis coignetiae) has been known to contain abundant nutrients compared to grape (Vitis vinifera L.) and as a source of stilbenes, a kind of polyphenol. This study was performed to investigate the antioxidative activity of WG extracts by measuring electron donating ability (EDA), nitrite scavenging ability (NSA), super oxide dismutase (SOD)-like activity and total polyphenol content (TPC). The extracts were obtained using microwave-assisted extraction (microwave power: 90 W, extraction time: 5 min, extractant: water, 50 and 100% ethanol). EDAs, SOD-like activities and TPCs were the highest at 50% ethanol extracts while conversely lowest at 100% ethanol extracts. EDAs of 50% and 100% ethanol extracts at 1.6 g/dL concentration, $97.70{\pm}0.55\;and\;98.05{\pm}0.36%$, were higher than those of 0.1 and 1% L-ascorbic acids, $101.44{\pm}0.98%\;and\;99.43{\pm}0.78%$, respectively. At the concentration of 1.6 g/dL, 50% ethanol extract showed lower NSA (pH 1.2) than water extract unlike EDA, TPC and SOD-like activity. Regarding TPCs of WG extracts, the activities were the highest at 50% ethanol extracts (1.6 g/dL: $38.76{\pm}0.23$ mM gallic acid equiv.) followed by water and 100% ethanol extracts. The results suggest the usefulness of developing functional foods using antioxidative active compounds of WG with high polyphenol contents.

Improvement Effect of Corn Silk, Perilla Leaf and Grape Stem Extract Mixture against UVB-Induced Skin Damage and Compound 48/80-Induced Pruritus (옥수수수염, 들깻잎 및 포도줄기 복합추출물의 UVB 조사 피부 손상 및 Compound 48/80 유도 가려움증 개선 효과)

  • This study was conducted to evaluate the synergistic protective effects of mixtures of corn silk, perilla leaf and grape stem extract (CPG mixture) against UVB-induced skin damage and compound 48/80-induced pruritus in mice. The results showed that treatment with CPG mixture exhibited much stronger suppressive effect on erythema and melanin index as well as melanin formation than treatment with ascorbic acid (AA) in UVB-irradiated mice. Moreover, the treatment with CPG mixture showed ameliorative effect on immune cell infiltration and collagen fiber destruction in UV-irradiated mice. The treatment with CPG mixture inhibited glutathione (GSH) depletion, lipid peroxidation and production of pro-inflammatory cytokines in UVB-irradiated mice. Furthermore, the treatment with CPG mixture inhibited compound 48/80-induced scratching behavior and histological changes in mice. Taken together, these results indicated that CPG mixture has potentials as functional and therapeutic materials against skin damage and itch-related skin diseases.
