  • The purpose of this study is to develop a culture and leisure resources scale and examine the validity of the scale. The culture and Leisure resources scale was developed and identified its validity by exploratory factor analysis, confirmatory factor analysis, and correlation analysis. The culture and leisure resources scale consisted of four factors, which were 'personal resources', 'support staff resources', 'facility resources', and 'program resources'. Those four factors indicated the reasonable fit indices from confirmatory factor analysis. In addition, this scale showed convergent validity with the satisfaction of cultural and leisure environment and the satisfaction of one's cultural and leisure life. The result of scale score comparison between rural and urban areas showed that culture and leisure resources in rural areas were less abundant than those in urban areas. Furthermore, while the variables which affected the satisfaction of urban residents' were mainly 'personal resources', the variables in case of rural residents' were mainly 'program resources' and 'facility Resources'. The culture and leisure resources scale can be used for policymakers practicably to evaluate regional level of culture and leisure resources, to compare between regions and to find policy priorities for improving the quality of leisure, especially for disadvantaged areas with less resources.

  • This study was carried out to investigate the effects of agitation, aeration and scale-up on the mycelial growth, exo-polysaccharide (EPS) production, and mycelial morphology in the liquid culture of Ganoderma lucidum. A correlation between roughness and operating variables was also studied to scale-up the liquid culture of G. lucidum in a jar fermenter. When the agitation speed or aeration rate increased, the morphological form was changed from rough pellet to smooth pellet form. Increase of the agitation and aeration reduced the mycelial roughness. On the other hand, in the case of pellet size, it was not affected by aeration. The higher EPS production was obtained at approximately 17% of roughness and mycelial pellet size of 3~5 mm. The morphology at each fermenter was closely correlated with kLa value, and it was found that similarity of morphology would be used as a criteria of scale-up for liquid culture of G. lucidum.

  • Photosynthetic bacteria (PSB) attract considerable interest as useful microorganisms; nevertheless, a generalized culture technique has not been previously reported owing to difficulty in their cultivation. Therefore, a simple culture technique suitable for public use was investigated. Among the PSB tested, the strain Rhodobacter azotoformans EBN-7 was the most suitable for scale-up production because it showed the highest specific growth rate (0.20 h-1) on basal medium. In scale-up cultivation (500 L), R. azotoformans EBN-7 showed 4.50 × 1010 colony-forming units mL-1 (number of viable cells), dry cell weight of 26.8 g/L, and a specific growth rate of 0.15 h-1. Cultivation using this final culture broth (as seed culture) in a 15 L simple reactor was successful, with maintenance of cell activity evident. For use as seed culture, the maximum allowable preservation period of R. azotoformans EBN-7 at 4℃ was 3 months. When R. azotoformans EBN-7 cultivated in a simple technique was applied to shrimp aquaculture water, NH4+-N was reduced from 0.61 mg/L to 0.24 mg/L (by 60.7%) in 4 days in comparison with the control. Thus, this simple culture technique using R. azotoformans EBN-7 has the potential for a good removal efficiency of NH4+-N, making seed culture easier and suitable for public use.

  • In an attempt to establish the mass proliferation system, Eucalyptus pellita, a 5-year-old plus tree, was cultured with three different culture types in 1L vessels: solid culture without ventilation (conventional culture), liquid culture without ventilation and open-type liquid culture with forced ventilation. Then the culture scale was subsequently increased from 1L to 10L in vessel volume. After 4 weeks of 1L-scale culture, the best growth was obtained by culturing plantlets on open-type liquid culture, suggesting that the in vitro plantlets growth can be enhanced by liquid medium and ventilation. In open-type large scale culture in 10L vessel, plantlets growth resulted in a 370% increase in the number of nodes, 3.6 times increase in leaf expansion, and 3.3 times increase in shoot length, while the conventional culture suppressed shoot growth due to the callusing on the leaves and lack of $CO_2$. The results indicated that the open-type large scale culture system was effective for enhancing productivity by improving growth of the plantlets in clonally proliferated plus tree, Eucalyptus pellita.
