클로람페니콜 내성 플라스미드 pKH7의 Rep 단백질과 CAT 단백질의 염기서열 분석 (Nucleotide Sequences of Rep and CAT Proteins encoded by Chloramphenicol-Resistance Plasmid pKH7)

  • The nucleotide sequence of Xbal-Mbol fragment of pKH7, a chloramphenicol-resistant($Cm^{r}$) plasmid isolated from multidrug-resistant S. aureus SA2, has been determined. Xbal-Mbol fragment of pKH7 was found to contain two ORFs. One ORF encoded Rap and the other encoded CAT protein. The deduced amino acid sequences of Rep and CAT of pKH7 were compared to those of pUB112 and pC221. Comparisons revealed that there was one amino acid difference in CAT between pKH7 and pUB112. CAT of pKH7 exhibited 98.6% amino acid identity to that of pC221. In case of Rep proteins, a slightly lower homology of 96.4% and 86.7% in amino acid sequences was observed between pKH7 and pUB112 and between pKH7 and pC221, respectively.

식육중 잔류항균물질 비교 조사 -서울지역 도축 소와 돼지를 중심으로- (Comparison of residual antibiotic materials in meet -Slaughtered cattle and swine in Seoul-)

  • This study was carried out to compare the residual antibiotic materials in muscles of slaughter cattle and swine from slaughterhouses in Seoul from 2000 to 2001 by EEC-4-plate method, Charm II and HPLC method. 1. Residual antibiotic materials were detected from 95 samples(0.8%) by EEC-4-plate and 57 samples(10.2%) by Charm II. The final HPLC method determined the positives are 43(45.3%) and 27(47.3%) respectively. 2. The detection ratios were 45% by EEC-4-plate and 47% by Charm II. 3. Seventy samples were classified as tetracyclines 56(75.7.4%), sulfonamides 10(14.9%), $\beta$-lactam 6(8.1%) chloramphenicol 1(1.4%). Three of them were confirmed to be positive simmultaneously for tetracyclines, sulfonamides and chloramphenicol. 4. The highest residual concentration of chlortetracycline, oxytetracycline, sulfamethazine, sulfadimethoxine, sulfaquinoxaline, penicillin, ampicillin and chloramphenicol were 0.34, 11.29, 68.16, 0.13, 4.0, 0.12, 0.4 and 0.04ppm, respectively.

Spinel Nanoparticles ZnCo2O4 as High Performance Electrocatalyst for Electrochemical Sensing Antibiotic Chloramphenicol

  • In this study, ZnCo2O4 nanoparticles were synthesized via the coprecipitation method using different annealing temperatures from 200℃ to 800℃. By varying the treatment temperature, the morphology changed from amorphous to tetragonal, and finally to polygonal particles. As temperature increased, the sizes of the nanoparticles also changed from 5 nm at 200℃ to approximately 500 nm at 800℃. The fabricated material was used to modify the working electrode of a screen-printed carbon electrode (SPE), which was subsequently used to survey the detection performance of the antibiotic, chloramphenicol (CAP). The electrochemical results revealed that the material exhibits a good response to CAP. Further, the sample that annealed at 600℃ displayed the best performance, with a linear range of 1-300 μM, and a limit of detection (LOD) of 0.15 μM. The sensor modified with ZnCo2O4 also exhibited the potential for utilitarian application when the recovery in a real sample was above 97%.

다양한 산화공정을 이용한 수중의 Chloramphenicol, Salicylic Acid 및 Ketoprofen 의약물질 제거 : 산화 동력학 평가 (Removal of Chloramphenicol, Salicylic Acid and Ketoprofen using Various Oxidation Processes : Oxidation Kinetic Evaluation)

  • 본 연구에서는 다양한 산화공정에서 chloramphenicol, salicylic acid 및 ketoprofen에 대한 제거특성을 평가하였다. 염소 투입농도에 따른 의약물질 3종의 제거특성을 살펴본 결과 chloramphenicol과 ketoprofen은 0.5~5.0 mg/L의 염소 투입농도에서 접촉시간 60분 동안 전혀 제거되지 않았으나 salicylic acid는 1.0 mg/L 이상의 염소 투입농도에서 제거경향이 뚜렷이 나타났며, 접촉시간 및 염소 투입농도가 증가할수록 제거율은 증가하였다. 오존 투입농도에 따른 의약물질 3종의 제거특성을 살펴본 결과 chloramphenicol과 ketoprofen은 0.2~2.0 mg/L의 오존 투입농도에서는 제거되지 않았으며, salicylic acid는 1.0~5.0 mg/L의 오존 투입농도에서 약 30~70%의 제거율을 보였다. 오존/과산화수소 투입농도별로 접촉시간에 따른 의약물질 3종의 제거특성을 살펴본 결과 오존단독 공정보다는 오존/과산화수소 공정에서의 제거율이 높았다. 염소, 오존 및 오존/과산화수소 투입농도별 의약물질 3종에 대한 산화분해 속도상수와 반감기를 살펴본 결과 염소, 오존 단독 투입에 비해 오존/과산화수소 공정에서의 산화분해 속도상수가 큰 것으로 나타났으며, 과산화수소/오존의 비가 1 이상에서는 산화분해 속도상수의 증가율이 둔화되었다. Salicylic acid의 경우는 염소와 오존처리에 의해서도 비교적 큰 산화분해 속도상수를 나타내어 산화공정에서 쉽게 제거가 가능하였다.

동물유래(動物由來) 병원세균(病源細菌)의 각종(各種) 항생물질(抗生物質)에 대한 감수성조사(感受性調査) (Antmicrobial Drug Susceptibility of Pathogenic Bacteria Isolated from Animals in Korea)

  • The antimicrobial drug susceptibility of 439 isolates of animal pathogens recovered from various clinical cases during 1978-79 has been investigated by the use of disk diffusion technique. The majority of 308 strains of Eschericihia coli were highly resistant to bacitracin, erythromycin, penicillin, streptomycin and tetracyclinon while only 0.3 per cent of them were resistant to gentamicin and 3.2 per cent to colistin. The percentages of strains resistant to ampicillin, carbenicillin, cephalothin, chloramphenicol and neomycin were 30.5%, 24.7%, 11:4%, 28.2% and 26.2% and repectively. However, none of E. coli cultures of ovine origin were resistant to ampicillin, carbenicillin, chloramphenicol, colistin, gentamicin, kanamycin, and neomycin. A total of 39 patterns of multipe drug1 resistance of 308 strains E. coli against 9 drugs in general use such as ampicillin, cephalothin, chloramphenicol, colistin, gentamicin, kanamycin, neomycin, streptomycin and tetracycline were observed and the most common multiple resistance patterns were SM, TC pattern (20.5%) and AM, CP, KM, NM, SM, TC pattern (9.7%). None of the 43 cultures of salmonella organism from pigs and chickens were resistant to ampicillin, carbenicillin, cephalothin, colistin, gentamicin and kanamycin; and the majority of the cultures were susceptible to chloramphenicol (90.0%), neomycin (97.7%) and tetracycline (93.0%). All the cultures were found to be resistant to bacitracin and penicillin and the rate of resistant strains to erythromycin and s treptomycin being 79.1% and 41.9% respectively. It was found that the majority of 63 cultures of staphylococcal isolates were resistant to lincomycin, penicillin, streptomycin and tetracycline. The percentages of 63 staphylococcal isolates susceptible to gentamicin, nitrofurantoin, cephalothin, ampicillin, methicillin, bacitracin and chloramphenicol were 98.4%, 98.4%, 95.2%, 93.7%, 93.7%, 92.1% and 92.1% respectively. The 25 cultures of streptococcal isolates were resistant in order of prevalence to streptomycin(88.0%), kanamycin(68.0%), gentamicin (44.0%), tetracycline (44.0%) and methicillin (40.0%) wihle the majority of them were sensitive to ampicillin, bacitracin, chloramphenicol and penicillin.

  • 3$0^{\circ}C$에서 대수증식기 세포를 42$^{\circ}C$에서 1시간동안 배양시킨 다음 3$0^{\circ}C$로 다시 이동시켰을 때 DNA합성은 Chloramphenicol의 존재하에서도 재 개시되었다. 그리고 Chloramphenicol을 처리하였을 때 세포 생존력은 크게 증가하지 않았고, 세포의 모양은 정상세포보다 더 길어졌다.

  • The author was carried out bacteriological identification, and in order to evaluate the sensitivity of the different chemotherapeutic agents including chloramphenicol to Vibrio parahaemolyticus isolated from the stool of the patient's diarrhea. The results obtained were as follows: 1) Biochemical properties of Vibrio parahaemolyticus strains isolated from patients with diarrheal food poisoning was showed Table 1. 2) The sensitivity pattern of the isolated strains of Vibrio parahaemolyticus were sensitive to chloramphenicol, sulfonamide, kanamycin and colistin. But tetracycline, penicillin and leucomycin were resistant.

  • The resistance inhibitory activities of 54 odorant mixtures (essential oil) from 41 Korean aromatic herbs were tested against multi-drug resistant Staphylococcus aureus SA2, which has resistances to 10 usual antibiotics including chloramphenicol. As results, combinations of 28 kinds of samples from 21 herbs and chloramphenicol have resistance inhibitory activities in dose dependent manner.

Synergistic Effect of Bacteriophage and Antibiotic against Antibiotic-Resistant Salmonella Typhimurium

  • 본 연구는 항생제 내성 Salmonella Typhimurium CCARM 8009을 저해하기 위한 phage와 항생제 조합처리의 효과를 평가하였다. 디스크 확산법과 액체배지 희석법에 의해 phage와 항생제의 상승 저해효과를 측정하였고 배양을 통한 항생제 내성 유도를 평가하였다. Phage를 처리한 cefotaxime, chloramphenicol, ciprofloxacin, erythromycin의 디스크의 저해 구역은 각각 13.6%, 19.3%, 12.7%, 78.8%로 증가되었다. Phage와 항생제 조합 처리에 의해 tetracycline, chloramphenicol, ciprofloxacin, erythromycin, streptomycin의 최소생육억제농도는 각각 64, 4, 0.0078, 64, 256 mg/mL으로 감소되었다. Phage와 항생제의 조합 처리는 항생제 내성 S. Typhimurium CCARM 8009을 효과적으로 저해하였다 (4 log reduction). 본 결과는 phage와 항생제의 조합처리는 항생제 내성균을 제어하기 위한 방법으로 충분히 응용가치가 높음을 보여주고 있다.

Isolation and Characterization of Transcriptional Elements from Corynebacterium glutamicum

  • A promoter-probe shuttle vector pSK1Cat was constructed for the isolation of transcriptional signal sequences from Corynebacterium glutamicum. Besides conferring resistance to kanamycin in Escherichia coli and C. glutamicum, the vector carried a promoterless cat gene to confer resistance to chloramphenicol upon insertion of the appropriate transcriptional signals in the multiple cloning site. By utilizing the vector, a series of transcriptionally active fragments were isolated from the genome of C. glutamicum. The clones, ranging from 200 bp to 1 kb in size, were grouped into 3 classes of strong, medium, and weak, based on the chloramphenicol acetyltransferase (CAT) activity and sensitivity to the chloramphenicol of the clone-carrying C. glutamicum cells. C. glutamicum cells carrying the $P_{19}$ clone, a representative in the strong class, were able to grow on minimal agar plates containing over $40 mg/mell$ chloramphenicol, and showed CAT activity of 10 m㏖/mgㆍmin, performing slightly better than the cells carrying $P_{tac}$ , a strong E. coli promoter. Subcloning analysis of the $P_{19}$ clone identified a 180 bp intergenic fragment ($P_{180}$), which was located upstream of a gene encoding a hypothetical membrane protein. The expression conferred by $P_{180}$ was not affected by either the kinds of carbon sources or changes in temperature. These properties make the $P_{180}$ clone useful for the deregulated expression of biosynthetic genes in C. glutamicum during amino acid fermentation.
