본 연구는 남자초등학교 4학년생 중 태권도 유품자($1{\sim}2$품)로서 태권도칼슘섭취군(8명), 태권도군(8명), 대조군(8명) 총 24명을 대상으로 하였다. 태권도와 칼슘 섭취가 남자초등학생의 체격, 성장호르몬 및 IGF-1에 미치는 영향을 규명하기 위하여 운동 강도 RPE ($7{\sim}15$), 운동빈도 주 5회, 1회 50분의 지속시간으로 12주간 실시하였다. 칼슘은 1일 1회 저녁에 750 mg을 섭취시킨 결과 다음과 같은 결론을 얻었다. 신장은 집단 내 세 그룹 모두 유의하게 증가하였지만 집단 간에서는 대조군에 비해 태권도군과 태권도칼슘섭취군이 유의하게 증가하였다. 성장호르몬은 대조군이 유의하게 감소하였으며 태권도군과 태권도칼슘섭취군은 대조군에 비해 유의하게 증가하였다. IGF-1은 집단 내 태권도군과 태권도칼슘섭취군이 유의하게 증가하였으며 집단 간에서는 대조군에 비해 태권도군과 태권도칼슘섭취군이 유의하게 증가하였다. 이상의 결과를 통해 초등학생에게 태권도프로그램과 칼슘섭취는 신장과 체중, 성장호르몬 및 IGF-1에 긍정적인 영향을 미쳤으며, 향후 적절한 태권도프로그램개발과 운동처방 및 영양섭취에 대한 지속적인 연구가 요구된다.
우리나라의 전통 발효 된장으로부터 균체외 효소로 mannanase를 생산하는 세균 2주가 분리되었다. 분리균 WL-6과 WL-11은 형태적 특성, 생화학적 성질 및 16S rDNA의 염기서열에 따라 Bacillus subtilis로 확인되었다. 이들 두 균주로부터 각각 mannanase 유전자를 대장균에 클로닝하여 염기서열을 결정한 결과 mannanase 유전자는 362 아미노산으로 구성된 단백질을 코드하며 1,086 뉴클레오티드로 동일하게 이루어졌다. WL-6과 WL-11 mannanase (Man6, Man11)의 아미노산 잔기 배열은 서로 8개 잔기가 다르며 GH family 26에 속하는 B. subtilis의 mannanases와 매우 상동성이 높았다. Man6과 Man11의 아미노 말단의 26개 아미노 잔기가 signal peptide로 예측되었다. 재조합 대장균로부터 각각 생산된 Man6과 Man11은 94~95% 정도가 균체내에 존재하였고, mannotriose, mannotetraose, mannopentaose, mannohexaose와 같은 만노올리고당과 locust bean gum을 유사하게 분해하여 주된 반응산물로 mannobiose와 mannotriose를 생성하였다. Man6는 55℃와 pH 6.0, Man11은 60℃와 pH 5.5에서 각각 최대 반응활성을 보였으며, Man11이 Man6에 비해 열안정성이 높았다.
Objectives : This study was designed to evaluated the effect of electroacupuncture on Quality of life of patients with urinary incontinence Methods : Subjects were voluntarily recruited by newspapers and internet. Electroacupuncture was performed three times a week for 3 weeks. Acupuncture point for EA group was B32, Electrical stimulation frequency was 2Hz, duration 20 minutes, and intensity was up to pain threshold according to patients. The patients's symptoms were assessed before, after 3 weeks of treatment by QOL item of International Prostate Symptom Score(IPSS), Medical Outcomes Study(MOS) 36-Item Short-Form Health Survey(SF-36). Results : QOL score of IPSS were significantly improved after 3 weeks(p<0.05) compared to the pre-treatment. There were significant changes in Social functioning(SF), role-physical(RP), role emotional(RE), mental health(MH), bodily pain(BP) score of SF-36 after 3 weeks(p<0.05), but there were no significant changes in physical functioning(PF), vitality(VT), general health(GH) score of SF-36. Conclusion : This study suggests that electroacupuncture treatments can be applicable to improve symptoms in patients with urinary incontinence.
POU1F1 is a positive regulator for GH, PRL and TSH${\beta}$ and its mutations associate with production traits in ruminant animals. We described a DdeI PCR-RFLP method for detecting a silent allele in the goat POU1F1 gene: TCT (241Ser)>TCG (241Ser). Frequencies of $D_1$ allele varied from 0.600 to 1.000 in Chinese 801 goats. Significant associations of DdeI polymorphism with production traits were found in milk yield (*p<0.05), litter size (*p<0.05) and one-year-old weight (*p<0.05) between different genotypes. Individuals with genotype $D_1D_1$ had a superior performances when compared to those with genotype $D_1D_2$ (*p<0.05). Hence, the POU1F1 gene was suggested to the potential candidate gene for superior milk performance, reproduction trait and weight trait. Genotype $D_1D_1$, characterized by a DdeI PCR-RFLP detection, was recommended to geneticists and breeders as a molecular marker for better performance in the goat industry.
The present study was planned to investigate the role of sex hormone receptor gene expression in the pathogenesis of hepatocellular carcinoma (HCC). Adult male Wistar rats were divided into seven groups. Group (1) was negative control. Groups (2), (5), (6), and (7) were orally administered with N-nitrosodiethylamine for the induction of HCC, then group (2) was left untreated, group (5) was orally treated with curcumin, group (6) was orally treated with carvacrol, and group (7) was intraperitoneally injected with doxorubicin, whereas groups (3) and (4) were orally administered only curcumin and carvacrol, respectively. The HCC group showed significant upregulation in the androgen receptor (AR) and the estrogen receptor-alpha ($ER{\alpha}$) gene expression levels in the liver tissue. On the contrary, HCC groups treated with either curcumin or carvacrol showed significant downregulation in AR and $ER{\alpha}$ gene expression levels in the liver tissue. In conclusion, the obtained data highlight that both AR and $ER{\alpha}$ but not estrogen receptor-beta ($ER{\beta}$) gene expression may contribute to the male prevalence of HCC induced in male rats. Interestingly, both curcumin and carvacrol were found to have a promising potency in alleviating the male predominating HCC.

A DNA fragment containing 2,079 base pairs from Bacillus circulans CGMCC 1416 was cloned using degenerate PCR and inverse PCR. An open reading frame containing 981 bp was identified that encoding 326 amino acids residues, including a putative signal peptide of 31 residues. The deduced amino acid sequence showed the highest identity (68.1%) with $endo-{\beta}-1,4-D-mannanase$ from Bacillus circulans strain K-1 of the glycoside hydrolase family 5 (GH5). The sequence encoding the mature protein was cloned into the pET-22b(+) vector and expressed in Escherichia coli as a recombinant fusion protein containing an N-terminal hexahistidine sequence. The fusion protein was purified by $Ni^{2+}$ affinity chromatography and its hexahistidine tag cleaved to yield a 31-kDa ${\beta}$-mannanase having a specific activity of 481.55U/mg. The optimal activity of the purified protein, MANB48, was at $58^{\circ}C$ and pH 7.6. The hydrolysis product on substrate locust bean gum included a monosaccharide and mainly oligosaccharides. The recombinant MANB48 may be of potential use in the feed industry.

Paenibacillus xylanilyticus KJ-03 was isolated from soil samples obtained from a field with Amorphophallus konjac plants. A gene encoding xylanase was isolated from KJ-03 and cloned using a fosmid library. The xynA gene encodes xylanase; it consists of 1,035 bp and encodes 345 amino acids. The amino acid sequence deduced from the P. xylanilyticus KJ-03 xylanase showed 81% and 69% identities with those deduced from the P. polymyxa E681 and Paenibacillus sp. HPL-001 xylanases, respectively. The xynA gene comprises a single domain, consisting of a catalytic domain of the glycosyl hydrolase (GH) 10 family. The xynA gene was expressed in Escherichia coli BL21 (trxB), and the recombinant xylanase was purified by Niaffinity chromatography. The purified xylanase showed optimum activity with birchwood xylan as a substrate at $40^{\circ}C$ and pH 7.4. Treatment with $Mg^{2+}$ and $Li^+$ showed a slight decrease in XynA activity; however, treatment with 5 mM $Cu^{2+}$ completely inhibited its activity. The results of the thin layer chromatography analysis indicated that the major hydrolysis product was xylobiose and small amounts of xylose and xylotriose. XynA showed increased activity with oat spelt xylan and birchwood xylan, but showed only slight activity with locust bean gum.
An endoxylanase gene (PoxynA) that belongs to the glycoside hydrolase (GH) family 11 was cloned from a xylanolytic strain, Penicillium oxalicum B3-11(2). PoxynA was overexpressed in Trichoderma reesei QM9414 by using a constitutive strong promoter of the encoding pyruvate decarboxylase (pdc). The high extracellular xylanase activities in the fermentation liquid of the transformants were maintained 29~35-fold higher compared with the wild strain. The recombinant POXYNA was purified to homogeneity, and its characters were analyzed. Its optimal temperature and pH value were $50^{\circ}C$ and 5.0, respectively. The enzyme was stable at a pH range of 2.0 to 7.0. Using beechwood as the substrate, POXYNA had a high specific activity of $1,856{\pm}53.5$ IU/mg. In the presence of metal ions, such as $Cu^{2+}$, and $Mg^{2+}$, the activity of the enzyme increased. However, strong inhibition of the enzyme activity was observed in the presence of $Mn^{2+}$ and $Fe^{2+}$. The recombinant POXYNA hydrolyzed birchwood xylan, beechwood xylan, and oat spelt xylan to produce short-chain xylooligosaccharides, xylopentaose, xylotriose, and xylobiose as the main products. This is the first report on the expression properties of a recombinant endoxylanase gene from Penicillium oxalicum. The properties of this endoxylanase make it promising for applications in the food and feed industries.

Background: The understanding of back-pain as one of the commonest clinical presentations, made the path to the present study. More than three-quarters of the world's population experience back-pain at some time in their lives Purpose: The present study has evaluated the selected Naturopathic therapeutic procedures on individuals with low back-pain. Methods: Thirteen patients were selected for the study and randomly divided into two groups, viz., Group A, Naturopathic group and Group B, Control group. One patient discontinued. Naturopathic group was treated with mud pack, aromatherapy massage with Eucalyptus oil, acupressure, steam, wet trunk pack, exercises, relaxation and breathing exercises. Advice for life style and diet was given for patients of both the groups. The effect of the therapy was assessed subjectively and objectively. Particular scores drawn for Naturopathic group and control group were individually analyzed before and after treatment and the values were compared using standard statistical protocols. Results: Naturopathic intervention revealed 100% relief in both subjective and objective parameters (i.e., 6 out of 14 parameters showed statistically highly significant P < 0.01 results, while 8 showed significant results P < 0.05). Conclusion: Statistically highly significant results of this study point out the successful management of the multi factorial low back-pain with a multi focused Naturopathic treatment approach. Such encouraging results may pave the way for a future study on a large sample in a longer duration incorporating clinical investigations.
꿀벌(Apis mellifera)의 장으로부터 xylan분해능이 우수한 세균 HY-20을 분리하였고, Bacillus 속으로 동정하였다. 분리균주 HY-20의 세포 외 GH11 xylanase (XylP) 유전자 (687-bp)는 25,522 Da의 분자량과 9.33의 pI 값을 갖는 228개의 아미노산으로 구성된 단백질을 인코딩하는 것으로 확인되었으며, 지금까지 특성규명이 이루어지지 않은 B. pumilus xylanase (GenBank accession no.: AY526092)의 아미노산 서열과 97%의 상동성을 보였다. pET-28a(+)/xylP를 포함하고 있는 Escherichia coli BL21 균주에서 과발현 된 His-tagged XylP (rXylP)는 cation exchange 및 gel permeation chromatography를 통해 순수하게 분리 정제 되었으며, $50^{\circ}C$의 온도와 pH 6.5 조건에서 반응할 때 birchwood xylan에 대해 가장 높은 가수분해 활성을 나타내었다. rXylP는 15분간 $55^{\circ}C$에 노출될 때 본래 활성의 약 50%를 잃어버리는 전형적인 중온성 효소의 특성을 보였으며, $Hg^{2+}$ (1mM)와 N-bromosuccinimide (5mM)에 노출될 때 완전히 불활성화 되었고, $Mn^{2+}$ (1 mM), $Fe^{2+}$ (1mM) 및 sodium azide (5 mM)에 의해서는 활성이 약 10% 증가될 수 있는 것으로 밝혀졌다. 한편, rXylP는 xylose유래 다당류는 효율적으로 분해하였지만 PNP-sugar 유도체 및 glucose 유래 다당류는 분해하지 못하였다.
