Enhanced Antioxidant Enzymes Are Associated with Reduced Hydrogen Peroxide in Barley Roots under Saline Stress

  • Antioxidant enzymes are related to the resistance to various abiotic stresses including salinity. Barley is relatively tolerant to saline stress among crop plants, but little information is available on barley antioxidant enzymes under salinity stress. We investigated temporal and spatial responses of activities and isoform profiles of superoxide dismutase (SOD), catalase (CAT), ascorbate peroxidase (APX), non-specific peroxidase (POX), and glutathione reductase (GR) to saline stress in barley seedlings treated with 200 mM NaCl for 0, 1, 2, 5 days, respectively. In the control plant, hydrogen peroxide content was about 2-fold higher in the root than in the shoot. Under saline stress, hydrogen peroxide content was decreased drastically by 70% at 2 d after NaCl treatment (DAT) in the root. In the leaf, however, the content was remained unchanged by 2 DAT and increased about 14 % at 5 DAT. In general, the activities of antioxidant enzymes were increased in the root and shoot under saline stress. But the increase was more significant and consistent in the root. The activities of SOD, CAT, APX, POX, and GR were increased significantly in the root within 1 DAT, and various elevated levels were maintained by 5 DAT. Among the antioxidant enzymes, CAT activity was increased the most drastically. The significant increase in the activities of SOD, CAT, APX, POX, and GR in the NaCl-stressed barley root was highly correlated with the increased expression of the constitutive isoforms as well as the induced ones. The hydrogen peroxide content in the root was most highly correlated with the CAT activity, indicating an increased role of CAT in hydrogen peroxide detoxification under salinity stress. In addition, the results suggest the significance of temporal and spatial regulation of each antioxidant isoform in determining the competence of the antioxidant capacity under saline stress.

Cancer Chemopreventive Effects of Korean Seaweed Extracts

  • Cancer chemopreventive effects can be exerted through the induction of phase II detoxification enzymes and the inhibition of inflammatory responses. In this study, the cancer chemopreventive effects and anti-inflammatory responses of 30 seaweed extracts were examined. The extracts of Dictyota coriacea and Cutleria cylindrica exhibited the high chemoprevention index, having 4.36 and 4.66, respectively. They also activated antioxidant response element at $100\;{\mu}g/mL$ by about 3-fold while did not activate xenobiotic response element. Seven seaweed extracts, Ishige okamurae, Desmarestia ligulata, Desmarestia viridis, Dictyopteris divaricata, D. coriacea, Sargassum horneri, and Sargassum yezoense, showed significant inhibition on nitric oxide (NO) and prostaglandin $E_2$ ($PGE_2$) production in a dose-dependant manner in $5-20\;{\mu}g/mL$. These seaweed extracts could be used as food materials for cancer chemoprevention. D. coriacea could contain potential chemopreventive agents not only that regulate genes via an ARE-dependent mechanism but also prevent the inflammation through inhibition of NO and $PGE_2$ production.

The effect of glutathione S-transferase M1 and T1 polymorphisms on blood pressure, blood glucose, and lipid profiles following the supplementation of kale (Brassica oleracea acephala) juice in South Korean subclinical hypertensive patients

  • BACKGROUND/OBJECTIVES: Glutathione S-transferase (GST) forms a multigene family of phase II detoxification enzymes which are involved in the detoxification of reactive oxygen species. This study examines whether daily supplementation of kale juice can modulate blood pressure (BP), levels of lipid profiles, and blood glucose, and whether this modulation could be affected by the GSTM1 and GSTT1 polymorphisms. SUBJECTS/METHODS: 84 subclinical hypertensive patients showing systolic BP over 130 mmHg or diastolic BP over 85 mmHg received 300 ml/day of kale juice for 6 weeks, and blood samples were collected on 0-week and 6-week in order to evaluate plasma lipid profiles (total cholesterol, triglyceride, HDL-cholesterol, and LDL-cholesterol) and blood glucose. RESULTS: Systolic and diastolic blood pressure was significantly decreased in all patients regardless of their GSTM1 or GSTT1 polymorphisms after kale juice supplementation. Blood glucose level was decreased only in the GSTM1-present genotype, and plasma lipid profiles showed no difference in both the GSTM1-null and GSTM1-present genotypes. In the case of GSTT1, on the other hand, plasma HDL-C was increased and LDL-C was decreased only in the GSTT1-present type, while blood glucose was decreased only in the GSTT1-null genotype. CONCLUSIONS: These findings suggest that the supplementation of kale juice affected blood pressure, lipid profiles, and blood glucose in subclinical hypertensive patients depending on their GST genetic polymorphisms, and the improvement of lipid profiles was mainly greater in the GSTT1-present genotype and the decrease of blood glucose was greater in the GSTM1-present or GSTT1-null genotypes.

  • NAD(P)H:quinone reductase (QR) is one of the protective phase II enzymes against toxicity that accomplishes the capacity of detoxification by modulating the effects of mutagens and carcinogens. The detoxification mechanism is that quinone reductase promotes the 2-electron reduction of quinones to hydroquinones which are less reactive. This study is to search new inducers of quinone reductase from natural products, which can be used as cancer chemopreventive agents. Plant extracts were evaluated by using quinone reductase generating system With Hepa 1c1c7 murine hepatoma cell lines for enzyme inducing properties and crystal violet staining method for the measurement of cytotoxicity provoked. We have tested approximately 106 kinds of natural products after partition into n-hexane, ethyl acetate and aqueous layers from 100% methanol extracts of natural products. The ethyl acetate fractions of Vitex rotundifolia $(fruits,\;2FC:\;12.7\;{\mu}g/ml)$, Cnidium officinale $(aerial\;parts,\;2FC:\;10.5\;{\mu}g/ml)$, Chrysanthemum sinese $(flowers,\;2FC:\;17.4{\mu}g/ml)$ and the hexane fractions of Angelica gigas $(roots,\;2FC:\;13.2\;{\mu}g/ml)$, Smilax china $(roots,\;2FC:\;l1.9\;{\mu}g/ml)$, Sophora flavescens $(roots,\;2FC:\;16.3\;{\mu}g/ml)$ revealed the significant induction of quinone reductase in a murine hepatic Hepa 1c1c7 cell culture system.

제초제 검출 키트 개발을 위한 식물 해독효소 고정화 연구 (Study on the immobilization of plant glutathione S-transferase for development of herbicide detection kit)

  • Glutathione S-transferase는 식물의 해독작용에 중추적인 역할을 하는 화학 효소이다. 본 연구에서는 제초제 검출 키트 개발에 응용을 위하여 식물 해독작용에 중추적인 역할을 하는 glutathione Stransferase의 고정화 방법을 연구하였다. Chloroacetanilide계 제초제에 높은 효소 활성을 보이는 벼 유래 OsGSTF3에 공유결합을 통한 polystyrene-alkylamine 비드와 리간드결합을 통한 agarose-aminoalkyl 비드,포괄법을 통한 Na-alginate 비드를 이용하여 고정화를 실시하였다. 정제된 OsGSTF3 10 mg을 사용하여 고정화 하였을 때 0.62 mg/g 비드로 polystyrene-alkylamine에 가장 효율적으로 고정화 되었다. 고정화 된 OsGSTF3의 효소 활성은 야생형의 30%를 나타내었으며, 재사용에 의한 효소활성 측정시 3회 까지 처음 활성의 80% 이상을 유지하였다.

  • BACKGROUND/OBJECTIVES: Glutathione s-transferase (GST) is involved in the formation of a multigene family comprising phase II detoxification enzymes, involved in the detoxification of reactive oxygen species. This study evaluated whether daily supplementation with kale juice could modulate levels of plasma antioxidant vitamins and oxidative stress-related parameters. We further examined whether this modulation was affected by combined GSTM1 and T1 polymorphisms. SUBJECTS/METHODS: Totally, 84 subclinical hypertensive patients having systolic blood pressure (BP) over 130 mmHg or diastolic BP over 85 mmHg, received 300 mL of kale juice daily for 6 weeks. Blood samples were drawn before start of study and after completion of 6 weeks. RESULTS: After supplementation, we observed significant decrease in DNA damage and increase in erythrocyte catalase activity in all genotypes. Plasma level of vitamin C was significantly increased in the wild/null and double null genotypes. The plasma levels of ${\beta}-carotene$, erythrocyte glutathione peroxidase activity, and nitric oxide were increased only in the wild/null genotype after kale juice supplementation. CONCLUSIONS: The effect of kale juice was significantly greater in the GSTM1 null genotype and wild/null genotype groups, suggesting possibility of personalized nutritional prescriptions based on personal genetics.

살진균제인 캡탄 처리 후 갈색거저리의 해독효소 유전자 발현 (Gene Expression of Detoxification Enzymes in Tenebrio molitor after Fungicide Captan Exposure)

  • 최근 살진균제는 세계 식량 안보에 없어서는 안될 필수 요소이며, 그 사용량은 증가하고 있다. 살진균제는 직접적 또는 간접적으로 곤충에 영향을 미쳐 유전자 및 분자 수준의 변화를 일으킨다. 곤충은 다양한 해독 매커니즘을 통해 살진균제를 포함한 농약으로부터 유발되는 활성산소(ROS) 독성을 제거한다. 본 연구는 살진균제 캡탄의 비치명적 투여량(0.2, 2, and 20 ㎍/µL)을 주입 후 갈색거저리의 유충에서 해독효소의 mRNA 발현량을 분석했다. 갈색거저리의 전사체 분석을 통해 해독 매커니즘 관련 유전자인 퍼옥시다제(POX), 카탈라제(CAT), 슈퍼옥사이드 디스뮤타제(SOD) 및 글루타티온-S-트랜스퍼라제(GST)를 발굴하였다. 처리 24시간 후 TmPOX5 mRNA가 유의하게 증가한 것으로 나타났다. 처리 3 시간 후 TmSOD4의 mRNA가 유사하게 증가하였다. 또한 2 ㎍/µL 처리 24시간 후 TmCAT2의 mRNA 가 유의하게 증가하였다. 캡탄 노출 후 TmGST1 및 TmGST3의 mRNA 발현량도 증가하였다. 결론적으로, TmPOX5 및 TmSOD4 유전자는 갈색거저리에서 캡탄 노출에 대한 바이오마커 또는 생체이물 센서로 작용할 수 있음을 시사한다.

에탄올 및 사염화탄소의 긴 손상에 미치는 흰점박이꽃무지 추출물의 영향 (Influence of Protaetia brevitarsis Extract on Liver Damage Induced by Carbon Tetrachloride and Ethanol in Rats)

  • 흰점박이꽃무지(Protaetia brevitarsis)는 간 질환의 치료를 위해 민간요법 뿐만 아니라 한방에서는 이용하는 전통약물로 알려져 있다. 본 연구는 사염화탄소와 에탄올로 간 손산을 유도한 흰쥐에서 흰점박이꽃무지 추출물(PBE)의 영향을 조사하였다. 사염화탄소는 이물질대사효소(cytochrome P_450, cytochrome b$_{5}$, cytochrome b$_{5}$ reductase), TBARS, 혈청 AST와 ALT, 간 무게비에서 유의한 변화가 확인되는 간독성이 유발하였으며, 에탄올은 혈청 AST와 TBARS의 변화는 유도하지 않았으나 이물질대사효소, ALT, 체중 당 무게비가 유의한 변화를 나타내었다. PBE로 전 처리한 실험군은 이물질대사효소(cytochrome P_450, cytochrome b$_{5}$, cytochrome b$_{5}$, reductase), 혈청 TBARS, AST와 ALT, 간무게비에서 어떠한 유의적인 변화도 관찰되지 않았다. 이러한결과는 적어도 이 실험조건에서는 PBE가 사염호탄소 혹은 에탄올로 유도한 간 독성에 대하여 보호 또는 회복효과가 없다고 판단된다.

기주식물 방어물질에 대한 담배나방의 생화학적 적응 (Biochemical Adaptation of the Oriental Tobacco Budworm, Helicoverpa assulta, to Host-plant Defensive Compounds)

  • 식물의 이차대사산물은 곤충-식물 상호관계에서 중요한 역할을 한다. 식식성 곤충은 식물의 방어물질에 대처하는 다양한 전략을 가지고 있다. 광식성 곤충은 넓은 범위의 다양한 식물들을 섭식하고 그 해독 기작도 보다 복잡한데, 이는 많은 종류의 식물유래 화합물에 반응하는 경향이 있기 때문으로 보인다. 이와는 달리 협식성 곤충은 몇몇 유사한 식물에 국한되어 살아가며 보다 효율적인 적응 방식을 지니고 있을 것으로 여겨진다. 이러한 협식성 곤충의 적응은 식물의 방어물질에 대한 해독효소를 다량 생산하거나 방어물질 또는 그 대사산물을 격리하는 전략을 마련하였기 때문으로 보인다. 담배나방은 담배와 고추 등 주로 가지과의 몇몇 식물만을 가해하는 협식성 곤충이다. 담배나방의 기주식물 적응성을 이해한다면, 이 해충에 의한 작물의 피해를 줄이는 방법을 개발하는데 도움을 줄 수 있을 뿐만 아니라, 담배나방과 같은 협식성 곤충의 생리, 생태, 진화를 연구하는 데에도 중요한 단서를 제공할 것이다. 본 종설에서는 담배나방의 기주식물 범위, 유충과 기주식물의 상호작용, 그리고 기주식물에서 특이적으로 나오는 니코틴과 캡사이신에 대한 곤충의 반응과 해독 메카니즘을 중심으로, 지난 반세기 동안의 연구결과를 요약하고 앞으로의 전망을 제시하고자 한다.

Comparative Investigation of Glutathione S-Transferases, Glyoxalase-I and Alliinase Activities in Different Vegetable Crops

  • Glutathione S-transferases(GSTs, EC 2.5.1.18), glyoxalase-I(EC 4.4.1.5) and alliin lyase(alliinase, EC 4.4.1.4) are important enzyme systems in plant bodies. The first two are mainly detoxifying enzymes that utilize glutathione(GSH) in the defense mechanism, and the last one is mainly involved in secondary metabolism and relevant to sulfur compounds derived from GSH. The activities of the three enzymes have been investigated in soluble extracts of vegetable crops, including pumpkin, cabbage, broccoli, radish, carrot, potato, sweet potato, mungbean, and onion. GST activities were detected in all of the vegetables, and the extract of onion bulb exhibited the highest specific activity(648 nmol/min/mgP). The putative GSTs of most of the vegetables were found to be induced by ethanol. The activities of GSTs in onion bulb were found to be markedly inhibited by S-hexyl glutathione and were also inhibited by S-butyl glutathione and S-propyl glutathione. The anti-CmGSTF1 antiserum recognized a thick band for putative onion GST. The estimated glyoxalase-I activity level was also high in onion bulb(4540 nmol/min/mgP), indicating that the thick band detected by Western blot analysis might result from partial recognition of glyoxalase-I by the antiserum. The specific activities for glyoxalase-I were moderate in radish and carrot, and the extracts of other vegetables had rather low levels of activities. The extract of onion also showed the highest specific activity level for alliinase(2069nmol pyruvate/mgP). The extracts of other vegetables also had alliinase activities, although the estimated values were much lower than that of onion.
