지누아리(Grateloupia filicina) 추출물의 항산화 및 항염능 (Antioxidant and Anti-inflammatory Activity of Grateloupia filicina extract)

  • 본 연구에서는 최적의 조건에서 추출된 지누아리 추출물의 항산화능과 항염능을 확인하였다. 다양한 ethanol 수용액으로 지누아리의 유용물질을 추출하여 DPPH, ABTS, nitrite 라디컬 소거능을 측정하고, 폴리페놀 및 플라보노이드 함량을 분석하였다. 20% ethanol 수용액으로 추출한 경우에 라디컬 소거능이 가장 우수하였고, 폴리페놀과 플라보노이드의 함량 또한 가장 높았다. 이로부터 20% ethanol 수용액이 지누아리의 항산화 및 항염 물질을 효율적으로 추출하였음을 확인하였다. 한편, RAW 264.7 세포주를 이용하여 지누아리 추출물의 세포독성과 항염능을 확인하였다. 지누아리 추출물은 200 ㎍/mL의 농도에서 독성을 나타내지 않았고, 이 농도에서 50% 이상의 NO 생성 억제능을 나타냈다. 이러한 결과로 지누아리 추출물은 항산화 및 항염 특성을 지니는 기능성 화장품 원료로 사용 가능함을 확인하였다.

쇠비름 물, 에탄올 추출물의 항산화 및 항염증 활성 (Antioxidant and anti-inflammatory activities of water extracts and ethanol extracts from Portulaca oleracea L.)

  • 본 연구에서는 쇠비름의 활용가치를 높이고 기능성 소재로서의 이용 가능성을 타진하기 위해 쇠비름 물 및 에탄올 추출물의 항산화와 항염증 활성을 탐색하였다. DPPH 라디칼 소거능과 FRAP을 이용한 총 항산화 효능 평가 결과 에탄올 추출물이 물 추출물보다 항산화 활성이 우수한 것을 확인하였고, ABTS 라디칼 소거능은 $1,000{\mu}g/mL$의 농도에서만 쇠비름 물 추출물의 활성이 더 높았다. 한편, 세포독성을 살펴보기 위하여 쇠비름 물 및 에탄올 추출물을 이용해 대식세포에 MTT assay를 수행한 결과, $4,000{\mu}g/mL$의 이하 농도까지는 세포의 생존율에 영향을 미치지 않는 것을 확인하였다. $1.0{\mu}g/mL$ LPS로 염증반응을 유도시킨 세포에 대해 쇠비름 물 추출물에 비해 에탄올 추출물이 더 효과적으로 농도의존적인 NO와 ROS 생성 억제 효과가 있었다. 추가적으로 세포의 항염증 효능의 지표 단백질로 알려진 TNF-${\alpha}$, IL-$1{\beta}$, IL-6의 발현을 확인하였을 때 LPS 단독 처리군 대비 쇠비름 에탄올 추출물에서 3가지 단백질 모두의 발현양이 감소하는 것을 확인하였다. 이상의 결과를 종합하여 볼 때 쇠비름 에탄올 추출물은 물 추출물에 비해 높은 항산화와 항염증 활성을 가지는 것으로 확인되었으며, 추후 쇠비름의 유효성분 추출을 통한 항염증 물질의 연구와 염증 억제 성분의 분리 및 그 작용기전 연구에 기초 자료가 될 것으로 사료된다.

한약복합추출물(NI-01)의 항염증 및 소양감 억제 효과 (Anti-inflammatory and Anti-itching Effects of Herbal Medicine Complex Extracts(NI-01))

  • The purpose of this study was to investigate the possibility of herbal medicine complex extract (NI-01), which were prepared from 6 natural materials (Cinnamomum cassia Blume, Lonicerae Flos, Paeonia suffruticosa Andrews, Arctium lappa Linne, Schzandra chinesis Bailon, Elsholtzia ciliata Hylander), as a functional material for inhibition of atopic dermatitis. anti-oxidative activity was confirmed by measuring DPPH electron donating ability and ABTS+ radical scavenging ability. Cytotoxicity and NO inhibition were measured using RAW 264.7 cells to confirm anti-inflammatory efficacy. The test substance was orally administered to the pruritus-induced ICR mice to confirm the inhibition of pruritus. The bovine cornea opacity and permeability (BCOP) assay was performed to confirm safety for irritation. NI-01 showed high antioxidant activity in DPPH and ABTS+ methods. In the anti-inflammatory effect tests with RAW 264.7 cells, NO production was inhibited at NI-01 concentrations of 50 (14.9%) and 100 (4.2%) ㎍/mL, which indicated that the anti-inflammatory effect was increased in a concentration-dependent manner. NI-01 also showed anti-itching effect after inducing of itching by compound 48/80 in ICR mice. NI-01 was proved to be a non-irritant substance in BCOP assay. The results of this study suggested that the herbal medicine combined extract (NI-01) has high antioxidant, anti-inflammatory and anti-itching effects, and safety for irritation. Therefore, herbal medicine complex extract (NI-01) is thought to be highly applicable for the inhibitory ingredients of the atopic dermatitis.

Antioxidant, anti-inflammatory, and antibacterial activities of a 70% ethanol-Symphyocladia linearis extract

  • Research on the potential biological activity of red alga Symphyocladia spp. has been limited to Symphyocladia latiuscula, which is widely used as a food ingredient in Korea. Here, we examined the biological activity of another species, Symphyocladia linearis, which is found in Korea and was reported as a new species in 2013. The aim of this study was to evaluate the antioxidant, anti-inflammatory, and antibacterial properties of a 70% ethanol extract of S. linearis. Antioxidant activity, which was evaluated using radical scavenging assays, revealed half maximal inhibitory concentration values for 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) of 34.57 and 11.70 ㎍/mL algal extract, respectively. Anti-inflammatory activity of the S. linearis ethanolic extract was evaluated using RAW 264.7 cells by measuring the inhibition of lipopolysaccharide-induced nitric oxide (NO) and prostaglandin E2 (PGE2) production. The potential cytotoxicity of NO and PGE2 was first examined, confirming no toxicity at concentrations ranging from 10-100 ㎍/mL. NO production was inhibited 61.1% and 78.0% at 50 and 100 ㎍/mL S. linearis extract, respectively; and PGE2 production was inhibited 69.1%, 83.2%, and 94.8% at 25, 50, and 100 ㎍/mL S. linearis extract, respectively. Thus, the S. linearis extract showed very strong efficacy against PGE2 production. The cellular production of reactive oxygen species, measured using 2',7'-dichlorofluorescin diacetate fluorescence, was inhibited 48.8% by the addition of 100 ㎍/mL S. linearis extract. Antibacterial activity was evaluated using the disc diffusion method and minimum inhibitory concentration (MIC). S. linearis was effective only against gram-positive bacteria, exhibiting antibacterial activity against Staphylococcus aureus with a MIC of 256 ㎍/mL extract and against Bacillus cereus with a MIC of 1,024 ㎍/mL extract. Based on these results, we infer that a 70% ethanolic extract of S. linearis possesses strong anti-inflammatory properties, and therefore has the potential to be used in the prevention and treatment of inflammatory and immune diseases.

베리류 에탄올 추출물의 항산화 활성 (Antioxidant Activities of Various Berries Ethanolic Extract)

  • Edible berries are rich in anthocyanins and phenolic acids, compounds that possess antioxidant, anti-inflammatory, and other biological activities. Antioxidant and anti-inflammatory activities of five berries including acaiberry (Euterpe oleracea Mart.), Aronia/black chokeberry (Aronia melanocarpa), blueberry (Vaccinium angustifolium), black currant (Ribes nigrum L.), and cranberry (Vaccinium macrocarpon) were assessed. The Aronia G (prepared by GreenField s.c.) exhibited the highest antioxidant activities as shown in total phenolic (138.81 mg CAE/g), flavonoid (3.68 mg QE/g), and anthocyanin (20.31 mg/g) contents compared to the other berries. It also showed the strongest scavenging activities such as DPPH (69.69 mg vitamin C/g) and ABTS radical scavenging activity ($757.79{\mu}mol$ trolox/g). Aronia G exhibited strong ferric reducing antioxidant power ($553.98{\mu}mol$ vitamin C/g), and oxygen radical absorbance capacity ($820.92{\mu}mol$ trolox/g). In addition, black currant and Aronia showed stronger inhibition of nitric oxide production in lipopolysaccharide-stimulated RAW 264.7 cell than the other berries. According to the above results, the Aronia and other edible berries have notably high level of antioxidant activities and they could be used as a potential source of natural antioxidants.

하늘타리(Trichosanthes kirilowii Maxim.) 부위별 추출물의 항산화 및 항염증 활성 (Anti-Oxidant and Anti-Inflammatory Activities of Various Organ Extracts from Trichosanthes kirilowii Maxim.)

  • In this study, antioxidant and anti-inflammatory activities of the extract of different organs such as seed, fruit and root from Trichosanthes kirilowii Maxim. (TKM) were investigated in vitro. Among organs of TKM, the methanol extract of seed showed weak 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities with $10.4{\pm}0.1%$ and $25.5{\pm}1.1%$ at $50{\mu}g/mL$ and $100{\mu}g/mL$, respectively, and strong 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulphonicacid) (ABTS) radical scavenging activities with $72.4{\pm}6.3%$ and $96.4{\pm}2.3%$ at $50{\mu}g/mL$ and $100{\mu}g/mL$, respectively. Anti-inflammatory effects of the extracts were investigated by measuring nitric oxide (NO) production, mRNA expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) in LPS-induced Raw 264.7 macrophage cells. The methanol extracts of seed and fruit at $50{\mu}g/mL$ potently suppressed LPS-stimulated production of NO and reduced the expression of iNOS mRNA. The methanol extracts of seed and fruit also reduced the expression of COX-2 mRNA remarkably. Therefore, seed and fruit of TKM may be utilized for a wide range of health benefits associated with antioxidant and anti-inflammatory activities.

물오리나무 유래 고함량 Oregonin 추출방법 및 생리활성 연구 (Extraction Method and Physiological Activity of High Content Oregonin Derived from Plant of Alnus sibirica Fisch. ex Turcz)

  • Oregonin is known as a representative active substance of plants of the Alnus species. In this study, we will show the results of establishing a method for extracting trace amounts of useful substances in plants. Additionally, a variety of physiological activity studies were conducted with extracts containing high content of useful substances. In this study, anti-oxidant, anti-inflammatory and anti-allergic activities of supercritical fluid residue 60% EtOH extracts of Alnus sibirica (ASCFR) were investigated in vitro. To explore anti-oxidant and anti-inflammatory characteristics of supercritical extract, DPPH radical scavenging, and ABTS radical scavenging activity, inhibition of nitric oxide production in LPS-stimulated RAW 264.7 cells and ${\beta}$-hexosaminidase release in IgE-stimulated RBL-2H3 cells were examined, respectively. Results showed a concentration-dependent increase in DPPH and ABTS radical scavenging activity. Cells stimulated with LPS produced more nitric oxide than normal control cells. However, cells treated with supercritical fluid residue 60% EtOH extract decreased production in a concentration-dependent manner. In anti-allergic activity studies, ASCFR showed concentration-dependentanti-allergic activity. Based on results and strong antioxidant activity, ASCFR has potential as basic research materials for development of therapeutic supplements-based medicines or functional cosmetics related to chronic inflammatory skin immunity diseases.

목질진흙버섯 Phellinus linteus HN1009K의 항산화 활성과 항염증효과 (Antioxidant and Anti-inflammatory Effects of Phellinus linteus HN1009K)

  • 최근 농가에서 재배하고 있는 상황버섯 종에 비하여 균사생장이 빠르고 자실체 발생과 생육이 빠른 P. linteus HN1009K (HK1009)가 개발되었다. 본 연구는 HK1009의 균사체와 자실체 에탄올 추출물의 항산화 및 항염증효과를 기존에 재배되고 있는 P. linteus (PL, Korea Sangwhang)과 P. baumii (PB, Jangsu Sanghwang)를 대조구로 하여 비교분석 하였다. HK1009는 균사체와 자실체 추출물 모두 PL과 PB에 비하여 항산화 활성이 높았으며 xanthine oxidase분석에서는 PL, HK1009, PB순으로 항산화 활성이 높게 나타났다. RAW264.7세포를 이용한 상황버섯 균사체 추출물 PB(EX), PL(EX), HK1009(EX)시료의 NO assay 결과는 PB, PL, HK1009 모두 농도 의존적으로 원액 $200{\mu}g/ml$ 에서는 $7{\sim}8{\mu}M$ 정도로 염증 억제 효과가 뛰어났다. RAW 264.7 세포에 대한 MTT assay 결과에서 PB(EX), PL(EX), HK1009(EX) 추출물의 원액처리구 에서도 세포의 생존능이 70% 이상을 유지하여 세포 독성이 없는 것으로 판단되었다.

Isolation and Biological Activities of an Alkaloid Compound (3-methylcanthin-5, 6-dione) from Picrasma quassiodes (D. Don) Benn.

  • An alkaloid, 3-methylcanthin-5, 6-dione, was isolated from the stem of Picrasma quassioides (D. Don) Benn. and characterized by comprehensive analyses of its 1D and 2D NMR spectra. It was also evaluated for its cytotoxic activity in vitro against three human cancer cell lines (MDA-MB-231, HT-29 and NCI-N87), using MTT assays. We found that 3-methylcanthin-5, 6-dione exhibited significant anti-inflammatory activity via inhibiting NO production induced in LPS-stimulated murine macrophage RAW264.7 cells. The antioxidant activity of 3-methylcanthin-5, 6-dione was measured by DPPH free radical scavenging assays, hydroxyl radical scavenging assays and reducing power assays. Our results showed that 3-methylcanthin-5, 6-dione has significant biological activities.

노루궁뎅이 버섯 열수추출물의 항산화 활성과 항염증 효능 평가 (Evaluation of the Antioxidant Activity and Anti-Inflammatory Effect of Hericium erinaceus Water Extracts)

  • This study was conducted to determine the antioxidative activity of Hericium erinaceus (HE) and we investigated that HE extract contributes to cell proliferation and to anti-LPS-induced inflammation. HE extract showed high DPPH free radical scavenging activity. However, the reducing power activity slightly increased in compare with control. Nitrite scavenging activity, ABTS radical scavenging activity, SOD-like activity of HE were elevated in dose-dependent. The level of total phenolic and flavonoid showed 30.06 mg/100 g and 33.86 mg/100 g, respectively. Linoleic acid oxidation inhibition had reached a maximum level on the fourth day and started to drop from the fifth day. HE extract contributes to cell proliferation on the RAW 264.7 cell. Our finding demonstrated that water extracts of HE possess significant antioxidant activities and may be suggested a new potential source of anti-inflammatory medicines.
