Genetic diversity and structure of natural populations of Picea jezoensis in South Korea

  • Picea jezoensis (Sieb. et Zucc.) Carriere is one of the major and widespread components of the cold-temperate and boreal forests in Russian Far East, northeast China, Korea, and Japan. However, it is restricted to a highly fragmented range in South Korea with small populations. Mean expected heterozygosity $(H_e)$ based on 22 loci in 11 isozyme systems was 0.077 for four sampled populations that covered the whole distribution range of P. jezoensis in South Korea. This value is within the range reported for conifers, but it is very low compared to that of other spruce species as well as that of P. jezoensis populations in Russian Far East. Most populations had a slight excess of heterozygotes and the Wright's $F_{IS}$ (-0.019) was comparable to that previously reported for other spruce species. In all of the four populations, the Wilcoxon sign-rank test indicated no greater heterozygosity than that expected for populations at mutation-drift equilibrium, suggesting that the populations have not been bottlenecked recently. Despite a fragmented range and isolated populations, population differentiation was not high $(F_{ST}=0.047)$ and the number of migrants per generation was 5.09. Nei's genetic distances were also small $({\bar{D}}=0.005)$ but strongly related to geographic distances between populations, suggesting an Isolation by Distance. The northernmost isolate, Mt. Gyebang population was genetically distinct from the other three populations. Implications for the conservation of genetic variation of P. jezoensis in South Korea were discussed.

  • The Korean plum yew (Cephalotaxus koreana Nakai) is a shade-tolerant, coniferous shrub. The seeds have been used as a folk medicine in Korea, and an alkaloid extract (HTT) is known to have anticancer properties. We estimated the genetic diversity of 429 trees in 16 populations in South Korea using 194 polymorphic amplicons from seven combinations of AFLP primer-restriction enzymes. The average number of effective alleles and the percentage of polymorphic loci were 1.37 and 79.4%, respectively. Shannon's diversity index and the expected heterozygosity were 0.344 and 0.244, respectively. We divided 16 populations into four groups on the UPGMA dendrogram and the PCA biplot. The first two principal components explained 84% of the total genetic variation. Genetic differentiation between populations explained 14% of total genetic variation, and the remaining 86% came from difference between individuals within populations, as determined by an analysis of molecular variance (AMOVA). However, the genetic differentiation did not correlate with the geographic distance between populations from the Mantel test. The Bayesian statistics, which are comparable to Wright's $F_{ST}$ and Nei's $G_{ST}$, were ${\theta}^I=0.406$ and ${\theta}^{II}=0.172$, respectively. The population genetic diversity was slightly lower, and the strength of genetic differentiation was much weaker, than the average of those plants having similar life histories, as assessed using arbitrary marker systems. We discuss strategies for the genetic conservation of the plum yew in Korea.

Isolation and inheritance of microsatellite loci for the oily bittering (Acheilognathus koreensis): applications for analysis of genetic diversity of wild populations

  • The oily bittering Acheilognathus koreensis is a freshwater species that is endemic to Korea and is experiencing severe declines in natural populations as a result of habitat fragmentation and water pollution. For the conservation and restoration of this species, it is necessary to assess its genetic diversity at the population level. We developed 13 polymorphic microsatellite loci that were used to analyze the genetic diversity of two populations collected from the Kum River and the Tamjin River in Korea. All loci exhibited Mendelian inheritance patterns when examined in controlled crosses. Both populations revealed high levels of variability, with the number of alleles ranging from 3 to 20 and observed and expected heterozygosities ranging from 0.500 to 0.969 and from 0.529 to 0.938, respectively. None of the loci showed significant deviation from Hardy-Weinberg equilibrium, and one pair of loci showed significant linkage disequilibrium after Bonferroni correction. Pairwise $F_{ST}$ and genetic distance estimation showed significant differences between two populations. These results suggest that the microsatellites developed herein can be used to study the genetic diversity, population structure and conservation measure of A. koreensis.

  • 북아메리카 원산의 나래가막사리(Verbesina alternifolia)는 국화과의 다년생 식물로, 우리나라에는 1980년대 이후에 들어온 것으로 알려져 있다. 나래가막사리는 전국의 산지 5개 지역, 도로 12개 지역, 하천변 5개 지역에서 분포가 조사되었고, 치악산국립공원, 무등산도립공원 등 생태계 중요지역에도 집중 분포하였다. 나래가막사리는 도로 및 계곡을 따라 산지 및 하천변으로 확산되며, 생명력이 강해 음지, 양지 모두에서 잘 자라는 것으로 나타났다. 군락을 형성한 곳에서는 서서히 분포역을 넓혀 주변 토착식생의 정착을 간섭하며 산지 및 하천변으로 확산되고 있다. 나래가막사리는 특히 자연식생 보전이 필요한 지역에서 분포를 제한할 수 있는 관리가 필요하며, 지하부의 유묘발달로 지상부 관리보다 뿌리제거를 통한 작업이 이루어질 수 있도록 제거방법을 보완할 필요가 있다.

Molecular analysis of genetic diversity, population structure, and phylogeny of wild and cultivated tulips (Tulipa L.) by genic microsatellites

  • Tulip (Tulipa L.) is one of the most important ornamental geophytes in the world. Analysis of molecular variability of tulips is of great importance in conservation and parental lines selection in breeding programs. Of the 70 genic microsatellites, 15 highly polymorphic and reproducible markers were used to assess the genetic diversity, structure, and relationships among 280 individuals of 36 wild and cultivated tulip accessions from two countries: Iran and the Netherlands. The mean values of gene diversity and polymorphism information content were 0.69 and 0.66, respectively, which indicated the high discriminatory power of markers. The calculated genetic diversity parameters were found to be the highest in wild T. systola Stapf (Derak region). Bayesian model-based STRU CTU RE analysis detected five gene pools for 36 germplasms which corresponded with morphological observations and traditional classifications. Based on analysis of molecular variance, to conserve wild genetic resources in some geographical locations, sampling should be performed from distant locations to achieve high diversity. The unweighted pair group method with arithmetic mean dendrogram and principal component analysis plot indicated that among wild tulips, T. systola and T. micheliana Hoog exhibited the closest relationships with cultivated tulips. Thus, it can be assumed that wild tulips from Iran and perhaps other Middle East countries played a role in the origin of T. gesneriana, which is likely a tulip species hybrid of unclear origin. In conclusion, due to the high genetic variability of wild tulips, they can be used in tulip breeding programs as a source of useful alleles related to resistance against stresses.

Assessing Trees Diversity in Jebel Elgarrie Forest Reserve in the Blue Nile State, Sudan

  • The study aims to examine population indices of mature trees in Jebel Elgarrie forest, Blue Nile State, Sudan. We used remote sensing techniques to stratify the forest into vegetation classes depending on tree density. We distributed 97 circular sample plots (0.1 ha) proportionally to the area of the vegetation classes. In each sample plot we identified, counted and recorded all mature trees (DBH ≥10 cm). We calculated frequency, density, abundance, richness, evenness and diversity for each species and we drew abundance rank curve of mature trees. We used One-Way ANOVA to test for differences (α=0.05) in mean density (No./ha) of mature trees between vegetation classes. Results revealed that the forest was conveniently sub-divided into high density (C1), medium density (C2), low density (C3) and bare farm land (C4) classes. We identified fifteen tree species that belong to 10 families and 14 genera. Combretaceae and Fabaceae were the common families while Anogeissus leiocarpa was the most frequently occurring species. While species diversity varied between vegetation classes, diversity of the forest as a whole is low. While mean density of mature trees in C1, C2, C3 and C4 it was 100, 74, 10, and 0, respectively, it was 54 for the whole forest indicating low stocking, Following One-Way ANOVA, multiple comparisons revealed significant differences in mean density of mature trees between C1 & C3 and C2 & C3. The study provided empirical results on population indices of mature tree species, which would be of importance for successful management and conservation of the forest.

IPA를 활용한 농촌다움에 대한 거주민의 인식 및 평가 - 고창군 지역 리더그룹을 중심으로 - (Residents' Perception and Evaluation of New Ruralism Using IPA - Focusing on the Regional Leader Group in Gochang-gun -)

  • This study measured the importance and satisfaction of the values related to 'New Ruralism' using the IPA(Importance-Performance Analysis) technique by the rural leaders' group. In the result, the satisfaction value was evaluated lower in the importance of 'landscape and ecological resource conservation', 'settlement environment', and 'population' related items. On the other hand, the items of 'tourism resource management', 'landscape damage', and 'landscape and ecological resource conservation' showed high satisfaction. Therefore, the leaders' group of Gochang-gun perceived the importance of local tourism image, landscape damage prevention, settlement environment, and rural landscape in 'New Ruralism' management. However, the leaders' group valued lower in the items of population, ecological value and natural and historical tourist sites. This seems to be a characteristic of the leaders' group mainly engaged in agriculture. Through factor analysis, the six items excluded from the analysis were considered important factors in 'New Ruralism' in Gochang-gun through a review of previous studies. Still, the leaders' group of Gochang-gun did not recognize them as important items in the rural resource. However, these items will be able to contribute to the development of 'New Ruralism' in Gochang-gun, and it is necessary to improve the awareness of the leader group. Even if local governments or experts present an ideal goal for rural development, it is difficult to achieve the goals if there is a difference from the perception of residents. Therefore, it is important to understand the perception of 'New Ruralism' of the leaders' group who plays an important role in regional development projects.

Population structure and regeneration of Himalayan endemic Larix species in three high-altitude valleys in Nepal Himalaya

  • Background: The Himalayan forests are of great importance to sustain the nature and community resource demands. These forests are facing pressures both from anthropogenic activities and ongoing global climatic changes. Poor natural regeneration has been considered a major problem in mountainous forests. To understand the population structure and regeneration status of Larix (Larix griffithiana and Larix himalaica), we conducted systematic vegetation surveys in three high-altitude valleys namely Ghunsa (Kanchenjunga Conservation Area, KCA), Langtang (Langtang National Park, LNP), and Tsum (Manaslu Conservation Area, MCA) in Nepal Himalaya. The average values of diameter at breast height (DBH), height, and sapling height were compared for three sites and two species using Kruskal-Wallis test. Population structure was assessed in terms of proportion of seedlings, saplings, and trees. Regeneration was analyzed using graphical representation of frequencies of seedlings, saplings, and trees in histograms. Results: The results showed that the population structure of Larix in terms of the proportion of seedling, sapling, and tree varied greatly in the three study areas. KCA had the highest record of seedling, sapling, and tree compared to other two sites. Seedlings were the least among three forms and many plots were without seedlings. We found no seedling in MCA study plots. The plot level average DBH variation among sites was significant (Kruskal-Wallis χ2 = 7.813, df = 2, p = 0.02) as was between species (Kruskal-Wallis χ2 = 5.9829, df = 1, p = 0.014). Similarly, the variation in average tree height was significant (Kruskal-Wallis χ2 = 134.23, df = 2, p < 0.001) among sites as well as between species (Kruskal-Wallis χ2 = 128.01, df = 1, p < 0.001). All the sites showed reverse J-shaped curve but more pronounced for KCA and MCA. In comparing the two species, Larix griffithiana has clear reverse J-shaped diameter distribution but not Larix himalaica. Conclusion: The varied responses of Larix manifested through regeneration status from spatially distinct areas show that regeneration limitations might be more pronounced in the future. In all the three studied valleys, regeneration of Larix is found to be problematic and specifically for Larix griffithiana in MCA and Larix himalaica in LNP. To address the issues of disturbances, especially serious in LNP, management interventions are recommended to sustain the unique Himalayan endemic conifer.

설악산국립공원 멸종위기 산양(Naemorhedus caudatus) 개체군 크기와 서식지 이용 현황 (Current status of population size and habitat selection of the long-tailed goral(Naemorhedus caudatus) in Seoraksan National Park)

  • 본 연구에서는 설악산국립공원에 서식하는 멸종위기종 산양과 서식지의 보전 및 관리를 위해 산양의 분변 및 카메라트랩을 이용한 개체수와 서식지 이용 분석을 2010~2014년 동안 수행하였으며 생태학적 조사 방법(흔적조사, 카메라트랩조사)을 사용하였다. 분변 조사와 카메라트랩핑 분석 결과 각각 166개체 및 251개체의 산양이 설악산국립공원에 서식하고 있는 것으로 나타났다. 흔적조사(분변)를 통한 서식지 이용 특성 분석에서 산양은 경사도 $35^{\circ}{\sim}60^{\circ}$, 고도 600~700m, 향 북동, 수계와의 거리 0~50m, 도로와의 거리 300~600m, 활엽수림을 가장 선호하는 것으로 나타났다. 카메라트랩 조사를 통한 산양 개체군 구성, 주간(07-18시)이 56.5%, 야간(18-07시)이 43.5%로 산양의 활동성을 파악하였다. 이러한 산양의 개체수와 서식지 이용 특성 분석은 향후 그들이 살아가는 서식지의 보전과 서식지의 관리를 위한 중요한 기초 자료로 활용될 수 있을 것이다.

AFLP 마커를 이용한 단양쑥부쟁이 개체군의 유전다양성 보전을 위한 최소개체군의 크기산정 (Assessment of the Minimum Population Size for ex situ Conservation of Genetic Diversity in Aster altaicus var. uchiyamae Populations Inferred from AFLP Markers)

  • 본 연구는 멸종위기식물인 단양쑥부쟁이(Aster altaicus var. uchiyamae)의 개체군을 대상으로 유전다양성을 유지하는데 필요한 최소개체수를 산정하기 위하여 수행되었다. 단양쑥부쟁이가 분포하고 있는 네 지역에서 각각 유전다양성 및 유전적 분화도를 분석하였다. AFLP(amplified fragment length polymorphism) 마커를 이용한 유전적 변이의 분석결과, 총 4개의 프라이머 조합에 대해서 936개의 밴드가 확인되었으며, 그 중 934개의 밴드(99.8%)가 다형성을 보여주었다. 단양쑥부쟁이 개체군 내에서 유전다양성(PPB = 45.3%, h = 0.104, I = 0.168, hs = 0.108)은 높은 수준으로 나타났으며, 개체군 간 유전적 분화도($G_{ST}$ = 0.075, ${\theta}^B$ = 0.079)는 낮은 수준이었다. AMOVA(Analysis of molecular variance)분석 결과에서도 전체 유전적 변이 중 91%가 개체군 내에서 보이는 반면, 9%는 개체군 간 변이에 기인한 것으로 나타났다. 단양쑥부쟁이 개체군에서 보이는 유전적 특성은 개체군 간의 빈번한 유전자 이동에 기인한 것으로 사료된다. 최대화 전략법에 의하여 경기도 여주일대의 3개 개체군을 대상으로(굴암, 도리섬, 삼합) 개체군 내 최소개체수를 산정한 결과 도리섬개체군에서는 17개체, 삼합개체군에서는 16개체, 굴암개체군에서는 11개체로 파악되었다. 단양쑥부쟁이 개체군의 최소개체수에 대한 정보는 효율적인 현지 외 보전을 위한 가이드라인을 제시해 줄 수 있다.
