Chemical Compositions and Antioxidant Activities of Leaves and Stalks from Different Sweet Potato Cultivars (고구마 품종별 잎과 잎자루의 성분특성과 항산화활성)

  • This study was conducted to evaluate the vegetative value of sweet potato leaves and stalks by investigating their chemical composition and antioxidant activity. The major minerals were found to be Ca and K. The crude protein content of the leaves ranged from 15.57% in Geonpungmi to 20.34% in Yeonhwangmi, while that of stalks was between 3.71% in Yulmi and 5.97% in Sinchunmi. The total phenolics and flavonoids content of the leaves were about 3.0~4.0 times higher than those of stalks. The levels of polyphenols and flavonoids were high in Geonpungmi leaf and in Sinchunmi stalk. ABTS radical activities were high in Geonpungmi leaf (58.75 mg AA eq/g) and in Daeyumi stalk (14.95 mg AA eq/g). DPPH radical scavenging was high in Geonpungmi and Jeungmi leaves ($IC_{50}$, 0.11 mg/mL) and in Sinchunmi stalk ($IC_{50}$, 1.09 mg/mL).

Physiological activitive of Grifola frondosa by log cultivation and bottle cultivation (원목재배와 병재배 잎새버섯(Grifola frondosa)의 생리활성)

  • This research was carried out to determine the differences of physiological activites between Grifola frondosa of log cultivation(LC) and Grifola frondosa of bottle cultivation(BC). Total flavonoids content, total phenolics content, electron donating ability(EDA), nitrite-scavenging ability(NSA), SOD-like activity and inhibitory effect of Xanthine oxidase were examined. The highest value of total flavonoid content is $5.96{\pm}0.81mg/g$ in water extract from Grifola frondosa of log cultivation at $40^{\circ}C$ (LC-W40) but, one of total phenolics compound is $44.53{\pm}0.89mg/g$ in water extract from Grifola frondosa of bottle cultivation at $40^{\circ}{\cdots}$ (BC-W40). The EDA using DPPH of BC-W40 extract showed the highest value of $97.14{\pm}0.71%$. Nitrite-scavenging ability was $62.55^{\circ}{\ae}0.36%$ in extract from Grifola frondosa of BC-W40 at pH 1.2. The value was SOD-like activity showed the highest value of $18.95^{\circ}{\ae}1.39%$ in extract from LC-W40. Xanthine oxidase inhibitory activity was the highest value of $54.31{\pm}0.40%$ in extract from Grifola frondosa BC-W40, and dependent on concentrations. These results showed that a the antioxidant effects of Grifola frondosa is excellent. However, physiological activities of Grifola frondosa were not depend on caltivation method regulary, and were different according to kind of solvents, concentraitions and physialogical factors examined such as EDA, SOD-like activity and NSA.

Anti-inflammatory Activities of Methanolic Extracts from Different Rose Cultivars (품종별 장미꽃 메탄올 추출물의 항염증 효과)

  • The genus Rosa (Rosaceae) is an abundant source of phenolics and is traditionally used as a food supplement and as herbal medicine. Various plant phenolics are known to have anticancer, antioxidant, and anti-inflammatory properties. In this study, we investigated the anti-inflammatory effects of rose methanolic extracts (RMEs) from four different rose cultivars (Macarena, Onnuri, Oklahoma, and Colorado) in lipopolysaccharide (LPS)-activated RAW 264.7 cells. Our results demonstrated that pretreatment of REMs ($500{\mu}g/mL$) significantly reduced NO production by suppressing iNOS protein expression in LPS-stimulated cells. Anti-inflammatory effects by RMEs were observed in the following order: Oklahoma > Colorado > Onnuri > Macarena. Consistent with this finding, RMEs inhibited the translocation of $NF-{\kappa}B$ from the cytosol to the nucleus via the suppression of $I{\kappa}B{\alpha}$ phosphorylation and also inhibited LPS-stimulated $NF-{\kappa}B$ transcriptional activity. These findings suggest that RMEs exert anti-inflammatory actions and help to elucidate the mechanisms underlying the potential therapeutic values of RMEs. Therefore, RMEs could be regarded as a potential source of natural anti-inflammatory agents.

The Effects of Anti-Inflammatory Activities and Active Fractions Analysis of Ethanol Extract from Red Rose Petals (붉은 장미꽃잎 에탄올 추출물의 활성 분획물 분석 및 항염증 활성 효과)

  • Red rose petals are usually disposed but they are an abundant source of phenolics and traditionally used as food supplement and as herbal medicine. Of the Various phenolics, they are known to have anticancer, antioxidant, and anti-inflammatory properties. In this study, we investigated the anti-inflammatory effects of red rose ethanolic extracts(GRP) on lipopolysaccharide (LPS)-activated RAW 264.7 cells. Anti-melanogenesis in B16F0 Melanoma Cells by Extract of Fermented Cordyceps militaris Containing High Cordycepin (Cordycepin-고함유 동충하초(Cordyceps militaris) 발효 추출물의 미백효과)

  • To find a novel skin whitening agent, the effect of cordycepin-enriched Cordyceps militaris (CM${\alpha}$) extract fermented by fungi on anti-melanogenesis in B16F0 mouse melanoma cells was investigated. Fermented CM${\alpha}$ was prepared with fungi, including Monascus purpureus (Mp), Aspergillus oryzae (Ao), Aspergillus kawachii (Ak), and Rhizopus oryzae (Ro), respectively. When the content of the phenolics and the flavonoids and the activities of the antioxidant and the mushroom tyrosinase inhibition were measured in the CM fermented by Ak (AkF-CM), the highest content of the phenolics was 46 mg/g dry weight and the highest content of the flavonoids was 0.93 mg/g; the highest activity of the DPPH radical scavenging was 62.74% and the highest activity of the mushroom tyrosinase inhibition was 79.97% CM${\alpha}$CM${\alpha}$. From this result, AkF-CM${\alpha}$ exhibited the highest mushroom tyrosinase inhibitory activity and so it was used in subsequent anti-melanogenesis. B16F0 melanoma cells were treated with 1-10 mg/ml concentrations of AkF-CM${\alpha}$ and 200 ${\mu}M$ arbutin as the positive control. The melanin content and cell viability of the melanoma cells by arbutin treatment decreased to 43% and 92% of the control, respectively. AkF-CM${\alpha}$ treatment at 1, 3, and 5 mg/ml concentrations decreased the extracellular melanin release induced by IBMX treatment by 35%, 45%, and 53%, respectively. AkF-CM${\alpha}$ showed inhibitory activity against both intracellular tyrosinase in melanoma cells and mushroom tyrosinase. AkF-CM${\alpha}$ reduced the protein level of tyrosinase in the IBMX-stimulated cells. These results indicate that AkF-CM${\alpha}$ suppressed the activity and protein content of cellular tyrosinase and decreased the total melanin content in cultured B16F0 melanoma cells.

Screening of Biological Activities of Extracts from Rhododendron mucronulatum Turcz. Flowers (진달래꽃(Rhododendron mucronulatum Turcz. Flower) 추출물의 생리활성 탐색)

  • Extracts from Rododendron mucronulatum Turcz. flowers were tested for antioxidant and their inhibitory activities of ${\alpha}$-amylase, ${\alpha}$-glucosidase and angiotensin converting enzyme (ACE). Total contents of phenolics were found as $30.6{\pm}0.14mg/g$ (60% EtOH extract) and $23.2{\pm}0.21mg/g$ (water extract). Electron donation ability (EDA), ABTS [2,2azinobis(3-ethylbenzothiazoline-6-sulfonic acid)] radical decolorization, Antioxidant protection factor (PF) and thiobarbituric acid reactive substance (TBARs) were measured for the antioxidative activity of the extracts from Rododendron mucronulatum Turcz. flowers. The water extract were determined as 97.5% at ethanol extract showed 83.2% and 60% EtOH extract were 89.7% in EDA. The water extract showed higher antioxidant activity than 60% EtOH extract when evaluated by ABTS radical decolorization and antioxidant PF. The TBARS of water extracts and 60% EtOH extracts were shown as $0.29{\times}10^2{\mu}M\;and\;0.28{\times}10^2{\mu}M$, respectively, and were lower than control. ACE inhibitory activity in water extract (67.6% inhibition) was higher than that of 60% EtOH extract (46.7% inhibition) at $200{\mu}g/mL$. Water extracts had higher inhibitory activities on ${\alpha}$-amylase and ${\alpha}$-glucosidase than 60% EtOH extracts. The result suggests that the water extract from Rododendron mucronulatum Turcz. flowers will be useful as natural antioxidants and functional foods.

Evaluation for the flowers of compositae plants as whitening cosmetics functionality (국화과 꽃의 미백 화장품 기능성 검색)

  • 18 flowers of Compositae family were collected and extracted in aqueous methanol (MeOH). The concentrated extract was partitioned into n-hexane, ethyl acetate (EtOAc), n-BuOH, and water fractions. The extract and fractions were evaluated for total phenolics, total flavonoids, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity, and tyrosinase inhibition activity. n-Hexane and EtOAc fractions of Aster yomena, n-hexane fraction of Cosmos bipinnatus White, n-hexane and EtOAc fractions of C. bipinnatus Pink showed high total phenolics. And EtOAc fractions of A. yomena, C. bipinnatus White, C. bipinnatus Red, C. morifolium Froggy, and C. morifolium Himaya exhibited high total flavonoids. EtOAc fractions of A. yomena, C. bipinnatus White, C. bipinnatus Pink, C. morifolium Yellowmable, and MeOH extract of C. morifolium Rosa significantly scavenged DPPH radical. EtOAc fractions of C. chinensis, C. bipinnatus White, C. bipinnatus Red, C. morifolium Himaya, and C. morifolium Hongsim highly inhibited the tyrosinase activity. A. yomena, C. bipinnatus White, C. bipinnatus Pink, C. bipinnatus Red and C. morifolium Himaya are evaluated as good source for whitening cosmetics materials.

Effect of dietary supplementation of quercetin on antioxidant activity and meat quality of beef cattle (Quercetin의 급여가 우육의 항산화 효과 및 품질에 미치는 영향)

  • This study was carried out to investigate the effects of dietary supplementation of quercetin ($Kocetin^{TM}$, QR) on antioxidative activity and meat quality of beef cattle (Holstein-Friesian). Beef cattle were divided into 3 groups; dietary supplementation of QR at 21 (n=4) and 42 ppm (n=3), and non-supplemented control (n=4). The QR comprised of 10% of quercetin. After slaughtering the beef cattle, loins were obtained and analyzed. Dietary supplementation of QR at 42 ppm showed significantly higher final pH of loin but did not affect the water holding capacity, drip loss, cooking loss, surface color, total phenolics content, 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity. Dietary QR showed no difference in both 2-thiobarbituric acid reactive substances and volatile basic nitrogen values. Textural characteristic results also showed no difference, except for cohesiveness. Cohesiveness was significantly higher in loin from beef cattle treated by dietary QR at 42 ppm when compared to control. Results suggest that dietary QR, which has only 10% of quercetin is not sufficient to have positive biochemical effects on beef meat quality.

Evaluation of crude protein, crude oil, total flavonoid, total polyphenol content and DPPH activity in the sprouts from a high oleic acid soybean cultivar

  • Soybeans [Glycine max (L.) Merill] are a rich source of antioxidants and other phytonutrients. Soybean sprouts contain many biologically active secondary metabolites and are rich in polyphenols, flavonoids, and phenolic compounds. In the present study, two soybean cultivars, Hosim, with high oleic acid (- 80% in total seed oil), and Pungsannamul, with normal oleic acid (- 23%) in seed, were examined for changes in the content of crude protein, crude oil, total flavonoids, total phenolics, and DPPH (1,1-diphenyl-2-picryl-hydrazyl) during the sprouting duration of 5 days. The protein content in both the varieties was found to increase by the days of sprouting. The crude oil content of Pungsannamul sprouts was found to be maximum on day 1 (16.9%, w/w) and decreased thereafter to reach to the level of 14.8% on day 5. No significant differences in the crude oil content of Hosim sprouts from day 1 to 5 were observed. Flavonoid content was found to increase up to day 4 and then dropped on day 5, in both the cultivars. Total polyphenol content showed a tendency to increase up to day 3 and started to decrease significantly from day 4. DPPH activity was found to increase up to day 5 in both the varieties. All the components studied in the high oleic acid soybean sprouts showed a change in content during the sprouting process similar to the change that would occur in normal oleic acid soybeans. The study showed that the contents of antioxidant, flavonoid, and polyphenol significantly increase during the sprouting.

Quantitative Assessment of the Relative Antineoplastic Potential of the n-butanolic Leaf Extract of Annona Muricata Linn. in Normal and immortalized Human Cell Lines

  • Natural products have been the target for cancer therapy for several years but there is still a dearth of information on potent compounds that may protect normal cells and selectively destroy cancerous cells. The present study was aimed to evaluate the cytotoxic potential of n-butanolic leaf extract of $Annona$ $muricata$ L. on WRL-68 (normal human hepatic cells), MDA-MB-435S (human breast carcinoma cells) and HaCaT (human immortalized keratinocyte cells) lines by XTT assay. Prior to cytotoxicity testing, the extract was subjected to phytochemical screening for detecting the presence of compounds with therapeutic potential. Their relative antioxidant properties were evaluated using the reducing power and $DPPH^*$radical scavenging assay. Since most of the observed chemo-preventive potential invariably correlated with the amount of total phenolics present in the extract, their levels were quantified and identified by HPLC analysis. Correlation studies indicated a strong and significant (P<0.05) positive correlation of phenolic compounds with free radical scavenging potential. The results revealed that the extract was moderately cytotoxic to normal cells with a mean IC50 value of 52.4 ${\mu}g$ when compared with those obtained for cancerous cells (IC50 values of 29.2 ${\mu}g$ for MDA-MB-435S and 30.1 ${\mu}g$ for HaCaT respectively). The study confirms the presence of therapeutically active antineoplastic compounds in the n-butanolic leaf extract of $Annona$ $muricata$. Isolation of the active metabolites from the extract is in prospect.
