Effects of Korean ginseng berry on skin antipigmentation and antiaging via FoxO3a activation

  • Background: The ginseng berry has various bioactivities, including antidiabetic, anticancer, antiinflammatory, and antioxidative properties. Moreover, we have revealed that the active antiaging component of the ginseng berry, syringaresinol, has the ability to stimulate longevity via gene activation. Despite the many known beneficial effects of ginseng, its effects on skin aging are poorly understood. In this study, we investigated the effects of ginseng and the ginseng berry on one of the skin aging processes, melanogenesis, and age-related pigment lipofuscin accumulation, to elucidate the mechanism of action with respect to antiaging. Methods: The human melanoma MNT1 cell line was treated with ginseng root extract, ginseng berry extract, or syringaresinol. Then, the cells were analyzed using a melanin assay, and the tyrosinase activity was estimated. The Caenorhabditis elegans wild type N2 strain was used for the life span assay to analyze the antiaging effects of the samples. A lipofuscin fluorescence assay was performed during 10 passages with the syringaresinol treatment. Results: A 7-d treatment with ginseng berry extract reduced melanin accumulation and tyrosinase activity more than ginseng root extract. These results may be due to the active compound of the ginseng berry, syringaresinol. The antimelanogenic activity was strongly coordinated with the activation of the longevity gene foxo3a. Moreover, the ginseng berry extract had more potent antiaging effects, caused a life span extension, and reduced lipofuscin accumulation. Conclusion: Taken together, our results suggest that these antimelanogenic effects and antiaging effects of ginseng berry mediate the activation of antioxidation-FoxO3a signaling.

고려인삼의 혈압에 미치는 영향에 대한 이해 (Toward a More Complete Understanding of the Effects of Korean Ginseng (Panax ginseng) on Blood Pressure)

  • There is no doubt that the effect of ginseng on blood pressure could be different depending upon the type of ginseng employed for the experiment and methodology, thereby can exert bilateral modulatory activity on blood pressure. It has been reported that ginseng induced no significant change in blood pressure in those subjects with normal blood pressure, but had a normalizing effect on the subjects with abnormal blood pressure. Especially, experimental evidence indicates that ginsenoside Rg3, a major component of red ginseng, has been found to lower blood pressure, which is mediated by release of endothelium-derived NO, enhancing the accumulation of cGMP in the rat aorta. This clinical results further support the beneficial effect of Korean ginseng on blood pressure elucidated by animal experiment. As expected, a multicentric non-controlled clinical study shows that the effect of ginseng consumption has been found to normalize blood pressure in hypertensive or hypotensive individuals as compared to virtually no effect in normotensives. In addition, ginseng has been known to exhibit blood pressure decreased with no significant side effect and deteriorated QOL during the combination therapy of ginseng and anti-hypertensive drugs. This review provides a comprehensive overview on the effects of Korean ginseng on blood pressure.

  • This study is conducted to evalute the effect of ginseng fraction component (ginseng extract solution, GES; ginseng protein, GP; ginseng saponin, GSA; ginseng residue, GR) upon hyperlipidemia and fatty liver induced by high fat administration. In doing so, the serum, liver and epididymal adpoid tissue have been examined for lipid components level and lipoprotein fraction. Feces bile acid and neutral sterol excretion have been also measured. 1'he results obtained from this study are as follows. 1. Serum, liver, epididymal lipid components of GP and GSA group were significantly lower than the controlgroup. 2. During the feeding experiment, VLDL and LDL increase while HDL decrease in all group. However the degree of VLDL and LDL increase and HDL decrease were signficantly small in GP and GSA group compared with control group. 3. In the excretion of bile acid and neutral sterol, all experiment group showed increased excretion in the comparison of control group.

RAPD Pattern of Ginseng(Panax ginseng C.A. Meyer) Lines Containing High Level of Ginsenoside

  • The important component for medical effect in ginseng is ginsenoside. Korea Ginseng & Tobacco Research Institute contains approximately 200 lines produced by inbred selection. It is assumed that ginseng lines containing high level of ginsenoside should be included in those lines. Besides, new breeding methods such as cell line selection in vitro and hairy root were recently developed. Therefore, this study was carried out to detect genes related to ginsenoside, and to use it for selection marker to select and distribute lines containing high level of ginsenoside. DNA was extracted from both ginseng roots and hairy roots, and the difference between the line containing high ginsenoside(KG101) and normal ginsenoside(KG103) were analysed. As a result, 28 out of 36 primers showed bands, and many primers showed band difference between ginseng lines. It is considered that the bands should be analysed using DNA sequence comparison to check if those are related to ginsenoside. In case of hairy roots of ginseng, almost no differences were found between two lines.

Study on life span extension efficacy by Korean Red Ginseng

  • The backbone structure of ginsenosides, active ingredients of Panax ginseng, is similar with that of sterol, especially cholesterol. Caenorhabditis elegans (c. elegans) is one of free living nematodes and is well-established animal model for biochemical and genetic studies. C. elegans cannot synthesize de novo cholesterol, although cholesterol is essential requirement for its growth and development. In the present study, we investigated the effects of Korean red ginseng total extract (KRGE), ginseng total saponins (GTS) on life span of C. elegans in cholesterol-deprived and -fed medium. Cholesterol deprivation caused damages on life span of worms throughout F1 to F3 generations. KRGE or GTS supplement to cholesterol-deprived medium restored the life span of worms as much as cholesterol alone-fed medium. In study to identify which ginsenosides are responsible for life span restoring effects of KRGE, we found that ginsenoside Rc supplement not only restored life span of worms grown in cholesterol-deprived medium but also prolonged life span of worms grown in cholesterol-fed medium. These results show a possibility that ginsenosides could be utilized by C. elegans as a sterol substitute and further indicate that ginsenoside Rc is the effective component of Korean red ginseng that prolongs the life span of C. elegans.

Solvent Free Solid Injector (SFSI)를 이용한 수삼, 홍삼, 백삼의 향기성분 분석방법 (The Analysis of Volatile Components of Fresh Ginseng, Red Ginseng and White Ginseng by Solvent Free Solid Injector (SFSI) Techniques)

  • 인삼의 휘발성 성분 분석 시 SFSI를 이용한 분석법의 최적화를 일하여 실험을 수행하였다. 최적화를 위하여 세 가지 시료조제방법인 생체시료, oven건조와 동결건조를 검토하였고, GC injector 온도는 100, 150, 200, 250, $300^{\circ}C$에서의 휘발성분 분석효율을 검토하였으며, 모세관에서 시료의 기화 시간인 preheating time은 3, 5, 7, 10, 15 min의 조건에서 검토한 결과, 시료 조제방법은 생체시료를 그대로 사용 시, GC injector 온도는 $250^{\circ}C$일 때 preheating time은 10min일 때 가장 분석효율이 높았다. 휘발성분 분석을 위해 사용된 인삼은 금산에서 재배한 6년근 삼으로 수삼, 백삼, 홍삼을 사용하였으며, SFSI가 장착된 CC/MS에 의해 휘발성분을 분석, 동정하였다. 분석결과 수삼에서는 33종, 백삼에서는 36종, 홍삼에서는 38종의 휘발성분이 분석되어져 SFSI에 의한 분석법이 인삼의 휘발성분을 분석, 동정하는 방법으로 이용될 수 있음을 보여주고 있다.

Remarkable impact of steam temperature on ginsenosides transformation from fresh ginseng to red ginseng

  • Background: Temperature is an essential condition in red ginseng processing. The pharmacological activities of red ginseng under different steam temperatures are significantly different. Methods: In this study, an ultrahigh-performance liquid chromatography quadrupole time-of-flight tandem mass spectrometry was developed to distinguish the red ginseng products that were steamed at high and low temperatures. Multivariate statistical analyses such as principal component analysis and supervised orthogonal partial least squared discrimination analysis were used to determine the influential components of the different samples. Results: The results showed that different steamed red ginseng samples can be identified, and the characteristic components were 20-gluco-ginsenoside Rf, ginsenoside Re, ginsenoside Rg1, and malonyl-ginsenoside Rb1 in red ginseng steamed at low temperature. Meanwhile, the characteristic components in red ginseng steamed at high temperature were 20R-ginsenoside Rs3 and ginsenoside Rs4. Polar ginsenosides were abundant in red ginseng steamed at low temperature, whereas higher levels of less polar ginsenosides were detected in red ginseng steamed at high temperature. Conclusion: This study makes the first time that differences between red ginseng steamed under different temperatures and their ginsenosides transformation have been observed systematically at the chemistry level. The results suggested that the identified chemical markers can be used to illustrate the transformation of ginsenosides in red ginseng processing.

백삼 및 백미삼 추출물의 초단파 및 식초 처리에 의한 인삼 사포닌 성분 변화 (The Change of Ginsenoside Composition in White Ginseng and Fine White Ginseng Extract by the Microwave and Vinegar Process)

  • The purpose of this study is to develop a new preparation process of ginseng extracts having high concentrations of ginsenoside $Rg_3$, $Rg_5$ and $Rk_1$, a special component of Red ginseng. Chemical transformation from ginseng saponin glycosides to prosapogenin was analyzed by the HPLC. Extracts of White ginseng (Panax ginseng) and Fine White ginseng were processed under several treatment conditions including microwave and vinegar (about 14% acidity) treatments. Results of those treatments showed that the quantity of ginsenoside $Rg_3$ increased by over 0.6% at 4 minutes of pH 2~4 vinegar and microwave treatments. The results of processing with MWG-4 indicate that the Microwave and vinegar processed white ginseng extracts (about 14% acidity) that had gone through 4-minute treatments were found to contain the largest amount of ginsenoside $Rg_3$ (0.626%), $Rg_5$ (0.514%) and $Rk_1$ (0.220%). Results of treatments with MFWG-5 showed that the Fine White ginseng extracts that had been processed with microwave and vinegar (about 14% acidity) for 5 minutes were found to contain the largest amount of ginsenoside $Rg_3$ (4.484%), $Rg_5$ (3.192%) and $Rk_1$ (1.684%). It is thought that such results provide basic information in preparing White ginseng and Fine White ginseng extracts with functionality enhanced.

전자코를 이용한 홍삼 농축액의 원산지 판별을 위한 향기패턴 분석 (Analysis of Aroma Pattern for Geographical Origin of Red Ginseng Concentrated by Electronic Nose)

  • 중국산과 국내산 홍삼 농축액의 혼합비율에 따는 원산지 판별 가능성을 검토하기 위해 전자코를 이용하여 향기 패턴을 분석하였다. 중국산 홍삼 농축액과 국내산 홍삼 농축액의 원산지 판별이 가능하였고 중국산 홍삼농축액의 혼합비율이 증가할수록 검출되어지는 향기 성분의 패턴은 감소하는 경향을 나타내었다. Frequency pattern, derivative pattern을 Vapor printTM 으로 도형화 하여 비교한 결과 서로 다른 패턴을 보여주어 중국산 홍삼 농축액 첨가비율에 따른 차이는 물론 원산지의 차이도 뚜렷하게 나타났다.

人蔘이 햄스터의 血色素와 A/G率에 미치는 영향에 관하여 (The Effect of Ginseng on the Hamster's Hemoglobin A/G Ratio in the Serum)

  • 햄스터에 대하여 水蔘을 1日 1g 씩 25日간 給與하였던 바, 다음과 같은 實驗結果를 얻었다. 1) Hb 量은 人蔘投與 10日부터 급격한 有意性 增加를 보여준다. 2) 人蔘을 給與한 뒤 15日부터 血淸總蛋白의 含量은 有意性 증가를 나타낸다. 3) 人蔘을 給與한 뒤, 10日부터 A/G率은 차차로 增加 되는 것 같이 보이지만 有意性 增加는 25日부터 나타나고 있다. 이는 globulin 보다 albumin의 含量이 더욱 增加됨을 의미한다. 요컨데 햄스터에 대하여 人蔘을 給與한 뒤, 25日간의 결과를 보명 Hb, 血淸總蛋白量 및 A/G率이 增加됨을 알게된 것이다.
