Anthocyanin and origomeric proanthocyanidin (OPC) fractions showing antioxidizing activity are present in grape extracts. Grape extracts are widely used in cosmetic applications as functional ingredients. The aim of our study is to isolate the antioxidant fraction from waste of grape products. The extraction was done using soxhlet apparatus. Next, the extraction was subjected for identification of antioxidants by using HPLC. Antioxidant assay was performed by using 2,2-diphenyl-picryl-hydrazyl (DPPH) and superoxide dismutase (SOD) method. Antioxidizing activity was found to be higher in grape seed extract when compared to grape skin extract. Using the extract, a novel formula with multi lamella emulsion structure has been developed and its safety and stability were confirmed by standard protocols.
As a part of chemical study on the bioactive metabolites from marine organisms, we have investigated cytotoxicity using brine shrimp bioassay for each solvent fractions of the marine algae(12 species), marine sponges(3 species), coelenterates(2 species), echinoderms(4 species), marine molluscs(17 species), and ascidians(2 species), respectively. As the results, chloroform extract of Stichopus japonicus (LC$_{50}$ : 274 $\mu\textrm{g}$/ml), ethyl acetate extract of Anthocidaris crassispina(LC$_{50}$ : 121 $\mu\textrm{g}$/ml), n-butanol extract of Unda), ia Pinnatifida (LC$_{50}$ : 178 $\mu\textrm{g}$/ml), and water extract of Thais clavigera (LC$_{50}$ : 61 $\mu\textrm{g}$/ml) displayed the most significant cytotoxic activity against brine shrimp. Among the marine organisms tested, echinoderms and marine molluscs were thought to be the most active Phylums on screening of new bioactive compounds.
In order to find antiviral compounds against Herpes simplex virus type I (HSV-1) and II (HSV2) from natural products, a convenient virus-induced cytopathic effect (CPE) inhibition assay was introduced. More than 300 fractions were prepared by solvent fractionation from sixty collected plants or purchased herbal medicines, and their anti-herpetic activities were evaluated. Among them, several medicinal plants showed potent anti-herpetic activity. Selective indexes (SI) of the EtOAc extract of Caraganae Radix (Caragana sinica) against HSV-1 and HSV-2 were more than 8.06 and 24.79, SI of the MeOH extract of Acer okamotoanum leaves were 3.92 and 3.51, SI of the $CH_{2}Cl_{2}$ extract of Veratri Rhizoma et Radix (Veratrum patulum) were 5.49 and 1.31 and SI of the MeOH extract of aerial part of Osmundae Rhizoma (Osmunda japonica) were more than 3.45 and 1.25, respectively.
This study was carried out to investigate the structure of antioxidative constituents and the free radical scavenging effect of the main ingredients from Galla Rhois(Rhus javanica Linne). Antioxidative activities of n-hexane, EtOAc and BuOH extracts of Galla Rhois were similar or even higher than that of natural (tocopherol) or synthetic antioxidant (BHA). It is suggested that major fractions for the antioxidative activity of Galla Rhois were the n-hexane, EtOAc and BuOH extract compartments. In the subsequently experiment, one active compound from n-hexane extract, three active compounds from EtOAc extract and one active compound from BuOH extract were isolated. Their chemical structures were identified as syringic acid, protocatechuic acid, gallic acid methylester, gallic acid and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ on the basis of the speculation of spectral data and chemical reaction. Among the compounds, protocatechuic acid, gallic acid methylester and $1,2,3,4,6- penta-O-galloyl-{\beta}-D-glucose$ showed most potent radical scavenging effect using DPPH method.

Gardenia jasminoides (GJ) is a widely used herbal medicine with anti-inflammatory properties, but its effects on the ORAI1 channel, which is important in generating intracellular calcium signaling for T cell activation, remain unknown. In this study, we investigated whether 70% ethanolic GJ extract (GJEtOH) and its subsequent fractions inhibit ORAI1 and determined which constituents contributed to this effect. Whole-cell patch clamp analysis revealed that GJEtOH (64.7% ± 3.83% inhibition at 0.1 mg/ml) and all its fractions showed inhibitory effects on the ORAI1 channel. Among the GJ fractions, the hexane fraction (GJHEX, 66.8% ± 9.95% at 0.1 mg/ml) had the most potent inhibitory effects in hORAI1-hSTIM1 co-transfected HEK293T cells. Chemical constituent analysis revealed that the strong ORAI1 inhibitory effect of GJHEX was due to linoleic acid, and in other fractions, we found that genipin inhibited ORAI1. Genipin significantly inhibited IORAI1 and interleukin-2 production in CD3/CD28-stimulated Jurkat T lymphocytes by 35.9% ± 3.02% and 54.7% ± 1.32% at 30 μM, respectively. Furthermore, the same genipin concentration inhibited the proliferation of human primary CD4+ T lymphocytes stimulated with CD3/CD28 antibodies by 54.9% ± 8.22%, as evaluated by carboxyfluorescein succinimidyl ester assay. Our findings suggest that genipin may be one of the active components of GJ responsible for T cell suppression, which is partially mediated by activation of the ORAI1 channel. This study helps us understand the mechanisms of GJ in the treatment of inflammatory diseases.
In recent years, many agricultural scientists are studying on eco-friendly farming methods. Among of lots of the methods, the natural insecticides are highly motivated and interested due to their safety and biodegradable issues, and readily available source of bioinsecticides. In this study, the crude extract of custard apple (Annona reticulata L.) seed and its three fractions which were separated based on polarity indexes were examined for their insecticidal activities against Myzus persicae Sulzer and Nilaparvata lugens S.. The crude extract (Aceton/MeOH) showed strong insecticidal activities against both insects at 3.00 mg/mL ($LD_{50}$=0.45mg/mL for M. persicae S. and 1.42 mg/mL for N. lugens S.). Furthermore, simple fractionation with hexane, chloroform, and water lead to increase three-folds insecticidal activity on chloroform fraction ($LD_{50}$=0.13mg/mL for M. persicae S. and 1.14 mg/mL for N. lugens S.). The results suggest that A. reticulata extracts might be used to control for M. persicae effectively.
This study aimed at evaluating the potential of makgeolli, which is widely consumed as beverage, as a functional cosmetic ingredient, based on analyses on its antioxidant activity and tyrosinase activity inhibitory effect. The sample was extracted by concentrating the suspension obtained after adding MeOH (3 L) to the residue of a commercial makgeolli and then adding EtOAc (3 L) to the concentrate, which was subjected to fractionation. The upper layer of the fractions was used as the final sample. In MTT assay assessments, no cytotoxicity was observed at a concentration range of 10 to $1,000{\mu}g/mL$; the antioxidant activity of the extract showed a concentration-dependent tendency and it had a high activity with an $EC_{50}$ of 7.008 mg/mL. Also, in a tyrosinase activity inhibitory effect assessment, the extract showed an $IC_{50}$ value of 39.22 mg/mL. These results confirmed that this sample has potential as a functional cosmetic ingredient.

Ethylacetate and butanol fractions of leaf extracts (OLE) showed the higher contents of total phenolic compounds than hexane and water fractions. Oleuropein contents were $4.21{\pm}0.57,\;3.92{\pm}0.43,\;0.32{\pm}0.03,\;5.76{\pm}0.32$, and $32.47{\pm}0.25mg$/100g for ethanol extract, and hexane, chloroform, ethyl acetate, and butanol fraction, respectively. Treatment of ultraviolet-B (UVB) irradiated cells with 3 OLEs prepared by using ethylacetate and butanol at concentrations 0.001, 0.005, and 0.01% respectively showed significant recovery of cell viabilities. Treatment of dexametason 1 mM reduced tumor necrotic factor (TNF)-${\alpha}$ secretion by about 40%. UVB irradiated immortalized human keratinocytes (HaCaT) cells were treated with 3 different OLEs at the same concentrations. Ethylacetate fraction showed the strongest inhibition activity with respect of reduction of the elevated (TNF)-${\alpha}$. Cytotoxicity of OLEs on the B16-F1 cells was evaluated through thiazolyl blue tetrazolium bromide (MTT) assay. Ethylacetate fraction has no cytotoxicity in the range of 0.005-0.01%. A slight cytotoxicity was observed at the concentration of 0.1% butanol fraction of OLE that caused 10% decrease in cell viability.

Rice plant extracts of brown planthopper (BPH) resistant rice varieties, Jangseongbyeo (JSB) and Hwacheongbyeo (HCB) at different growth stages (seedling, tillering, heading and ripening) were sequentially fractioned using hexane, ethyl ether, ethyl acetate, butanol, and distilled water. The extracts were applied to BPH susceptible rice variety, Dongjjnbyeo (DJB), to investigate the insecticidal and repellent effects against BPH. BPH insecticidal effects were not clearly observed with almost all of the extract fractions obtained from both JSB and HCB varieties for 12 h, whereas the ethyl ether and hexane extract fractions showed about 10 to 30% of BPH mortality in 24 to 48 h of application periods. An effective BPH repellent activity was found with the applications of ethyl ether extract fractions obtained from JSB variety. The extract fractions obtained from HCB variety did not show any different repellence among the various fractions. The BPH repellent effects of the extract fractions obtained at different growth stages of either JSB or HCB varieties did not show any correlations. The effect of ethyl ether fraction on BPH repellent was continually increased by 30 h after treatment and thereafter decreased. In addition, the first sub-fraction separated by a flash column chromatography eluted with chloroform:methanol (9:1, v/v) from the BPH effective ethyl ether faction in JSB variety might be meaningful to repel BPH from BPH susceptible target rice plants. The results indicated that the ethyl ether fraction obtained from JSB was higher in repellent activity than in insecticidal activity, and suggesting that there might be specific substance(s) in the first sub-fraction (sF1) of the ethyl ether fraction in JSB that could provide repellent activity against BPH.
Succinate dehydrogenase was activated by ethyl acetate extract from the methanol extract of white ginseng previously treated with petroleum ether and ethyl ether to remove all highly nonpolar components, and the residual aqueous solution from the ethyl acetate extraction. Also, all of the extracted fractions of red ginseng except the ether extract gave positive results. On the contrary to some suggestions by other workers that alkaloids of ginseng may enhance the succinate degydrogenase activity, our results show that the alkaloids may have exhibited some inhibitory eject on this enzyme.
