  • The constituents of Sanguisorba hakusanensis leaves (Rosaceae), of which the roots have been used as an astringent, hemostatics and antiphlogistics, were studied phytoche mically. From water fraction of the MeOH extract, gallic acid 3-O-${\beta}$-D-(6'-O-galloyl)-glucopyranoside(I), quercetin-3-O-${\beta$-D-galactopyranoside(II), quercetin-3-O-${\alpha}$-L-arabinopyranoside(III) and $2{\alpha},\;3{\beta},\;19{\alpha}$, 23-tetrahydroxyurs-12-en-28-oic acid 28-O-${\beta}$-D-glucopyranoside(IV) were isolated by column chromatographic separation using Amberlite XAD-2, ODS-gel and Sephadex LH-20. The structure of these compounds were elucidated by spectroscopic parameters of $^1H$-NMR, $^{13}C$-NMR, $^1H-^1H$ COSY, $^{13}C-^1H$ COSY, EI-Mass, FAB-Mass, IR, UV and by comparison with authentic samples.

  • The ethyl acetate (EtOAc) layer of Canavalia gladiata (sword bean) methanol extracts showed higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging activity than other layers. Four phenolic compounds were isolated from the EtOAc layer by silica gel column chromatography and prep-HPLC using a guided DPPH radical-scavenging assay. The isolated compounds were identified as methyl gallate (1), gallic acid (2), 1,6-di-O-galloyl ${\beta}$-$\small{D}$-glucopyranoside (3), and 1,4,6-tri-O-galloyl ${\beta}$-$\small{D}$-glucopyranoside (4) based on MS and NMR analyses. Among the four compounds, no. 4 was isolated from this plant for the first time. Their DPPH radical-scavenging activities based on $SC_{50}$ decreased in the following order: 4 (6.9 ${\mu}M$)>3 (8.3 ${\mu}M$)>2 (10.0 ${\mu}M$)>1 (10.3 ${\mu}M$).

Their structures were identified as genistein (1), sophoricoside (2), genistein-4'-O--L-rhamnopyranoside (3), $genistein-4'-O-{\alpha}-L-rhamnopyranosly-(1{\rightarrow}2)-{\beta}-D-glucopyranoside$ (4), $kaempferol-3-O-{\alpha}-D-sophoroside$ (5), $kaempferol-3-O-{\beta}-D-glucopyranosyl-(1{\rightarrow}2)-{\alpha}-L-rhamnopyranosly-(1{\rightarrow}6)-{\beta}-D-glucopyranoside$ (6), rutin (7) and gallic acid $4-O-{\beta}-D-(6'-O-galloyl)-glucopyranoside$ (8) by chemical and spectroscopic analysis and comparisons with previously reported spectral data. Compounds 3 and 8 were isolated for the first time from this plant. Anti-oxidative activity was evaluated for the isolated compounds. 8 exhibited potent anti-oxidative activity against the radical scavenging ability of DPPH with the $IC_{50}$ value of $17.1\;{\mu}g/ml$.

  • As part of ongoing study focused on the discovery of natural antioxidants from Korean plants by measuring the DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging effect and superoxide quenching activity, ethanol extract of the aerial parts of Curcuma longa (Zingiberaceae) was found to show potent antioxidant activities. Subsequent activity-guided fractionation of the ethanol extract led to the isolation of two phenolic compounds, 1,2,3,4,6-penta-O-galloyl-${\beta}$-D-glucopyranoside (1) and gallic acid (2), as antioxidant compounds. Their structures were elucidated by spectroscopic studies. Compounds 1 and 2 were isolated for the first time from this plant. These compounds showed the significant antioxidative effects during the DPPH free radical scavenging test, and the riboflavin- and xanthine-originated superoxide quenching activity tests.
