The Radioprotective Effect and Mechanism of Captopril on Radiation Induced-Heart Damage in Rats (방사선 조사 후 발생한 흰쥐 심장손상에서 Captopril의 방어역할과 기전)

  • Purpose : Captopril (angiotension converting enzyme inhibitor) is known to have a radioproptective effect in the lungs, intestines and skin, but its effect in the heart is unclear. To investigate the radioprotectlve efiect and mechanism of captopril on the heart, the histopathological changes and immunohistochemical stains were compared with radiation alone, and radiation combined with captopril, in the rats. Materials and Methods : The histopathological changes and immunohistochemical stains ($TNF{\alpha}$, $TGF{\beta}1$, PDGF and FGF2) were examined in the radiation alone and the combined captopril and radiation groups, 2 and 8 weeks after irradiation. Each group consisted of 8 to 10 rats (Sprague-Dawley). Irradiation (12.5 Gy) was given to the left hemithorax in a single fraction. Captopril (50 mg/Kg/d) mixed with water, was given orally and continuously from the first week prior to, up to the 8th week of the experiment. Results : In the radiation alone group, the ventricle at 2 weeks after irradiation showed prominent edema (p=0.082) and fibrin deposit (p=0.018) compared to the control group. At 8 weeks, the edema was decreased and fibrosis increased compared to those at 2 weeks. The histopathological changes of the combined group were similar to those of the control group, due to the reduced radiation toxicity at 2 and 8 weeks. The endocardial fibrin deposit (p=0.047) in the atrium, and the interstitial fibrin deposit (p=0.019) and edema (p=0.042) of the ventricle were reduced significantly in the combined group compared to those in the radiation alone group at 2 weeks. The expressions of $TNF-{\alpha}$, $TGF-{\beta}1$, PDGF and FGF-2 in the radiation alone group were more increased than in the control group, especially in the pericardium and endocardium of the atrium at 2 weeks. At 8 weeks, the pericardial $TNF-{\alpha}$ and $TGF-{\beta}1$ in the radiation alone group continuously increased. The expressions of $TNF-{\alpha}$, $TGF-{\beta}1$ and PDGF were decreased in the combined group at 2 weeks. At 8 weeks, the expressions of $TNF-{\alpha}$ in the atrial and ventricular pericardia were markedly reduced (p=0.049, p=0.009). Conclusion : This study revealed that the early heart damage induced by radiation can be reduced by the addition of captopril in a rat model. The expressions of $TNF-{\alpha}$, $TGF-{\beta}1$ and PDGF were further decreased in the combined compared to the radiation alone group at both 2 and 8 weeks. From these results, it may be concluded that these cytokines probably play roles in the radioprotective mechanism of captopril from the radiation-induced heart toxicity, similarly to in other organs.

Effects of Dietary Salt Restriction on the Development of Renal Failure in the Excision Remnant Kidney Model (식이 sodium 제한 및 식이 sodium 제한에 따른 항고혈압제의 투여가 만성신부전증의 진행에 미치는 영향에 관한 실험적 연구)

  • Purpose: To evaluate whether or not sodium restriction had its own beneficial effect and increased the efficiency of the anti-hypertensive drugs on the progression of renal failure. Methods: We studied using the excision remnant kidney model. Treatment groups were as follows: 5/6 nephrectomy and a 0.49% (normal-high) sodium diet (NN); 5/6 nephrectomy and a 0.25% (normal-low) sodium diet (LN); 5/6 nephrectomy, a 0.49% sodium diet and enalapril (NNE); 5/6 nephrectomy, a 0.49% sodium diet and nicardipine (NNN); 5/6 nephrectomy, a 0.25% sodium diet and enalapril (LNE); 5/6 nephrectomy, a 0.25% sodium diet and nicardipine (LNN). Both diets were isocaloric and had the same content of protein, phosphorus and calcium. Proteinuria, remnant kidney weight, mesangial expansion scores, and glomerular volume were assessed. Results: Blood pressure tended to be lower in LN compared to NN (P<0.05). NN developed progressive hypertension. LNE, LU, NNE, and NNN reduced blood pressure. LNE, LNN, NNE, NNN, and LN had significantly less proteinuria than NN at 16 weeks (P<0.05). At 24 weeks, LN developed proteinuria (82 mg/day), which were lessened in LNE (54 mg/day) and not lessened in LNN (76 mg/day). Mesangial expansion scores were significantly less in LN rats compared to those in NN rats. Glomerular volumes at 24 weeks in LN rats were significantly less compared to those at 16 weeks in NN rats. Mesangial expansion scores and glomerular volumes at 4, weeks, 12 weeks, and 24 weeks were not different among LN, LNE, and LNN groups. Conclusion: Dietary salt restriction lessens renal damage, at least in part, by inhibiting compensatory renal growth and reducing blood pressure. Enalapril was particularly successful in reducing proteinuria and glomerular injury when combined with dietary salt restriction.

  • Landfill require special care due to the dangers of nearby surface water and underground water pollution caused by leakage of leachate. The leachate does not leak due to the installation of the geomembrane but sharp wastes or landfill equipment can damage the geomembrane and therefore a means of protecting the geomembrane is required. In Korea, in accordance with the waste control act being modified in 1999, protecting the geosynthetics liner on top of the slope of landfill and installing a drainage layer to fluently drain leachate became mandatory, and technologies are being researched to both protect the geomembrane and quickly drain leachate simultaneously. Therefore, this research has its purpose in studying the drainage functions of leachate and protection functions of the geomembrane in order to examine the application possibilities of Geo-Multicell-Composite (GMC) as a Leachate Collection Removal and Protection System (LCRPs) at the slope on top of the geomembrane of landfill by observing methods of inserting filler with high-quality water permeability at the drainage net. GMC's horizontal permeability coefficient is $8.0{\times}10^{-4}m^2/s$ to legal standards satisfeid. Also crash gravel used as filler respected by vertical permeability is 5.0 cm/s, embroidering puncture strength 140.2 kgf. A result of storm drain using artificial rain in GMC model facility, maxinum flow rate of 1,120 L/hr even spray without surface runoff was about 92~97% penetration. Further study, instead of crash gravel used as a filler, such as using recycled aggregate utilization increases and the resulting construction cost is expected to savings.

Protective effects of Gastrodia rhizoma and steamed & fermented Gastrodiae rhizoma with anti-oxidant efficacy and suppression of NFκB signaling pathway on LPS-induced liver injury (LPS로 유발한 간손상 마우스에서 항산화 및 항염증 효능을 통한 천마와 증숙 발효 천마의 간보호 효과)

  • This study is aimed to evaluate the protective effect of Gastrodiae rhizoma and steamed, dried & fermented Gastrodiae rhizoma on Lipopolysaccharide (LPS)-induced hepatic injury in the mice model. Sample was selected to GR0F0 (not processed gastrodia rhizome) and GR6F4 (fermented with Saccharomyces cerevisiae before steamed and dried 6 times) based on 1,1-diphenyl-2-picrylhydrazyl, 2,2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid, and High-performance liquid chromatography analysis. Mice were randomly divided into 4 groups - Normal group, vehicle group (LPS treated), GR0F0 group (fed GR0F0 before LPS treated) and GR6F4 group (fed GR6F4 before LPS treated) with 6 mice in each group. GR0F0 group and GR6F4 group were fed each extract 200 mg/kg/day during 8 days. LPS 20 mg/kg injected to the experimental groups as abdominal injection. We measured aspartate aminotransferase, alanine amino-transferase in serum. GR0F0 and GR6F4 showed a significant decrease compared to the vehicle group. As a result of measuring the ROS, GR6F4 group showed a significant reduction in both the serum and liver tissues compared to the vehicle group. GR0F0 group showed a significant reduction only in the liver tissues. Activator protein-1, cyclooxygenase-2, and Inducible nitric oxide synthase were significantly decreased GR0F0 group and GR6F4 group. But tumor necrosis factor alpha only showed a significant reduction in GR6F4 group. GR0F0 and GR6F4 groups against liver damage in mice with LPS. That showed significant effects on anti-oxidant and anti-inflammatory action. The effects of GR6F4 group showed superior results compared to GR0F0 group. Therefore, Steamed, dried & fermented Gastrodia rhizoma was might have a protective effect on liver injury.

Prediction of Dispersal Directions and Ranges of Volcanic Ashes from the Possible Eruption of Mt. Baekdu

  • To predict the influence of volcano eruption on agriculture in South Korea we evaluated the dispersal ranges of the volcanic ashes toward the South Korea based on the possibilities of volcano eruption in Mt. Baekdu. The possibilities of volcano eruption in Mt. Baekdu have been still being intensified by the signals including magmatic unrest of the volcano and the frequency of volcanic earthquakes swarm, the horizontal displacement and vertical uplift around the Mt. Baekdu, the temperature rises of hot springs, high ratios of $N_2/O_2$ and $_3He/_4He$ in volcanic gases. The dispersal direction and ranges and the predicted amount of volcanic ash can be significantly influenced by Volcanic Explosivity Index (VEI) and the trend of seasonal wind. The prediction of volcanic ash dispersion by the model showed that the ash cloud extended to Ulleung Island and Japan within 9 hours and 24 hours by the northwestern monsoon wind in winter while the ash cloud extended to northern side by the south-east monsoon wind during June and September. However, the ash cloud may extent to Seoul and southwest coast within 9 hours and 15 hours by northern wind in winter, leading to severe ash deposits over the whole area of South Korea, although the thickness of the ash deposits generally decreases exponentially with increasing distance from a volcano. In case of VEI 7, the ash deposits of Daejeon and Gangneung are $1.31{\times}10^4g\;m^{-2}$ and $1.80{\times}10^5g\;m^{-2}$, respectively. In addition, ash particles may compact close together after they fall to the ground, resulting in increase of the bulk density that can alter the soil physical and chemical properties detrimental to agricultural practices and crop growth.

  • Establishing conservation programs to protect and maintain populations of endangered species are not only a global trend, but also a pursuit endorsed by the Korean Environmental Conservation Act. This study evaluates the feasibility of alternative habitat patches for the endangered butterfly Parnassius bremeri. A portion of habitat of P. bremeri is expected to be fragmented and damaged due to the scheduled construction of the Sangju-Yongduk Highway. A trans fer of the habitat patches of P. bremeri is also scheduled. In order to select an alternative habitat patch, the Mark-Release-Recapture (MRR) method was used to simulate a patch transfer model. The connectedness between habitat are as and the survival of local populations were evaluated for each candidate habitat. It was found that metapopulations with patch distances of <250m showed a 50% connectedness and survival rate in local populations. P. bremeri were expected to migrate at an average distance of 300m. In addition, P. bremeri formed a metapopulation that exhibited intimate patch dynamics that promoted persistence among these patches. Possible candidate habitats including those recommended by local governing bodies were evaluated along with habitats that may counter problems arising from the damage done to the original habitat and habitats that may have a compensatory value equal to that of the original habitat. Based on these criteria, Ansa-myeon township office was selected due to its high scores. This scoring was based on a consideration of a wide range of variables that mark a successful transfer of habitat. These include the amount of funding available, the governing bodies of the possible alternative habitat, and the Expected collaborative effort of local citizens. This decision was collaborated on by incorporating the expertise of various fields of study including biology, ecology, biogeography, ecological engineering, landscape architecture, and social sciences. Therefore, it is suggested that in order to evaluate an alternative habitat for organisms, many social issues as well as ecological issues must be considered.

Determinants of IPO Failure Risk and Price Response in Kosdaq (코스닥 상장 시 실패위험 결정요인과 주가반응에 관한 연구)

  • Recently, failure rates of Kosdaq IPO firms are increasing and their survival rates tend to be very low, and when these firms do fail, often times backed by a number of governmental financial supports, they may inflict severe financial damage to investors, let alone economy as a whole. To ensure investors' confidence in Kosdaq and foster promising and healthy businesses, it is necessary to precisely assess their intrinsic values and survivability. This study investigates what contributed to the failure of IPO firms and analyzed how these elements are factored into corresponding firms' stock returns. Failure risks are assessed at the time of IPO. This paper considers factors reflecting IPO characteristics, a firm's underwriter prestige, auditor's quality, IPO offer price, firm's age, and IPO proceeds. The study further went on to examine how, if at all, these failure risks involved during IPO led to post-IPO stock prices. Sample firms used in this study include 98 Kosdaq firms that have failed and 569 healthy firms that are classified into the same business categories, and Logit models are used in estimate the probability of failure. Empirical results indicate that auditor's quality, IPO offer price, firm's age, and IPO proceeds shown significant relevance to failure risks at the time of IPO. Of other variables, firm's size and ROA, previously deemed significantly related to failure risks, in fact do not show significant relevance to those risks, whereas financial leverage does. This illustrates the efficacy of a model that appropriately reflects the attributes of IPO firms. Also, even though R&D expenditures were believed to be value relevant by previous studies, this study reveals that R&D is not a significant factor related to failure risks. In examing the relation between failure risks and stock prices, this study finds that failure risks are negatively related to 1 or 2 year size-adjusted abnormal returns after IPO. The results of this study may provide useful knowledge for government regulatory officials in contemplating pertinent policy and for credit analysts in their proper evaluation of a firm's credit standing.

Uplift Bearing Capacity of Spiral Steel Peg for the Single Span Greenhouse (온실용 나선철항의 인발저항력 검토)

  • This study examined the uplift bearing capacity of spiral steel pegs according to the degree of soil compaction and embedded depth in a small-scaled lab test. As a result, their uplift bearing capacity increased according to the degree of soil compaction and embedded depth. The uplift bearing capacity under the ground condition of 85% compaction rate especially recorded 48.9 kgf, 57.9 kgf, 86.2 kgf and 116.6 kgf at embedded depth of 25 cm, 30 cm, 35 cm and 40 cm, respectively, being considerably higher than under other ground conditions. There were huge differences in the uplift bearing capacity of spiral steel pegs according to the compaction conditions of ground. Their maximum uplift bearing capacity was 116.6 kgf under the ground condition of 85% compaction rate and at embedded depth of 40 cm, and it is very high considering the data of spiral steel pegs. It is thus estimated that wind damage can be effectively reduced by careful maintenance of ground condition surrounding spiral steel pegs. In addition, spiral steel pegs will be able to make a contribution to greenhouse structural stability if proper installation methods are provided including the number and interval according to the types of greenhouse as well as fixation of plastic film. The findings of the study indicate that the optimal effects of spiral steel pegs for greenhouse can be achieved at embedded depth of more than 35cm and compaction degree of more than 85%. The relative density of the model ground in the test was 67% at compaction rate of 85%.

Estimation of fire Experiment Prediction by Utility Tunnels Fire Experiment and Simulation (지하공동구 화재 실험 및 시뮬레이션에 의한 화재 설칠 예측 평가)

  • The utility tunnels are the important facility as a mainstay of country because of the latest communication developments. However, the utilities tunnel is difficult to deal with in case of a fire accident. When a cable burns, the black smoke containing poisonous gas will be reduced. This black smoke goes into the tunnel, and makes it difficult to extinguish the fire. Therefore, when there was a fire in the utility tunnel, the central nerves of the country had been paralyzed, such as property damage, communication interruption, in addition to inconvenience for people. This paper is based on the fire occurred in the past, and reenacting the fire by making the real utilities tunnel model. The aim of this paper is the scientific analysis of the character image of the fire, and the verification of each fire protection system whether it works well after process of setting up a fire protection system in the utilities tunnel at a constant temperature. The fire experiment was equipped with the linear heat detector, the fire door, the connection water spray system and the ventilation system in the utilities tunnel. Fixed portion of an electric power supply cable was coated with a fire retardant coating, and a heating tube was covered with a fireproof. The result showed that the highest temperature was $932^{\circ}c$ and the linear heat detector was working at the constant temperature, and it pointed at the place of the fire on the receiving board, and Fixed portion of the electric power supply cable coated with the fire retardant coating did not work as the fireproof. The heating tube was covered with the fireproof about 30 minutes.

Anti-oxidative and Cytoprotective Effect of Ursodeoxycholic Acid, an Active Compound from the Bear's Gall, in Mouse Microglia (생쥐 뇌소교세포주에서 웅담추출활성성분(우르소데옥시콜린산)의 항산화 및 세포보호효과)

  • The in vitro cytoprotective and anti-oxidative effects of ursodeoxycholic acid, a major active compound from bear's gall were investigated in mouse brain microglia. In the present study, we wished to scrutinize the potential role of UDCA as an anti-neurodegenerative agent in neurodegenerative disease such as Alzheimer's disease. This concept was supported by the multiple preliminary studies in which UDCA has an anti-inflammatory effect in microglial cells. In the study, we found that $7.5\;{\mu}g/mL$ UDCA was effective in the protection of cells from $H_2O_2$ damage, a reactive oxygen, and the resuIt was coincided with the anti-apoptotic effect in DAPI staining. Moreover, the metal-catalyzed oxidation study showed that UDCA has antioxidant effect as much as ascorbic acid at $50{\sim}100\;{\mu}g/mL$. In conclusion, these study results suggested that neuro-degenerative diseases such as Alzheimer's disease probably caused by over-expressed beta amyloid peptide in elderly people can be controled by UDCA through an anti-inflammatory, anti-oxidative and anti-apoptotic effect. The evidences showed in the study may be references for more in-depth in vivo and clinical studies for a candidate of anti-neurodegenerative therapy in the near future.
