This study was performed to investigate the potential of pear (Pyrus pyrifolia) as a immune-modulating functional food by assay of splenocytes proliferation and induction of cytokines (IFN-${\gamma}$, IL-4) in vitro. When mouse splenocytes were exposed to various concentration (0.16, 0.31, 0.63, 1.25, 2.50 mg/ml) of pear methanol extracts (P-M) without mitogens, splenocytes proliferation (SP) was significantly increased. Also, SP to mitogens, concanavalin A (Con A) and lipopolysaccharide (LPS) were significantly increased by P-M when compared with controls. When splenocytes were cultured with P-M in the presence of Con A, cytokine (IFN-${\gamma}$, IL-4) levels in culture supernatant were significantly enhanced in a dose-dependent manner except 2.5 mg/ml when compared with control group. Therefore, our study suggest that the pear has the potential of being an immune-modulating functional food.

Antitumor and immunomodulatory activities of an aqueous extract (GF100) of Acanthopanax senticosus was examined. In experimental lung metastasis of colon26-M3.l carcinoma cells, intravenous (i.v.) administration of GF100 2 days before tumor inoculation significantly inhibited lung metastasis in a dose-dependant manner. The i.v. administration of GF100 also exhibited the therapeutic effect on tumor metastasis of colon26-M3.1 cells, when it was injected 1 day after tumor inoculation. In an in vitro cytotoxicity analysis, GF100 enhanced the responsiveness to a mitogen, concanavalin A (ConA), of splenocytes in a dose-dependent manner. (omitted)

Near infrared (NIR) spectroscopy is a powerful technique for non-destructive analysis that can be obtained in a wide range of environments. Recently, NIR measurements have been utilized as probe for quantitative analysis in agricultural, industrial, and medical sciences. In addition, it is also possible to make practical application on NIR for molecular structural analysis. In this work, Fourier transform near infrared (FT-NIR) measurements were carried out to utilize extensively in the relative amounts of different secondary structures were employed, such as Iysozyme, concanavalin A, silk fibroin and so on. Several broad NIR bands due to the protein absorption were observed between 4000 and $5000\;^{-1}$. In order to obtain more structural information from these featureless bands, second derivative and Fourier-self-deconvolution procedures were performed. Significant band separation was observed near the feature at $4610\;^{-1}$ ,. Particularly the peak intensity at $4525\;^{-1}$ shows a characteristic change with thermal denaturation of fibroin. The structural information can be also obtained by mid-IR and CD spectral. Correlation of NIR spectra with protein structure is discussed.

The morphological characteristics of newly isolated Cordyceps hepialidicola were characterized, and the phylogenetic relationships with other Cordyceps species were investigated using a sequence analysis of the internal transcribed spacer (ITS). The PCR product of 592 bp showed a homology of 92 and 91% with C. militaris and C. nutans, respectively, In an in vitro model using mouse peripheral blood mononuclear cells (PBMC), a methanol extract of C. hepialidicola induced multiple cytokines, including IFN-${\gamma}$ IL-4, and IL-18. The extract also enhanced the percentages of the CD4$\^$+/ and CD8$\sub$+/ T cells in the healthy murine PBMCs to 56.1% and 13.0%,respectively. The percentages of CD4$\^$+/ and CD8$\^$+/ in the untreated controls were 28.4 and 7.3%, and concanavalin A-treated positive controls were 62.4 and 18.3%, respectively.

Phospholipase C is a key enzyme of signaling pathways hydrolyzed phosphatidylinositol 4,5-bisphosphate to generate 2 second messengers. Among the PLC, $PLC-{\beta}$ subfamily consisted of 4 isoforms, $PLC-{\beta}$ 1~4. Here, we studied the tissue specific expression of $PLC-{\beta}3$ in olive flounder (Paralichthys olivaceus) following external stimulation like lipopolysaccharide (LPS), concanavalin A (ConA) and environmental stress compared with the inflammatory cytokines IL-1b. $PoPLC-{\beta}3$ gene transcripts has the effect in stimulated tissue compared to control. These results provide what we sure to be a important role for $PLC-{\beta}3$ activity in tissue and verify $PLC-{\beta}3$ as potential immune enzyme for signal transduction.

Brain-derived neurotrophic factor (BDNF) induced a significant neurite extension of antennal lobe (AL) neurons from the silk moth Bombyx mori in culture on lamini/ concanavalin A-coated dishes, in comparison with smaller effect of 20-hydroxyecdy-sone (20-HE). But the effect fur neurite extension by 5-hydroxytryptamine (5-HT) could not be found. A significant increase in the number of new primary branches from the principal neurites of AL neurons was also shown in culture with BDNF and 5-HT, but not with 20-HE. (omitted)

The purpose of this research was to investigate the effects of 50% ethylalcohol extracts of Lespedeza cuneata G. Don (LE) on the activity of murine immune cells. LE was administered orally once a day for 7 days at the dose of 250 mg/kg. LE increased the cell viability of thymocytes, splenocytes and peritoneal macrophages in vitro and in vivo system, but decreased the cell viability of thymocytes and splenocytes in the presence of concanavalin A in vivo system. Also, the administration of LE was increased the production of ${\gamma}$-interferone, but did not affect the production of interleukin-2 and interleukin-4. Furthermore, LE decreased the phagocytic activity of peritoneal macrophages in vitro and in vivo system, but enhanced the production of nitric oxide. These results suggest that LE has a immuno-regulative action via stimulation of immune cells proliferation.

Lectin from mistletoe (Viscum coloratum) were obtained by salt fractionation, gel filtration and anion exchange chromatography. A molecular weight of about 60,000D has been determined by SDS-PAGE and two basic subunits which have molecular weights of 33,000D and 28,000D are linked by a disulfide bond. The partially purified mistletoe lectins agglutinated human B erythrocytes. Agglutinating activity was relatively stable at varied pH $(3.77{\sim}8.71)$ and at temperature range of $0{\sim}40^{\circ}C$ and not affected by 9 metal ions. Galactose, lactose and N-acetyl-D- galactosamine inhibited agglutinating activity of lectin. Lectin from mistletoe was more mitogenic to murine lymphocytes than concanavalin A.

Guinea pigs were administrated with T-2 toxin at a rate of 1 and 0.6mg/kg body weight per day for 21 days to study the immunological and pathological effects of T-2 toxin in guinea pigs. Blood was collected before T-2 treatment and on days 7, 14 and 21 of the trial for hematological and biological examinations and for the mitogen assay using lymphocytes. Myeloid: erythroid ratios were examined from the fernur bone marrow samples taken a day before T-2 toxin treatment began, on day 12 and at death. Guinea pigs received with 1mg/kg body weight of T-2 toxin daily showed leukopenic, lymphopenic and anemic signs on day 7 and 14. The mitogenic responses to the T-cell mitogen, Concanavalin A and B-cell mitogens, lipopolysaccharide were significantly depressed on day 7. Histologically, marked cellular damages including karyorrhexis and depletion of lymphocytes were observed in the actively dividing cells of the gastrointestinal tract, lymph node, spleen and bone marrow of guinea pigs.

Low molecular weight endo-glucanase fraction of cellulase from Trichoderma reesei was purified using Sephadex G-100 and concanavalin A-Sepha-rose 4B affinity chromatography. Its biochemical characteristics including pH profile, temperature profile and kinetic behavior were studied. The optimum conditions for enzymatic reaction were pH 6.0 and $5^{\circ}C.$ The activation energy for CMC (carboxymethylcellulose) was 10,800 cal/mole. Its adsorption to amorhous and crystalline cellulose was observed. Adsorption to amorphous cellulose was more rapid and greater than that of crystalline cellulose. Reconstitution study was performed. Significance of low molecular weight endo-glucanase on cellulose hydrolysis will be further discussed.
