Odorant Receptors Containing Conserved Amino Acid Sequences in Transmembrane Domain 7 Display Distinct Expression Patterns in Mammalian Tissues

  • Mammalian genomes are well established, and highly conserved regions within odorant receptors that are unique from other G-protein coupled receptors have been identified. Numerous functional studies have focused on specific conserved amino acids motifs; however, not all conserved motifs have been sufficiently characterized. Here, we identified a highly conserved 18 amino acid sequence motif within transmembrane domain seven (CAS-TM7) which was identified by aligning odorant receptor sequences. Next, we investigated the expression pattern and distribution of this conserved amino acid motif among a broad range of odorant receptors. To examine the localization of odorant receptor proteins, we used a sequence-specific peptide antibody against CAS-TM7 which is specific to odorant receptors across species. The specificity of this peptide antibody in recognizing odorant receptors has been confirmed in a heterologous in vitro system and a rat-based in vivo system. The CAS-TM7 odorant receptors localized with distinct patterns at each region of the olfactory epithelium; septum, endoturbinate and ectoturbinate. To our great interests, we found that the CAS-TM7 odorant receptors are primarily localized to the dorsal region of the olfactory bulb, coinciding with olfactory epithelium-based patterns. Also, these odorant receptors were ectopically expressed in the various non-olfactory tissues in an evolutionary constrained manner between human and rats. This study has characterized the expression patterns of odorant receptors containing particular amino acid motif in transmembrane domain 7, and which led to an intriguing possibility that the conserved motif of odorant receptors can play critical roles in other physiological functions as well as olfaction.

Cloning and Sequence Analysis of the Full-length cDNA of Coxsackievirus B3 Isolated in Korea (한국에서 분리된 콕사키 바이러스 B3 cDNA의 클로닝 및 전체 염기서열 분석)

  • We have determined and analyzed the full-length cDNA sequence of a coxsackievirus B3 (CVB3) Korean isolate (CVB3-Korea/97) which has been known as a general human pathogen. The whole genome contains 7,400 nucleotides and has a single large open reading frame with 6,555 nucleotides that encodes a potential polyprotein precursor of 2,185 amino acids. The genome also contains a 5' non-coding region (NCR) of 741 bases and a 3' NCR of 104 bases followed by poly(A) tail. Sequence homologies of nucleotides and deduced amino acids between the CVB3-Korea/97 strain and the prototype (Nancy strain) were 81.7% and 91.5%, respectively. The genes encoding the functional proteins including viral protease and RNA dependent RNA polymerase showed higher homology than those encoding the structural proteins. We have further analyzed the sequences of 5' NCR, VP1 and VP2 of CVB3-Korea/97, which are known as cardiovirulent determining factors at the nucleotide and amino acid levels. Although the CVB 3-Korea/97 strain was isolated from an aseptic meningitis patient without cardiomyopathy, its 234th nucleotide and 165th amino acid were uracil and Asn as same as those of other cardiovirulent strains one. However, the 155th amino acid of VP1, which closely associated with cardiovirulence, was replaced with $Arg^{155}$ by single nucleotide substitution from $A^{2916}$ to $T^{2916}$. Moreover, additional amino acid substitutions were observed in the flanking region of $Asp^{155}$. Taken together, amino acid(s) substitution in VP1 may playa critical role in determining cardiovirulence of the CVB3-Korea/97 strain rather than individual nucleotide replacements in the 5' NCR and/or an amino acid substitution in VP2.

Changes in Nutrient Distribution, Cycling, and Availability in Aspen Stands after an Intensive Harvesting (집약적(集約的)인 벌채(伐採)로 인한 미국(美國)사시나무림내 양분(養分)의 분포(分布), 순환 (循環) 및 가용성(可溶性)의 변화(變化))

  • Aspen demand has increased recently in the Great Lakes region in the United States. Since aspen has moved into the region in late 1800's, its growing stock has increased so as to change forestry industry of the Lake States. Intensive timber harvesting and biomass removal may cause nutrient depletion, especially on nutrient-poor sites. Forest nutrients and nutrient cycling were investigated in aspen stands of 7-10, 27-33, and 41-42 year-old growing on sandy soils in Minnesota. Nutrients added to the aspen stands by atmospheric deposition and soil weathering were efficiently absorbed and stored in the tree biomass. Aboveground biomass increased from $24.4t{\cdot}ha^{-1}$ at young stands to $139.2t{\cdot}ha^{-1}$ at mature stands. Nutrients accumulated in the tree biomass showed same magnitude of difference. Nutrients added to the site through atmospheric deposition were in the order of Ca, N, K, Mg, and P. Annual litterfall was greater in older stands. However, the amount of nutrients returned by litterfall was not significantly different among stand ages due to the greater nutrient contents in the litterfall of young stands. Litter decomposition and nutrient release rates were greater at young stands than at older stands. Likewise, nutrient availability was higher in young aspen stands and became lower as the stands grew older. Nutrient leaching loss was minimal at all stand ages. Soil N mineralization was greater at young stands than at older stands. Nutrient cycling process was facilitated in young aspen stands with an increased level of available nutrients, Based on the estimations of nutrient balance and nutrient removal by harvesting, Ca was the most critical element which was likely to be depleted if aspen stands are intensively harvested with short rotations.

Progress and Prospect of Research on North Korea in Korean Human Geography (한국 인문지리학 분야에서 북한 연구의 동향과 과제)

  • This study is to review research issues on North Korea and unified territory in terms of topics and approach method in Korean human geography. The conclusion of this paper is as follows. Before 1980's, topics on political geography, such as geopolitics or unified land, were the main stream in research. In 1990's with the end of the cold war and the access to material which was published in North Korea, scope of research was widened especially in geography education. After 2000's with the expansion of cooperation between South and North Korea, the scope of topics were more expanded in all field of human geography, for example, critical geopolitic in political geography, Gaeseong Industrial Complex, Najin-Seonbong region in economic geography, place names, Geumgangsan, North Korean defectors in social and cultural geography. The approach method of toward North Korea is fall into two categories. One is regional geography and the other is the unified land. In the latter approach, topics on the regional structure after reunification or on the life adaptation of defectors in South Korea etc. were studied. After unification of land, new Korean Studiea will be established and human geographers should make ready for this. Before unification, research on the land in north Korea should be proceeded in terms of historical geography.

Study on the stress distribution depending on the bone type and implant abutment connection by finite element analysis (지대주 연결 형태와 골질에 따른 저작압이 임프란트 주위골내 응력분포에 미치는 영향)

  • Oral implants must fulfill certain criteria arising from special demands of function, which include biocompatibility, adequate mechanical strength, optimum soft and hard tissue integration, and transmission of functional forces to bone within physiological limits. And one of the critical elements influencing the long-term uncompromise functioning of oral implants is load distribution at the implant- bone interface, Factors that affect the load transfer at the bone-implant interface include the type of loading, material properties of the implant and prosthesis, implant geometry, surface structure, quality and quantity of the surrounding bone, and nature of the bone-implant interface. To understand the biomechanical behavior of dental implants, validation of stress and strain measurements is required. The finite element analysis (FEA) has been applied to the dental implant field to predict stress distribution patterns in the implant-bone interface by comparison of various implant designs. This method offers the advantage of solving complex structural problems by dividing them into smaller and simpler interrelated sections by using mathematical techniques. The purpose of this study was to evaluate the stresses induced around the implants in bone using FEA, A 3D FEA computer software (SOLIDWORKS 2004, DASSO SYSTEM, France) was used for the analysis of clinical simulations. Two types (external and internal) of implants of 4.1 mm diameter, 12.0 mm length were buried in 4 types of bone modeled. Vertical and oblique forces of lOON were applied on the center of the abutment, and the values of von Mises equivalent stress at the implant-bone interface were computed. The results showed that von Mises stresses at the marginal. bone were higher under oblique load than under vertical load, and the stresses were higher at the lingual marginal bone than at the buccal marginal bone under oblique load. Under vertical and oblique load, the stress in type I, II, III bone was found to be the highest at the marginal bone and the lowest at the bone around apical portions of implant. Higher stresses occurred at the top of the crestal region and lower stresses occurred near the tip of the implant with greater thickness of the cortical shell while high stresses surrounded the fixture apex for type N. The stresses in the crestal region were higher in Model 2 than in Model 1, the stresses near the tip of the implant were higher in Model 1 than Model 2, and Model 2 showed more effective stress distribution than Model.

A Critical Assesment on the Shin-hwal-ryuk Policy as a New Regional Development Policy in Korea (신활력사업계획 수립 및 추진과정에 대한 평가와 개선 방안)

  • This paper aims to evaluate the new regional development policy which is entirely focused on the lagging rural regions in Korea. The new regional development, called the shin-hwal-ryuk policy is to reflect the radical change in the idea and philosophy of rural development. Although traditional rural development policies were top-down-based and physical infrastructure-centered, the new rural development policy idea is based on bottom-up and soft infrastructure which is related to the promotion of regional innovation capacities. However, it is revealed that the new rural policy involves a variety of problems in the process of establishing and making progress the policy plan in a local level. In the operating process of the policy plan, the central government has shown too quick-tempered for achieving visible outcomes, while many of local government suffer from the lacks of professional capabilities to carry out the plan. I see that as a result of the mixture of these problems the new policy is regarded as 'passively localized process' by the central government rather than 'actively localizing process' by the region for building regional innovation capacity.

Han River Pollution Studies (한강의 오염도)

  • The Han River is an important water source in Seoul and neighbouring districts, for public and industrial supply, and for agriculture and fishery. Nowadays, more than six million inhabitants are supplied withe water from this river. The total length of the river is 470km, and has 17 10$\^$9/㎥ an average annual flow. The hydrographic characteristics at Seoul are 653㎥/sec in an average flow, 4,608㎥/sec in the maximum average flow, and 201㎥/sec in the minimum average flow. These are influenced in some degree by snowmelt in early spring, and greatly by the flood during summer. For the pollution problems, the periods of low flow are critical ones. As a rule they occur around the months November through June. Nowadays, most of the sewage from towns and industries is discharged untreated. Apart from domestic and industrial sewages, there are some discharges of mineral matter by mines in the upriver region. In general, water quality of the Han River is kept very clean and healthy until Kwangnaru of the upper region of Seoul. A large pollution, however, is received in the downstream by the domestic and industrial sewages of Seoul. It can be seen that dissolved oxygen, COD and BOD$\sub$5/ diminish markedly, and the intensity of almost every water parameter of the river continues to increase. Comparison of the figures for 1971 derived from a sampling point 40km downstream of Kwangnaru leads to the conclusion that hardness, Ca and Mg were no changed; alkalinity, Si and soluble- Fe were slightly increased; CO$\sub$2/, acidity, Cl, NO$\sub$2/-N, Cu, Zn and Al were increased in 2 and 3 times; total residue, total ignitious residue, COD, BOD$\sub$5/, NH$\sub$4/-N, PO$\sub$4/-P, Mn, Pb and total-Fe were increased in 4 to 7 times; and SO$\sub$4/, particulate-Fe and Cd were increased in 10 to 11 times. On the other hand, coliforms were increased in 650 times; fecal coliforms in 365 times; enterococci and total plate counts in 30 times, respectively. In view points of water quality standards, the down Han River water is now leveling out in Cd, coliforms and fecal coliforms for the agricultural use; in dissolved oxygen and some trace elements (Cu, Zn, Pb and Cd) for the fishery use; in ammonia, COD, BOD$\sub$5/, and Cd for the drinking use.

  • Rainfall infiltration through the unsaturated zone is influenced by a range of factors including topography, geology, soil, rainfall intensity, temperature and vegetation; the actual infiltration varies largely in time and space. The infiltration capacity of soil is a critical factor in identifying groundwater recharge and leakage of surface water. It may differ depending on soil types and geological features of a particular basin or territory as well as on the usage of the land. This study was conducted in forest and farmland region of the mid-mountain area (EL. 50~300 m) of Jeju Island to test soil infiltration capacity of the area where rainfall contributes to groundwater. Results were analyzed using the four soil group classification methods presented by Jeong et al. (1995) and NAS (2007) to discover that the method offered by NAS (2007) is more reliable in the mid-mountain area of Jeju Island. The study compares and reviews the existing classification methods using the results of infiltration capacity tests executed on different soil groups throughout the whole region of the Jeju mid-mountain area. It is expected that this work will serve as a guideline for evaluating surface water recharge and hydraulic characteristics of Jeju Island.

Genetic Characterization of H7-subtype Avian Influenza Viruses (H7 아형 조류인플루엔자 바이러스의 유전자 특성)

  • Based on their virulence, the avian influenza viruses (AIVs) are classified into two pathotypes: low pathogenic avian influenza (LPAI) virus and highly pathogenic avian influenza (HPAI) virus. Among the 16 HA subtypes of AIV, only the H5 and H7 subtypes are classified as HPAI. Some AIVs, including H5 and H7 viruses, can infect humans directly. Six H7 subtype isolates from wild birds of the H7N7 (n=4) and H7N1 (n=2) subtypes were characterized in this study. Phylogenetic analysis showed that eight viral genes (HA, NA, PB2, PB1, PA, NP, M, and NS) of the H7 isolates clustered in the Eurasian lineage, the genetic diversity of which is indicated by its division into several sublineages. The Korean H7 isolates had two motifs, PEIPKGR and PELPKGR, at the HA cleavage site, which have been associated with LPAI viruses. Six H7 isolates encoded glutamine (Q) and glycine (G) at positions 226 (H3 numbering) and 228 of HA, suggesting avian-type receptor-binding specificity. None of the Korean H7 isolates had the amino acid substitutions E627K in PB2 and I368V in PB1, which are critical for efficient replication in human cells. The Korean H7 isolates showed no deletions in the NA stalk region and in NS. These results suggest that the Korean H7 isolates from wild birds are different from the H7N9 influenza viruses isolated in China in 2013, which are capable of infecting humans.

A Critical Review of Korean Politics Before and After by Three Kim's Politics: Political Effects and Distortions (3김정치 전후 한국정치의 비판적 검토: 정치적 효과와 왜곡)

  • After the democratization movement in 1987, korean politics was transformed into three kim's politics by y Kim Yeong-sam, Kim Dae-jung and Kim Jong-pil. Before the time of three kim's politics, korean politics lasted for one long term, but three kim's politics made possible the peaceful regime change through political party integration and party coalition. The evaluation of three kim's politics coexist both positive and negative. The Positive political effects are diversification of political party composition and stabilization of regime change. Three kim's politics transformed the political party composition of Korea from a two-party system to a multi-party system, made possible a peaceful regime change through the unification of three parties and the DJP coalition. However, the negative political distortions of three kim's politics are the shortening of political parties and the concentration of political ideology. In three kim's politics, political party in Korea has a very short life due to the creation of political parties, the dissolution of political parties, the reorganization of political parties and the integration of political parties. Conservatism and progressive tendencies related three kim's politics were stabilized through Yeongnam region and Honam region. Therefore, three kim's politics means that the proportion of Korean politics is very high. Political effects and distortions derived from three kim's politics have become a challenge for Korean politics to overcome.
