Studies on Constituents of the Higher Fungi of Korea(XXXVII) - Antitumor Components of Armillariella mellea - (한국산(韓國産) 고등(高等) 균류(菌類)의 성분(成分) 연구(硏究)(제(第)37보(報)) - 뽕나무버섯의 항암(抗癌) 성분(成分) -)

  • To find antitumor components in Korean basidiomycetes, the carpophores of Armillariella mellea which were collected in Gyeong Gi Province were extracted with distilled water at $90{\sim}100^{\circ}C$ for eight hours. The hot water extract was concentrated under reduced pressure, mixed with three-fold volumes of ethanol and allowed to stand at $4^{\circ}C$ overnight. The precipitate was centrifugated and lyophilized to yield a protein-polysaccharide fraction. It was examined for antitumor activity against sarcoma 180 implanted in ICR mice. The fraction showed 75.7%, 83.9%, and 94.1% of tumor inhibition ratios at the doses of 10, 20 and 50 mg/kg/day, respectively. The chemical analysis of the fraction showed that it contained a polysaccharide(41.3%) and a protein (35.0%). The hydrolyzates of the polysaccharide moiety contained fucose (4.5%), xylose (1.1%), galactose (17.4%), glucose (55.4%), mannose(19.4%), and one unknown monosaccharide. The protein moiety contained seventeen amino acids. The protein-polysaccharide from A. mellea was administered, i.p., to mice and caused an influx of polymorphonuclear leukocytes (PMN) at $5{\sim}24$ hours which was followed by an accumulation of macrophages and disappearance of the PMN at $48{\sim}72$ hours.

Comparison of Protein Binding Polysaccharide from Agaricus blazei Murill Prepared by Ultrafiltration and Spray-Drying Process

  • Chemical properties of spray-dried powders separated based on molecular weight from crude protein binding polysaccharide (CP-SD) of Agraricus blazei were examined. Contents of ${\beta}$-glucan in SD-1, SD-2 and SD-3 were 18.67%, 48.24%, and 37.15% respectively, and SD-2 (10-150 kDa) showed the highest molecular weight. Obtained ${\beta}$-glucans were not pure glucan, but was determined to be an acidic proteo-heteroglycan with a large amount of glucose (74.46-80.05%), galactose (8.91-15.2%), and mannose (4.9-5.46%). Composition of their amino acids was mainly aspartic and glutamic acids. FT-IR spectrum revealed SD-1, SD-2 and SD-3 were structures of ${\beta}$-1,3-glucans and ${\alpha}$-1,6-glucans at 890 and 930 $cm^{-1}$, respectively, signals of ${\alpha}$-1,6-glucans for CP-SD was not found. Useful CP-SD was recovered from A. blazei for preparation of three powder types as food materials.

  • To investigate antitumor components of Korean higher fungi, the carpophores of Schizophyllum commune and Auricularia auricula-judae collected in Kyeong Buk Province were extracted with hot water or 0.1N-NaOH solution. The concentrated extracts were precipitated by addition of ethanol, and the precipitates were purified by dialyzing through visking tube and polysaccharide fractions were obtained. They were found to show antitumor activity against sarcoma 180 implanted in mice. Especially, the inhibition ratio of the extract of Auricularia adicula-judae was 90.8% in the doses of 100mg/kg/day for the period of ten days. The tumor in five of the eight mice was completely regressed. The components of these aqueous extracts were found to be polysaccharide and protein. The hydrolysis of the respective polysaccharide yielded four monosaccharides. After hydrolysis of the protein fraction, 15 amino acids were identified in the respective fraction of S. commune and A. auricula-judae.

  • To find pharmacologically active components of Agrocybe cylin(Iracea, its basidiocarps were extracted with water. The extracts were separated by DEAE cellulose column chromatography, Sepharose CL-4B gel filtration, and Concanavalin A-Sepharose 4B affinity chromatography. Among the obtained fractions from A, cylinclracea, fraction IN which was the neutral proteinbound-polysaccharide fraction exhibited a marked antitumor activity and it was tentatively named "Cylindan". It showed about 70% of tumor inhibition against the solid form of sarcoma 180 when a dose of 30 mg/kg/day was intraperitoneally injected into ICR mice. When each fraction was examined by chemical analysis, Cylindan consisted of 85% polysaccharide, 3% protein and 1% hexosamine. Its polysaccharide moiety contained glucose, mannose, fucose and galactose and its protein moiety contained the comparatively large amounts of aspartic acid and glycine, and other 11 amino acids.

  • Lysozyme was covalentyl conjugated with galactomannan through a amino-carbonyl reaction between the lysine $\varepsilon$-amino groups of lysozyme and the reducing ends of galactomannan at a relative humidity of 79% and 6$0^{\circ}C$. The resulting lysozyme-galactomannan conjugate (LGC) was investigated for its antibacterial activity against Escherichia coli. Lysozyme alone did not exhibit antibacterial activity against E. coli. in contrast , significant bactericidal effect was observed for LGC, depending on the reaction temperature. The degree of conjugation between lysozyme and galactomannan was dependent on the incubation time, which affected the antibacterial efficiency against E. coli. This study demonstrated that the amino-carbonyl reaction between lysozyme and galactomannan could be a potential tool to modify lysozyme toward broadening its antibacterial spectrum to Gram-negative bacteria.

  • To separate and quantitate antitumor protein-bound polysaccharides of Coriolus versicolor, the constituents were obtained from the submerged culture of the mycelia(C) and from the extract of the carpophores of the wild fungus(N). The polysaccharides were degraded by methanolysis. The neutral monosaccharides were separated and quantitated by HPLC using microbondapak carbohydrate analysis column, refractive index detection and water-acetonitrile acetic acid elution. The analyses of these constituents by HPLC showed that the natural constituent(N) consisted of glucose, xylose and mannose, the average amount being 96.44, 2.16 and 1.73%, respectively. The fermentation constituent(C) consisted of mannose, glucose, xylose and galactose, the average amount being 61.30, 14.00, 12.95, and 11.75%, respectively. The analyses of these constituents by an amino acid analyzer showed that both C and N contained 17 amino acids.

  • A protein-polysaccharide fraction was prepared from the carpophores of Laetiporus sulphureus. This fraction suppressed growth of sarcoma 180 in A-strain mice when administered i. p. To investigate the mechanism of antitumor action of this fraction, plaque assay was conducted by administrating i. p. to the mise at a dose level of 50mg/kg for five days. Ten days later, the mice were immunized with 1 * 10$^{7}$ sheep red blooc cells. The number of hemolytic plaque forming cells was significantly greater than that of the control mice. Three monosaccharides and fifteen amino acids were identified in the protein-polysaccharide fraction.

  • Chitosan derivatives, 6-amino-6-deoxychitosan (6A6DC) was successively prepared as a reactive carbohydrate for the stabilization of epidermal growth factor (EGF) by the reactions of chitosan with tosyl chloride, sodium azide, and lithium aluminum tetrahydride. The structure of 6A6DC was confirmed by EA, FT-IR, $^1H-NMR$ and $^{13}C\{^1H\}-NMR$. The degree of substitution (ds) of amino groups in 6A6DC was determined to be 0.7. 6A6DC did not show any cytotoxicity on the normal human dermal fibroblast (NHDF) proliferation at least in the range tested (0.3 g/mL 600 g/mL) and was considered as a suitable material for the stabilization of EGF against proteolytic degradation due to its non-cytotoxicity and high reactivity.

Sustained Release Properties of Vitamin C in Chitosan Molecular network (키토산 분자네트 워크속에서 비타민 C의 지속적 방출 특성)

  • Chitosan is a dietary fiber because of a linear polysaccharide composed of $\beta-(1{\rightarrow}4)$-linked 2-amino-2-deoxy-D-glucopyranose. In this study, control release system of vitamin C has been estimated in chitosan molecular network as a vitamin C carrier of controlled release. The amount of released vitamin C were decreased in higher amount of chitosan concentration. Especially, vitamin C were slowly released from chitosan solution in dialysis membrane when compared with vitamin C solution alone in dialysis membrane. These result assumed that chitosan driving force is dependent on chitosan molecular weight and cationic property of amino group with anionic property of vitamin C.
