Effect of Use of SPS Agar and SGS Broth and Storage of Specimen on the Isolation of Vibrio vulnificus (Vibrio vulnificus 분리율에 대한 SPS Agar와 SGP Broth의 사용 및 검체 저장의 영향)

  • Vibrio vulnificus septicemia is nor-rare diease in Korea. Carriage rate of the orgaism by shellfish is not well known. In this study performance of SPS agar and SGP broth and effect of storage of specimen in the isolaton was determined using the shellfish specimens collected from the coast and market of Koonsan city. Isolation rate was similar with TCBS and with SPS, but the rate became much highher after enrichment in SGP broth. 80% of oyster speimes were positve when inoculated immediately, but the rate dropped rapidly after storage of specimens at freezing temperature for sometime. All of the isolates fermented lactose in 2 days. A few isolates were not identfiable with API 20E system, because of acid prduction from melibiose. Serover 04 was the frequent isolates.

Isolation of Anaerobic Bacteria from Oral Pyogenic Infections (구강 화농성 감염에서 혐기성 세균의 배양분리)

  • Strict anaerobic procedures and anaerobic chamber were employed in order to improve the isolation of obligate anaerobes from oral pyogenic infections. Also different culture media were utilized to maximize bacterial recovery. Two blood media: nalidixic acid Tween blood agar (NATB) and plain blood agar (BA), two non-blood media: nalidixic acid Tween agar (NAT) and Gifu anaerobic medium (GAM) were used and compared for their isolation efficacy. Specimens from seven patients were collected with syringe. After collection, the needle was sealed with sterilixed silicone rubber and brought to labortory. The time spent from specimen collection to its processing in anaerobic chamber usually was 15 min. Identification of isolated bacterial strains was done with the API-20A system. Increase in isolation of anaerobic vacteria was achieved. Obligate anaerobic bacteria isolated were 3.3 strains per specimen. This figure falls within the range of 1.9-4.4 strains per specimen reported in other countries. With respect to the media utilized, blood media were superior to non-blood media. NATB medium was effective especially for the isolation of Gram-positive cocci. A total of 15 species of Gram-negative rods was isolated and 12 of them belonged to Bacteroides.

Laboratory Study on the Electrical Resistivity Characteristics with Contents of Clay Minerals (점토광물의 함유량에 따른 전기비저항 특성에 관한 실험적 연구)

  • This study considers to electrical resistivity characteristics for clay minerals types and contents in fractured and fault zone. The electrical resistivity is measured for an artificial agar specimen with clay minerals instead of a natural rock. The artificial agar specimen with clay minerals was special worked in study. The clay minerals used are Kaolinite and Montmorillonite in test, the clay mineral contents increases until $0\~40\%$ to the same specimen. As results, the electrical resistivity of the specimen decreased gradually as the clay mineral contents increases for all types of clay minerals. Montmorillonite shows remarkably lower resistivity than Kaolinite, although its clay content is fewer than that of Kaolinite. Also, a proposed experimental expression shows a good correlation coefficient as high as 0.89 or more in all clay minerals.

Changes in quality characteristics of raw ginseng(Panax ginseng C.A. Meyer) pudding during storage (수삼 푸딩의 저장중 품질특성 변화)

  • Healthy ginseng pudding was made with different doses of two gelling agents and stored at $37^{\circ}C$ for eight weeks. And then quality characteristics and palatability were measured. Sweetness and pH level increased in newly made ginseng puddings but decreased during storage. Acid taste decreased but increased during storage in A (pectin 0.2%+agar 0.6%). But it increased but subsequently decreased in B (agar 0.8%). Acidity in C (agar 1%) remained unchanged until six weeks but slightly increased at 8 weeks of storage. Texture increased in all specimens but subsequently decreased. Chromaticity value of L decreased in all specimens before increasing while the values of a and b increased and subsequently decreased. In texture examination, hardness and adhesion were higher in C (agar 1%) and lower in A (pectin 0.2%+agar 0.6%). There were no differences in elasticity among three specimens. Cohesion was higher in C (agar 1%) while it showed a similar level in A (pectin 0.2%+agar 0.6%) and B(agar 0.8%). Stickiness and chewiness were higher in C(agar 1%) but lower in A(Pectin 0.2%+agar 0.6%), compared with C(agar 1%) specimen. In sensory test, overall palatability was higher in A (Pectin 0.2%+agar 0.6%). In conclusion, ginseng pudding with 0.6% agar and 0.2% pectin exhibited better quality. And ginseng pudding has potential to be developed as healthy dessert considering ample physiological and functional properties.

  • A low resistivity zone is found in many places such as a fractured fault zone, weathered zone and aquifer. The electrical resistivity is influenced mainly by pore fluid as well as the clay mineral types and contents, Hence, it is very important to understand the relationship between the electrical resistivity and clay contents associated with the low resistivity zone for geotechnical applications such as civil engineering. This study examines the characteristics of clay mineral types and contents to electrical resistivity through sample measurements, and proposes an expression relating the resistivity and clay content. The electrical resistivity is measured for an artificial agar specimen with clay minerals instead of a natural rock. The clay minerals used are Kaolinite and Montmorillonite. Montmorillonite shows remarkably lower resistivity than Kaolinite, although its clay content is fewer than that of Kaolinite.   • In this study, the method of evaluation of moisturizing ability of cosmetics using specimens was studied as an alternative method of clinical tests. Cosmetic spreadability, surface drying, and moisture loss rate of four specimens (Japanese cake (Maru mochi), Garaetteok, wheat flour, and agar) were evaluated. Also, the water loss rate of the specimens and the transepidermal waterloss were analyzed for 10 kinds of lip make-up products (5 kinds of lipstick, 3 kinds of lip balm, 2 kinds of lip gloss). The moisture loss rate (moisture evaporation sensitivity) of the agar specimen was highest in the order of agar> Garaetteok> Japanese cake> wheat flour. Agar specimens are the most suitable in terms of spreadability, surface crack, and raw material supply. Containers for the preparation of agar specimens are suitable for plastic materials with low heat transfer, which can produce convex, smooth surface specimens. In the evaluation of moisturizing ability of lip makeup products, there was a strong correlation between the water loss rate and the transepidermal waterloss rate measured with agar specimens. These results show that the proposed method can be used as one of the useful alternative test methods.

  • This research was study the accuracy of three-dimensional stereotactic breast biopsy, using a core Needle Biopsy and to assess the accuracy of Stereotactic biopsy and Sono guided biopsy. Using Stereotactic QC phantom to measure the accuracy of the 3D sterotactic machine. CT Scan and equipment obtained in the measured X, Y, Z and compares the accuracy of the length. Using Agar power phantom compare the accuracy of the 3D sterotactic machine and 2D ultrasound machine. Z axis measured by the equipment to compare the accuracy and reliability. Check the accuracy by using visual inspection and Specimen Medical application phantom. The accuracy of the 3D sterotactic machine measured by Stereotactic QC phantom was 100%. Accuracy as compared to CT, all of X, Y, Z axis is p > 0.05. The accuracy of the two devices was 100% as measured by Agar powder phantom. There was no difference between t he t wo d evices as C T and p > 0.05. 3D sterotactic machine of the ICC was 0.954, 2D ultrasound machine was 0.785. 2D ultrasound machine was different according to the inspector. Medical application phantom experiments in 3D sterotactic machine could not find the Sliced boneless ham. 2D ultrasound machine has not been able to find a small chalk powder group. The reproducibility of the three-dimensional stereotactic breast biopsy was better than effect of Sono guided biopsy.

Changes in the Oral Microflora in Patients with Acute Myeloid Leukemia during the Period of Induction Therapy (항암 화학요법중인 급성 골수성 백혈병 환자의 구강내 세균변화에 관한 연구)

  • To investigate the changes in aerobic and facultative anaerobic oral microflora during remission-induction chemotherapy in patients with acute myeloid leukemia, 10 consecutive patients were studied during a period of 28 days. One day before, during and after the induction therapy, patients were given 10% Betadine solution for mouthrinses after breakfast and kept from eating and drinking. After 3 hours, paraffin-stimulated whole saliva was obtained for 2 minutes and transported to the laboratory. The samples were dispersed and homogenized by use of vortex mixer for 20 seconds. From these samples 10-fold serial dilutions (from 10-1 through 10-3) were prepared. Each dilution of 0.1 ml was plated on duplicate set of one nonselective medium (Blood agar) and four selective media (Sabourauds dextrose agar, Mannitol salt agar, Mac-Conkey agar, SF medium ) using applicator woods. All agar plate were incubated at 37$^{\circ}C$ for 48 hours. The total number of microorganisms was calculated and the percentage distribution of the various microorganisms from each specimen was drawn. 1. The salivary flow rate decreased by 66%, going from 5.38 ml/2min to 1.81 ml/2min over two days during the chemotherapy. 2. The total number of microorganisms in saliva increased by 22%, going from 4.88$\times$105/ml to 6.00$\times$105/ml over two days during the chemotherapy. 3. The salivary flow rate and the total number of microorganisms in saliva were recovered within 28 days after the chemotherapy. 4. The quantitative alteration in oral Enterobacteria, Enterococci, Staphylococci, Cndida during the chemotherapy had no statistical significance. 5. In saliva of the patients with acute myeloid leukemia who ahd intraoral ulcer, Enterobacteria was quantitatively predominent. Our study suggests that chemotherapy-induced transient xerostomia may induce acute oral infection. Consequently, the use of saliva substitute, the removal of intraoral infection source and the consistent oral hygiene care seem to be required to avoid the transmission of potential pathogenes in this group of patients.

  • Ceotrichum candidum was identified from 2 dogs among 52 dogs which had various dermatological problems. One was 5-month-old male Labrador and the other was 7-month-old female German Shepherd. The diagnosis was confirmed by direct microscopy of the specimen as well as cultured isolation of the fungus from the cutaneous lesions. The. organism showed septate hyphae and rectangular to cylindrical arthrospores and no blastospores. Urea was not utilized and no assimilation of maltose was observed. Clinical improvement was occurred by application of 2% gentian violet and 2% miconazole solutions. It is suggested that this organism should be included in the differential diagnosis of canine dermatitis.

  • The properties of ideal root canal sealers include the ability of sealing the total root canal system and no toxic effects to periradicular tissues. Cytotoxicity test using cell culture is a common screening method for evaluation of the biocompatibility of root canal sealers. The purpose of this study was to investigate the cytotoxic effect of newly developed resin-based sealer (Adseal 1, 2, and 3) comparing with those commercial resin-based sealers (AH26 and AH Plus), ZOE-based sealers (Tubliseal EWT, Pulp Canal Sealer EWT) and calcium hydroxide based sealer (Sealapex), An indirect contact test of cytotoxicity by agar diffusion was performed according to the international standard ISO 10993-5. L929 fibroblast cells were incubated at $37^{\circ}C$ in humidified 5% $CO_2-containing$ air atmosphere. The freshly mixed test materials were inserted into glass rings of internal diameter 5 mm and height 5 mm placed on the agar. After the 24 hrs incubation period, the decolorization zones around the test materials were assessed using an inverted microscope with a calibrated screen. A Decolorization Index was determined for each specimen. Adseal 1. 2, and 3 did not exert any cytotoxic effects, whereas AH26, AH Plus, Tubliseal EWT, Pulp Canal Sealer EWT, and Sealapex produced mild cytotoxicity.
