The Experimental Study on Anti-inflammation and Anti-oxidation of Sopungdojeok-tang(Xiaofengdaochi-tang) and Samulsopungsan(Siwuxiaofeng-san) (소풍도적탕(消風導赤湯)과 사물소풍산(四物消風散)의 항염 및 항산화 효과에 대한 실험적 연구)

  • Objective: This experimental study was performed to investigate the anti-inflammation and anti-oxidant effects of Sopungdojeok-tang(SPDJT) and Samulsopung-san(SMSPS) which were used to treat patient with atopic dermatitis Methods: Anti-inflammation and anti-oxidant effects of SPDJT and SMSPS were measured by the inhibitory ability of Nitric oxide(NO) production and the scavenging for 1,1-diphenyl-2-picrylhydrazyl(DPPH) radical. Results: 1. SPDJT and SMSPS were not showed cell toxicity. 2. In inhibitory effects against NO production, all groups of SPDJT were showed anti-inflammation, but all groups of SMSPS were not showed anti-inflammation. There were statistical significances between $20{\mu}g/m\ell$ and 4, $10{\mu}g/m\ell$ in SPDJT groups. 3. In DPPH radical scavenging activity, all groups of SPDJT and all groups of SMSPS were showed anti-oxidant effects. There were statistical significances between $20{\mu}g/m\ell$ SPDJT group and 4, $10{\mu}g/m\ell$ SMSPS groups. Conclusion: These results suggest that the SPDJT groups are better than the SMSPS groups in antiinflammation and anti-oxidant effects.

  • Portulaca oleracea L. (PO) or Purslane is an annual grassy plant that is distributed in many parts of the world, especially the tropical and subtropical areas. PO has some pharmacological properties such as analgesic, antibacterial, skeletal muscle-relaxant, wound-healing, anti- inflammatory and a radical scavenger. This review article is focused on the anti-inflammatory, immuno-modulatory, anti-oxidant and anti-tumor activities of the PO. Anti-inflammatory, immuno-modulatory, anti-oxidant and Anti-tumor effects of PO were searched using various databases until the end of August 2018. The online literature was searched using PubMed, Science Direct, Scopus, Google Scholar and Web of Science. Our review showed that PO exerts its effects through anti-inflammatory properties and balancing the adaptive and innate immune system depending on situations. PO acts as immune-modulator and anti-oxidant agent in both inflammatory states by the dominance of Th2 response such as asthma, cancer and atopic dermatitis and evoked Th1 disorders including hepatitis and multiple sclerosis.

Anti-Oxidant and Anti-Inflammatory Complex Health Functional Analysis of Chinese Resource Plant Boehmeria, Carlemannia and Clausena (중국 자원식물 보메리아, 칼라마니아, 클로세나의 항노화 항염 복합 건강관리 기능성 연구)

  • Ageing has been known to be deeply related to oxidative activities. Furthermore, inflammation is a response initiated by conditions such as infection and injury. It has been known that complex connections occur between oxidative stress and the inflammatory response, however underlying mechanism needs to be investigated. Chinese resource plants have been widely used as functional food and medicine for a long time, however it is not studied well how many of these resource plants work. We first decided testing anti-oxidant and anti-inflammatory activities with 95% ethanol extracts of the chinese resource plants, Boehmeria pilosinscula (Blume) Hassk., Carlemannia tetragona Hook f. and Clausena emarginata C.C.Huang. To measure anti-oxidant activity, we performed DPPH assay in Raw264.7 cells with 95% ethanol extracts. Clausena extracts showed significantly higher anti-oxidant activity than those of Boehmeria and Carlemannia. We then performed Nitric Oxide assay to measure the inflammation suppression levels with 95% ethanol extracts of these plants. Clausena extracts showed significantly higher suppression of nitric oxide production than those of Boehmeria and Carlemannia. This indicates that inflammation levels are significantly reduced by Clausena. After measuring anti-oxidant and anti-inflammatory activities, we then performed MTT assay to measure the cell survival rate of Raw264.7 cells treated with these extracts. Boehmeria showed much more cell survival rate than Carlemannia and Clausena. Taken together, this result suggests that Clausena extracts have more anti-oxidant and antiinflammatory activities than Boehmeria and Carlemannia, while Boehmeria extracts have more cell survival rate than Carlemannia and Clausena.

Anti-oxidant, Anti-inflammatory, and Anti-obesity Effects of Ethanol Extracts from Ulmus divididiana var. japonica

  • In this study, the anti-oxidant, anti-inflammatory, and anti-obesity activities of an ethanol extracts of Ulmus divididiana var. japonica (UDE) were investigated in vitro and in vivo. UDE anti-oxidant activity was evaluated with an Electron Spin Resonance (ESR) spectrometer, which measured 1, 1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activity. Cell viabilities were estimated using 3-[4,5-Dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay. LPS-stimulated BV-2 microglia were used to study the production of nitric oxide (NO). Cells stimulated with LPS produce more NO than normal control cells. However, cells treated with the UDE decreased this production in a concentration dependent manner (100, 250, 500, $1,000{\mu}g/mL$). Also, we investigated the anti-obese activity of UDE in SD rats. The SD rats were randomly divided into five groups: 10% low fat diet (N), 45% high fat diet (H), 45% high fat diet + garcinia extracts 200 mg/kg/day (HG200), high fat diet + UDE 200 mg/kg/day (HU200), high fat diet + UDE 400 mg/kg/day (HU400). UDE was found to lower whole body and abdominal and epididymal adipose tissue weights and lowered plasma levels of triglyceride (TG), compared to those in H group. Histological analyses of the liver and fat tissues of rat treated with UDE revealed significantly decreased number of lipid droplets and decreased size of adipocytes compared to the H group. These results suggest that UDE might be used to develop potent anti-oxidant, anti-inflammatory, and anti-obesity agents, and may be useful as ingredients for related new functional raw materials.

Anti-oxidant and Anti-aging Activity on Saxifraga stolonifera MEERBURGH Ethanol Extract (바위취 에탄올 추출물의 항산화 및 항노화 작용)

  • To investigate the effect of ethanol extract of Saxifraga stolonifera MEERBURGH on skin care, we measured anti-oxidant and anti-aging activity. S. stolonifera ethanol extract itself had anti-oxidant activity in a dose-dependent manner in DPPH radical scavenging. Silica dose-dependently increased the intracellular ROS generation in RAW 264.7 cells. S. stolonifera ethanol extract inhibited silica-induced intracellular superoxide anion generation, $H_2O_2$ and hydroperoxide generation in RAW 264.7 cells. S. stolonifera ethanol extract significantly inhibited both hyaluronidase and elastase activity, also significantly inhibited MMP-1(collagenase) activity as well. In NIH 3T3 fibroblast cells, S. stolonifera ethanol extract significantly increased collagen-like polymer synthesis, which suggesting the S. stolonifera ethanol extract might be used as hydration and anti-wrinkle agents. From the above results, it is suggested that the main ingredients of S. stolonifera ethanol extract play an important role in anti-oxidant and anti-aging activity.

Anti-Inflammatory Effects of Tongbi-san(通痺散) Extract on RAW264.7 Macrophages (통비산(通痺散) 열수추출물의 항염증반응 및 항산화활성에 대한 연구)

  • Objectives : This study is to investigate the anti-inflammatory and anti-oxidant effects of Tongbi-san extract (TS) on RAW264.7 macrophages using by cell cytotoxicity, Nitric Oxide (NO) and Prostaglandin $E_2$ ($PGE_2$) production and 1,1-diphenyl-2-picryl ghdrazyl (DPPH) free radical scavenging capability. Methods : Cell cytotoxicity was measured by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay. The production of NO was measured by Griess assay. The production of $PGE_2$ was measured by immunoassay. And the anti-oxidant activity was measured by the DPPH method. Results : TS did not increased significantly compared to the TS untreated group in the cell cytotoxicity. TS inhibited NO and $PGE_2$ production in lipopolysaccharide-stimulated RAW 264.7 cells. TS had the DPPH free radical scavenging capability. Conclusion : The anti-inflammtory and anti-oxidant effects of TS may be use for a treatment of anti-inflammatory diseases.

Anti-inflammatory and Anti-Oxidant Effects of Oxypaeoniflorin, Paeoniflorin and Paeonia lactiflora cv. 'Red Charm' Flower Petal Extracts in Macrophage Cells

  • The root extracts of Paeonia lactiflora cv. 'Red Charm' has been studied by many groups, however, little attention has been paid to its flower petal. Paeonia is the genus in the Paeoniaceae family. 'Red Charm' Paeonia is a soft-stemmed herbaceous peony hybrid of P. officinalis and P. lactiflora. We previously showed the flower petal extract of Red Charm might have anti-oxidant and anti-inflammatory activities, however, it was not clear which components might be involved in this activity. Bioinformatics analysis previously indicated these extracts have potential anti-oxidant materials. One of them is identified as paeoniflorin, which is major component in root extract of Red Charm. In this study, we compared paeoniflorin and oxypaeoniflorin using DPPH assays to measure its anti-oxidant activities. Oxypaeoniflorin showed higher levels of radical scavenging activity, similar to ascorbic acid control, whereas paeoniflorin did not. Furthermore, nitric oxide assay showed they have similar anti-inflammatory effects. Taken together, these results suggest oxypaeoniflorin may play a more important role in the anti-oxidant activity of the flower petal and root extracts of Red Charm, compared to paeoniflorin. Further studies may be able to provide a platform to develop potential dual effects therapeutics for oxidant-mediated and inflammation-mediated disease in the near future.

Anti-microbial Effect on Streptococcus mutans and Anti-oxidant Effect of the Butanol Fractions of Sanguisorbae Radix (지유(地楡) 부탄올 분획물의 Streptococcus mutans 항균활성 및 항산화 효과)

  • Objective : Sanguisorbae Radix(SO). that belong to Rosaceae is widely distributed in Asia including Korea, Japan and China. It has been used as traditional medicine from old times as a treatment for anti-inflammatory drugs. This study was designed to investigate the effects of n-BuOH fractions of SO on anti-oxidant effect and anti-microbial effect on $Streptococcus$ $mutans$ ($S.$ $mutans$). Methods : The anti-microbial effect of $n$-BuOH fractions of SO. was assessed by the paper disk diffusion method and anti-oxidant effect was assessed by the DPPH radical scavenging effect, Superoxide anion radical scavenging effect and SOD like ability. Results : DPPH radical scavenging of the $n$-BuOH fraction from SO in 50 ug/mL was shown to be Fr-2 (89.9%), Fr-3 (88.7%), Fr-4 (76.3%), Fr-1 (59.4%), Fr-5 (56.2%). Superoxside anion radical scavenging activity of the n-BuOH fraction from SO. in 50 ug/mL for Fr-3 was shown to be 78%. SOD-like activity of n-BuOH fraction from the SO in 1000 ug/mL for Fr-3 were shown to be 76.1% respectively. The $n$-BuOH fraction from the SO had high anti-microbial effect on $S.$ $mutans$. Conclusion : As a result, the $n$-BuOH fraction from SO. has good anti-microbial and anti-oxidant effects in a concentration-dependent manner.

Anti-oxidant and anti-inflammatory effect of Allium Hookeri water extracts in RAW 264.7 cells (삼채(三菜) 물추출물이 RAW 264.7 세포의 항산화 및 염증반응에 미치는 영향)

  • Objectives : Allium hookeri is a well-known traditional herbal remedy and its root used for treatment of inflammation and tumor. However, the mechanism of anti-inflammatory effect of Allium hookeri is still unknown. This study aims to examine the mechanism of anti-inflammatory effect of Allium hookeri on mouse macrophage cell line, RAW 264.7 cells. Methods : Anti-oxidant effect of water extract of Allium hookeri (WEAH) was measured by 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulphonic acid (ABTS) assay. 3- (4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium (MTS) assay was performed to determine the effect of WEAH on cell viability in RAW 264.7 cells. In addition, anti-inflammatory effect of WEAH was investigated in RAW 264.7 cells. Inflammation of RAW 264.7 cells induced by lipopolysarccharide (LPS) treatment and expression levels of inflammatory cytokine interleukin 1 β (IL-1β) and interleukin 6 (IL-6) gene were analyzed using quantitative reverse transcription PCR (qRT-PCR) analysis. Furthermore, the phosphorylation of inhibitor of nuclear factor kappa B (IκBα) after LPS treatment with WEAH-treated RAW 264.7 cells was confirmed by immunoblot analysis. Results : WEAH showed a strong anti-oxidant effect and no cytotoxicity to RAW 264.7 cells up to 2 mg/㎖ concentration. The LPS-induced mRNA expression levels of IL-1β and IL-6 were decreased by WEAH treatment. Furthermore, the LPS-induced phosphorylation of IκBα is attenuated by WEAH treatment. Conclusions : Through experimental demonstration of anti-oxidant and anti-inflammatory effects of WEAH, we suggest that Allium hookeri is a valuable material for prevention and treatment of various inflammatory diseases.

Study on Effects of Anti-oxidant and Viscoelastic on Emulsion by the Extract of Astragalus membranaceus (황기(黃芪)추출물의 항산화 효능과 에멀션 점탄성도에 미치는 영향에 관한 연구)

  • Objectives : The Astragalus membranaceus has been used to diuretic, tonic, anti-viral, anti-oxidant activities in oriental herb medicine, and in each experiment, proved in this effects. Root of A. membranaceus includes according to flavonoid, saponin, polysaccharide, thus it has been studied for anti-oxidant, anti-inflammatory, anti-cancer activities. However, previous researches have been performed with being limited mainly to food industry. The present study was conducted anti-oxidative activity of A. membranaceus extracts. In addition, The objective of the present study was to determine the possibility for the application of emulsions in the development of A. membranaceus for cosmetics. Methods : In this study, aimed SOD like activity of A. membranaceus extract, effects of A. membranaceus extract on viscoelastic properties of emulsion was measured using rheometer. Results : In the results, 1, 3-butylene glycol extract of A. membranaceus efficiently reduce the active oxygen, SOD like activity of more than 40% decrement depend on concentration. And A. membranaceus extract increase the modulus of elasticity of emulsion, therefore A. membranaceus extract have not influence on stability loss. As such, the A. membranaceus extracts will remain stable when applied to emulsions or skin products. Conclusions : In consequence of this study, A. membranaceus extract can be effectively used in cosmetic emulsions when the relation between natural product extracts and formulation of cosmetics is elucidated. It is highly recommended that rheology be applied to determine the optimal extract concentration for cosmetic formulations.
