인체분변으로부터 분리한 유산균 Lactobacillus pentosus Miny-148의 생균제 특성 연구 (Probiotic Property of Lactobacillus pentosus Miny-148 Isolated from Human Feces)

  • 우수한 생균제를 개발하기 위하여 안전성이 알려진 유산균을 대상으로 인체의 분변으로부터 300여 균주를 분리하고 내산성, 내담즙성, 내열성, 항균력, 항암 및 항바이러스 효과를 가지는 균주들을 선발하여 생균제 특성을 나타내는지를 알아보기 위해 본 실험을 수행하였다. 인체에서 분리한 여러 균주 중 Miny-148 균주는 낮은 pH 및 높은 담즙산에 대한 내성, 열처리에 대한 열안정성을 지녀 기초적인 probiotic 특성을 가진 균주로 선발되어, Lactobacillus pentosus (99.9% 상동성)로 동정되었다. 항균력 실험에서 Escherichia coli O157:H7을 비롯한 Shigella flexneri, Bacillus anthracis, Staphylococcus aureus, E. coli, Vibrio cholerae, V. vulnificus, Salmonella typhimurium, 그리고 및 methicillin-resistant S. aureus (MRSA) 균주 8종 등 총 16종의 병원성세균을 억제하였다. 또한 Miny-148은 결장암 세포인 HT-29 cell을 억제하였을 뿐 아니라, transmissible gastroenteritis virus의 생육을 저해하여 세포변성 억제효과를 가진 우수한 probiotic 특성을 지닌 균주로 분석되었다.

  • 본 연구에서는 어린감과 성숙감의 탄닌 함량 및 담즙산과의 결합능력을 비교 조사하였다. 또한 사람의 장내 세포인 Caco-2 cell에 어린감과 성숙감 추출물을 처리한 후 고농도의 DCA로 자극시켜 생성되는 NO의 수준과 TEER의 차이를 확인하였다. 탄닌 함량과 담즙산의 결합능력은 성숙감에 비해 어린감에서 유의적으로 높은 것으로 나타났으며 어린감 산성 에탄올 추출물에서 유의적으로 가장 높은 탄닌 함량 및 담즙산 결합능력을 보였다. NO 생성 억제능으로 확인한 염증반응에서도 어린감 산성 에탄올 추출물이 유의적으로 가장 높은 효과를 보였으며 DCA에 의해 유도된 장세포 간극의 느슨함을 유의하게 막아줄 수 있음이 확인되었다. 본 연구 결과를 토대로 어린감의 산성 에탄올 추출물은 고지방 식사를 통해 고농도로 축적되는 담즙산에 의해 형성된 유해한 장내 환경을 건강하게 유지시켜줄 수 있는 기능성 소재로 개발할 가능성이 있음이 확인되었다.

한국인 유아 분변에서 분리한 Lactobacillus acidophilus KY1909의 프로바이오틱 특성 (Probiotic Characteristics of Lactobacillus acidophilus KY1909 Isolated from Korean Breast-Fed Infant)

  • 발효 및 유산균 제품에 모유만을 먹은 생후 3개월된 유아의 분변으로부터 bromocresol pule을 함유한 MRS 배지에서 노란색의 집락을 형성하는 균주들을 내산성 및 담즙산 내성이 우수한 13종의 Lactobacillus 균주들을 분리하였으며 당발효성 및 생화학적 실험을 토대로 분리된 유산균들을 동정하였다. 최종적으로 선발한 균주를 L. acidophilus로 KY1909로 명명하였다. Lactic acid 생성 능력을 평가한 후 이성질체를 분석한 결과 상업적으로 이용되는 L. acidophilus NCFM, L. acidophilus CH5, L. acidophilus NOCKS 균주들은 전체 lactic acid 중에서 L(+) form이 차지하는 비율이 각각 52.49, 40.63 및 63.13%로 나타났으나, L. acidophilus KY1909는 L(+) lactic acid의 비율이 93.96%로 나타났다.

연맥 사일리지에서 분리된 Lactobacillus plantarum의 균특성에 관한 연구 (Studies on the characteristics of Lactobacillus plantarum isolated from oat silage)

  • The growth characteristics and the cellular protein patterns of the Lactobacillus plantarum isolated and identified from oat silage were examined in order to confirm whether it will be used practically as probiotics or not. L plantarum was identified by morphological and biochemical tests including of final conforming by API 50CHL kit. The cultivation in MRS broth of the strain under the condition of different temperature, proved that they grew into $2.0{\times}10^{9}$ in $25^{\circ}C$, into $1.4{\times}10^{9}$ in $35^{\circ}C$ but they decreased into $4.5{\times}10^{5}$ growth in $45^{\circ}C$. The comparison of the growth by measurement of O.D600nm value after 24 hour cultivation between L plantarum and commercial probiotics, showed that the strain had a higher growth than commercial as 1.841 : 1.623. The measurement of it under bile acid's existence, indicated that this isolation was not influenced by bile acid and the tolerance was $3.2{\times}10^{9}$, $3.9{\times}10^{9}$ and $3.2{\times}10^{9}$, respectively, when each of 0%, 1%, and 2% oxigall existed. The examination of their antibiotics susceptibility by disk diffusion test, proved that L plantarum showed resistance against danofloxacin(5mcg), gentamycin(10mcg), kanamycin(30mcg), neomycin(30mcg) and streptomycin(10mcg). Based upon the test of the bacteriocin formation of this L plantarum, it was found out that the inhibition zone was not formed. In growth of L plantarum and E coli in nutrient broth, all E coli died out within 6 hours after cultures.

백합(Meretrix meretrix)식해에서 분리한 Pediococcus pentosaceus SH-10의 생균제적 특성 (Probiotic Properties of Pediococcus pentosaceus SH-10 Isolated from the Hard Clam Meretrix meretrix Shikhae)

  • This study examined the suitability of characteristics of potential strains of probiotic bacteria. Among 25 lactic acid bacteria isolated from Korean traditional fermented food, the Hard Clam Meretrix meretrix Shikhae, the SH-10 strain, which exhibited superior resistance to low pH and bile salts, was selected as a potential probiotic bacteria. By examining carbohydrate utilization, morphological properties, and the 16S rRNA gene sequence, the SH-10 strain was identified as Pediococcus pentosaceus (hereafter, P. pentosaceus SH-10). P. pentosaceus SH-10 was resistant to amikacin, cefotetan, ciprofloxacin, gentamicin, kanamycin, nalidixic acid, streptomycin, and vancomycin. Tests of antimicrobial activities against pathogens such as Bacillus cereus, Listeria monocytogenes, Salmonella choleraesuis, and Staphylococcus aureus, indicated that P. pentosaceus SH-10 inhibited the growth of pathogenic bacteria. These results suggest that P. pentosaceus SH-10 can be developed as a probiotic bacteria.

Fermentation Characteristics of Exopolysaccharide-Producing Lactic Acid Bacteria from Sourdough and Assessment of the Isolates for Industrial Potential

  • Lactic acid bacteria (LAB) with antimicrobial activity and high exopolysaccharide (EPS) production ability isolated from sourdough were studied for their fermentation characteristics as potential new starter cultures. The values of pH, titratable acidity, and viable cell counts were $4.06{\pm}0.009-4.50{\pm}0.015,\;0.787{\pm}0.020%-1.172{\pm}0.018%,\;and\;8.78{\pm}0.08-8.98{\pm}0.06$ log CFU/ml, respectively. In order to select probiotics with a high survival rate in the gut, isolates were tested to assess resistance against the artificial gastric acid and bile juice. Viable LAB counts were significantly (p<0.05) affected by the acidity. At pH 2.0, the total declines in the initial bacterial counts were 4.52$\pm$0.07 log for S. thermophilus St-Body-1, >7.98$\pm$0.03 log for E. flavescens DU-10, >7.95$\pm$0.05 log for E. faecium DU-12, and 3.15$\pm$0.06 log for L. amylovorus DU-21. Among the strains, L. amylovorus DU-21 was the only strain that had bile tolerance under simulated gastrointestinal conditions. In order to improve EPS production by L. amylovorus DU-21, the influence of carbon source was studied. When glucose was used as a carbon source, EPS production dramatically increased to 17.19$\pm$0.28 g/l (p<0.05). The maximum cell growth (10.012$\pm$>0.012 log CFU/ml) and EPS production (18.71$\pm$0.19 g/l) were achieved when 15 g/l of glucose was employed as the carbon source.

  • A lactic acid bacterial strain resistant to several antibiotics such as oxytetracycline, tylosin, neomycin and sulfathiazole, which have been often used as a therapeutic agent in livestock, was isolated from the porcine gastrointestinal tract. The isolate YB-55 was identified as belonging to the genus Pediococcus with the highest similarity to P. pentosaceus on the basis of its 16S rRNA sequence and biochemical properties. The isolated strain showed viability of over 85% at pH 3.0 and was resistant to bile salt. The strain produced lactic acid of 12.3 g/L by jar fermentation and maintained its viability in the presence of antibiotics at dosage related therapeutic effect, suggesting P. pentosaceus YB-55 may of ffr potential as a probiotics for livestock.

  • The use of probiotics for human and animal health is continuously increasing. The probiotics used in humans commonly come from dairy foods, whereas the sources of probiotics used in animals are often the animals' own digestive tracts. Increasingly, probiotics from sources other than milk products are being selected for use in people who are lactose intolerant. These sources are non-dairy fermented foods and beverages, non-dairy and non-fermented foods such as fresh fruits and vegetables, feces of breast-fed infants and human breast milk. The probiotics that are used in both humans and animals are selected in stages; after the initial isolation of the appropriate culture medium, the probiotics must meet important qualifications, including being non-pathogenic acid and bile-tolerant strains that possess the ability to act against pathogens in the gastrointestinal tract and the safety-enhancing property of not being able to transfer any antibiotic resistance genes to other bacteria. The final stages of selection involve the accurate identification of the probiotic species.

Bacteroides Fragilis군의 분리 및 동정에 관하여 (Classification of Bacteroides Fragilis Group)

  • Six strains of Bacteroides fragilis group were isolated from clinical specimens, and these isolates were classified by the resistance to bile and kanamycin, and by catalase and indole reaction. Of 6 strains, 4 strains were belonged to B. fragilis and 2 were B. uniformis. Among 6 strains, only 2 of B. fragilis harbored plasmids. But these plasmid showed no correlation with phenotypic expression. There were some differences in susceptibility to 23 ${\beta}$-lactam antibiotics, also, marked susceptibility differences were found between 2 species, so these results may be contributed to the treatment of infection due to this micro-organism and the identification of these species.

Probiotic Characteristics of Lactobacillus plantarum FH185 Isolated from Human Feces

  • Lactobacillus plantarum FH185 was isolated from the feces of healthy adults. In our previous study, L. plantarum FH185 was demonstrated that it has anti-obesity effect in the in vitro and in vivo test. In order to determine its potential for use as a probiotic, we investigated the physiological characteristics of L. plantarum FH185. The optimum growth temperature of L. plantarum FH185 was 40℃. L. plantarum FH185 showed higher sensitivity to novobiocin in a comparison of fifteen different antibiotics and showed higher resistance to polymyxin B and vancomycin. It also showed higher β-galactosidase and N-acetyl-β-glucosaminidase activities. Moreover, it was comparatively tolerant to bile juice and acid, and inhibited the growths of Salmonella Typhimurium and Staphylococcus aureus with rates of 44.76% and 53.88%, respectively. It also showed high adhesion activity to HT-29 cells compared to L. rhamnosus GG.
