Immune Stimulation and Anti-Metastasis of Crude Polysaccharide from Submerged Culture of Hericium erinaceum in the Medium Supplemented with Korean Ginseng Extracts (수삼추출물 첨가 배지에서 배양된 노루궁뎅이버섯 균사체 심부발효물 조다당획분의 면역 및 암전이 억제활성)

  • To find the new use of Korean ginseng and mushroom, crude polysaccharides were prepared from submerged cultures of Hericium erinaceum in the medium supplemented with Korean ginseng extracts. When we fractionated crude polysaccharides (HE-GE-CP-1, 3, and 5) from hot-water extracts of submerged cultures of H. erinaceum with ginseng extracts (1%, 3%, and 5% addition of total medium), the yields of HE-GE-CP-1, 3, and 5 were identified at 5.7, 5.1, and 4.8%, respectively. Among crude polysaccharide fractions, HE-GE-CP-5 was significantly higher (1.89-fold of the saline control) than those of HE-GE-CP-1 (1.64-fold) or HE-GE-CP-3 (1.76-fold) on mitogenic activity of splenocytes. HE-GE-CP-5 also had the more potent bone marrow cell proliferation (1.83-fold) rather than HE-CP or HE-GE-CP-1 or HE-GE-CP-3 (1.59- or 1.44- or 1.69-fold, respectively), and anti-metastatic activity as anti-cancer effect showed the highest prophylactic value (72.4% inhibition of tumor control) in 5% supplementation of ginseng extract. However, the lysosomal phosphatase of macrophage was significantly stimulated after HE-GE-CP-3 treatment (2.03-fold). In addition, the immunostimulating and anti-metastatic crude polysaccharide, HE-GE-CP-5, contained mainly neutral sugars (63.2%) with considerable amounts of uronic acid (19.3%) and a small amount of proteins (8.8%). HE-GE-CP-5 can stimulate immune system to inhibit tumor metastasis, and its anti-tumor metastasis may be associated with macrophages, splenocytes and Peyer's patch cells activation.

Effects of Number of Seeds per Hill in Dibbling on Growth Character, Yield and Feeding Value in Jeju Native Sorghum (제주재래수수의 점파립수에 따른 사료수량 및 조성분 분석)

  • Jeju native sorghum was grown at six spot seeding rates (1, 2, 3, 4, 5 and 6) from April 3 to september 6, 2000 in Jeju island to determine influence of number of seeds per hill in dibbling on agronomic characters, forage yield and qualities. Days to heading was increased as number of seeds per hill was increased. Plant height with three seeds per hill (206.7 cm) was the longest, while with six seeds per hill (175.2 cm) was the shortest. Fresh forage, dry matter and crude protein yield and total digestible nutrient (TDN) were the greatest at the three seeds per hill (48.1, 10.1, 0.9, 5.1 MT/ha). Crude protein, crude fat and nitrogen-tree extract (NFE) increased with the number of seeds per hill increased but crude fiber and crude ash decreased.

  • The chemical components in freeze dried Korean Gastrodia elata blume powder were analyzed. The proximate composition was composed of moisture(2.12%), crude protein (4.41%), crude fat(1.20%), crude ash(2.25%), crude fiber(4.60%) and N-free extract(85.15%). Free sugar was composed of glucose(1,314mg%), sucrose(1,081mg%) and fructose(869mg%). Total organic acid content was 2,095mg% and composed of succinic acid(1,238mg%), citric acid, malic acid, oxalic acid and fumaric acid in the order. Among the 11 fatty acids detected, linoleic acid was the most predominant one and unsaturated fatty acid was 80.55 area percent. 18 and 39 amino acids were detected from total and free amino acid, respectively. Arginine, asparagine, proline and sarcosine were the major components of free amino acid. The content of crude saponin and total phenol were 3,768mg% and 821mg%, respectively. Ascorbic acid(6.4mg%), vitamin B1(1.2mg%), vitamin B2(1.7mg%) and niacinamide(0.2m%) were detected as soluble vitamins of the powder. The content of vitamin A and vitamin E was 1.22mg% and 0.32%, respectively.

  • This experiment was conducted to observe the effects of fermented feed by Aspergillus oryzae and Aspergillus niger on the improvement of feed value and the effect of fermented feed additive for meat production of broiler. The results of fermented feed on the improvement of feed value were as follows; I; The effects of fermented feed value improvement were as follaws; 1) There were little difference between fermented feeds by Asp. oryzae and Asp. niger, compared with wheat bran, crude protein contents of Koji was highly increased and its nitrogen free extract and crude fat contents were decreased, but crude fiber and ash were little difference. 2) Total amino acids were highly increased as to fermented feeds but proline in Asp. niger koji feed, and proline and valine in Asp. oryzae koji feed were decreased and other amino acid were increased 2) The effect of fermented feeds on meat production of broiler were as follows; 1) Fermented feeds groups appeared higher weight (p<0.01)than weight of control on end of experimental period, but little difference were recognized between 5% and 10% fermented feed groups. 2) On the weight gain per day, highly significant were recognized(p<0.05) between control and test groups, 10% Asp. oryzae koji group was highest ($12.15{\pm}0.46g$) between all groups. 3) On the yield of carcass, there were significant highly difference (p<0.01) between control and test groups but little difference were recognized between each of 5% groups and 10% groups of fermented feeds. 4) Fermented feed groups appeared higher carcass yield (p (0<0.05) than control. But between all fermented feed groups were a little difference in partly. 5) On the influence of fowl meat composition, amount of moisture contents was a little decrease in fermented feed groups, and crude protein and crude fat were increased. 6) Feed conversion rate resulted a little amount decreasing. Specially, 10% Asp oryzae koji group was lowest (2.89) compare with control (3.35) 3. As a result of economical analysis appeared highest low income in koji groups. Low income were more gained percent of 40.22 in 10% Asp oryzae koji and 33.19 in 10% Asp. niger koji than control.

Antioxidative and Cytotoxicity Activities against Human Colon Cancer Cells Exhibited by Edible Crude Saponins from Soybean Cake (대두박 식용사포닌의 항산화 및 대장암세포 성장 억제효과)

  • To develop soybean cake as a functional food material, the anti-oxidative and cytotoxic activities against human colon cancer cells of crude saponins isolated from 70% (v/v) ethanol extracts of cake were investigated. The Diaion HP-20 adsorption method was used for isolation of crude saponins, which were then eluted with 100% ethanol. The non-saponin fraction was removed by elution with $H_2O$ and 20% (v/v) ethanol. The results of thin layer chromatography (TLC) analysis confirmed that crude saponins were present in the 100% ethanol extract of soybean cake. The hydrogen-donating properties of saponins were more than 60% at a concentration of $1,000\;{\mu}g/mL$. malondialdehyde(MDA) production was $1,200\;{\mu}mol\;MDA/g$ in mouse liver homogenate treated with crude saponins at the concentration of $1,000\;{\mu}g/mL$. This value was lower than that of the control, which was $3,700\;{\mu}mol\;MDA/g$. Saponins inhibited the growth of colon cancer cells in a dose- and time-dependent manner. Saponins also resulted in a decrease in the proportion of cells in the G1 phase of the cell cycle, whereas the cell proportion in G2/M phase was increased with $1,000\;{\mu}g/mL$ saponins. Thus, we conclude that saponins may induce G2/M cell cycle arrest.

Effect of Seeding Date on Forage Yield and Chemical Composition of Kenaf in Jeju (제주지역에서 파종기에 따른 양마의 사료수량 및 조성분 변화)

  • This study has been conducted from April 25, 1999 to June 25, 1999 in Jeju Province in order to determine the influence of seeding date on growth, yield and chemical composition of Kenaf (Hibiscus cannabinus L.) Delaying the seeding date from April 25 to June 25 decreased the plant height of Kenaf from 278.5 cm to 205.7 cm. As for the number of leaves, number of withering leaves, number of branches, and stem diameter decreased as the seeding date was delayed. As the seeding date was delayed, fresh forage yield decreased from 98.5MT/㏊ to 45.9MT/㏊, dry matter yield from 20.7MT/㏊ to 8.2MT/㏊, crude protein (CP) yield from 2.9MT/ha to 1.3MT/ha and total digestible nutrients (TDN) yield form 1l.6MT/㏊ to 5MT/㏊. However, both leaf yield and stem yield were nearly the same tendency. Delaying the seeding date from April 25 to June 25 increased crude protein contents of leaves from 21.5% to 24.4%, crude fat contents from 5.2% to 6.1%, nitrogen free extract (NFE) contents form 39.9% to 41.2% and TDN contents from 64.3% to 69.7%. However, delaying seeding date decreased crude fiber contents from 20.8% to 17.5%, and crude ash contents from 7.9% to 7.0%. Based on the these findings, optimum seeding date for forage production of Kenaf seems to be about 25 April in atmospheric phenomena and volcanic ash soils of Jeju island.

Studies on the Composition and Antioxidative Effect of Leaves form Korean Rosa davurica Pall. (한국산 생열귀나무(Rosa davurica Pall.) 잎의 성분 및 항산화 효과에 관한 연구)

  • The objective of this study was carried out to investigate nutritional characteristics and biological activities effects of Korean leaf of Rosa davurica Pall. in vitro. They were extracted with methanol and then further fractionated to n-hexane, chloroform, ethyl-acetate, n-butanol and water from methanol extracts. Methods of the antigenotoxic used in this experiment were UVA/UVB absorption property and DPPH radical scavenge. The proximate compositions of leaves of Rosa dauvrica Pall were 67.5% of crude Moisture, 0.7% of crude fat, 6.8% of crude protein, 6.1% of crude ash, and 20.8% of crude fiber. The major minerals were K (1637.2 mg%), Ca (219.5 mg%), P (182.1 mg%), and Mg (135.1 mg%). Most of the fractions of methanol extract which leaves of Rosa dauvrica Pall. have strong absorbency at UVB region (308 nm) and UV A region (350nm). These fractions have a good absorbency property as synthetic filter and could be served as substitutes for synthetic UV sunscreen agents. All fractions (n-hexane, ethyl-acetate, n-butanol and water) from methanol extracts except chloroform fraction exhibited DPPH radical scavenging activity with IC$\_$50/ of 35.3, 6.0, 14.0, and 18.0 $\mu\textrm{g}$/mL.

Influence of High Hydrostatic Pressure Treatment after Germination on Anti-proliferation Effects of Soyasaponin-rich Fraction in Black Soybean (Glycine max L.) (발아와 고압처리가 검정콩 사포닌 추출물의 암세포주 증식억제에 미치는 영향)

  • The objective of this study was to determine the effect of high hydrostatic pressure (HHP) treatment on proliferation of human cancer cell lines (MCF-7, HCT-116, PC-3 and AGS) of crude soyasaponin extracts in germinated black soybean. Black soybean was germinated and subjected to HHP, followed by preparation of crude soyasaponin extracts. Cell treatments done with extracts less than $400{\mu}g/mL$ concentrations had no significant effect on 3T3-L1 adipocyte cell viability. The inhibitory effect of crude soyasaponin extracts with germination periods and applied pressure on breast cancer cell (MCF-7), human colon cancer cell (HCT-116), human gastriccancer cell (AGS) and prostate cancer cell (PC-3) growth were investigated using MTT assay. The highest anti-proliferation of human cancer cell line of crude soyasaponin extracts was observed at 150 MPa treatment after germination for 4 days (150 MPa-Day 4). The cell viability on MCF-7, HCT-116, PC-3 and AGS cell lines of crude soyasaponin extract in 150 MPa-Day4 was 48.82%, 57.37%, 39.89% and 23.94% at $400{\mu}g/mL$, respectively. These results suggest that soyasaponin extracts from black soybean subjected to HHP after germination may mediate physiological activity.

Quality Characteristics of Low-Fat Sausage Containing Curcumin Extract during Cold Storage (울금 추출물 함유 저지방 소시지의 냉장저장 중 품질 특성)

  • The effects of curcumin extract on the physicochemical, microbial and sensory properties of low-fat sausages during refrigerated storage were studied. Sausage products were produced with three different formulations including 0%, 2.5%, and 5.0% curcumin extract. Low-fat sausages made with the addition of curcumin extract had lower (p<0.05) crude fat content, pH and TBARS values than the control sample. The addition of curcumin extract did not affect the water holding capacity, cooking loss, shear force, meat color, texture profile, and total bacterial count in low-fat sausages duringstorage (p>0.05). With regard to sensory evaluation, 2.5% curcumin extract added to low-fat sausages resulted in a high overall acceptability (p<0.05). In conclusion, low-fat sausages with added 2.5% curcumin extract had a higher acceptability and lipid oxidative stability during storage than products without curcumin extract.

The Effect of Maesil(Prunus mime) Extract on the Acid Production and Growth of Yoghurt Starter (매실(Prunes mime) 착즙액이 Yoghurt Starter의 산생성 및 증식에 미치는 영향)

  • This experiment was carried out to investigate the effect of Maesil extract on the acid production and growth of yoghurt starter in the skim milk medium. The Maesil extract was added to skim milk medium fur 1% to 9% and the medium was fermented by single or mixed culture of 4 types of lactic acid bacteria(Streptococcus thermophilus, Lactobacillus acidophilus, Lactobacillus, bulgaricus, Lactobacillus casei). The chemical composition of Maesil, the changes in acid production (titratable acidity, pH) and number of viable cells of the medium during lactic fermentation in skim milk added with Maesil extract, and the keeping quality of curd yoghurts containing Maesil extract have determined. The composition of Maesil were 0.4% crude ash, 4.1% dietary fiber, 4.66%l citric acid, 0.264% total sugars and 405.34mg% vitamin C. The addition of Maesil extract stimulated the acid production and propagation of the lactic acid bacteria. Among the treatments tested, the addition of 3% Maesil extract with the mixed culture of Streptococcus thermophilus, Lactobacillus acidophilus and Lactobacillus casei produced the highest amount of acid(1.23%) and showed the highest number of viable cell counts(3.6$\times$10$^{11}$ cfu/mL). When the curd yoghurts containing 3% Maesil extract with the mixed culture of the lactic acid bacteria were kept at 4$^{\circ}C$ and 2$0^{\circ}C$ for 30 days, it was showed that the changes of titratable acidity, pH and number of viable cell counts of the lactic acid bacteria were not significantly different during storage. Therefore the keeping quality of the curd yoghurts adding 3% Maesil extract showed relatively good at the shelf-life.
