Effects of Egg Shell Membrane Hydrolysates on Anti-Inflammatory, Anti-Wrinkle, Anti-Microbial Activity and Moisture-Protection

  • This study was conducted to examine the effects of eggshell membrane hydrolysates (ESMH) on the anti-inflammatory, anti-wrinkle, anti-microbial activity, and moisture-protection for cosmetic use. Whole ESMH (before fractionation), and fraction I (>10 kDa), fraction II (3-10 kDa), and fraction III (<3 kDa) of the hydrolysates were assessed in this experiment. As lipopolysaccharide (LPS) and IFN-${\gamma}$ caused the inflammation on Raw264.7 cell, whole ESMH and fraction I showed to be effective in inhibiting the induction of cell inflammation depending on the concentration, and also showed outstanding effect to suppress the skin inflammation. Fraction I inhibited collagenase and elastase activities to a greater extent than the other fractions, while all fractions had antibiotic effects at concentrations of 10 mg/disc and 20 mg/disc. In addition, it showed the moisture protection effects of skin on the holding amount and losing amount of moisture in upper-inner arm of the human body with a relatively low loss rate in skin, which confirmed that the hydrolyzed fractions of ESM helps to form the superior protective layer of moisture. It was concluded that ESMH fractions with different molecular weights, especially the 10 kDa fraction, have anti-lipopolysaccharide, anti-IFN-${\gamma}$-induced inflammation, anti-collagenase and elastase activities, and thus can be used as a cosmetic agent to protect skin.

전도산(顚倒散)의 추출용매에 따른 항염 및 항산화 비교 연구 (The Comparative Study of Anti-inflammation and Anti-oxidation in Accodance with Extraction Solvents of Jeondo-san)

  • Objective : The purpose of this study was to compare anti-Inflammation and anti-oxidation of Jeondo-San(JDS) extracted with two kinds of solvents, ethanol and water. Methods : Two kinds of JDS extractions were prepared 20, 50, $100\;{\mu}g/mg$. The Cytotoxicity was measured by MTT assay in Raw 264.7 cell. The anti-inflammation effects were measured by inhibitory efficacy on $PGE_2$, NO, TNF-$\alpha$, COX-2 and iNOS in Raw 264.7 cell. The anti-oxidation effects were measured by ROS inhibitory efficacy, intracellular GSH synthesis and DPPH Radical scavenging in HaCaT cell. Results : 1. All of JDS extraction groups had no cytotoxicity in Raw 264.7 cell. 2. All of JDS extraction groups showed significantly inhibitory effect on production of $PGE_2$. Inhibitory efficacy increased in accodance with concentration. 3. All of JDS extraction groups showed significantly inhibitory effect on production of NO. Inhibitory efficacy increased in accodance with concentration. 4. All of JDS extraction groups did not show significantly inhibitory effect on production of TNF-$\alpha$. 5. $100\;{\mu}g/ml$ JDS extracted with ethanol and $50\;{\mu}g/ml$, $100\;{\mu}g/ml$ JDS extracted with water showed inhibitory effect on iNOS expression. 6. All of JDS extraction groups showed significantly inhibitory effect on production of ROS. Inhibitory efficacy increased in accodance with concentration. Ethanol extractions were better than water extractions. 7. $100\;{\mu}g/ml$ JDS extracted with ethanol only produced GSH of $32{\pm}5.2%$. 8. All of JDS extraction groups showed significantly scavenging effect of DPPH radicals. Inhibitory efficacy increased in accodance with concentration. Ethanol extractions were better than water extractions. Conclusion : Two kinds of JDS extractions have not cytotoxicity and inhibit production of NO. JDS extracted with water was effective in anti-inflammation, JDS extracted with ethanol was effective in anti-oxidation.

약침(藥鍼)의 연구 동향에 대한 분석 - 항염(抗炎) ${\cdot}$ 진통(鎭痛) ${\cdot}$ 비만(肥滿) 및 안전성(安全性)에 관한 연구를 중심으로 - (An Analysis of Tendencies of Studies on Herbal Acupuncture - Focusing on domestic theses since 2001 about anti-inflammation, pain relief and anti-obesity effects, including safety-)

  • Objective : To research Trends of studies about anti-inflammation and pain relief, obesity, and safety of herbal acupuncture therapy by analyzing domestic theses, published since 2001, about herbal acupuncture therapy. Methods : Domestic theses, published since 2001, mentioning anti-inflammation or pain relief, obesity, or safety of herbal acupuncture therapy were reviewed and analyzed. These theses were then classified by university, year, and subject. Results : The following results were obtained in this study. 1. Among published theses related to anti-inflammation effects of herbal-acupuncture, studies about arthritis comprised the most part, 52. In theses relating to causes of arthritis, 16 were about adjuvant, which was the most, followed by Type II collagen, LPS and carrageenan. Blood test, reactions of inflammation and revelation of cytokine and immune cellswere methods for evaluating anti-inflammation effect. The tendency of experimental methods was to focus on molecular biologic method. 2. In theses related to pain relief, many clinical attempts with herbal injection were carried out, and Carthami Flos and Scolopendrawere used most. Observing reduction of pain inducing factor and checking behavioral change were methods for evaluating pain relief. 3. In theses related to obesity, research focused on effects in association with spots on the body suitable for acupuncture. There were also attempts comparing effectiveness between single injections and complex injections. Astraball Radix, Angelica Gigantis Radix, Coicis Semen and Taeumjowetang were used. Evaluation of anti-obesity effects were by weight loss, food efficiency, blood lipid profile and evaluation of liver function. 4. In theses related to safety of herbal-acupuncture, Herba Chelidonii Chaenonelis Fructus, Clematis Florida Thunb, Corydalidis Tuber, Paeoniae Radix, and Carthami Flos which marked 2 theses each were most studied. Methods of evaluating safety were mostly by observing liver and kidney functions based on blood test, and by applying herbal injections to clinical treatment. Conclusion : Herbal acupuncture is being used in various ways associating with its anti-inflammation, pain relief and anti-obesity effect. Studies on efficacy and mechanism of herbal acupuncture are being conducted even at the molecular biology level.

Intestinal anti-inflammatory activity of Sasa quelpaertensis leaf extract by suppressing lipopolysaccharide-stimulated inflammatory mediators in intestinal epithelial Caco-2 cells co-cultured with RAW 264.7 macrophage cells

  • BACKGROUND/OBJECTIVES: Inflammatory bowel disease (IBD), including Crohn's disease and ulcerative colitis, involves chronic inflammation of the gastrointestinal tract. Previously, Sasa quelpaertensis leaves have been shown to mediate anti-inflammation and anti-cancer effects, although it remains unclear whether Sasa leaves are able to attenuate inflammation-related intestinal diseases. Therefore, the aim of this study was to investigate the anti-inflammatory effects of Sasa quelpaertensis leaf extract (SQE) using an in vitro co-culture model of the intestinal epithelial environment. MATERIALS/METHODS: An in vitro co-culture system was established that consisted of intestinal epithelial Caco-2 cells and RAW 264.7 macrophages. Treatment with lipopolysaccharide (LPS) was used to induce inflammation. RESULTS: Treatment with SQE significantly suppressed the secretion of LPS-induced nitric oxide (NO), prostaglandin $E_2$ ($PGE_2$), IL-6, and IL-$1{\beta}$ in co-cultured RAW 264.7 macrophages. In addition, expressions of inducible nitric oxide synthase (iNOS), cyclooxygenase (COX)-2, and tumor necrosis factor (TNF)-${\alpha}$ were down-regulated in response to inhibition of $I{\kappa}B{\alpha}$ phosphorylation by SQE. Compared with two bioactive compounds that have previously been identified in SQE, tricin and P-coumaric acid, SQE exhibited the most effective anti-inflammatory properties. CONCLUSIONS: SQE exhibited intestinal anti-inflammatory activity by inhibiting various inflammatory mediators mediated through nuclear transcription factor kappa-B (NF-kB) activation. Thus, SQE has the potential to ameliorate inflammation-related diseases, including IBD, by limiting excessive production of pro-inflammatory mediators.

추출용매에 따른 산초 (Zanthoxylum schnifolium)열매 추출물의 라디칼소거능과 항염증 효과 (Radical Scavenging and Anti-inflammation Activities from Different Extracts of Zanthoxylum schnifolium Fruits)

  • This study was presented to the physiological activities such as metal chelating capacity, superoxide dismutase-like activity, anti-inflammation, cytotoxicity from Zanthoxylum schnifolium fruit. The values for metal chelating capacity of 1 mg/mL of 70% EtOH and 70% MeOH extracts were 68% and 67%, respectively, it was shown that metal chelating capacity in a dose-dependent manner. The 100% EtOH and 100% MeOH extract exhibited strong superoxide dismutase-like activity. The highest SOD-like activity obtained from 100% EtOH was 51% at a concentration of 4 mg/mL. The all extracts of Zanthoxylum schnifolium fruit except $H_2O$ extract significantly decreased the concentration of LPS-induced NO in Raw 264.7 cells. The 100% EtOH, 100% MeOH and 70% EtOH extract represented the highest activity in the anti-inflammation properties in vitro. However, the 100% EtOH and 100% MeOH extracts has cytotoxicity in Raw 264.7 cells and it may affect cell viability. Conclude that 70% EtOH extract was most suitable for anti-inflammation. Our results revealed that the Zanthoxylum schnifolium fruit expected to physiological activities.

도화(桃花, Prunus persica Flos)의 항산화 및 항염증 활성 (Anti-oxidant and Anti-inflammation Activities of Prunus persica Flos)

  • 도화 추출물의 항산화능 및 항염증 효과를 검증하여 기능성 화장품 소재로서의 가능성을 검증하였다. 도화의 항산화능 측정으로 전자공여능을 측정한 결과 도화 에탄올 및 아세톤 추출물 500ppm에서 90.0% 이상의 효과를 나타내어, 합성 항산화제로 알려진 BHT와 유사한 효과를 나타내었으며, SOD 유사활성능 측정 결과 도화 에탄올 및 아세톤 추출물 1,000ppm에서 40.0% 이상의 활성을 나타내었다. Xanthine oxidase 저해활성능 측정 결과 도화 에탄올 및 아세톤 추출물 1,000 ppm에서 vitamin C와 유사한 30.0%의 효과를 나타내어, 도화 열수 추출물에 비해 도화 에탄올 및 아세톤 추출물의 효과가 높게 나타났다. 항염증 효과 측정으로 hyaluronidase 저해활성을 측정한 결과 도화 열수, 에탄올 및 아세톤 추출물에서 35.0%의 저해활성을 나타내었으며, macrophage 세포를 이용한 nitric oxide (NO) 저해활성을 측정한 결과 도화 추출물 모든 농도에서 12시간 이후부터 NO 생성량이 억제됨을 확인할 수 있었다. 또한 피부염증 유발균인 Staphylococcus epidermidis와 Propionibacterium acnes의 항균효과를 측정한 결과 도화 에탄올 추출물에서 항균 효과를 나타내었다.

이비인후과 염증질환에 대한 데옥시리보뉴클레아제-브로멜라인정의 효과 조사 (The Effect of Deoxyribonuclease-Bromelain Tablet in Patients with Inflammation of Otorhinolaryngology)

  • The author has tested the clinical efficacy and safety of an anti-inflammatory drug in patients with inflammation of the otnrhinolaryngology. Deoxyribonuclease prepared from bovine pancreas destroyed fibers and reduced the viscosity of the exudates. Bromelain is a group of proteolytic enzymes which are known to have an effect on inflammation, swelling, and pain in inflammatory disorders. The mucolytic and anti-inflammatory properties of deoxyribonuclease associated with bromelain in inflammation of otorhinolaryngology were assessed in 67 patients. A standard dose of 3 tablets of deoxyribonuclease bromelain tablet was given for not less than 7 days. In all tile groups of patients considered, the drug showed a considerable therapeutic effectiveness. At the end of 7-28 days treatment, more than $89.6\%$ of treatment was improved. These findings suggest that it could be permitted to use deoxyribonuclease-bromelain tablet in patients with inflammation of the otorhinolaryngology.

Anti-Hyperalgesic Effects of Meloxicam Hydrogel via Phonophoresis in Acute Inflammation in Rats; Comparing Systemic and Topical Application

  • The aim of this study was to determine if a meloxicam hydrogel could be administered in vivo via phonophoretic transdermal delivery using pulsed ultrasound by examining its anti-hyperalgesic effects in a rat carrageenan inflammation model. Carrageenan (1%) was injected into the plantar surface of the right hindpaw, and meloxicam hydrogel was administered via phonophoretic transdermal delivery. Changes in the mechanical and thermal hyperalgesia, as well as swelling, showed that phonophoretic delivery of meloxicam exhibited significantly better anti-hyperalgesic and anti-inflammatory effects than pulsed ultrasound. Topical and systemic application of meloxicam hydrogel using phonophoresis showed similar anti-hyperalgesic effects. These findings suggest that the transdermal administration of a meloxicam hydrogel using phonophoresis by pulsed ultrasound might be useful for treating acute inflammation.

Anti-Oxidant, Pro-Oxidant and Anti-Inflammatory Effects of Unpolished Rice Relevant to Colorectal Cancer

  • Colorectal cancer (CRC) is a major worldwide health problem owing to its high prevalence and mortality rates. Carcinogenesis in the colon is a multistage and multifactorial process. An imbalance between free radical exposure and anti-oxidant defense systems may leads to oxidative stress and attack of macromolecules which can alter signal transduction pathways and gene expression. Consequently, oxidative damage can lead to cellular dysfunction and contribute to pathophysiological processes in a variety of diseases including CRC. One factor tightly associated with CRC is chronic inflammation, which can be present from the earliest stage of tumor onset. Unpolished rice is an attractive chemoprevention in CRC due to their anti-oxidant and anti-inflammatory activities. The aim of this paper is to review evidence linking oxidative stress and inflammation to CRC and to provide essential background information for understanding future research on oxidative stress and inflammation on CRC. Mechanisms of action of unpolished rice in CRC carcinogenesis are also discussed.

Effect of Astaxanthin on Anti-Inflammatory and Anti-Oxidative Effects of Astaxanthin Treatment for Atopic Dermatitis-induced Mice

  • Background: This study sought to determine whether the antioxidant effects of astaxanthin (AST) could have an anti-inflammatory effect to reduce inflammation caused by atopic dermatitis (AD). Methods: Using a mouse model of AD induced by phtalic acid (PA), the levels of inflammation, inflammatory agents, and evidence of antioxidant activity were examined in PA treated mice (n = 3), PA-AST treated mice (n = 3), and a control group of mice (n = 3). This included measurements of ear thickness, levels of mast cells, IgE, inflammatory cytokine, malondialdehyde (MDA), hydrogen peroxide, HO-1, and GPx-1. Results: AST treatment significantly prevented inflammation as measured by ear thickness (p < 0.05), mast cell count (p < 0.001), and IgE concentration in the blood (p < 0.001). Levels of TNF-α (p < 0.001), IL-1β (p < 0.001), IL-6 (p < 0.001), and MDA (p < 0.05) were also significantly lower. In addition, GSH levels increased significantly (p < 0.001), and the level of hydrogen peroxide significantly reduced (p < 0.01). The expression of HO-1, GPx-1 increased. Conclusion: In this small experimental study, AST acted on inflammatory mechanisms that induced AD, through anti-inflammatory and antioxidant mechanisms, and is a candidate of interest in the clinical treatment of AD.
