Yeast Surface Display of Capsid Protein VP7 of Grass Carp Reovirus: Fundamental Investigation for the Development of Vaccine Against Hemorrhagic Disease

  • VP7, an outer capsid protein of grass carp reovirus (GCRV), was expressed and displayed on the surface of Saccharomyces cerevisiae for developing an efficient vaccine against hemorrhagic disease of grass carp. The result of flow cytometry analysis indicated that protein VP7 could be displayed on the surface of yeast cells after inducing with galactose. The expression of VP7 was confirmed by western blot analysis and further visualized with confocal microscopy. The specific antibodies against VP7 generated from mice were detectable from all immune groups except the control group, which was immunized with untransformed yeast cells. The displaying VP7 on glycosylation-deficient strain EBYΔMnn9 was detected to induce a relatively low level of specific antibody amongst the three strains. However, the antiserum of EBYΔM9-VP7 showed relative high capacity to neutralize GCRV. Further neutralization testing assays indicated that the neutralizing ability of antiserum of the EBYΔM9-VP7 group appeared concentration dependent, and could be up to 66.7% when the antiserum was diluted to 1:50. This result indicates that appropriate gene modification of glycosylation in a yeast strain has essential effect on the immunogenicity of a yeast-based vaccine.

Chronic Toxicity of Cyanide on Survival, Feeding and Growth of Israel Carp, Cyprinus carpio (이스라엘 잉어, Cyprinus carpio의 생존, 섭이 및 성장에 미치는 시안의 만성적 독성)

  • Chronic toxicity of cyanide on survival, feeding and growth rate were examined in the Israel carp, Cyprinus carpio under a continuous-flow system. Survival rate of the Israel carp was significantly affected by $\ge151{\mu}$g/$\ell$ cyanide concentration, and feed efficiency and feeding rate were also reduced above $151{\mu}$g/$\ell$ cyanide concentration. Growth rate of the Israel carp exposed to $\ge73{\mu}$g/$\ell$ cyanide concentration were significantly decreased than that exposed to normal condition.

Skin Pigmentation of 0-age and 1-age Red- and White-colored Fancy Carp Cyprinus carpio var. koi Fed Diets Containing Different Amounts of Paprika (사료 내 파프리카 함량에 따른 당년생 및 1년생 비단잉어(Cyprinus carpio var. koi) 홍백 표피의 체색 변화)

  • This study investigated the effect of dietary paprika on the skin coloration of red- and white-colored fancy carp Cyprinus carpio var. koi, in two age groups: 0-age ($7.4{\pm}0.1$ g/fish) and 1-age ($164{\pm}2.9$ g/fish). Three isonitrogenous diets were formulated to contain 0, 5, and 10% paprika (Con, P5 and P10, respectively). Three replicate groups of 0-age fish and two replicate groups of 1-age fish were fed one of the experimental diets for 9 weeks. Survival, weight gain, and feed efficiency were not significantly affected by the dietary paprika level (P>0.05) at both fish sizes. The dietary paprika level influenced the redness ($a^*$), lightness ($L^*$), and yellowness ($b^*$) of fish skin. The $L^*$ value of the skin of the fish fed the P5 and P10 diets tended to decrease with feeding period. The skin $a^*$ value of 0-age fish (small) fed the diets containing paprika increased significantly with feeding period and was higher than that of fish fed the control diet after 3 weeks (P<0.05). However, the $a^*$ value of 1-age fish (large) fed the diets containing paprika tended to increase slightly with feeding period. At the end of the experiment, the skin total carotenoid level was increased significantly in fish of both sizes fed the P5 and P10 diets compared to fish fed the control diet. Therefore, 5% paprika powder in the diet increases the skin redness of red- and white-colored fancy carp, especially in small fish.

  • This study was conducted to investigate the growth and skin color of red- and white-colored fancy carp Cyprinus carpio var. koi juvenile reared at different water temperatures (21, 24, 27, and $30^{\circ}C$) for eight weeks. Two experimental diets were prepared to contain 10% Arthrospira platensis or 10% Arthrospira pacifica powder. After the feeding trial, survival was 100% in all group. Weight gain of fish reared at 24, 27, and $30^{\circ}C$ was higher than $21^{\circ}C$ group. Feed efficiency was not affected by water temperature and dietary Arthrospira sp. Daily feed intake tended to increase with water temperature. The $a^*$ value of skin coloration was affected by Arthrospira sp., but not by water temperature. Fish fed the diet containing A. pacifica powder reared at $27^{\circ}C$ had the highest $a^*$ value among the groups. Total carotenoids were highest in fish fed the diet containing A. pacifica powder reared at $21^{\circ}C$. Based on the results of this study, we conclude that 24-$30^{\circ}C$ water temperature was the optimal range for growth, and dietary inclusion of A. pacifica improved the skin redness of red- and white-colored fancy carp reared at $27^{\circ}C$.

An Efficient Cooperative Web Caching Scheme (효율적인 협동적 웹캐슁 기법)

  • Nowadays, Internet is used worldwide and network traffic is increasing dramatically. Much of Internet traffic is due to the web applications. And I propose a new cooperative web caching scheme, called DCOORD which tries to minimize the overall cost of Web caching. DCOORD reduces the communication cost by coordinating the objects which are cached at each cache server. In this paper, I compare the Performance of DCOORD with two well-known cooperative Web caching schemes, ICP and CARP, using trace driven simulation. In order to reflect the cost factor in the network communication, I used the CSR(Cost-Saving Ratio) as our performance metric, instead of the traditional hit ratio. The performance evaluations show that DCOORD is more cost effective than ICP and CARP.

The response of peripheral blood lymphocytes against in vivo stimulation with mitogen in carp, Cyprinus carpio (Mitogen 투여에 대한 잉어 순환혈액 림프구의 반응)

  • This work was carried out to investigate the functional heterogeneity of peripheral blood lymphocytes(PBLs) in carp, Cyprinus carpio. PHA, Con A, LPS and BCG were injected intraperitoneally into carp to determine the blastogenic response and rosette formation activity. In each group of fish treated with stimulators, the cell numbers and DNA contents of lymphocytes were higher than those of untreated control group and reached the highest level between 1 week and 2 weeks after injection with mitogens. These results showed that BCG and Con A were strong stimulators of proliferation compared to PHA and LPS. However, PHA-treated fish twice showed the highest rosette formation response among the consecutive stimulations with the same mitogen. Alase, the results on consecutive mitogen stimulation revealed that carps reinjected by different mitogens led to an increased stimulation higher than the one reinjected after 1 week with same mitogen. It seems that different mitogens may stimulate different cell populations and implies functionally separated subpopulations of lymphocytes in carp.

Growth and Excretion of Nitrogen and Phosphorus of Israeli Strain of Carp(Cyprinus carpio) Fed a Low Pollution Diets (저오염 사료의 급여에 의한 잉어의 성장과 질소 및 인 배설량)

  • Growth performance and excretions of nitrogen (N) and phosphorus (P) were investigated with carp (Cyprinus carpio) with carp (Cyprinus carpio) grower fed a low pollution diets (A and B) and commercial ones (Com-1 and Com-2). A recirculated rearing system (Exp. I) and a floating net cage system (Exp. II) were employed for two feeding trials in which fish having an initial body weight of 152g and 193g were fed for 41 an 39 days, respectively. The highest weight gain, daily growth rate and protein efficiency ratio were found (P<0.05) in fish fed diet A containing 10% fish meal and 2% monocalcium phosphate (MCP) for both experiments. They also showed the lowest feed conversion ratio and daily feeding ration among treatments. Fish fed diet B containing 10% fish meal, 5% fish protein concentrate and 1% MCP showed weight gain and FCR more improved (P<0.05) than those fed commercial diets. In all groups, whole body cmpositions were not greatly different among treatment, and protein and P contents in final fish ranged from 14.3 to 15.6% and from 0.39 to 0.48%, respectively. Fish fed diet A excreted the least N which were 38.3 in Exp. I and 39.6g/kg gain in Exp. II. However, the values found in fish fed two commercial diets amounted to 59.1 and 58.9g, respectively. A significant decrease in P excretion was also found in fish groups fed diets A and B. In Exp. I, a reduction of 53.4% was shown in fish fed diet A, compared to the averaged value (18.5g P/kg gain) of two commercial groups. In Exp. II conducted using the floating net cages, fish fed diet B excreted the least P (8.6g/kg gain) among the treatment, which was followed by fish fed diet A, showing 48.6% reduction compared to the average value (18.5g/kg gain) for fish fed two commercial diets. The present results clearly showed that N and P excretions from fish culture could be significantly reduced by using the low pollution diet.

Induced Spawning of the Israeli Strain of Common Carp, Cyprinus carpio by Water Temperature Control (수온조절에 의한 이스라엘잉어의 산난유발)

  • A Series of trials for the spawning induction of the Israeli strain of common carp were carried out at the fish culture research station of National Fisheries University of Pusan from 1978 to 1981. 1, During 3 years up to the first periods in 1980 carp pituitaries and/or commercial gonadotropic hormone was used to induce the brood fish to spawn without success. In the trials in the later periods in 1980 and in 1981, water temperature increase method was used with success in 40 females of 16 trials leaving a single failure with 2 females. 2. Brood fish had been kept in a small circular tank ($7\;m^2$) continuously supplied with cool well water enough to keep the water temperature in the tank relatively cool, ranging from 16.9 to $20.2^{\circ}C$, and the water in the spawning tank ($15\;m^2$) was 3.0 to $6.5^{\circ}C$ higher than in the brood fish holding tank. 3. It could be said that the spawning of Israeli strain of common carp in Korea hardly occur without water temperature increase even though treated with hormone.

Genetic Similarity and Variation in the Cultured and Wild Crucian Carp (Carassius carassius) Estimated with Random Amplified Polymorphic DNA

  • Random amplified polymorphic DNA (RAPD) analysis based on numerous polymorphic bands have been used to investigate genetic similarity and diversity among and within two cultured and wild populations represented by the species crucian carp (Carassius carassius). From RAPD analysis using five primers, a total of 442 polymorphic bands were obtained in the two populations and 273 were found to be specific to a wild population. 169 polymorphic bands were also produced in wild and cultured population. According to RAPD-based estimates, the average number of polymorphic bands in the wild population was approximately 1.5 times as diverse as that in cultured. The average number of polymorphic bands in each population was found to be different and was higher in the wild than in the cultured population. Comparison of banding patterns in the cultured and wild populations revealed substantial differences supporting a previous assessment that the populations may have been subjected to a long period of geographical isolation from each other. The values in wild population altered from 0.21 to 0.51 as calculated by bandsharing analysis. Also, the average level of bandsharing values was $0.40{\pm}0.05 $ in the wild population, compared to $0.69{\pm}0.08$ in the cultured. With reference to bandsharing values and banding patterns, the wild population was considerably more diverse than the cultured. Knowledge of the genetic diversity of crucian carp could help in formulating more effective strategies for managing this aquacultural fish species and also in evaluating the potential genetic effects induced by hatchery operations.

Genetic Distances of Crucian Carp Populations analyzed by PCR Approach

  • Genomic DNAs isolated from crucian carp of four rivers, belonging to the family Cyprinidae was amplified by seven oligonucleotides primers. In the present study, we employed hierarchical clustering method in order to reveal genetic distances and variations. Crucian carp was acquired from Hangang river (CAH), Geumgang river (CAG), Nakdonggang river (CAN) and Yeongsangang river (CAY). The primer BION-12 generated the most loci (a total of 50) with an average of 10 in the CAY population. The primer BION-10 generated the least loci (a total of 19), with an average of 3.8 in the CAG population, in comparison to the other primers used. Seven oligonucleotides primers made 16.7 average no. per primer of specific loci in the CAH population, 7.4 in the CAG population, 8.6 in the CAN population and 0.9 in the CAY population, respectively. The specific loci generated by oligonucleotides primers revealed inter-individual-specific characteristics, thus disclosing DNA polymorphisms. The dendrogram obtained by the seven oligonucleotides primers indicates four genetic clusters. The genetic distance that displayed significant molecular differences was between individuals no.06 and no.08 from the CAG population (genetic distance = 0.036), while the genetic distance among the five individuals that displayed significant molecular differences was between individuals no.08 and no.09 from the CAG population (genetic distance = 0.088). With regard to average bandsharing value (BS) results, individuals from CAY population ($0.985{\pm}0.009$) exhibited higher bandsharing values than did individuals from CAH population ($0.779{\pm}0.049$) (P<0.05). Relatively, individuals of CAY population were fairly closely related to that of CAN location (genetic distance between two populations<0.016).
