Daraesoon (shoot of hardy kiwi) mitigates hyperglycemia in db/db mice by alleviating insulin resistance and inflammation

  • BACKGROUND/OBJECTIVES: Mitigating insulin resistance and hyperglycemia is associated with a decreased risk of diabetic complications. The effect of Daraesoon (shoot of hardy kiwi, Actinidia arguta) on hyperglycemia was investigated using a type 2 diabetes animal model. MATERIALS/METHODS: Seven-week-old db/db mice were fed either an AIN-93G diet or a diet containing 0.4% of a 70% ethanol extract of Daraesoon, whereas db/+ mice were fed the AIN-93G diet for 7 weeks. RESULTS: Consumption of Daraesoon significantly reduced serum glucose and blood glycated hemoglobin levels, along with homeostasis model assessment for insulin resistance in db/db mice. Conversely, Daraesoon elevated the serum adiponectin levels compared to the db/db control group. Furthermore, Daraesoon significantly decreased both serum and hepatic triglyceride levels, as well as serum total cholesterol levels. Additionally, consumption of Daraesoon resulted in decreased hepatic tumor necrosis factor-α and monocyte chemoattractant protein-1 expression. CONCLUSIONS: These results suggest that hypoglycemic effect of Daraesoon is mediated through the improvement of insulin resistance and the downregulation of pro-inflammatory cytokine expression in db/db mice.

Proteomic Analysis of Proteins Increased or Reduced by Ethanol of Lactobacillus plantarum ST4 Isolated from Makgeolli, Traditional Korean Rice Wine

  • LAB were isolated from makgeolli locally produced around Jinju, Gyeongnam, S. Korea during spring of 2011. Randomly selected 11 isolates from MRS agar plates were identified first by API CHL 50 kits and then 16S rRNA gene sequencing. All 11 isolates were identified as Lactobacillus plantarum. Among them, ST4 grew in MRS broth with ethanol up to 10%, showing the highest alcohol resistance. L. plantarum ST4 was moderately resistant against acid and bile salts. When cellular proteins of L. plantarum ST4 under ethanol stress were analyzed by two-dimensional gel electrophoresis (2DE), the intensities of 6 spots increased, whereas 22 spots decreased at least 2-fold. Those 28 spots were identified by peptide mass fingerprinting (PMF). FusA2 (elongation factor G) increased 18.8-fold (6% ethanol) compared with control. Other proteins were AtpD (ATP synthase subunit beta), DnaK, GroEL, Tuf (elongation factor Tu), and Npr2 (NADH peroxidase), respectively. Among the 22 proteins decreased in intensities, lactate dehydrogenases (LdhD and LdhL1) were included.

Comparative Studies on Polymorphism and Fithess between Two ADH Alleles in Drosophila melanogaster (Deosophila melanogadter의 ADH Polymorphism 과 두 유전자 사이의 적응성에 관한 비교 연구)

  • Tne present studies were camied out to ~nvestigate the allele frequency variations of alcohol dehydrogenase (ADH) in natural populat~ons of Drosophiio melonogoster and the correlations of iwo ADH alleles between fitness and ethanol. ADH alleles were found to be polymorphic in natural populations of D. rnelanogaster. The frequencies of FF, FS and SS genotypes were 47.66, 42 18, and 10.16%. respectively, therefore the F gene frequency (68.75) was shown to be hlgher than the S gene (31.25 %). The FF genotype was slightly superior to the SS genotype in both fecundiiy and eclaslon. The frequency of AdhF allele in the small alt>fic~apl opulaliow originated from natural populations was increased for 20 generations on normal media at 25$^{\circ}$C In resistance to ethanol, the FF genotype was supenor to the SS genotype, too. It meant that ethanol as environmental factor might be the selective factor on ADH locus in natural populat~ons of D meionogoster.

Biocontrol of Biofilm-forming Bacillus cereus by Using Organic Acid, Ethanol, and Sodium Chloride (생물막 형성 Bacillus cereus에 대한 유기산, 에탄올 및 NaCl의 제어효과)

  • Food poisoning by Bacillus cereus is one of the common food-borne diseases and B. cereus is widely distributed in natural and commercial products owing to the strong resistance caused by biofilm or spore. The ethanol, NaCl, and organic acids of acetic acid, citric acid, and lactic acid for biocontrol of biofilm-forming B. cereus on glass wool were investigated. The biofilm on glass wool was observed in many developments after 48 h incubation. As the results of reduction of biofilm-forming B. cereus by sanitizers, reduction levels of each organic acid treatment ranged to 5-6 log CFU/g-glass wool. In case of combination treatments of 20% ethanol, 10% NaCl, and 1% of each organic acid for 1-5 min, the reduction level of biofilm-forming B. cereus was 7-8 log CFU/g-glass wool. Therefore, combination treatments of ethanol, NaCl, and an organic acid might effectively reduce biofilm-forming B. cereus in various food processes and industries.

Bimetallic Zeolitic Imidazolate Framework Derived Co3O4/CoFe2O4 Catalyst Loaded In2O3 Nanofibers for Highly Sensitive and Selective Ethanol Sensing (금속-유기 골격체 열분해를 통해 합성된 Co3O4/CoFe2O4 첨가 In2O3나노섬유를 이용한 고감도 고선택성 에탄올 센서)

  • In this study, pure and Co3O4/CoFe2O4-loaded Indium oxide (In2O3) nanofibers were synthesized by the electrospinning of an Indium/Polyvinylpyrrolidone precursor solution containing cobalt and iron bimetallic zeolitic imidazolate frameworks and subsequent heat treatment. The ethanol, toluene, p-xylene, benzene, carbon monodxide, and hydrogen gas sensing characteristics of the solution were measured at 250-400 ℃. 0.5 at%-Co3O4/CoFe2O4-loaded In2O3 nanofibers exhibited extreme response (resistance ratio - 1) to 5 ppm of ethanol (210.5) at 250 ℃ and excellent selectivity over the interfering gases. In contrast, pure In2O3 nanofibers exhibited relatively low responses to all the analyte gases and low selectivity above 250-400 ℃. The superior response and selectivity toward ethanol is explained by the catalytic roles of Co3O4 and CoFe2O4 in gas sensing reaction and the electronic sensitization induced by the formation of p (Co3O4/CoFe2O4)-n (In2O3) junctions.

Antibacterial Activity and Inhibition of Resistance in Methicillin-resistant Staphylococcus aureus by Maneung-hwan Ethanol Extract (만응환(萬應丸) 에탄올 추출물의 메티실린 내성 포도상구균에 대한 항균활성 및 내성억제 효과)

  • Objectives In this study, we investigated the antimicrobial activity of a 70% ethanol extract of Maneung-hwan (MEH), which is prescribed by practitioners of oriental medicine for use against methicillin-resistant Staphylococcus aureus (MRSA). Methods The antibacterial activity of MEH against MRSA strains was evaluated using the disc diffusion method, broth microdilution method (minimal inhibitory concentration, MIC), checkerboard dilution test, and time-kill test. The mechanism of action of MEH was investigated by bacteriolysis using detergents or ATPase inhibitors Additionally, mRNA and protein expression were investigated by quantitative reverse transcription-polymerase chain reaction and western blot assay, respectively. Results The MIC of MEH was 25~1,600 ㎍/mL against all the tested bacterial strains. We showed that MEH extract exerts strong antibacterial activity. In the checkerboard dilution test, the fractional inhibitory concentration index of MEH in combination with antibiotics indicated synergism or partial synergism against S. aureus. The time-kill study indicated that the growth of the tested bacteria was considerably inhibited after a 24-h treatment with MEH and selected antibiotics. To measure the cell membrane permeability, MEH (3.9 ㎍/mL) was combined with Triton X-100 (TX) at various concentrations N,N-dicyclohexylcarbodimide (DCCD) was also tested as an ATPase inhibitor. TX and DCCD cooperation against S. aureus exhibited synergistic action. Accordingly, the antimicrobial activity of MEH in the context of cell membrane rupture and ATPase inhibition was assessed. Additionally, the expression of genes and proteins associated with resistance was reduced after exposing MRSA to MEH. Conclusions These results suggest that MEH possesses antibacterial activity and acts as a potential natural antibiotic against MRSA.

  • Thermal performance and flow patterns inside the closed loop pulsating heat pipe (CLPHP) were experimentally investigated. For investigating the effect of working fluids, CLPHP was filled with various working fluids including methanol, acetone and ethanol. The thermal resistance was calculated by temperatures in evaporator and condenser and flow patterns were visualized by a digital camera. The thermal resistances for all fluids were decreased as the heat increases. Flow patterns change from static slug to elongated slug flows, bulk circulation and annular flows as the heat increases. Dry-out occurs after annular flows. For reasonable comparison of thermal performances, normalized CHF, Kutateladze number (Ku), was compared. Even though ethanol has smallest CHF, Ku of ethanol is similar with that of methanol. In addition, acetone has the highest Ku that means CLPHP with acetone provides the higher thermal performance compared with CLPHP with other fluids.

  • This paper is aimed to study the effect of Pd activator, the annealing temperature, and operating temperatures on the response characteristics of the $SnO_2/Al_2O_3$ sensor. The resistance of device has shown minimum value when annealing temperature and operating temperature of device are $550^{\circ}C$ and $350^{\circ}C$ respectively in ethanol gas. And the response characteristics of the device showed the best results when lwt% Pd was added to SnOz especially in low concentration of ethanol gas.

Comparison of Ga-doped and Ag-doped ZnO Nanowire Gas-sensor Sensitivity and Selectivity

  • Pure ZnO, ZnO nanowires doped with 3 wt.% Ga (3GZO) and doped with 3 wt.% Ag (3SZO) were grown by a hot-walled pulse laser deposition (HW-PLD) technique. The optical and chemical properties of Ga and Ag doped nanowires was analyzed. Nanowires were determined to be under 200 nm in diameter and several μm in length. Change of significant resistance was observed and the gas detection sensitivities of ZnO, 3GZO and 3SZO nanawires were compared. The sensitivities of ZnO, 3GZO, and 3SZO nanowire sensors were measured at 300℃ for 1 ppm of ethanol gas at 97%, 48%, and 203%, respectively.

Antibacterial Activity and Enhancing Antibiotic Effect of Extract and Fractions from Curcuma longa against MRSA Strain (MRSA에 대한 울금 추출 및 분획물의 항균활성과 항생제 증강 효과)

  • Curcumin content of butanol fraction from C. longa was found to be 22.4942% of the highest content. However, in DPPH radical scavenging ability and antibacterial activity against methicillin resistance Staphylococcus aureus(MRSA, CCARM3696), ethylacetate fraction contained 2.5791% of curcumin was exhibited highest activity. In comparison of enhancing antibiotic(ampicillin) effect against MRSA, ethanol extract contained 1.7838% of curcumin showed more strong activity. This indicates that the ethanol extract and some fractions from C. longa can have antibacterial activity and enhancing antibiotic effect possibly without curcumin. Appropriate use of antimicrobial agent was important point prior to the development of new antibiotics. And in that sence, extract and fractions of C. longa were worth using as synergist of antibiotics and natural antimicrobial agent.
