Effect of Ginseng Components (Ginsenosides and Fat Soluble Fraction) on Rat Liver Glucokinase Activity (쥐의 간 Glucokinase 활성에 미치는 인삼 성분의 영향)

  • Effect of ginsenoside mixture, ginsenoside $Rb_1$,$Rb_2$,$Rg_1$ and the fat soluble fraction of the roots of Panax ginseng C.A. Meyer on the activity of glucokinase (GK) in vitro has been observed and found that GK activity was increased about 15c1c at the concentration of ginsenoside mixture and/or the fat soluble fraction being $10^{-7}$,$10^{-5}$%. It was also observed that glucose uptake by rat liver was increased in the presence of either ginsenoside mixture or the fat soluble fraction by perfusion technique. Ginsenoside mixture stimulated various enzymes related to glucose metabolism, however, both ginsenoside mixture and the fat soluble fraction did not stimulate GK activity as expected. Primary culture of liver cells showed that the ginsenoside mixture and the fat soluble fraction increased GK activity significantly and they stimulated the GK activity synergistically in the co-presence of insulin.

Effects of Dietary Fat to Carbohydrate Ratio on Hepatic Glucokinase Activity in Rats (식이 중 지방과 탄수화물 비율이 쥐 간이 Glucokinase의 활성도에 미치는 영향)

  • This study was undertaken to determine the effect of dietary fat to carbohydrate ratio on plasma glucose. free fatty acid level and hepatic glucokinase activity in normal or insulin treated diabetic rats. Sprague-Dawley male rats were fed with 3 different but isocaloric diets for 5 weeks. Diet 1 made to have low fat(4% corn oil and 65.8% corn starch wt/wt) : diet 2 medium fat (12% : 47.8%) : diet 3 high fat (20% : 29.8%) In the normal rats an apparent increase of GK activity was observed from the animal fed low fat diet when compared with other groups. GK activities were decreased in all the alloxan-diabetic rats than the normal rats. When insulin was injected the GK activities in all the livers of alloxan-diabetic rats restored to normal level and GK activity was highest in low fat group. In the entire group significant relationships were seen between the plasma glucose and GK activities(r=-0.6, p<0.001) FFA levels and GK activities(r=-0.63 p<0.001) Both in normal and insulin treated diabetic rats significantly depressed level of hepatic GK activity was observed in the livers of animals fed high fat diet for 5 weeks and depressed level of GK activity may be related to insulin resistance.

Effects of Cordyceps militaris on Key Enzymes of Carbohydrate Metabolism (Cordyceps militaris 성분이 당대사 관련 효소 활성에 미치는 영향)

  • The present study was designed to investigate the antidiabetic effect of Cordyceps militaris on activities of hepatic glucose-regulating enzymes, such as glucokinase (GK), pyruvate dehydrogenase (PDH) and acetyI-CoA carboxylase (ACC). We have isolated the active compounds, CM-A and CM-B from C. militaris and the extracts are under investigation to determine their mechanism of action. Hepatic GK, PDH and ACC activities were significantly (p < 0.05) increased compared with the control. Treatment with CM-A led to a rise in percentage of enzymes by 380$\%$,396$\%$ and 286$\%$, respectively, relative to control levels. CM-B were 329$\%$, 312$\%$ and 239$\%$, respectively. The increase in GK and ACC activities was linearly proportional with increased ratio of CM-A. Our findings suggest that C. militaris exerted antihyperglycemic potency, which is thought to be mediated through activation of GK, PDH and ACC enzymes related to glucose metabolism directly or indirectly and therefore C. militaris is promising as antidiabetic functional foods.

Antidiabetic Effect of Ethanol Extract on Phelladindron amurense Rupr (황백 에탄올 추출물의 혈당강하작용)

  • This study was performed to investigate the antidiabetic effect of ethanol extract of Phelladrindron Amurense Rupr (P.A) in Streptozotocin(STZ) induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg,b.w. dissolved in citrate buffer. The ethanol extract of P. A was orally administrated once a day for 7 days at a dose of 1,000mg/kg. The content of serum glucose, was significantly decreased in P.A treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucokinase(GK) and glucose-6-phosphate dehydrogenase(G-6-PDH) were significantly increased(p<0.05), and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased(p<0.05) in P.A treated group compared to those of STZ-control group, These results indicated that ethanol extract of P.A have antidiabetic effect in STZ-induced diabetic rats.

Effects of Acanthopanax senticosus Water Extract on Glucose-Regulating Mechanisms in HepG2 Cells (가시오갈피 물 추출물이 간세포에서 포도당 이용 대사에 미치는 영향)

  • This study aimed to investigate glucose uptake mechanisms and metabolic mechanisms for absorbed glucose in HepG2 cells treated with Acanthopanax senticosus water extract (ASW). A colorimetric assay kit was used to measure polyphenol content, glucokinase (GK) activity, glucose uptake, glucose consumption in cell culture medium, and glycogen content. RT-PCR and western blotting were performed to examine changes in the expression levels of glucose transporter 2 (GLUT2), hepatocyte nuclear factor $1{\alpha}$ ($HNF-1{\alpha}$), phosphatidylinositol 3-kinase (PI3k), protein kinase B (Akt), phospho-AMP-activated protein kinase (AMPK), phosphoenolpyruvate carboxykinase, GK, and glycogen synthase kinase $3{\beta}$ ($GSK3{\beta}$). Increased glucose uptake upon ASW treatment was confirmed to result from increased expression of $HNF-1{\alpha}$, which is one of the transcription factors acting on the GLUT2 promoter. From the measurements of GK activity, we observed that ASW had an effect on glucose phosphorylation, and we also confirmed that increased AMPK phosphorylation promoted glycolysis and suppressed gluconeogenesis. We confirmed that the increase in glycogen upon ASW treatment was induced by activation of Akt by PI3k, followed by phosphorylation of $GSK3{\beta}$. This study demonstrates that ASW activates glucose metabolic mechanisms in liver cells and is therefore a potential candidate to alleviate diabetes.

Anti-diabetic Effects of Mixed Extracts from Lycium chinense, Cordyceps militaris, and Acanthopanax senticosus (지골피, 동충하초, 가시오가피 혼합 추출물의 항당뇨 작용)

  • In this study, we examined the anti-diabetic activity in vitro by the mixed Korean herb water extracts(JDG) composed of Lycium chinense, Cordyceps militaris, and Acanthopanax senticosus on hepatic glucose-regulating enzyme activities such as glucokinase(GK), acetyl-CoA carboxylase(ACC), and inhibition activity of $\alpha$-glucosidase. The hepatic cytosol fraction of a type II diabetic animal(Goto-Kakizaki rat) was used in GK and ACC activity assays. JDG mixed water extracts significantly increased the hepatic GK and ACC activity. The highest anti-$\alpha$-glucosidase activity was observed in JDG 5 water extract when compared to the acarbose control, inhibitor of $\alpha$-glucosidase. We suggest that Lycium chinense, Cordyceps militaris, and Acanthopanax senticosus mixed water extracts may exert an anti-diabetic activity by enhancing the glucose metabolism and may be used as natural $\alpha$-glucosidase inhibitors in type 2 diabetic conditions.

  • This study investigated the anti-diabetic effects of the mixed water extract (JDG 100) composed of Lycii Cortex, Acanthopanax senticosus and Cordyceps militaris on glucose-regulating key enzymes such as glucokinase (GK), acetyl-CoA carboxylase (ACC). In the current study, HepG2 cells were exposed to pathological condition such as hyperglycemic condition (4.5 g glucose/L) with JDG 100 and then experiments such as RT-PCR and Western blotting were carried out. JDG 100 treated cells increased to $168{\pm}0.04%$ and $182.4{\pm}0.03%$ in GK mRNA and protein expressions, respectively, compared to control. Treatment of the JDG 100 up-regulated ACC mRNA ($127.3{\pm}0.02%$) and protein ($126.7{\pm}0.24%$) of HepG2 cells in the high glucose media. These observations suggest that JDG 100 mixed water extract may have a potential as an anti-diabetic agent in type 2 diabetes mellitus.

Antidiabetic Effect of ethanol extract of Forsythia Koreana in Streptozotocin-Induced Diabetic Rat (Streptozotocin에 의해 유도된 당뇨쥐에서 연교의 에탄올 추출물의 당뇨 개선 효과)

  • This study was carried to investigate the antidiabetic effect of ethanol extract in Streptozotocin(STZ)-induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 45mg/kg dissolved in citrate buffer. The ethanol extract of Forsythia Koreana(F.K) was orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in F.K treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased, and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in F.K treated group compared to the those of STZ-control group, but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was not significantly increased, These results indicated that ethanol extract of F.K would have antidiabetic effect in STZ-induced diabetic rats.

Diabetes was induced by intravenous injection of STZ at a dose 45mg/kg.b.w. dissolved in citrate buffer(pH4.5). The ethanol extract of Codonopsis lanceolata root was orally administrated once a day for 7 days. The contents of serum glucose, triglyceride(TG) and total cholesterol were significantly decreased(p<0.05) in Codonopsis lanceolata root treated group compared to the those of STZ-control group. Also the content of hepatic glycogen and activities of glucose-phosphate dehydrogenase(G-6-PDH) and glucokinase(GK) were significamtly increased(p<0.05). These results indicated that ethanol extract of Codonopsis lanceolata root would have antidiabetic effect in STZ-induced diabetic rats.

The Effect of Platycodon grandiflorum Root Ethanol Extract on Blood Glucose, Lipid, Activities of Carbohydrate Metabolism Related Enzyme in Streptozotocin-Induced Diabetic Rats (도라지 뿌리 에탄올 추출물이 streptozotocin으로 유발된 흰쥐의 혈당지질, 당대사에 미치는 영향)

The ethanol extract of Platycodon grandiflorum root was orally administrated once a day for 7 days. The contents of serum glucose, triglyceride(TG) and total cholesterol were significantly decreased(p<0.05) in Platycodon grandiflorumt root treated group compared to the those of STZ-control group. Also the contents of hepatic glycogen and HDL-cholesterol, the activities of glucose-phosphate dehydrogenase(G-6-PDH) and glucokinase(GK) were significamtly increased (p<0.05). These results indicated that ethanol extract of Platycodon grandiflorum root would have antidiabetic effect in STZ-induced diabetic rats.
