도토리 가루의 성분분석과 항산화능 평가 (Studies for Component Analysis and Antioxidative Evaluation in Acorn Powders)

  • 도토리 가루를 이용하여 기능성 식품개발을 목적으로 도토리 가루의 화학성분, 항산화 활성을 조사하였다. 일반성분은 탄수화물이 87.29%, 수분 10.57%, 조지방 1.18%, 조단백 0.84%, 회분 0.12% 순으로 나타났다. 도토리 가루에 가장 많은 무기성분으로는 K이 40.68mg/100g으로 나타났으며, 그 외 P(8.95mg/100g), Ca(8.64mg/100g), Mg(4.67mg/100g), Na(3.94mg/100g), Fe(1.41mg/100g) 순으로 나타났다. 구성당은 총 7종이 분리 동정되었으며, 그 중 glucose가 대부분으로 나타났고 rhamnose, galactose, arabinose, mannose, fucose, xylose 순으로 함유되어 있었다. 주요 지방산 조성은 oleic acid(C18:1)가 31.87%로 가장 높았고, linoleic acid(C18:2)는 18.72%, linolenic acid(C18:3)는 16.67% 그리고 palmitic acid(C16:0)가 11.32% 순으로 나타났다. 항산화 효과는 물 추출물에서 $IC_{50}$$19.0{\mu}g/mL$로 가장 높게 측정되었고 75% 에탄올 추출물$(IC_{50}:\;21.4{\mu}g/mL)$ 및 메탄올 추출물$(IC_{50}:\;45.2{\mu}g/mL)$순으로 항산화 효과가 측정되었다.

  • 본 연구는 가시여지 잎 에탄올 추출물(ALE) 및 조다당 추출물(ALP)의 면역 활성에 관하여 비교하기 위하여, 선천 및 적응면역에서 중추적인 역할을 수행하는 큰포식세포에 ALE 및 ALP를 처리하여 세포 증식률, 산화질소 분비능, 사이토카인(종양괴사인자, IL-6, $IL-1{\beta}$) 분비능 및 기전 분석을 통한 신호전달에 관하여 관찰하였다. ALE 및 ALP를 큰포식세포에 처리하여 면역활성에 관하여 비교하였을 때, 큰포식세포 면역활성의 바이오-마커인 산화질소, 사이토카인의 분비능 및 iNOS의 세포내 발현이 ALP 처리구에서 유의적으로 증가되는 것으로 관찰되었으며, 기전분석결과, ALP의 처리는 MPAKs의 인산화 및 $NF-{\kappa}B$의 핵내 이동성을 증가시켜 면역활성을 증가시키는 것으로 관찰되었다. 따라서, ALP의 높은 면역활성은 MPAKs의 인산화 및 $NF-{\kappa}B$의 활성과 밀접한 관련이 있는 것으로 판단된다.

매생이 추출물의 생리 활성과 항산화 활성에 관한 연구 (A Study on Physiological Activity and Antioxidative Activity of Maesangi(Capsosiphon fulvescens) Extract)

  • Physiological activity and antioxidative activity of Maesangi(Capsosiphon fulvescens) extracts with distilled water or 95% ethanol were investigated. For the evaluation of physiological and antioxidative activities, some evaluation assay methods such as measurement of Hunter color value, chlorophyll a/b value, total phenolics, reducing power and thiobarbituric acid(TBA) value of soybean oil were used. Proximate composition and mineral contents of Masaengi were orders of crude protein>crude fiber>moisture>crude ash>crude lipid, and K>Ca>Mg>Na>P>Fe>Zn, respectively. In ethanol extract, the content of total phenolic compounds in Maesangi was determined to half times of that in Dasima(Laminaria). The reducing power of Maesangi-ehtanol extract was about 5 % of vitamin C and was lower than that of Dasima-ethanol extract. The TBA value of Maesangi-ethanol extract on soybean oil oxidation was about 47 % and 68.4 % to control in three and eight days oxidation, respectively. But TBA value difference was not observed significantly with the dosage below 5mL of ethanol extract.

젖소 사료로서의 맥콜박 이용 가능성에 관한 연구 (Studies on the Possibility of the Utilization of Macol Meal as Feedstuffs for Cattle)

  • This experiment was conducted to evaluate the feed value of Macol meal, which is one of the byproducts from the extraction of liquid syrup from barley grain, on lactating dairy cows. All of the lactating dairy cows feeding in eight dairy farms were used in feeding experiment and fed concentrates plus Macol meal 5Kg as wet basis. The milk yields and milk fit during the periods of 10 months were investigated and blood picture, blood chemistry and diseases occuring frequency before and after feeding Macol meal were analyzed. The results obtained were as follows ; 1. In the proximate compositions, the contents of moisture, crude protein, crude fat, crude fiber, crude as and nitrogen free extracts was 68.8%, 7.43%, 1.04%, 7.08%, 1.56% and 14.09% respectively. On the other hand, the contents of NDF, ADF and cellulose was 36.68%, 35.71% and 10.85%, as dry matter basis, respectively. 2. The daily milk yield per head was 22.84kg during two months before feeding Macol meal and 23.89 kg during the periods of ten months after feeding Macol meal. 3. The blood picture and blood chemistry were not affected by feeding Macol meal. 4. The diseases occuring frequency were not significantly affected by feeding Macol meal. from the results of this experiment it could be concluded that Macol meal can be used in lactating dairy cows rations without any adverse effects.

Antinociceptive and Antiinflammatory Effect of a Diterpene Isolated from the Aerial Part of Siegesbeckia pubescens

  • The aerial part of Siegesbeckia pubescens (Compositae) has been used to treat rheumatoid arthritis and hypertension in the Oriental medicine. This crude drug has been used without process (SP-0) or with three times-process of steaming and drying (SP-3) or the nine times of that process (SP-9). To search for the antinociceptive anti-inflammatory components from this crude drug, activity-directed fractionation was performed on this crude drug. Since the $CHCl_3$ extract was shown to have a more potent effect than other extracts, it was subjected to silica gel & ODS column chromatography to yield two diterpene compounds (1). Compound 1 was structurally identified as ent-16 (H, 17-hydroxykauran-19-oic acid, which were tentatively named siegeskaurolic acid A. A main diterpene, siegeskaurolic acid A was tested for the antiiflammatory antinociceptive effects using both hot plate- and writhing anti-nociceptive assays and carrageenan-induced anti-inflammatory assays in mice and rats. Pretreatment with siegeskaurolic acid A (20 and 30mg/kg) significantly reduced the stretching episodes, action time of mice and carrageenan-induced edema. These results support that siegeskaurolic acid is a main diterpene responsible for antinociceptive and antiiflammatory action of S. pubescens. In addition, the assays on SP-0, SP-3 and SP-9 produced the experimental results that SP-9 had more significant effects than other two crude drugs. These results suggest that the processing on the original plant may lead to the higher pharmacological effect.

당귀와 승검초의 기능성 성분과 아질산염 소거능 (Effective Components and Nitrite Scavenging Ability of Root and Leaves a Angelica gigas Nakai)

  • This research was conducted to examine the usability of Dangui (Angelica gigas Nakai, Root) and Seungumcho (Angelica gigas Nakai, Leaf) as functional food in aspects of their functional components and nitrite scavenging ability. Analysis of proximate composition showed that Dangui contains more moisture, crude lipid and crude protein than Seungumcho. On the other hand, Seungumcho contains more than twice mont of crude ash and crude fiber than Dangui. Dangui showed higher contents in phosphorus, iron, magnesium, and Seungumcho showed higher contents in copper, calcium, sodium, potassium compare to each other. Substances such as calcium, magnesium and iron that showed high contents in Dangui and Seungumcho are the most important inorganic substances. Total dietary fiber(TDF) of Dangui, sum of IDF and SDF, was 24.2%, and Seungumcho showed 28.18%. vitamin C contents of a 29.690.33 mg/100g appeared only in Seunggumcho. Total phenol contents of Dangui was 0.100${\pm}$002% and that of Seungumcho 0.0900${\pm}$008%. Nnitrite Scavenging ability of both water soluble and methanol soluble extracts were more than 90% at pH 1.2, and it decreased as pH level adjusted to pH 4.2, pH and 6.0.

생약으로부터 In vitro DNA 결합활성의 검색 (Screening of In vitro DNA-binding Activity from the Crude Drugs)

  • One hundred and seventeen crude drugs were screened for DNA-binding activity in vitro. The DNA-methyl green assay is a useful biochemical screen for the detection or development of biologically active compounds which bind to DNA. This assay is based upon the fact that free methyl green undergoes rapid spontaneous molecular rearrangement to its colorless carbinol base so that the liberation of the dye from DNA by displacement can be followed spectrophotometrically as a decrease in absorbance at 65Onm. Seven methanolic extracts of crude drugs including Cinnamomi Cortex spissus, Coicis Semen, Coptidis Rhizoma, Perillae Semen, Plantaginis Semen, Polygalae Radix and Zanthoxyli Fructus showed less than 2,000${\mu}$g/ml as their $IC_{50}$ values. The active principles of Plantaginis Semen and Zanthoxyli Fructus were transferred into organic solvents, which showed the $IC_{50}$ values with 588 and 574${\mu}$g/ml, respectively. These fractions have been selected for isolation of biologically active constituents.

Comparison of Antioxidant Capacity and Nutritional Composition of three Cultivars of Actinidia arguta

  • Actinidia arguta extracts obtained from three cultivars ('Sae-Han', 'Dae-Sung', and 'Chil-Bo') were assayed for their antioxidant properties and nutritional composition. Antioxidant activity of the extracts was evaluated using 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging method. Total phenolic contents of the extracts were determined by the Folin-Ciocalteu method. Vitamin C (L-ascorbic acid) content measured by a colorimetric method and reducing sugar content estimated by dinitrosalicylic acid (DNSA) method. Crude extracts from A. arguta 'Sea-Han' showed the most potent radical-scavenging activity showing 86.55% at 10 mg/ml. The DPPH radical scavenging activity of extracts and solvent fractions from Sea-Han cultivar was in decreasing order of EtOAc fraction>BuOH fraction>$CH_2Cl_2$ fraction>hexane fraction, among which EtOAc fraction showed the highest antioxidant activity (87.51 % at 5 mg/ml). Total phenolic contents in A. arguta 'Sae-Han', 'Dae-Sung' and 'Chil-Bo', were 32.93, 28.23, and 25.60 mg/g, respectively. Vitamin C contents of them were 840.57, 578.81 and 730.10 ug/g, respectively.

In vitro Peroxynitrite Scavenging Activity of Arctic Seaweed Extracts

  • In this study, antioxidant activities of acetone/dichloromethane and methanol extracts of Arctic seaweeds were investigated. The antioxidant properties of both extracts of arctic seaweed were evaluated using two different peroxynitrite tests, including scavenging power on authentic peroxynitrite and inhibitory activities on peroxynitrite generation from 3-morpholinosydnonimine (SIN-1) producing superoxide anion and nitric oxide simultaneously in vitro. At concentration of $10\;{\mu}g/ml$, the acetone/dichloromethane and methanol extracts of Odonthalia dentata exhibited 54.6 and 64.2% inhibition against peroxynitrite generation from SIN-1 while they exhibited 24.3 and 23.0% scavenging activities on authentic peroxynitrite, respectively. On the other hand, the acetone/dichloromethane extract of Polysiphonia stricta exhibited 61.8% inhibition only against the induced peroxynitrite from SIN-1. Furthermore, the crude extracts of Odonthalia dentata and Polysiphonia stricta were fractionated into n-hexane, 85% aq. MeOH, n-BuOH, and $H_2O$ fractions, successively, and only 85% aq. MeOH fraction exhibited the best inhibition.

Total Phenolic Contents and Biological Activities of Korean Seaweed Extracts

  • Crude extracts of thirty seaweeds collected in Korea were obtained using 50% ethanol, and total phenolic contents and antioxidant activities were compared. Two brown algae, Ecklonia cava (E. cava) and Sargassum siliquastrum (S. siliquastrum), showing high antioxidant activity based on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and superoxide anion radical scavenging activity were further investigated for their inhibitory effects on tyrosinase activity. The E. cava extract had the highest total phenolic content among the seaweeds extracts. Total phenolic contents were strongly correlated with antioxidant activity in the thirty seaweed extracts ($R^2\;=\;0.9169$). The E. cava and S. siliquastrum extracts exhibited higher inhibition to tyrosinase activity than butylated hydroxytoluene (BHT) and epigallocatechin gallate (EGCG).
