한약재추출물의 항균활성 (Antimicrobial Activities of Medicinal Herb Extracts)

  • 본 연구에서 18종의 한약재추출물을 제조하여 병원성세균에 대한 한약재의 항균활성을 측정하였다. 오배자, 오미자 및 소목 메탄올 추출물은 대부분 균주에 대해 5 mg/mL 수준에서 항균활성을 나타냈으나 15여 종의 한약재추출물들은 30 mg/mL 수준에서 항균활성을 나타냈다. 오배자의 S. epidermidis와 Bor. bronchiseptica에 대한 최소저해농도는 0.6 mg/mL로 나타났다. 우수한 항균활성이 확인된 오배자, 소목, 오미자, 황금을 대상으로 극성이 다른 5가지 용매로 순차 분획하여 항균활성을 검토한 결과 대부분의 균주에서 EtOAc층이 다른 분획층에 비해 성장억제효과가 높게 나타났다. 항균력이 우수한 한약재추출물의 미생물 증식억제 효과를 조사하기 위해 증식배지에 0, 100, 300 및 500 ppm의 메탄올 추출물을 첨가하였다. 한약재 추출물을 첨가하지 않은 대조구에서는 12시간 이후 급격한 균의 성장을 나타내고 있으나 오배자와 소목추출물의 첨가 농도가 높을수록 그람 양성균인 S. aureus, S. epidermidis 및 L. monocytogenes는 뚜렷한 억제효과를 나타냈다. 오배자추출물은 그람음성균인 Sal. Pullorum과 Sal. Choleraesuis의 성장을 저해하였고 Bor. bronchiseptica, E. coli serotype $O_8$은 오배자와 소목추출물은 낮은 농도에서 성장이 다소 저해되었음을 보여주었고, 추출물의 농도가 높을수록 균의 성장이 강하게 억제되었다.

양마에서 분리한 Kaempferol과 그 배당체의 항균 및 주름억제 효과 (Anti-Microbial and Anti-Wrinkle Effect of Kaempferol and Kaempferol Rhamnosides isolated from Hibiscus cannabinus L.)

  • In this study, kaempferol and its rhamnosides (${\alpha}$-rhamnoisorobin, afzelin, kaempferitrin) were isolated electively by bioconversion technology from Hibiscus cannabinus L. leaves to evaluate the anti-wrinkles effects and anti-microbial effects. In order to evaluate anti-wrinkles activity, reduction of expression matrix metalloproteinase-1 (MMP-1) protein and proliferation/ pro-collagen production were investigated. Kaempferol and ${\alpha}$-rhamnoisorobin showed inhibition activity of MMP-1 generated to compared to positive control. In HaCaT cell proliferation assay, kaempferol and ${\alpha}$-rhamnoisorobin significantly promoted cell proliferation in a concentration-dependent manner. In addition, procollagen synthesis assays (by HDF-N cell) showd that TGF-${\beta}$ induced procollagen production and also, all four kinds of experimental significantly promoted procollagen synthesis in a concentration-dependent manner. Kaempferol and ${\alpha}$-rhamnoisorobin exhibited strong antimicrobial activities on five of microbes, Staphylococcus aureus, Escherichia coli, Pseudomonas aeruginosa, Candida albicans and Aspergillus niger.

황토노각플러스 추출물의 항산화, 항염 및 항미생물 효능 (Anti-oxidant, Anti-inflammation and Anti-microbial Effects of Hoangtonogak Plus Extracts)

  • 본 연구에서는 황토노각플러스 추출물의 생리활성 평가를 통해 화장품 원료로서의 가능성을 조사하였다. 황토노각 플러스 추출물에 대한 항산화 효과는 DPPH/ABTS 라디컬 소거능 활성 및 FRAP 분석, 세포 생존율은 MTT assay, 항염증 효과는 nitric oxide(NO) 생성억제 및 iNOS 단백질 발현 수준, 항균 효과는 총 3종 균주 Staphylococcus aureus, Staphylococcus epidermidis 및 Escherichia coli에 대한 한천 확산법을 측정하였다. 메탄올추출물(MN)과 열수추출물(WN)의 총폴리페놀 함량은 각각 2.92±0.01 mgGAE/g, 1.67±0.02 mgGAE/g로 나타났다. MN 추출물의 DPPH, ABTS 및 FRAP 값은 모든 농도에서 WN 추출물보다 높게 나타났다. RAW264.7 세포에 대한 두 추출물의 세포 독성은 관찰되지 않았다. 더욱이 1 mg/mL농도에서 MN과 WN 추출물의 NO 생성량은 11.69 μM과 20.4 μM로 측정되었으며, MN 추출물의 iNOS 단백질 발현 수준은 추출물 농도 의존적으로 억제되었다. 또한 MN 추출물은 S. epidermidis 균주에서만 항균효능을 보였다. 이러한 결과들로 황토노각플러스 추출물은 항산화, 항염, 항균효능을 지닌 천연물질 후보로써 가능성이 높다고 판단된다.

오배산가미(五倍散加味)가 점막(粘膜) 및 피부질환(皮膚疾患)에 미치는 영향 (The effects of Ohbaesangami (OBSGM) on the mucosa and skin diseases)

  • Objectives : To investigate the effects of Ohbaesangami (OBSGM) on mucosa and skin diseases, anti-microbial and anti-inflammatory tests were performed using several in vitro test models. Results : In anti-microbial test, OBSGM showed the slight inhibitory effect against Propionibacterium acnes (P. acnes) and Staphylococcus aureus (S. aureus). In anti-oxidant test, OBSGM showed the potent radical scavenging activity. In anti-inflammatory test, OBSGM weakly inhibited the lipopolysaccharide (LPS)-induced nitric oxide(NO) release from the RAW 264.7 macrophage cells. OBSGM also inhibited the LPS-induced $interleukin-1{\beta}(IL-1{\beta})$ and cyclooxygenase-2 (COX-2) expressions. The inhibitory effects of OBSGM on macrophage activation was via the inhibition of $NF-{\kappa}B$, evidenced by transient transfection assay. Furthermore, OBSGM markedly inhibited the activation of Jun-N-terminal kinase (JNK) and p38 MAP kinase in RAW 264.7 cells. In skin wrinkle formation assay, OBSGM strongly inhibited collagnease and elastase, whose activities are tightly related with the wrinkle formation. In addition, OBSGM inhibited the activities of MMP-1, MMP-2 on the mRNA levels in RAW 264.7 cells. However, OBSGM did not show an inhibitory potential on tyrosinase activity and melanin synthesis, indicating that it could not be applicable for skin whitening. Conclusion : These results suggest that the anti-inflammatory effect of OBSGM may be due to its inhibitory potentials on the macrophage activation. And, the anti-wrinkle effects of OBSGM may be due to its inhibitory potential on the collagnease and elastase activities. Therefore, OBSGM could be applicable for the treatment of mucosa and skin diseases.

제조방법에 따른 강하주의 품질 특성 (Quality Properties of Gangha-ju Liquor According to the Preparation Method)

  • This studies were performed to develop a Korean traditional folk liquor namely Gangha-ju has been prepared at Bosung district in Korea, and manufacturing conditions and anti-oxidation activity and anti-microbial activity of Gangha-ju were investigated. Ethyl-alcohol 20% and 30% Gangha-ju were brewed with glutinous rice wine, distilled liquor and 6 herbs of ginger, cinnamon, etc. Chemical and physical properties of 30% Gangha-ju were acidity 0.22, pH 4.31, amino acidity 3.26, transmittance 59 and conductivity $911\;{\mu}s/m$, and 20% Gangha-ju were 0.43, 4.20, 6.26, 62 and $924\;{\mu}s/m$. Volatile flavor compounds of ethyl alcohol, acetic acid, butanol, n-amyl alcohol, iso-pentyl alcohol, ethyl acetate, butyl acetate, acetaldehyde and furfural were detected, and main aroma compounds of Gangha-ju were isopentyl alcohol and ethyl acetate. Anti-oxidation activity by DPPH method was evaluated 31.32%, and nitrite scavenging effect was 31.79%. Anti-microbial activity against several microorganisms was pronounced strong activity over a wide range of test organisms, and Leuconostoc mesenteroids and Salmonella Ttyphimurium, Staphylococcus epidermidis were found to be more sensitive to Gangha-ju than Escherichia coli and Aspergillus flavus.

비방탈명산(秘方奪命散)의 항산화 ${\cdot}$ 항암 ${\cdot}$ 항균 효과 연구 (Study on the Anti-oxidative, Anti-microbial and Anti-cancer effect of Bibangtalmyungsan.)

  • Objectives : This study was carried out to evaluate anti-oxidative, anti-microbial and anti-cancer effect for clinical application of Bibangtalmyungsan (BTS) Results : 1. The oxidative effects were measured by polyphenol, DPPH radical scavenging activity. BTS water extract was showed more effective than ethanol extract and also various solvent fractions from BTS water extract showed effective in the following order : ethyl acetate fraction > butanol fraction > hexane fraction ${\fallingdotseq}$ chloroform fraction ${\fallingdotseq}$ aqueous fraction 2. The results of anti-microbial effects were as follows. 1) Antibacterial activities of BTS extracts against Gram's negative and positive bacteria were ineffective 2) Antifungal activities of the BTS extracts against Aspergillus spp. , Trichohyton mentagrophyte KTCC 1077 were not effective. 3. The result of anti-cancer effects were as followings: 1) BTS ethanol extract was more effective than water extract against Caco-2, Calu-6, but it had cytotoxic effect against NIH3T3. 2) On the Caco-2, effective only in ethyl acetate $fraction(IC_{50}:$ 35.81 ${\mu}g/ml)$. 3) On the Calu-6, the most effective in ethyl acetate $fraction(IC_{50}:$ 189.65 ${\mu}g/ml)$ and effective In butanol $fraction(IC_{50}:$ 299.74 ${\mu}g/ml)$ and hexane $fraction(IC_{50}:$ 345.13 ${\mu}g/ml)$ 4) On the SUN-601, $IC_{50}$ value was within 80 ${\mu}g/ml$ in ethyl acetate fraction and hexane fraction. 5) On the HCT-1l6, the most effective in ethyl acetate $fraction(IC_{50}:$ 82.94 ${\mu}g/ml)$ and effective in hexane $fraction(IC_{50}:$ 374.56 ${\mu}g/ml)$. 6) On the AML-2/WT, the most effective in ethyl acetate $fraction(IC_{50}:$ 41.44 ${\mu}g/ml)$ and effective In hexane $fraction(IC_{50}:$ 303.01 ${\mu}g/ml)$. 7) On the NIH3T3, effective only in ethyl acetate $fraction(IC_{50}:$ 203.42 ${\mu}g/ml)$, but it was more ineffective than other cancer cells. Conclusion : These result suggest that BTS has antioxidative, antifungal activities and cytotoxic effects against Caco-2, Calu-6, SUN-601, HCT-116, AML-2/WT and NIH3T3, especially ethyl acetate fraction from water extract has more effective in antioxidative and anticancer effects.

해조 다당류 추출물의 항균성 및 항염증 기능성평가 (The Functional Effects of Anti-microbial Activity and Anti-inflammatory of Seaweed polysaccharide Extracts)

  • 본 연구는 해조 다당류 추출물을 화장품 원료로 사용할 수 있는지 평가하고자 하였다. 항균 효과를 평가하기 위해 해조 다당류 추출물로 부터 Staphylococcus aureus (S. aureus) 균주에 대한 항균활성을 측정하였다. 또한, RAW 264.7 세포에서 항염증 효과가 확인되었으며, 해조 다당류 추출물을 Sprague-Dawley (SD) 흰쥐의 등쪽 피부에 적용하여 단회투여독성을 평가하였다. 그 결과, 해조 다당류 추출물은 피부섬유 아세포에서 최대 $1,000{\mu}g/mL$ 농도에서 세포 독성을 나타내지 않았다. 또한, 1 % 해조 다당류 추출물로 처리되었을 때, $1.52{\pm}0.34cm$로 생육저해환이 형성되어 S. aureus 균주에 대한 항균 활성을 확인하였다. 해조 다당류 추출물을 RAW 264.7 세포에 처리하면 농도 의존적으로 산화 질소 (NO)의 생성이 감소하고 인터루킨 1 베타 ($IL1{\beta}$), 종양 괴사 인자 알파 ($TNF{\alpha}$)와 같은 염증 관련 사이토카인의 생성이 증가하였으며, 프로스타글란딘 E2 (PGE2)가 감소 하였다. 해조 다당류 추출물을 여러 농도로 흰쥐에 투여했을 때 증상은 14 일 이상 변하지 않았으며 췌장이나 장기 무게에는 변화가 없었다. 결론적으로 우리는 해조 다당류 추출물이 세포 독성을 나타내지 않았으며 항균 및 항염증 효과가 있음을 발견하였다. 따라서, 화장품 원료로 사용하기에 적합하다고 사료된다.

  • Objectives: The purpose of this study was to investigate the in vitro anti-inflammatory, anti-pruritic and antimicrobial effects of the three herbal prescription (EST, SCT, WDT), which has been traditionally used for treating leukorrhea induced by various infections in the female genital tract. Methods: In this experiment, the anti-inflammatory effects were evaluated by Nitric oxide (NO), $Interlukine-1{\beta}$ ($IL-1{\beta}$), Interlukine-2 (IL-2), Interlukine-6 (IL-6), Tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), Prostaglandin $E_2$ ($PGE_2$), Leukotriene $B_4$ ($LTB_4$) production amount and Inducible nitric oxide synthase (iNOS), Nuclear factor kappa B ($NF-{\kappa}B$), Cyclooxygenase-2 (COX-2) gene expression levels in RAW264.7 cells. And the anti-pruritic effects were evaluated by Histamine, Acetylcholine (ACh), Acetylcholinesterase (AChE), Substance P production amount in Mast cell/9 (MC/9) and Pheochromocytoma 12 (PC12) cells. The anti-microbial effect was measured by inhibition zone diameter on Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans and Aspergillus niger. Results: As a result of measuring anti-inflammatory efficacy, $IL-1{\beta}$, IL-2, IL-6, $TNF-{\alpha}$, $PGE_2$, and $LTB_4$ production amounts were significantly reduced in the EST, SCT, WDT extraction groups compared with the control group, and significantly decreased the amount of $NF-{\kappa}B$, iNOS, and COX-2 gene expression and the amount of Phospho-Inhibitor kappa B alpha ($p-I{\kappa}B-{\alpha}$)/Inhibitor kappa B alpha ($I{\kappa}B-{\alpha}$) and $NF-{\kappa}B$ p65 protein expression. In addition, As a result of measuring the anti-pruritic effect, the amounts of histamine, ACh and Substance P were significantly decreased, and AChE production was slightly decreased, but it's significance did not appear. Finally the anti-microbial effects of EST, SCT, WDT extraction groups against Pseudomonas aeruginosa, Candida albicans and Aspergillus niger was inhibited, however the growth of Escherichia coli and Staphylococcus aureus was not inhibited. Conclusions: These data suggest that EST, SCT, WDT can be used to treat patients with leukorrhea.

저식염 고추장 양조시 항균물질 혼합첨가의 영향 (Effect of Combined Use of various Anti-microbial Materials on Brewing of Low Salted Kochujang)

  • 소금농도를 9%에서 6%로 줄이고 항균물질로 알코올과 겨자, 키토산을 혼합첨가 하여 제조한 저식염 고추장의 숙성중의 효소활성도와 미생물상 및 이화학적 특성을 비교하였다. 숙성 중 수분활성도의 저하는 식염 9%인 대조구에서 심하였고, 숙성 중에 효모수는 증가하였으나 알코올-겨자 혼합 첨가로 효모수를 조절할 수 있었다. 고추장 숙성 중 ${\beta}-amylase$ 활성은 항균물질 첨가시 증가하였으나 ${\alpha}-amylase$와 protease 활성은 조금 감소하였다. 고추장은 숙성이 진행되면서 L차 a-, b-값이 증가하였으며 알코올 첨가 고추장이 $({\Delta}E)$값의 변화가 적었다. 적정산도는 항균물질의 첨가로 숙성 후기에 저하되었으며, 산화환원전위의 저하는 대조구에서 심하였다. 고추장 숙성 중 총당의 감소는 알코올-겨자 혼합 첨가구(EM)에서 적어 환원당량이 많았으며, 숙성 후기의 알코올의 생성은 알코올에 키토산을 혼합 첨가한 고추장(EC)에서 높았다. 고추장 숙성 중기까지 아미노태 질소는 증가하였고, 숙성중 암모니아태 질소의 생성은 알코올-겨자-키토산 혼합 첨가구(EMC)에서 적었다. 12주 숙성된 고추장을 관능 평가한 결과 색과 향기에서 알코올(E) 또는 알코올-겨자 혼합 첨가구(EM)가 좋아 저식염 고추장 제조시 알코올 또는 알코올-겨자를 혼합 첨가하는 것이 전체적인 기호도에서 양호하였다.

수종 생약의 Protein kinase C 저해활성 (Inhibitory Activity against Protein Kinase C of Some Medicinal Plants)

  • MeOH extract of twenty medicinal herbs were screened for their effects against protein kinase C (PKC) using bleb-forming assay and PKC enzyme assay. Smilax china and Sanguisorba officinalis showed potent anti-PKC activity. Campsis grandiflora and Galla Halepensis showed moderate inhibitory effect on PKC.
